        산소 장력이 치주인대에서 유래한 세포의 활성과 기능에 미치는 영향

        정치료를 위해 발거한 제 1 소구치에서 사람의 치주인대 세포를 채취하여 37℃, 5% CO₂, 100% 습도의 환경에서 배양한 후 5 내지 6 계대배양하여 실험에 사용하였다. Gaspack system에 0.2㎛ Millipore filter를 부착하여 세균 감염을 막았고, 저산소군에는 10% O₂, 5% CO₂, 85% N2 가스를, 과산소군에는 90% O₂, 5% CO₂, 5% N₂ 가스를 연결하여 산소 장력에 변화를 주었으며 대조군에는 5% CO₂, 95% 공기를 공급하였다. 37℃에서 2, 4, 6일간 배양한 후에 세포활성을 측정하기 위해 tetrazolium(MTT) assay를 시행하였고 sulforhodamine B(SRB) assay를 이용하여 총 단백질 합성을 알아보았으며, 4-hydroxyproline 측정으로 교원질 합성을 측정해 보았다. 또한 효소면역측정법으로 IL-1β, Il-6, TNF-α를 측정하여 다음과 같은 결과를 얻었다. 1. 저산소군에서 세포 활성과 총 단백질 합성은 대조군보다 다소 높거나 같은 정도이었다. 2. 과산소군에서 세포 활성은 대조군보다 낮았으며 총 단백질 합성도 다소 감소되었다. 3. 교원질 합성은 저산소군과 과산소군에서 대조군보다 유의하게 감소되었다가 배양기간이 길어짐에 따라 증가되어 대조군과 유사한 정도에 달했다. 4. 효소면역법 측정 결과 IL-6, TNF-α, IL-1β순으로 다량 검출되었다. 5. IL-6, TNF-α, IL-1β는 저산소군과 과산소군에서 배양기간이 길어짐에 따라 대조군에 비해 급격히 증가하였다. 6. IL-6, TNF-α는 과산소 조건에서 배양6일 후에는 대조군에 비해 유의하게 증가하였으며 이는 배양 2일 또는 4일 후보다도 유의하게 증가한 것이었다. This study was undertaken to investigate the effect of oxygen tension on the activity and function of the cells derived from human periodontal ligament by measuring cell activity, total protein synthesis, collagen synthesis, IL-1β, IL-6, TNF-α. Human periodontal ligament fibroblasts were collected from premolars extracted for orthodontic treatment and incubated in the environment of 37℃, 5% CO₂, 100% humidity. After the fifth to sixth passage they were used for the experiment. Gaspack system to which 0.2㎛ Millipore filter was attached was connected to mixed-gas tanks. The mixed gases were composed of 10% O₂, 5% CO₂, 85% N₂ in hypoxic group of 90% O₂, 5% CO₂, 5% N₂ in hyperoxic group and 5% CO₂, 95% air for control. After incubation in 37℃ for 2, 4, 6 days, cell activity was determined by tetrazolium(MTT) assay and total protein synthesis was assayed using sulforhodamine B(SRB). And measurement of 4-hydroxyproline was performed to assess collagen synthesis and IL-1β, IL-6 and TNF-α were measured by enzymeimmunoassay. The results were as follows. 1. The cell activity and total protein synthesis in hypoxic group were a little higher than or almost the same with those in control group. 2. In hyperoxic group, the cell activity was lower than that in control group and total protein synthesis was decreased. 3. Collagen synthesis was significantly decreased initially in both hypoxic and hyperoxic group and increased nearly to the level of control group as the duration of cell incubation was longer. 4. As a result of enzymeimmunoassay, the amount of cytokines was IL-6, TNF-α and IL-1β in order. 5. IL-6, TNF-α and IL-1β were increased more rapidly in both hypoxic and hyperoxic group than in control group as the duration of cell incubation was longer. 6. There were more Il-6 and TNF-α in hyperoxic group than in control group after 6 days, and there were more Il-6 and TNF-α after 6 days than after 2 or 4 days in hyperoxic group. These results suggested that oxygen tension might modulate the production of extracellular matrix and cytokines in the cells derived from human periodontal ligament.

      • 폐 강판으로부터 철분말 제조 기술에 관한 연구(I)

        This paper aims to study on manufacturing of steel powder and iron powder through carburizing and decarburizing the waste beverage steel cans of as-collected without melting. Metallographic characteristics were investigated by OM, SEM, EDS, XRD and emission spectrometer. The results obtained are as follows: 1. 2-piece and 3-piece waste beverage steel cans are mainly made of 99.5wt%Fe and 0.3wt%Mn, and their cover and tap are composed of AI-4.3wt%Mg. 2. The paints and Al alloy cover and tap of the waste beverage steel cans could be separated and removed by holding at 630℃ for 30min. in the rotary heat furnace and retort. 3. The waste beverage steel cans could be carburized in the atmosphere of air plus propane gas by holding at 930℃ for 60min. and decarburized in that of H20 plus H2 by 700℃ x 1hr treatment. 4. The apparent density, flow rate, green density.,and rattler value examined of the decarburized powder are 3.42g/㎤, 28.3s/50g, 6.67g/㎤ and 0.621, respectively.

      • 한국여성에서 자궁내막증의 발생위험도와 Estrogen Receptor-α 유전자 다형성과의 관련성에 관한 연구

        Objectives: To investigate whether polymorphism of gene encoding estrogen receptor-a is asso-ciated with the risk of endometriosis in Korean women. Material and Methods :We investigated 136 patients with histopathologically confirmed endo-metriosis rAFS stage III/IV and 251 control group women who were surgically proven to have no endometriosis. Polymerase chain reaction(PCR) and restriction fragment length polymorphism (RFLP) of PCR products were done to determine each participant's estrogen receptor-a genotype. Results : The distridution according to PvuII genetic polymorphism of estrogen receptor-a were as follows. PP, Pp and pp were 14.7%(20 women), 39.0%(53 women) and 46.3%(63 women) in the study group and 13.9%(35 women), 38.6%(98 women) and 47.4%(119 women) in the con-trol group, respectively. There was no significant difference between the study group and the control group. Conclusion : The results suggest that estrogen receptor-a genetic polymorphism may not be associated with the development of endometriosis in Korean women. 목적: 자궁내막증은 에스트로겐에 의존적인 질환이므로 에스트로겐의 합성, 대사 및 작용에 관여하는 유전자의 다형성이 자궁내막증의 발생기전에 중요한 역할을 할 수 있다. 이에 본 연구에서는 한국인 여성에서 에스트로겐 수용체-a의 유전자 다형성이 자궁내막증의 발생 위험도를 증가시키는 지에 대해 연구하고자 하였다. 방법: 1996년 9월부터 2003년 8월가지 본원 산부인과에서 수술을 통해 병리조직학적으로 자궁내막증 III기와 IV기를 확인한 한국인 여성 136명을 대상으로 하였다. 대조군은 자궁내막증 환자군과 연령이 비슷한 만삭 산모에서 제왕절개술을 시행하거나 양성 난소낭종으로 수술을 시행 하였을 때, 자궁내막증이 없음을 확인한 여성 251명 으로 하였다. 결과: ER-a 유전자의 PvuII 다형성의 분포는 자궁내막증 환자군에서 PP군이 20명(14.7%), Pp군이 53명(39.0%), pp군이 63명(46.3%) 이었고 대조군에서의 분포는 각각 35명(13.9%), 97명(38.7%), 119명(47.4%)으로 나타났으며 자궁내막증 환자군과 대조군 사이의 유의한 차이는 없었다. Pp, pp형을 가지는 경우가 자궁 내막증 환자의 85.3%(116명), 대조군의 86.1%(216명)로 나타났으며, 이 경우 자궁내막증이 발생할 odds ratio가 0.904(95% CI, 0.519~1.702)로 통계적으로 유의한 차이가 없었다. 결론: 이상의 결과로 볼 때, 한국인 여성에서 자궁내막증의 발생위험과 에스트로겐 수용체 a 유전자 다형성간에 연관성은 없는 것으로 나타났다.

      • 한반도 중서부 지각구조와 물성연구 : 부여록옥광산에서 산출되는 귀사문석의 광물학적 특성 Mineralogy of precious serpentine from the Booyo Green Jade Mine

        충남 부여군 외산면에 위치한 부여록옥광산은 원래 석면을 대상으로 가행되었던 광산이었다가 폐광된 후 최근 세계적으로도 매우 희귀한 보석으로 인정받고 있는 귀사문석이 산출되고 있음이 발견되어 현재 채광작업을 재개하고 있다. 부여산 귀사문석은 광산의 주변암체인 선캠브리아기의 편암을 관입압 사문암체를 후기에 이를 다시 관입한 그로슐라 석류석맥의 접촉부를 따라 발달하고 있다. 이번 연구에서는 부여산 귀사문석에 대한 광물학적, 보석학적 특성을 규명하기 위하여 편광현미경, 비중계, 굴절계, 경도계, X선회절분석기, 화학분석, 적외선분광분석법 등을 이용해 연구하였다. 부여산 귀사문석은 짙은 녹색을 띠고 아투명하며 높은 지방광택을 나타난다. 모스경도는 H=5∼6이며 인성은 매우 높은 편이다. 비중은 2.67, 스포트법으로 측정한 굴절율은 No=1.56이다. X선회절분말양상은 7.40(100), 4.64(25), 3,68(68), 2.757(69), 2.530(49), 2.459(32), 2.113(32), 1.710(21)으로 안티고라이트임을 알 수 있었으며, 모암인 사문암도 안티고라이트로 구성되어 있다. 그러나 귀사문석과 사문암의 투명도와 경도 등이 다른것은 구성광물결정의 조직차이가 큰 요인인 것으로 여겨진다. 귀사문석의 주요 화학성분은 42.49% SiO_2, 39.08% MgO, 3.85% Fe_2O_3, and 11.87% H_2O 등이며, 미량원소로는 Cr(2188 ppm), Ni(1110ppm), Co(58 ppm), and Ta(108 ppm) 등이 있다. 적외선분광분석결과는 3600, 1190, 1070, 980, and 610 cm^-1에 흡수 peak를 나타내었다. 부여산 귀사문석(안티고라이트)은 감람석과 휘석으로 구성된 초염기성암이 변질되어 형성된 것으로 해석되며 그 생성반응은 (Fe, Mg)_2SiO_4 (감람석)+(Mg, Fe)SiO_3 (휘석)+2H_O→M_3sI_2O_5(OH_4)(티고라이트)+Fe_3O_4 (자철석). The Booyo Green Jade mine, located in Oesan-myon, Booyo-gun, Chungnam-do, had originally been developed for asbestos. Recently it has re-opened after discovery of precious serpentine. The precious serpentine is a gem variety of serpentine and is regarded as a rare and valuable gem material in the world. The precious serpentine develops along the contact between grossular garnet vein within the host rock serpentinite that had intruded schist of Precambrian age in the area. In this study mineralogical and gemmologicl properties of the precious serpentine from Booyo have been investigated by using polarizing microscope, specific gravity balance, refractometer, hardness tester, X-ray diffractometer, XRF and ICP-MS analyers, and infrared absorption spectroscope. The Booyo precious serpentine is deep green in color an semi-transparent, and also yields a highly oily luster. Mohs's hardness is measured to be H=5-4 with excellent toughness. Specific gravity is determined to be 2.67, and refractive index N_D=1.56 by a distant-vision method. Xray diffraction data shows peaks of 7.40(100), 4.64(25), 3,68(68), 2.757(69), 2.530(49), 2.459(32), 2.113(32), 1.710(21), which corresponds well to that of antigorite. The higher transparency and hardness of the precious serpentine than the neighboring serpentinite are most probably due to textural arrangement of constituent antigorite crystals. The major chemical constituents of precious serpentine are 42.49% SiO_2, 39.08% MgO, 3.85% Fe_2O_3, and 11.87% H_2O with trace elements Cr(2188 ppm), Ni(1110ppm), Co(58 ppm), and Ta(108 ppm). Infrared absorption spectroscopic analysis shows peaks at 3600, 1190, 1070, 980, and 610 cm^-1, Formation of the precious serpentine(antigorite) from ultramafic rock, which was made up of olivine and pyroxene, may represented by the following reaction: (Fe, Mg)_2SiO_4 (olivine)+(Mg, Fe)SiO_3 (pyroxene)+2H_O→M_3sI_2O_5(OH_4)(antigorite)+Fe_3O_4 (magnetite).

      • 홍삼 엑기스를 함유한 분말주의 제조 및 평가

        To develop a dry alcohol containing red ginseng extract, dry alcohols composed of ethanol, water, dextrin and sodium lauryl sulfate were prepared using spray dryer, and their ethanol contents and encapsulation efficiencies were deter-mined. An optimal dry alcohol containing red ginseng extract was chosen and the feeling for its oral administration was eval-uated. Dextrin at dextrin/water weight ratios below 1.6/1 and ethanol at ethanol/water weight ratios below 1/1 remarkably increased both the ethanol contents and encapsulation efficiencies of dry alcohols. However, dextrin at dextrin/ water weight ratios above 1.6/1 and ethanol at ethanol/water weight ratio above 1/1 slightly decreased the both parameters. It might be due to the low solubility of dextrin in ethanol and limited diffusion coefficient of ethanol to the dextrin shell. Furthermore, 0.5%(w/w) sodium lauryl sulfate gave the maximum ethanol content of dry alcohol. The more increased amounts of red ginseng extract were added, the more increased amounts of ginsenoside Rb1 but the more decreased amounts of ethanol were encap-sulated in dry alcohols. A dry alcohol containing red ginseng extract was prepared with dextrin/ethanol/water(1/1/1, w/w/w) mixed solution, in which 0.5%(w/w) sodium lauryl sulfate and 20%(w/w) red ginseng extract were dissolved. It contained the ethanol contents of 31.17±1.33%(w/w) and ginsengoside Rb1 of 243.0±7.0㎍/g. It gave the moderate taste of red ginseng extract at its oral administration with or without water. Thus, the dry alcohol containing red ginseng extract can be further developed as a more convenient dosage form for red ginseng extract.

      • 중추신경계 낭미충증에 관한 임상적 고찰

        We reviewed clinical features of 133 cases with neurocysticercosis who admitted at Severance Hopital Yongdong Severance Hospital and Chosun University Hospital from March, 1985 to February, 1991 The clinical results were summarized as follows; 1. The ages ranged from 10 to 76 years(mean age 50.0 years)and most of them were 5th or 6th decades 2. Among 133 cases of neurocysticercosis, Parenchymal type was 83 cases (62.4%), mixed type was 25cases(18.8%), ventriculomeningeal type was 23 cases (17.3%) and spinal type was 2 cases (1.5%). 3. The most two frequent initial symptoms were seizure (53.4%) and headache(24.1%). And frequent neurologic symptoms were seizures(64.7%), headache(61.7%), focal neurologic deficit(30.8%), mental changes (13.5%), visual disturbances (12.0%), vertigo(9.8%), cranial nerve pa1sy(6.8%), behavioral symptoms(3.0%), and urinary incontinence (2.3%) in order. 4. There were statistically significant differences in CSF 1eucocytosis(P <0.01), CSF / serum glucose ratio(P < 0.05) between different types. But there were no differences in CSF pressure, protein and glucose level, and ELISA titer. 5. 108cases(81.2%) who received medical treatment only had good prognosis. The overall mortality was 3.7% (5 cases) and causes of death were postoperative infection and shunt malfunction, all occured in ventnculo-meningeal type.

      • 초고집적소자의 층간절연막용 polysilazane계 spin on glass (SOG)에 관한 연구

        초고집적소자의 층간절연막으로서의 polysilazane계 spin on glass (SOG)의 적용 가능성을 평가하였다. 상용화된 무기 SOG 계열의 polysilazane과 polysilazane에 산화제를 첨가하여 SiO_2로의 변환 능력을 향상시킨 물질을 spin coating하고 curing하여 SOG 막을 제조한 다음, 400℃에서 900℃까지의 온도범위에서 열에 SOG막을 형성하여 gapfill 특성을 비교하였으며, SOG막에 contact hole을 형성한 다음 습식 세정에 대한 저항성을 평가하였다. 산화제를 첨가함으로써 polysilazane polymer 구조에서 SiO_2로의 변환이 더욱 효과적으로 이루어져, 기존 SOG 막에 비해 밀도 및 습식세정 chemical에 대한 저항성이 우수하고거 gapfill 특성은 동등한 수준인 SOG 막을 제조할 수 있었다. 특히, 기존의 SOG 막의 경우 좁은 gap space 내에서 curing이 충분하게 이어나지 않아 습식 세정시 hole profile이 나빠지는 문제를 산화제 첨가를 통해 해결할 수 있었다. We have investigated the feasibility of spin on glass (SOG) film from polysilazane-type resin as a premetal dielectric (PMD) layer of the next-generation ultra-large scale integrated (ULSI) devices. A commercial polysilazane resin and a polysilazane-type resin with oxidzing agent were spin-coated and cured to form SOG films. In order to study the effect of oxidizing agent and annealing, the SOG films were characterized as cured and after annealing at 400℃ to 900℃. The density and the resistance against wet chemical of the SOG films were improved by the addition of oxidizing agent, because oxidizing agent enhanced the conversion from polysilazane polymer to siO_2. The hole profile issue associated with insufficient curing of polysilazane innarrow gaps was alao resolved by oxidizing agent, while the gapfill capability of SOG was not deteriorated by oxidizing agent.
