Growing demands for the structure determination of biological macromolecules using cryogenic electron microscopy (cryo-EM) necessitate expansion of national research infrastructure. In particular, better accessibility to transmission electron microscopes (TEMs) is needed for sample screening and high-resolution data collection as well as for newly emerging cryo-electron tomography (cryo-ET) analysis. Here we introduce recently established Cryo-EM Research Center at Pusan National University. The center is equipped with high-end TEM for data collection, lower-end TEM for sample screening and focused ion beam-scanning electron microscope (FIB-SEM) for sample milling at cryogenic temperature. Ancillary devices for sample vitrification and computational resources are also available for routine cryo- EM workflow. The center is anticipated to contribute to the development of structural biology field in Korea.
Structural assessment of the tetramerization domain and DNA-binding domain of CP2c

Although the transcription factor CP2c has been recently validated as a promising target for development of novel anticancer therapy, its structure has not been solved yet. In the present study, the purified recombinant protein corresponding to the tetramerization domain of CP2c appeared to be well folded, whereas the Elf-1 domain showed a largely unfolded conformation. Particularly, the Elf-1 domain, which contains the putative DNA-binding region, showed a conformational equilibrium between relatively less-ordered and well-ordered conformers. Interestingly, addition of zinc shifted the equilibrium to the relatively more structured conformer, whereas zinc binding decreased the overall stability of the protein, leading to a promoted precipitation. Likewise, a dodecapeptide that has been suggested to bind to the Elf-1 domain also appeared to shift the conformational equilibrium and to destabilize the protein. These results constitute the first structural characterization of the CP2c domains and newly suggest that zinc ion might be involved in the conformational regulation of the protein.

Effect of Chlorella vulgaris on Immune-enhancement and Cytokine Production in vivo and in vitro

The object of this study was to investigate the immune-enhancing effects of Chlorella vulgaris (CV) on a deteriorated immune function by a protein-energy malnutrition (PEM) diet. Unicellular algae, CV were used as a biological response modifier. Male C57BL/6J mice were fed for 15 days with standard diet or a PEM diet, which is associated with decreased host immune defense. After 8 days, mice in the PEM diet group were orally administered by 0.05, 0.1, and 0.15 g/kg body weight of CV or distilled water. Nutritional parameters, and interferon (IFN)-γ levels were significantly increased in the blood serum of the CV (0.15 g/kg)-treated group (29.6±2.8 pg/mL) compared to the non-treated PEM group (4.1±0.4 pg/mL, p<0.05). In addition, cell proliferation and production of cytokines were investigated via a CV (0.01, 0.1, and 1 mg/mL) treatment using a human T cell line MOLT-4 cell. The CV treatment (1 mg/mL) significantly increased the production of both IFN-γ and interleukin (IL)-2 (51.3±3.4 and 285.9±18.8 pg/mL, respectively) compared to the control (51.3±3.4 and 442.6±14.3 pg/mL, respectively), but did not affect the production of IL-4. These results suggest that CV may be useful in improving the immune function.

A series of novel terpyridine-skeleton molecules containing conformational rigidity, 14 containing benzo[4,5]-furo[3,2,-ь]pyridine core and 15 comprising chromeno[4,3-ь]pyridine core, were synthesized, and their biological activities were evaluated. 3-(4-phenylbenzo[4,5]furo[3,2-ь]pyridin-2-yl)phenol (8) was determined to be a nonintercalative topo I and II dual catalytic inhibitor and 3-(4-phenylchromeno[4,3-ь]pyridine-2yl)phenol (22) was determined to be a non-intercalative tope IIa specific catalytic inhibitor by various assays. These two catalytic inhibitors induced apoptosis in addition to G1 arrest in T47D human breast cancer cells with much less DNA toxicity than etoposide. Compounds 8 and 22 significantly inhibited tumor growth in HCT15 subcutaneously implanted xenografted mice. The modification of compounds 8 and 22 with the introduction of a methoxy instead of a hydroxy group enhanced endogenous topo inhibitory activity, metabolic stability in diverse types of liver microsomes and improved pharmacokinetic parameters in rat plasma such as augmentation of bioavailability (41.3% and 33.2% for 2-(3-methoxyphenly)-4-4phenylbenzofuro[ 3,2,] pyridine (8-M) and 3-(4-phenylchromeno-[4,3-ь]pyridine-2-yl)methoxybenzene (22-M), respectively).

이산화탄소 분압은 마취의 기본 감시 항목이며, 분압과 분압의 파형을 분석하여 호흡관리를 위한 목적으로 활용되고 있다. 본증례는 마취 장비의 문제를 이산화탄소 분압의 파형을 감시하여 발견한 증례로서 이산화탄소 분압의 감시가 환자의 호흡관리 이외에도 마취 준비의 무결성을 확인하기 위한 목적으로도 매우 유용하게 이용되었기에 보고하고자 한다. 16-year-old man underwent emergency operation for open reduction and internal fixation due to left femoral bone fracture. At end of anesthetic induction, capnogram changed to abnormal shape on anesthetic monitor after position transposition of operating table. He was shown normal mechanical breathing sound via stethoscope on both lung fields. We changed a D-FENDTM Water Trap on anesthetic monitor. However capnogram was not corrected. Then we changed anesthesia gas sampling line. Capnogram was corrected to normal shape. We report the case of ruptured anesthesia gas sampling line identified by abnormal capnogram on anesthesia monitor during position transposition of operating table.
