        Fracture Behavior of Locally Corroded Steel Pipeline in District Heating System Using the Combination of Electrochemistry and Fracture Mechanics

        To identify the effects of the stress concentration at the pit on the fracture of the district heating (DH) pipeline, the stressincrease with the wall thickness reduction was formulated. The formula includes the stress intensity factor with the pittingcorrosion. As a result of the application of the DH pipeline conditions to the ASME B31.3 code formula and corrosion test,the interplay with only uniform corrosion and stress due to the internal pressure can not be an immediate reason for the DHpipeline fracture. The KIvalue which is calculated by loss of thickness due to both uniform and localized corrosion exceededfracture toughness value (42 MPa√m) of the weldment pipeline steel (ASTM A106 Gr. B) under all of the studied conditions.

      • cDNA Cloning and Genomic Structure of Glyceraldehyde-3-Phosphate Dehydrogenase from the Entomopathogenic fungus, Paecilomyces tenuipes-Jocheon

        Fungi belonging to the Paecilomyces spp. have recently been used as food and herbal medicines in Korea and are greatly popular as commercially available powdered supplement or dried fruiting body. Despite this acceptance and its use, little is known of the genes related to its reactive agents. Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces spp. based on the previous identification of ITS1 and ITS2 at the molecular level and collected from Jocheon Miryang, Korea. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes- Jocheon was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDH is comprised of 1,014bp encoding 338 amino acid residues. The deduced protein sequence of Pt-GAPDH showed higher homology with Beauberia bassiana-GAPDH(93% amino acid identity). Hydropathy analysis revealed that Pt-GAPDH protein is hydrophilic. The major three amino acids in its composition of amino acid residues were alanine(11.54%), valine(9.47%) and glycine(8.88%). The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA. More investigation works including gene expression, immunological analysis etc. will be carried continuously without hesitation after this presentation.

The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA.

        Candida utilis에 의한 Cytidine 5'-diphosphate Choline의 발효생산

        CDP-choline is known as an intermediate of lecithin biosynthesis, and as an important drug for nervous diseases of the brain. For the bioconversion of CMP and choline to CDP-choline, ATP is required as an energy source. In these studies, the biosynthetic reaction of CDP-choline was coupled with ATP regenerating system by glycolysis. As a microorganism containing the highest conversion activity of CMP and choline to CDP-choline, Candida utilis ATCC 42416 was selected. The optimum reaction condition were 50mM choline chloride, 20mM CMP, 100mM potassium phosphate (pH8.0), 300mM glucose, 50mM MgSO₄, 10% dried cells with shaking incubation at 30℃. The reaction was thus performed for 10 hours under the above optimum conditions. The concentration of CDP-choline was 16mM (80% in conversion ratio).

        축대칭 다단계 딥드로잉 공정의 성형인자에 대한 연구

        Formability in deep drawing process depends not only on a drawability of workpiece material but also on process conditions such as die punch corner radius, lubricant conditions, punch-die clearance etc. For instance, bending resistance should be reduced by increasing die round appropriately, drawing load should be minimized by improving the lubricant condition between die and material, and blanking load should be increased by selecting a pertinent punch round and by augmenting the friction resistance in punch. In this study, a multi-stage deep drawing process is analyzed using ABAQUS. The effects of formability factors, such as die shoulder radius, punch-die clearance and coefficient, are investigated, and the results are also discussed in detail.

      • 제주재래마 특이 표지인자 개발

        본 연구는 제주재래마의 유전적 특성파악과 이를 통한 타품종과의 유전적 특이성을 가지는 DNA 표지인자를 개발하고자 PCR-RAPD기법을 이용하여 제주재래마와 더러브렛, 가고시마의 3품종에서의 다형 band에 대한 특성파악을 실시한 결과이다. 각 품종별로 10두의 혈액을 혼합한 후 분석하였고 200개의 random primer들 중 PCR증폭양상의 재현성을 기준으로 70개의 primer를 선별하였다. 또한 선별된 primer들중 다형성을 9개의 primer를 이용하여 개체별 DNA분석을 실시하였으며 9개의 prkmer에 의해 나타난 15개의 band는 제주재래마 특이 band, 4개는 더러브렛종에 그리고 4개는 가지고시마 재래마에 품종특이 RAPD band가 나타났다. 따라서 RAPD기법에 의해 품종간의 차이를 보이는 표지인자의 개발이 가능할 뿐 아니라 품종간의 특성차이를 보이는 유전인자 개발과 특성파악이 용이한 것으로 평가되었다. This study was carried out using PCR-RAPD technique to estimate genetic characteristics of Cheju Horse (Jorangmal) and to develop the DNA marker for bread identification in horse. Each three horse breeds was represented by pooled DNA from 10 individuals and tested for 200 different random primers for their reproducible PCR pattern. From them 70 primer were selected and 9 primers were used to detect the difference between breeds at the individual level. Fifteen highly polymorphic bands were observed between breeds using 9 primers . Additionally, the 15 most polymorphic bands showed great difference between breeds in their appearing frequencies, implicating their usefulness in the breed discrimination and in the characterization of genetic structure. Therefore, the polymorphism of RAPD could be used in characterizing genomic difference between breeds and some genetic mechanism that give rise to breed disctimination in horse.

        관계형 데이터베이스를 이용한 배낭문제 해법기의 구현

        Knapsack problems represent many business application such as cargo loading, project selection, and capital budgeting. In this research we developed a knapsack problem solver based on Martello-Toth algorithm using a relational database management system on the PC platform. The solver used the menu-driven user interface. The solver can be easily integrated with the database of decision support system because the solver can access the database to retrieve the data for the model and to store the result directly.

        A Reports were done on proriasis which were treated by medications of Sasang Constitution and Constitution-Acupunture in Dept. surgery, opthalmology &tolaryngology, college of Oriental Medicine, Kyungsan University, Pohang, Korea, from May 30. 2000 to JUN 30 2001. we gave score and checked involvement, plaque, erythematous popules, invasiveness according to psoriasis area and severity index. The results were as follows: 1. Sasang Constitution classification was Soyanggin 14(93.33%), Taeumgin 1(6.67%) in the 15 cases. Hyungbangsabaeksan is used in 9 cases(60%), Yanggeuksanhoatang is used in 3 cases(13.11%), Geopungchunggisan is used in 3 cases(20%), Hyungbangdojucksan is used in 1 case(6.57%). 2. In the 9 cases, we checked for over 4 weeks, clinical severity of involvement, plaque, total score(psoriasis area and severity index) was siginificantly decreased(p<05). clinical severity of erythematous popules, itching was decreased but lt is not siginificant(p>05). 3. The mean duration of treatmemt was 73.3 days, result of Excellent or Good improvement was showed on over 120 days. These resilts indicate that Sasang Constitution and Constitution-Acupuntuer treatment is effetive on psoriasis and the more study is needed.

      • 광탄성 실험법에 의한 크레인훅의 응력해석

        An experimental study for a crane hook was performed to investigate the stress distribution along a certain line where the maximum and minimum stresses to be developed. The crane hook was modeled into a 2-dimensional plate made of urethane rubber called 'Photoflex'. The materiel of Photoflex is very sensitive to a load and has low photoelastic fringe constant. The photoelastic techniques used in this experiment were the Tardy compensation method, the fringe thinning process and the 4-step phase shifting method. Experimental results by photoelasticity were compared with the stresses obtained from the simple curved beam theory and finite element analysis. All the results were close to each other.
