        Human epithelial cell cultures were initiated from explants of normal human oral mucosa. Growth parameter and cell type were assessed by DNA concentration and morphology of the cell lines. Tissues were excised and epithelial cells isolated by collagenase digestion. Spermine was used in order to inhibit overgrowth of the epithelial cells by fibroblast. The effects of different medium formulations were compared. Cultures maintained on the same volume of RPMI 1640 and Dulbecco's modified minimum essential medium containing 10 % fetal bovine serum, 10 ?g/ml of insulin, lng/ml of somatomedin and 10 ng/ml of epidermal growth factor showed optimal and maximal growth. For each cultured human oral epithelial cell the DNA content was in the- range of 5.7 pg to 6.6 pg. This culture system provides epithelial cells that can be used for a variety of dental and biochemical studies.

        The aim of this study is to develop a new and improved method of a dyeing process for nylon/cotton fabrics. The dyeing property was highly dependent on the characteristics of each fabrics, the affinities of the dyestuffs, the concentration of the salts, the levelling agents, and the reserving agents. Results indicated that optimum dyeing method for nylon/cotton fabric was the use of mixed acid dye and reactant-fixable dye with an appropriate concentration of the salt, the reserving agent, and the levelling agent.

        일본의 직업교육·훈련제도의 특성과 최근의 변화

        이 논문에서는 노동시장과의 관련하에서 일본의 직업교육훈련제도의 특징과 최근의 변화 양상 및 의의를 구명하였다. 분석 틀로서는 마스덴(D. Marsden)과 미야모토(宮本光鯖)의 고용제도 모형을 이용하였다. '지적숙련 논쟁'의 검토 등을 통해 고도성장기 및 오일쇼크기에 형성된 일본의 직업교육훈련제도의 본질적 특징이 '작업능력으로서의 숙련'이 제도화되지 않은 채 OJT 중심의 기업내 훈련을 통해 직능자격제도상의 내부 승진에 의해 어느 정도 높은 수준의 다기능공으로서 기능을 형성하는 데 있음을 지적하였다. 최근의 장기 불황하에서 이러한 내부노동시장은 커다란 도전을 받고 있는데 그 변화의 방향은 내부노동시장의 붕괴가 아니다. 확대되는 외부노동시장을 자격제도의 정비에 의해 직업별 노동시장으로 정비하고 그것에 의해 내부노동시장을 유지해 나가는 정책 방향인 것을 구체적인 정책분석을 통해 밝혔다. This paper investigates the characteristics of Japanese vocational education and training(VET) sy stem and its recent changes with regard to the labour market changes under t he long-term economic depression. The employment system model of Marsden(199 9) is used as an analytical framework. By the review of 'the debate of intellectu al skill', it is pointed out that the fundamental characteristic of Japanese VET system l ies in forming multi-functional workers by the internal promotion in connection with the OJT-focused enterprise training along the job-competency rank system. In this system 'the skill as work-competency' is not institutionalised. By the long- term depression this type of internal labour market(ILM) has been under serious changes. By the policy analyses it is made clear that the direction of its change is not toward the collapse of the ILM. A series of labor market and VET polici es does aim at sustaining the ILM by the introduction of the unified job quali fication system, which is expected to institutionalise the widening external labo r market into the craft-based labour market.

      • 연약지반 성토시 사면의 안정에 미치는 Geotextile의 효과

        Geotextiles have been extensively used in road constructions as effective and economical materials performing several functions such as separation, filtration, drainage, protection against frost break, and as reinforcement to improve the load bearing capacity and stability of road. In this study, a method for the analysis of embankment reinforced with geotextile is studied. Computer program based on the simplified Bishop method is modified in order to investigate quantitively the effect of gotextile on the slope stability of embankment on weak soils. Evaluation of the tensile strength required for the design safety factor is also possible utilizing the computer program in this study.

      • Stevioside의 浸出方法의 改善 및 糖質甘味의 正味性에 對한 硏究

        現在 多量으로 생산 보급되고 있는 인공감미료의 결점을 보완하기 위한 硏究가 활발히 진행되고 있는데 이의 대체 방안의 하나로써 天然甘味料의 개발이 그 中의 하나이다. 감미료는 식품뿐만 아니라 의약품에서도 그 重要性이 날로 증대되어 가고 있는 실정이다. 본 연구에서는 現在 先進國에서 硏究되고 있는 天然甘味料인 Stevioside의 浸出 方法 및 浸出된 Stevioside의 甘味를 구미에 맞게 改善하여 食品 및 의약용 첨가제로서 널리 활용해 보고자 하는데 그 目的을 두고 있다. 본 硏究를 위한 實驗 結果를 요약하면 다음과 같다. 즉 stevioside의 浸出에 적당한 溫度는 60℃ 이었으며 浸出時間은 6時間 정도가 가장 좋았다. 또 여기에서 얻은 stevioside의 甘味는 우수하였으나, Glucose나 fructose등과 같은 단당류를 혼용했을 경우에도 보다 더 우수하였다. 또 pH 안정도 실험에서는 stevioside 용액은 중성 및 산성에서 보다 안정하였으나 알카리성에서는 불안정하였다. 以上의 結果로 부터 stevioside는 人工甘味料의 대체 甘味料로서 최적일 것으로 생각되나 수요에 충족하기 위해서는 재배 및 생산기술에 대한 보다 더 깊은 연구가 요청된다. The time has come that the development of natural sweetening materials is urgent. Especially the development of stevioside is beginning to show signs of activity in the developed countries. In these days, the appearance of natural sweetening materials as food additives for health and hygiene becomes necessary. The aims of this study are the experiment of the past study, leaching and manufacturing the materials depending on our actual circumstances, and producing the most excellent additives. The result of this study are as follows; 1. The appropriate temperature for the leaching of stevioside was 60℃ and the time needed for the leaching was about 6 hours. 2. In spice of the mixed sweetening materials of stevioside, maltose was better than glucose. 3. The stevioside of the sweetening effect was larger than sugar.

      • 냉동 제대혈 세포의 체외 증폭

        Background : Cord blood(CB), which has no HLA restriction, is an alternative to bone marrow for hematopoietic stem cell transplantation. The use of cord blood, however, is limited by the number of progenitor/stem cells necessary to reconstitute the older child or adult. Therefore, ex vivo expansion of CB could have tremendous impact on diverse clinical settings. We studied the ex vivo expansion of isolated population of CD34_(+) cells from cryopreserved CB cells. Methods : CD34 cells were isolated from cryopreserved CB mononuclear cells. Purified cells were cultured with various combinations of hematopoietic growth factors including erythropoietin(EPO), stem cell factor(SCF), granulocyte-colony-stimulating factor(G-CSF), gra-nulocyte, macrophage-colony-stimulating factor(GM-CSF), interleukin-1β(IL-1β), 1L-3, and IL-6. After 7, 10 or 14 days of culture, the fold increases of colony-forming unit- granu-locyte, macrophage(CFU-GM), burst-forming unit-erythroid(BFU-E), colony-forming unit-mix (CFU-Mix), and high proliferative potential colony-forming cell(HPP-CFC) were evaluated. Results : Ten-day culture with the combination of EPO, SCF, G-CSF, IL-1β, and IL-3 resulted in a median of 60-fold increase of CFU-GM, which was greater than those with the combinations of less than 5 growth factors. The addition of IL-6 or GM-CSF to this combination did not enhance CFU-GM expansion. Ten-day culture was significantly superior to 7-day culture for CFU-GM expansion. Prolongation of culture to 14 days, however, revealed decreased expansion of CFU-GM compared to 10 days. BFU-E and CFU-Mix were expanded to 2~5 folds in 7-day culture with the combination of EPO, SCF, and G-CSF. Further expansion was not achieved in 10-day culture and colonies disappeared in 14-day culture. HPP-CFC was expanded to a median of 7.5 folds in 7-day culture with the combination of EPO, SCF, G-CSF, IL-1β, IL-3, and IL-6. Neither 10-day or 14 day-culture enhanced expansion of HPP-CFU. Conclusion : Cryopreserved cord blood cells maintain ex vivo expansion potential. In our system, 10-day culture with the combination consisting of EPO, SCF, G-CSF, IL-1β, and IL-3 seems to be adequate for hematopoietic progenitor/stem cell expansion from cryopreserved cord blood cells.
