        The Clinical Observation of Inflammation Theory for Depression: The Initiative of the Formosa Long COVID Multicenter Study (FOCuS)

        There is growing evidence that the coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is associated with increased risks of psychiatric sequelae. Depression, anxiety, cognitive impairments, sleep disturbance, and fatigue during and after the acute phase of COVID-19 are prevalent, long-lasting, and exerting negative consequences on well-being and imposing a huge burden on healthcare systems and society. This current review presented timely updates of clinical research findings, particularly focusing on the pathogenetic mechanisms underlying the neuropsychiatric sequelae, and identified potential key targets for developing effective treatment strategies for long COVID. In addition, we introduced the Formosa Long COVID Multicenter Study (FOCuS), which aims to apply the inflammation theory to the pathogenesis and the psychosocial and nutrition treatments of post-COVID depression and anxiety.

      • Latent tuberculosis infection increases in kidney transplant recipients compared with transplant candidates: a neglected perspective in tuberculosis control

        Background: The prevalence and incidence of latent tuberculosis infection (LTBI) in patients with kidney transplant remains unclear. Methods: In this prospective study, we enrolled kidney transplant candidates (KTCs) and recipients (KTRs) from 2014 to 2018. We defined LTBI as a positive result of QuantiFERON-TB Gold In-tube (QFT). We analyzed the predictors for LTBI acquisition and followed QFT test for 2 years among those initially without LTBI. Results: Of 425 patients enrolled, 305 (71.8%) patients belonged to the KTC group and 120 (28.2%) to the KTR group. The initial QFT showed positive results in 33 (10.8%) and 25 (20.8%) in the KTC and KTR groups, respectively (p=0.007). The QFT response value in LTBI patients was higher in the KTR group than in the KTC group (1.85 vs. 1.06 IU/ml, p=0.046). Multivariate logistic regression showed that old age, absence of Bacillus Calmette-Guerin scar, and KTR group were independent factors for positive LTBI. For participants with initial negative QFT, positive QFT conversion within 2-year follow-up was higher after kidney transplantation (20%) than in KTCs (5.5%) (p=0.032). Conclusion: This study is the first cohort to follow up LTBI status in patients with kidney transplant and shows its higher prevalence and incidence in those belong to KTR. It indicates that surveillance of LTBI after renal transplantation is important.

        Upregulating sirtuin 6 ameliorates glycolysis, EMT and distant metastasis of pancreatic adenocarcinoma with krüppel-like factor 10 deficiency

        Krüppel-like factor 10 (KLF10) is a tumor suppressor in multiple cancers. In a murine model of spontaneous pancreatic adenocarcinoma (PDAC), additional KLF10 depletion accelerated distant metastasis. However, Klf10 knockout mice, which suffer from metabolic disorders, do not develop malignancy. The mechanisms of KLF10 in PDAC progression deserve further exploration. KLF10-depleted and KLF10-overexpressing PDAC cells were established to measure epithelial-mesenchymal transition (EMT), glycolysis, and migration ability. A murine model was established to evaluate the benefit of genetic or pharmacological manipulation in KLF10-depleted PDAC cells (PDACshKLF10). Correlations of KLF10 deficiency with rapid metastasis, elevated EMT, and glycolysis were demonstrated in resected PDAC tissues, in vitro assays, and murine models. We identified sirtuin 6 (SIRT6) as an essential mediator of KLF10 that modulates EMT and glucose homeostasis. Overexpressing SIRT6 reversed the migratory and glycolytic phenotypes of PDACshKLF10 cells. Linoleic acid, a polyunsaturated essential fatty acid, upregulated SIRT6 and prolonged the survival of mice injected with PDACshKLF10. Modulating HIF1α and NFκB revealed that EMT and glycolysis in PDAC cells were coordinately regulated upstream by KLF10/SIRT6 signaling. Our study demonstrated a novel KLF10/SIRT6 pathway that modulated EMT and glycolysis coordinately via NFκB and HIF1α. Activation of KLF10/SIRT6 signaling ameliorated the distant progression of PDAC.         Preparation and characterization of Y-TZP powders coated with alumina

        In this study, alumina was coated onto yttria tetragonally stabilized zirconia (Y-TZP)core powder using aluminum nitrate with urea in an aqueous solution. Boehmite coating onto the surface of Y-TZP powders was confirmed using transmission electron microscopy (TEM), energy dispersive spectroscopy (EDS), Fourier transform infrared (FTIR) spectroscopy and zeta potential measurements. The sintering behaviors of Y-TZP powders with and without an alumina coating were investigated using a dilatometer and scanning electron microscopy (SEM). Boehmite coating onto the surface of Y-TZP powders significantly improved the densification of the resulting ceramic bodies. The relative density of the Y-TZP with a boehmite coating sintered at 1350oC can reach 97.3%.         Incongruent Expression of Brain-Derived Neurotrophic Factor and Cortisol in Schizophrenia: Results from a Randomized Controlled Trial of Laughter Intervention

        Objective Schizophrenia has been associated with dysfunction of the hypothalamic-pituitary-adrenal axis. Furthermore, alterations in neurotrophic factors might contribute to the pathogenesis of schizophrenia. We aimed to evaluate the effects of a simulated laughter intervention on the levels of cortisol and BDNF and to determine whether the effects associated with simulated laughter could be sustained after discontinuation of the intervention. Methods In this randomized controlled study, patients with schizophrenia according to DSM-IV clinical criteria were randomly assigned to receive either 8-week-long simulated laughter intervention (n=32) or treatment-as-usual group (control group, n=27). The serum levels of BDNF and cortisol were measured at baseline, week 8, and four weeks after discontinuation (week 12) of the intervention program. Results After an 8-week simulated laughter intervention, the laughter group had significantly higher levels of BDNF; however, four weeks after discontinuation of the intervention, the levels of BDNF significantly dropped. Interestingly, the levels of cortisol did not change significantly at week 8, but they were significantly elevated at week 12. The levels of BDNF and cortisol in the control group did not change significantly between week 0 and week 8. Conclusion These findings suggest that the simulated laughter intervention has an early effect on neurogenesis with a significant delayed effect on stress regulation in subjects with schizophrenia.

        Experience of patients with inflammatory bowel disease in using a home fecal calprotectin test as an objective reported outcome for self-monitoring

        Background/Aims: Fecal calprotectin (fC) level is a predictive marker of mucosal healing for patients with inflammatory bowel disease (IBD). Home fC tests are now available. We evaluated the performance of the smartphone-based IBDoc home testing system in patients with IBD and obtained their feedback as an objective patient-reported outcome. Methods: This prospective study enrolled consecutive patients with IBD in clinical remission. fC in the same stool sample was assessed by using both the laboratory test (Quantum Blue calprotectin test) and home test (IBDoc). The correlation between the 2 tests was analyzed using the Pearson method. In addition, the patients were asked to fill a questionnaire based on their experience. Results: Fifty-one patients with IBD (68 tests and 49 questionnaires) were included. The correlation between Quantum Blue test and IBDoc was good (r=0.776, P< 0.0001). After the test, 56% patients found IBDoc easy to perform, and 96% were satisfied with it. Thirty-nine patients (80%) had a strong ( >70%) probability to use it for future monitoring if the price was acceptable. By using 250 μg/g as the cutoff, the agreement between home test and laboratory results was 80%, and by using 600 μg/g as the cutoff, the agreement increased to 92%. Conclusions: The correlation between the laboratory and home tests was good. Most patients found the home test to be feasible and easy to use and preferred it over laboratory test and endoscopy for monitoring. Therefore, the home test could be used as an objective patient-reported outcome. (Intest Res 2018;16:546-553)

        Comparison of Transforming Growth Factor-Beta1 and Lovastatin on Differentiating Mesenchymal Stem Cells toward Nucleus Pulposus-like Phenotype: An In Vitro Cell Culture Study

        Study Design: In vitro cell culture study. Purpose: This study aims to investigate the impact of transforming growth factor-beta1 (TGF-β1) and lovastatin on differentiating human mesenchymal stem cells (MSCs) toward nucleus pulposus (NP)-like phenotype. Overview of Literature: MSCs offer a cell source to the cell-based therapy for intervertebral disc degeneration. TGF-β1 is used to induce MSCs to differentiate into NP-like cells; however, an undesired expression of collagen type I has been reported. Statins reportedly stimulate expression of bone morphogenetic protein-2 (BMP-2) and promote the chondrogenic phenotype to NP cells. However, the effects of statins with or without TGF-β1 on the differentiation of MSCs into NP-like cells remain unclear. Methods: Human MSCs were treated with TGF-β1 alone, lovastatin alone, and simultaneous or sequential treatment with TGF-β1 and lovastatin. After the proposed stimulation, the total RNA was extracted to assess the expression profile of NP cells-specific genes. Hematoxylin–eosin staining was used for examining the microscopic morphology. Furthermore, we detected the syntheses of S-100 protein, aggrecan , and collagen type II in the extracellular matrix using immunohistochemical staining. Results: Simultaneous or sequential treatment of TGF-β1 and lovastatin could further augment the BMP-2 overexpression compared with lovastatin-alone treatment. However, the mRNA expression of aggrecan and collagen type II was not compatible with the expression level of BMP-2 . Immunohistochemical studies revealed compatible production of aggrecan, collagen type II , and S-100 protein in all three groups treated with lovastatin. Cells in groups treated with lovastatin were less populated than that in the group treated with TGF-β1 alone. Conclusions: This study demonstrates a promising role of lovastatin in inducing human MSCs into NP-like cells. However, further optimization of cell density before lovastatin treatment, treatment duration, and combination with TGF-β1 are warranted to attain better stimulatory effects.

        Exploring the Influential Factors of Manufacturers’ Initial Intention in Applying for the Green Mark EcoLabel in Taiwan

        The Green Mark eco-label constitutes an essential program introduced by Taiwan’s Environmental Protection Administration to support environmental protection. In the manufacturing industry, the Green Mark eco-label is intended to enable differentiating between manufacturers that apply green processes and those that do not. This paper presents an assessment model for identifying manufacturers’ initial intention to apply the Green Mark eco-label. The model was derived from the certification process flow. A total of 200 structured questionnaires were distributed to manufacturers in Taiwan, and 172 responses were retrieved. The model was analyzed by applying structural equation modeling, and the correlation of review process and end certification process exhibited the strongest correlation. Improving the document verification stage is highly likely to increase the overall initial intention ofmanufacturers to participate in the Green Mark eco-label program. The overall certification process contributed 16% to the total initial intention of manufacturers to apply the Green Mark eco-label to their products.

        Study on the engineering and electricity properties of cement mortar added with waste LCD glass and piezoelectric powders

        This study used a volumetric method for design. The control group used waste Liquid Crystal Displayplay (LCD) glass powder to replace cement (0%, 10%, 20%, 30%), and the PZT group used Pd-Zr-Ti piezoelectric (PZT) powder to replace 5% of the fine aggregate to make cement mortar. The engineering and the mechanical and electricity properties were tested; flow, compressive strength, ultrasonic pulse velocity (UPV), water absorption and resistivity (SSD and OD electricity at 50 V and 100 V) were determined; and the correlations were determined by linear regression. The compressive strength of the control group (29.5-31.8 MPa) was higher than that of the PZT group (25.1-29 MPa) by 2.8-4.4 MPa at the curing age of 28 days. A 20% waste LCD glass powder replacement (31.8 MPa) can fill up finer pores and accelerate hydration. The control group had a higher 50 V-SSD resistivity (1870-3244 Ω.cm), and the PZT group had a lower resistivity (1419-3013 Ω.cm), meaning that the resistivity increases with the replacement of waste LCD glass powder. This is because the waste LCD glass powder contains 62% SiO2, which is a low dielectric material that is an insulator. Therefore, the resistivity increases with the SiO2 content.

        Fermentation process development for hyaluronic acid production by Streptococcus zooepidemicus ATCC 39920

        The development of a fermentation strategy for hyaluronic acid (HA) production by Streptococcus zooepidemicus ATCC 39920 has been explored. The specific HA productivity (YP/X) was affected by the medium carbon-tonitrogen (C/N) ratio rather than the specific growth rate of cells. Accordingly, HA fermentation should be performed in a balanced medium with an optimum C/N ratio of 2 : 1 in a batch culture. To improve the performance of the batch culture, the operation conditions for the fill-and-draw culture were investigated. It was found that the timing of medium exchange is critical for successfully performing fill-and-draw operations. Since streptococcal cells at the stationary phase might lose the capacity of HA synthesis, the displacement of the medium in a fill-and-draw culture should be started at the late exponential growth phase.
