Conservative therapy with metformin plus megestrol acetate for endometrial atypical hyperplasia

Objective: To compare the efficacy of metformin plus megestrol acetate (MA) with that of MA alone for treating endometrial atypical hyperplasia (EAH). Methods: This pilot study included 16 EAH patients who met at least one metabolic syndrome (MS) criterion and received either adjunctive metformin plus MA (MET group) or MA monotherapy (MA group). Each patient in the MA group received 160 mg of MA daily, whereas patients in the MET group received the same dose of MA plus 0.5 g of metformin thrice daily. Treatment response was assessed by histological examination of dilation and curettage specimens obtained after 12 weeks of therapy. Results: Each group had eight patients, and half of the patients in each group were diagnosed with MS. The complete response (CR) rate was 75% (6/8) in the MET group and 25% (2/8) in the MA group (p=0.105). Complications of MS did not affect the response rates in either group. In the MET group, 75% (3/4) of the patients had CR in the presence or absence of MS. In the MA group, 50% (2/4) of the patients with MS had CR, whereas no patient without MS had CR. No irreversible toxicities were observed. Conclusion: Metformin plus MA may be a potential alternative therapy for treating EAH, and the MS status of patients may have no effect on the efficacy of metformin plus MA therapy.

Adenocarcinoma of lung has high incidence and a poor prognosis, woith chemotherapy as the main therapeutic tool, most commonly with cisplatin. However, chemotherapy resistance develops in the majority of patients during clinic treatment. Mechanisms of resistance are complex and still unclear. Although annexin play important roles in various tumor resistance mechanisms, their actions in cisplatin-resistant lung adenocarcinoma remain unclear. Preliminary studies by our group found that in cisplatin-resistant lung cancer A549 cells and lung adenocarcinoma tissues, both mRNA and protein expression of annexins A1, A2 and A3 is increased. Using a library of annexin A1, A2 and A3 targeting combined molecules already established by ourselves we found that specific targeting decreased cisplatin-resistance. Taken together, the underlined effects of annexins A1, A2 and A3 on drug resistance and suggest molecular mechanisms in cisplatin-resistant A549 cells both in vivo and in vitro. Furthermore, the study points to improved research on occurrence and development of lung adenocarcinoma, with provision of effective targets and programmes for lung adenocarcinoma therapy in the clinic.
A False Positive 18F-FDG PET/CT Scan Caused by Breast Silicone Injection

We present here the case of a 40-year-old woman with a greater than 10 year prior history of bilateral breast silicone injection and saline bag implantation. Bilateral palpable breast nodules were observed, but the ultrasound scan was suboptimal and the magnetic resonance imaging showed no gadoliniumenhanced tumor. The 18F-FDG PET/CT scan showed a hypermetabolic nodule in the left breast with a 30% increase of 18F-FDG uptake on the delayed imaging, and this mimicked breast cancer. She underwent a left partial mastectomy and the pathology demonstrated a siliconoma.
Hot Stamping Die Design for Vehicle Door Beams using Ultra-High Strength Steel

The energy saving and safety is the eternal theme of development of automobile industry. Ultra-high strength steel automotive body structures of hot stamping have been widely used with these dual advantages that reducing vehicle weight while enhancing safety performance at the same time. The forming and quenching integration die was investigated in this paper with a vehicle door beam as an example, especially the whole die structure and hot stamping process were optimized through numerical simulation of die strength, cooling pipe arrangement and other related factors, and beam samples with tensile strength 1550Mpa, elongation rate 6.5% and shape accuracy of ±0.3mm were produced by this die. Moreover, the stiffness increased by 2.2 times than the original tube beam and 3.8 times of the intensity, which led to full score in C-NCAP Crash Test. By reducing the thickness cross section and the depth of drawing comparing with the original tubular beam,door beam weight could be reduced by 9.32%, which had effective energy-saving and emission-reduction effect.
Equalizing charger based on multi‑windings transformer and voltage‑doubler rectifiers

Energy storage batteries are currently composed of a large number of low-voltage battery cells. The state of charge of the cells is different due to the characteristic differences between cells. This causes some of the cells to be over-charged during charging and some of the cells to be over-discharging during discharging. To improve this phenomenon, this paper proposes a battery charger with an equalizing function that works during the charging and discharging processes. A performance comparison among the proposed charger and other similar topologies is carried out. The operational principle of the proposed strategy is analyzed and a control strategy with only a single voltage loop is proposed, which can guarantee two-stage charging. Experimental results show that the proposed charger can simultaneously achieve a high efficiency and the requirement of voltage equalization between cells.
The role of protein arginine-methyltransferase 1 in gliomagenesis

Protein arginine methyltransferase 1 (PRMT1), a type-I arginine methyltransferase, has been implicated in diverse cellular events. We have focused on the role of PRMT1 in gliomagenesis. In this study, we showed that PRMT1 expression was up-regulated in glioma tissues and cell lines compared with normal brain tissues. The knock-down of PRMT1 resulted in an arrest in the G1-S phase of the cell cycle, proliferation inhibition and apoptosis induction in four glioma cell lines (T98G, U87MG, U251, and A172). Moreover, an in vivo study confirmed that the tumor growth in nude mouse xenografts was significantly decreased in the RNAi-PRMT1 group. Additionally, we found that the level of the asymmetric dimethylated modification of H4R3, a substrate of PRMT1, was higher in glioma cells than in normal brain tissues and decreased after PRMT1 knock-down. Our data suggest a potential role for PRMT1 as a novel biomarker of and therapeutic target in gliomas. [BMB Reports 2012; 45(8): 470-475]

Commercial enological tannins from variousorigins have been widely used in modern wine making. Inorder to investigate the diverse quality of tannin products, aquick, accurate and simple UPLC method was developed,which could simultaneously determine 11 principle characteristiccomponents of hydrolysable tannins and condensedtannins. The optimum resolution of the tannins wasachieved on a Waters Acquity UPLC-BEH C18 column(2.1 mm 9 50 mm, 1.7 lm) at 280 nm with gradient elution. The method was validated to achieve desired specificity,linearity, precision, accuracy and stability. Thedeveloped method was successfully applied to 30 commercialenological tannins from different origins for theirquality evaluation. The 30 tannin samples were qualitativelydistinguished into hydrolysable, condensed or mixturetannins, and quantificationally classified into fourlevels of excellent, good, fair and poor products. Themethod could be used for quick evaluating the quality ofenological tannins in practical use.

Matrix metalloproteinase (MMP)-2 and MMP-9 are important proteases involved in invasion and metastasis of various tumors. Extra-gastrointestinal stromal tumors (EGISTs) are rare neoplasms. This study was performed to assess MMP-2 and MMP-9 expression in EGIST tissue samples for association with clinicopathological data from the patients. Twenty-one surgical EGIST tissue specimens were collected for analysis of MMP-2 and MMP-9 expression using immunohistochemistry. MMP-2 and MMP-9 proteins were expressed in all of the epithelial cell types of EGISTs, whereas they were only expressed in 75% of the spindle cell type, although there was no statistically significant difference (p>0.05). Expression of MMP-2 and MMP-9 proteins was associated with tumor size, mitotic rate, tumor necrosis, and distant metastasis (p<0.05). MMP-2 expression was linked with MMP-9 levels (p<0.05). However, there was no correlation between MMP-9 expression and age, sex, primary site, or cell morphology in any of these 21 EGIST patients (p>0.05). Moreover, expression of MMP-2 and MMP-9 proteins increased with the degree of EGIST risk. This study provided evidence of an association of MMP-2 and MMP-9 expression with advanced EGIST behavior.

The present study was conducted to evaluate the effect of a fraction of macroporous resin (FMR), a bioactive component of Smilax china L., on benign prostatic hyperplasia (BPH) in castrated rats induced by testosterone propionate. Rats were randomly divided into five groups: the negative control group (sham-operated), the model group, two FMR-treated groups (at doses of 300mg/kg and 600 mg/kg of body weight), and the positive control group (treated with finasteride at the dose of 3 mg/kg). Drugs were administered once a day for three consecutive weeks by gastric gavage. Prostates were weighed, testosterone and dihydrotestosterone (DHT) levels in serum were determined, and histopathological examinations were carried out. FMR treatment inhibited prostatic hyperplasia, reducing the DHT level in serum and improving the prostate gland morphology compared with the model group. The overall results of this study suggest that FMR is effective at inhibiting experimentally induced prostate enlargement, and it presents a valuable resource for the treatment of human BPH.
