Deiodinases and the Three Types of Thyroid Hormone Deiodination Reactions

Thyroid hormone (TH) signaling is strictly regulated by iodothyronine deiodinase activity, which both preserves the circulating levels of the biologically active triiodothyronine (T3) and regulates TH homeostasis at the local level, in a cell- and time-dependentmanner. Three deiodinases have been identified—namely iodothyronine deiodinase 1 (DIO1), DIO2, and DIO3—that differ in theircatalytic properties and tissue distribution. The deiodinases represent a dynamic system that changes in the different stages of life according to their functions and roles in various cell types and tissues. Deiodinase activity at the tissue level permits cell-targeted fineregulation of TH homeostasis, mediating the activation (DIO1 and DIO2) and inactivation (DIO3) of THs. Deiodinase homeostasisis the driving force that leads T3-target cells towards customized TH signaling, which takes into account both the hormonal circulating levels and the tissue-specific response. This review analyzes the complex role of deiodinases in physiological and pathologicalcontexts, exploring new challenges and opportunities deriving from a deeper knowledge of the dynamics underlying their roles andfunctions.
CEO Power and Decision-Making under Pressure

Are powerful CEOs more effective in responding to pressure from the economic environment? Concentrating decision-making power may facilitate rapid decision-making. However, the quality of decision-making may be compromised, with severe consequences for the firm if a powerful CEO is less likely to receive independent advice or to have his decisions scrutinized. We empirically investigate the relation between CEO power and decision-making under pressure by examining firm performance when industry conditions deteriorate. We focus on industry downturns since these represent an exogenous ‘shock’ to a firm’s environment. The decision making context is important and we focus on three settings where the net effect of CEO power is likely more consequential: when the firm is innovative and decision making is likely more complex, when the industry is competitive and poor decisions can be more serious in terms of firm value and when the industry is characterized by high managerial discretion. In these settings powerful CEOs are found to perform significantly worse than other CEOs during industry downturns -- suggesting contexts in which centralized decision-making is potentially of greater concern.
Robotized inspection and health monitoring in the Gran Sasso National Laboratory

The Gran Sasso National Laboratory (LNGS) is the largest underground research center in the world devoted to neutrino and astroparticle physics. It is located in galleries below about 1400 meters of rock mass. In this environment, inspection and monitoring actions are challenging for the maintenance and the safety of the infrastructures and they require a combined use of different strategies. The paper address issues related to the structural safety of the whole environment by proposing solutions for inspection and monitoring of different areas and elements, such as the gallery vaults, the structures of the experimental prototypes, the plants and the machinery. A generic framework is discussed to evidence the features of each specific solution and the interaction between different systems. Tunnel structural healthy is the most difficult to evaluate because the vaults are coated by not removable panels which waterproof and insulate the environment. Therefore, specific solutions are proposed for the inspection and monitoring of the vaults which are visible only in the interspace realized from such cladding panels. In this respect, different methodologies based on the use of robotic systems are presented and discussed in order to implement a suitable inspection and monitoring program. The complementary requirements to perform a mechatronic survey are defined also as basis of ongoing activities currently performed in LNGS.
Dynamical models of a suspension bridge driven by vibration data

The great availability of measurement systems permits to acquire quite easily data related to structural oscillations in operational conditions. This occurrence may permit to enhance our capability to data-driven computing using directly experimental data and pertinent constraints and conservation laws, such as compatibility and equilibrium, surely certain. In the paper, a methodology will be presented to furnish an analytical mechanical model of a suspension bridge in which the main parameters can be derived from vibration measurements. In this respect, Polymax and Enhanced Frequency Domain Decomposition identification procedures are used to determine a complete modal model which is used to evaluate an error function. Optimization algorithms are used to evaluate the function minima in the fundamental parameter space. The procedure will be validated by results coming from a sophisticated finite element model for which geometric measurements are included through a 3D point cloud geometrical model and a consequent Building Information model (BIM) constructed with images acquired by unmanned aerial vehicle (UAV). The case study of the pedestrian cable suspension Polvorines bridge (100 meters of span) is considered to demonstrate the procedure, due the test campaign conducted on March 2020.

Background/Aims: Endoscopic ultrasound-guided fine-needle aspiration (EUS-FNA) is the go-to method for obtaining samplesfrom gastrointestinal tract and pancreatic lesions. When the transduodenal approach is utilized, the use of a more flexible needle,such as a nitinol 19-gauge (G) needle, has been recommended. The aim of this study was to evaluate the feasibility and accuracy of19-G flexible aspiration needles in obtaining samples from solid lesions through a transduodenal approach. Methods: This was a retrospective analysis of prospectively collected data from eight Italian endoscopy centers. Consecutive patientswith solid lesions who underwent transduodenal EUS-FNA with a 19-G flexible needle were included. Results: A total of 201 patients were enrolled. According to histology, EUS, radiology and 12 months of follow-up, 151 patientshad malignant lesions and 50 patients had benign lesions. EUS-FNA was feasible in all cases. An adequate histologic sample wasobtained in all except eight cases (96.1%). The sensitivity of EUS-FNA was 92.1% (95% confidence interval [CI], 86.8%–95.7%), andthe specificity was 100% (95% CI, 90.5%–100%). The positive predictive value was 100% (95% CI, 93.4%–100%), and the negativepredictive value was 74% (95% CI, 62.8%–82.7%). The diagnostic accuracy was 93.5% (95% CI, 89.2%–96.5%). Conclusions: The transduodenal approach for obtaining samples from solid lesions using a 19-G flexible needle seems feasible andaccurate.
Quality assessment of cellular therapies: the emerging role of molecular assays

Cellular therapies are becoming increasingly important in treating cancer, hematologic malignancies, autoimmune disorders, and damaged tissue. These therapies are becoming more effective and are being used more frequently, but they are also becoming more complex. As a result, quality testing is becoming an increasingly important part of cellular therapy. Cellular therapies should be tested at several points during their production. The starting material, intermediate products and the final product are usually analyzed. Products are evaluated at critical steps in the manufacturing process and at the end of production prior to the release of the product for clinical use. In addition, the donor of the starting biologic material is usually evaluated. The testing of cellular therapies for stability, consistency, comparability and potency is especially challenging. We and others have found that global gene and microRNA expression analysis is useful for comparability testing and will likely be useful for potency, stability and consistency testing. Several examples of the use of gene expression analysis for assessing cellular therapies are presented.
Cariprazine Add-on in Inadequate Clozapine Response: A Report on Two Cases

Cariprazine is a novel antipsychotic drug that exerts partial agonism of dopamine D2/D3 receptors with preferential binding to the D3 receptor, antagonism of 5HT2B receptors, and partial agonism of 5HT1A. Currently, cariprazine has shown clinical efficacy in patients with schizophrenia and with bipolar disorder, as well as adjunctive treatment in patients with Major Depressive Disorder (MDD) and drug-resistant MDD. In the present case series, we report on two patients with treatment-resistant schizophrenia and partial response to clozapine who benefit from combination with cariprazine. The effects of cariprazine combination were remarkable also concerning the adverse metabolic effects of clozapine.
