      • TmSR-C, scavenger receptor class C, plays a pivotal role in antifungal and antibacterial immunity in the coleopteran insect Tenebrio molitor

        Scavenger receptors (SRs) constitute a family of membrane-bound receptors that bind to multiple ligands. The SR family of proteins is involved in removing cellular debris, oxidized low-density lipoproteins, and pathogens. Specifically, class C scavenger receptors (SR-C) have also been reported to be involved in phagocytosis of gram-positive and -negative bacteria in Drosophila and viruses in shrimp. However, reports are unavailable regarding the role of SR-C in antifungal immune mechanisms in insects. In this study, a full-length Tenebrio molitor SR-C (TmSR-C) sequence was obtained by 5'- and 3'-Rapid amplification of cDNA ends-polymerase chain reaction (RACE-PCR). The TmSR-C full-length cDNA comprised 1671 bp with 5'- and 3'-untranslated regions of 23- and 107-bp, respectively. TmSR-C encodes a putative protein of 556 amino acid residues that is constitutively expressed in all tissues of late instar larvae and 2-day-old adults, with the highest transcript levels observed in hemocytes of larvae and adults. TmSR-C mRNA showed a 2.5-fold and 3-fold increase at 24 and 6 h after infection with Candida albicans and β-glucan, respectively. Immunoassay with TmSR-C polyclonal antibody showed induction of the putative protein in the cytosols of hemocytes at 3 h after inoculation of C. albicans. RNA interference (RNAi)-based gene silencing and phagocytosis assays were used to understand the role of TmSR-C in antifungal immunity. Silencing of TmSR-C transcripts reduced the survivability of late instar larvae at 2 days post-inoculation of C. albicans, Escherichia coli, or Staphylococcus aureus. Furthermore, in TmSR-C-silenced larvae, there was a decline in the rate of microorganism phagocytosis. Taken together, results of this study suggest that TmSR-C plays a pivotal role in phagocytosing not only fungi but also gram-negative and -positive bacteria in T. molitor.

      • Measurements of surgeons’ exposure to ionizing radiation dose: comparison of conventional and mini C-arm fluoroscopy

        <P>This study was performed to measure the equivalent scattered radiation dose delivered to susceptible organs while simulating orthopaedic surgery using conventional and mini C-arm fluoroscopy. In addition, shielding effects on the thyroid, thymus, and gonad, and the direct exposure delivered to the patient's hands were also compared. A conventional and mini C-arms were installed in an operating room, and a hand and an operator phantom were used to simulate a patient's hand and a surgeon. Photoluminescence dosimeters were used to measure the equivalent dose by scattered radiation arriving at the thyroid, thymus, and gonad on a whole-body phantom in the position of the surgeon. Equivalent scattered radiation doses were measured in four groups: (1) unshielded conventional C-arm group; (2) unshielded mini C-arm group; (3) lead-shielded conventional C-arm group; and (4) lead-shielded mini C-arm group. Equivalent scattered radiation doses to the unshielded group were significantly lower in the mini C-arm group than those in the conventional C-arm group for all organs. The gonad in the lead-shielded conventional C-arm group showed the highest equivalent dose among operator-susceptible organs, and radiation dose was reduced by approximately 96% compared with that in the unshielded group. Scattered radiation was not detected in any susceptible organ in the lead-shielded mini C-arm group. The direct radiation dose to the hand phantom measured from the mini C-arm was significantly lower than that measured from the conventional C-arm. The results show that the equivalent scattered radiation dose to the surgeon's susceptible organs and the direct radiation dose to a patient's hand can be decreased significantly by using a mini C-arm rather than a conventional C-arm. However, protective lead garments, such as a thyroid shield and apron, should be applied to minimize radiation exposure to susceptible organs, even during use of mini C-arm fluoroscopy.</P>

        Interpretation of toxicological activity of ionic liquids to acetylcholinesterase inhibition via in silico modelling

        For designing environmentally friendly ionic liquids (ILs), their structural effects on the toxicity should be interpreted via modelling based on the quantitative-structure-activity-relationship (QSAR) concept. For the purpose, QSAR models for predicting IL toxicity in acetylcholinesterase activity were developed by using linear free-energy relationship (LFER) descriptors, whose chemical meanings are well defined. These are excess molar refraction (E<SUB>c or a</SUB>), dipolarity/polarizability (S<SUB>c or a</SUB>), H-bonding acidity (A<SUB>c or a</SUB>), H-bonding basicity (B<SUB>c or a</SUB>), McGowan volume (V<SUB>c or a</SUB>), and ionic interactions of cation (J<SUP>+</SUP>) and anion (J<SUP>-</SUP>). Since the experimentally determined LFER descriptors are not available, we calculated them based on density functional theory, conductor-like screening model and the open-source software, obprop. The toxicity values of imidazolium- and pyridinium-based ILs could be predicted by a combination of four descriptors (A<SUB>c</SUB>, B<SUB>c</SUB>, V<SUB>c</SUB> and S<SUB>a</SUB>) with an R<SUP>2</SUP> of 0.828, and (E<SUB>c</SUB>, A<SUB>c</SUB>, E<SUB>a</SUB> and S<SUB>a</SUB>) with an R<SUP>2</SUP> of 0.879, respectively. In prediction study using the overall dataset containing various IL structures, the six calculated terms (E<SUB>c</SUB>, S<SUB>c</SUB>, A<SUB>c</SUB>, J<SUP>+</SUP>, E<SUB>a</SUB>, and S<SUB>a</SUB>) were selected and correlated with the observed toxicity values in R<SUP>2</SUP> of 0.748 for the training set, R<SUP>2</SUP> of 0.711 for the test set and R<SUP>2</SUP> of 0.655 for external validation set. And this study explains how the selected terms are contributing to the prediction models, and their chemical meanings were understood.

      • Shewanella marina sp. nov., isolated from seawater

        <P>A motile, rod-shaped, pale-brown-pigmented bacterium, designated strain C4T, was isolated from seawater collected from the South Sea (Republic of Korea). Cells were Gram-negative, facultatively anaerobic, and catalase- and oxidase-positive. The major fatty acids were summed feature 3 (C16:1omega7c and/or iso-C15:0 2-OH; 19.4%), C16:0 (16.3%), C17:1omega8c (9.5%) and iso-C15:0 (7.7%). The DNA G+C content was 40.8 mol%. A phylogenetic tree based on 16S rRNA gene sequences showed that strain C4T formed a lineage within the genus Shewanella (92.7-96.1% sequence similarity to representative strains of the genus Shewanella) and was part of a distinct branch with the clade comprising Shewanella haliotis DW01T and Shewanella algae ATCC 51192T. Phenotypic characteristics enabled strain C4T to be distinguished from S. haliotis and S. algae. On the basis of the data presented in this study, strain C4T represents a novel species, for which the name Shewanella marina sp. nov. is proposed. The type strain is C4T (=KCTC 22185T=JCM 15074T).</P>

        Terrestrial n-alkanes and their carbon isotope records from the Hanon paleo-maar sediment, Jeju Island, Korea: Implications for paleoclimate and paleovegetation over the last 35 kyrs

        <P>The carbon isotope of total carbon (delta C-13(org)), long-chain n-alkanes, and their compound-specific carbon isotope ratios (delta C-13(ALK)) were investigated in the Hanon paleo-maar sediment, Jeju Island of Korea to understand paleoclimate variabilities and their paleovegetation linkages. Based on the organic geochemical data (TOC (%), TN (%), and their delta C-13(org) and delta N-15), the core column stratigraphy was divided into three units, namely from the bottom to 14.5 ka (Unit I), from 14.5 ka to 3.01<a (Unit II), and from 3.0 ka to the core top (Unit III), respectively. In particular, delta C-13(org) showed marked fluctuation from -17.31%o to -28.68%o, suggesting different organic carbon sources. A relatively narrow range of variation in delta C-13(org) was observed in Unit III, and drastic changes in delta C-13(org) were observed from Unit I to Unit II. This may indicate a predominance of C-4 plants in Unit I, and drastic changes from terrestrial C-4 to C-3 in Unit II and predominance of C-3 plants in Unit III. The distributions of n-alkane were characterized by a continuous predominance of odd-numbered n-alkanes, particularly nC(29) and nC(31), and by high fluctuation of the total n-alkanes concentration. The average chain length (ACL), carbon preferences index (CPI), and paleoplant proxy (Paq) showed high fluctuation and glacial-interglacial variations with distinctive a high and low ratio at about 9.2 ka, corresponding to the switching points of delta C-13(org) and a high concentration of nalkane distribution. Individual n-alkane odd-numbered isotopes of delta C-13(Auc) ranged between -11.80%o in delta C-13n(21) and -34.93%o in delta C-13n(31), suggesting different sources of n-alkanes. The distribution of n-alkanes and their individual delta C-13(Auc), in particular delta C-13n(21), support paleovegetation changes, and their time dependent variations matched well with glacial-interglacial paleoclimate variations. Therefore, organic geochemical proxies recorded in Hanon paleo-maar sediment reflects paleoclimate variabilities as well as paleovegetation changes for the last 35 kyr in Jeju Island, Korea.         Holocene environmental change at the southern coast of Korea based on organic carbon isotope (δ<sup>13</sup>C) and C/S ratios

        <P>To trace the environmental change that occurred at a coastal area during the Holocene, we performed radiocarbon dating and geochemical analyses using sedimentary cores recovered from the Beolgyo tidal flats on the south coast of Korea. The delta C-13 values of total organic carbon (TOC), which represent relative inputs of terrestrial (or freshwater) and marine organic sources, revealed a long-term increasing trend from -27.5 to -21.7 parts per thousand during the period between 10,000 and 3000 cal BP, suggesting an evolutionary series of fluvial-dominant to marine-dominant environments, coupled with weakened summer monsoon intensity. From 3000 cal BP to the present, the delta C-13(TOC) decreased gradually from -21.7 to -24 parts per thousand, indicating enhancement of freshwater input during this time. The observed long-term environmental change was divided into six stages based on the averages of the total sulfur content (TS%) and the organic carbon to sulfur ratio (C/S), proxies for paleosalinity. On multi-centennial to millennial timescales, higher C/S ratios and lower TS% were linked to higher delta C-13(TOC) values, indicating strong freshwater input at Stage 2 (similar to 8300-7400 cal BP), Stage 4 (similar to 4500-3300 cal BP), and Stage 6 (similar to 1100 cal BP to present). Strong freshwater discharge events synchronously occurred at 4300 cal BP and 3600 cal BP along the south coast of Korea. Further testing should be carried out in other East Asian coastal areas. This study suggests that sedimentary delta C-13(TOC) values and C/S ratios can be used in Asian coastal regions to trace past freshwater input events and summer monsoon intensity during relatively stable sea level periods (e.g., the middle to late Holocene).       • Electrocatalytic activity of chemically deposited Cu<sub>x</sub>S thin film for counter electrode in quantum dots-sensitized solar cells

        The compact (c-Cu<SUB>x</SUB>S) and the porous (p-Cu<SUB>x</SUB>S) with particle decorated films of coppers-ulfidearesynthesized using a chemical bath deposition technique, and the films are characterized using electrochemical techniques. In addition, the chemically deposited Cu<SUB>x</SUB>S films are investigated as a counter electrode in quantum dots-sensitized solar cells (QSSCs). The available redox active reaction sites of the p-Cu<SUB>x</SUB>S film are found to be 57.9% higher than those available in the c-Cu<SUB>x</SUB>S film. From the electrochemical impedance spectroscopy, the effective diffusion coefficients of the polysulfide electrolyte in the c-Cu<SUB>x</SUB>S and p-Cu<SUB>x</SUB>S films are estimated to be 3.67 x 10<SUP>-5</SUP> and 6.35 x 10<SUP>-5</SUP> cm<SUP>2</SUP> s<SUP>-1</SUP>, respectively. These results can be ascribed to the improvement in the available redox active reaction sites and the electrocatalytic activity of the Cu<SUB>x</SUB>S counter electrode. As compared to the c-Cu<SUB>x</SUB>S film, the p-Cu<SUB>x</SUB>S film as a counter electrode exhibits an enhanced photovoltaic performance of the QSSCs with the power conversion efficiency of 3.17%, short-circuit current of 11.89 mA c<SUP>-</SUP>m<SUP>2</SUP>, open-circuit voltage of 0.50 V, and fill factor of 53.29. The improved performance of the QSSCs is ascribed to the improvements on the available redox active reaction sites, electrocatalytic activity and the diffusion coefficients, which are directly related to the surface morphology of the sulfide films.

        In the result of the research on the interests to the physical strength code system of boy high school (Comparison) of advancing to higher education and nonadvancing to higher education), it was possible to deduce the following conclusion and the suggestions from the theoretical background and the assumptions mentioned above forwarded by this research. 1. The understanding in the system of P.S.C. in inefficient. 2. The system failed to provide to one's strengh buiding 3. The training for P.S.C. is always not practiced consistently but being exercised on the immediate of inspection. 4. The understanding on the itemized purpose of its inspection is relatively low. 5. The inspection has something to do with the entrance examination and conderned because of the directive score points. 6. The inspection system failed to show a fairness and a lot of contradictions have been pointed out in the course of its execution. 7. In a comparison of advancing to higher education and nonadvanoing to higher education, the latter has not considered a special necessity. 8. The relative importance for the entrance examination by current inspection of P.S. has shown as fitted one. 9. It is not well comed to practice only in the training of P.S.C. at the assigned time to P.S. From the above conclusion, the following suggestions are required. 1. It is required that the system of P.S.C. is to be equally alotted to the all physical curriculum so that the guidance with the possitive plans be undertaken in the ordinal time. 2. It should not be limited of the system of P.S.C. in reflecting to the result of entrance examination for the boys of advancing to higher educations, and but considered in the dimension of national aspects, and the systematic backing policy enabling to exercise with a interest to the system of P.S.C even for the group of non-advancing for higher education be necessary. 3. A objectivity making the inspection thorough and correct in the course of execution, of the system of P.S.C. is required. 4. The fairness of inspection should be maintained by a sufficient training in advance for the staffs of the physical strength inspection and the inconsistencies should be developed. 5. The items of sport(Swiming, physical exercise, shooting etc.) beside the items of current P.S. inspection is recommended to be added. In the end, by strengthening the grade of current P.S. inspection, it is necessary to let people know that it Is very hard to get a special grade without a consistent effort in ordinary day.

All rights reserved</P><P>Background</P><P>The presence of latent tuberculosis (TB) infection (LTBI) should be evaluated before kidney transplantation. Although a new T cell-based assay for diagnosing LTBI gave promising results, this assay has not yet been compared with the tuberculin skin test (TST) for diagnosing LTBI in renal transplant candidates before transplantation.</P><P>Patients and methods</P><P>All adult patients admitted to a single institute for renal transplantation over a 1-year period were prospectively enrolled. A clinically predictive risk of LTBI was defined as: (i) recent close contact with a person with pulmonary TB; (ii) abnormal chest radiography; (iii) a history of untreated or inadequately treated TB; or (iv) a new infection (i.e., a recent conversion of TST).</P><P>Results</P><P>Of 209 renal recipients, 47 (22%) had a positive TST≥5 mm, 21 (10%) had a positive TST≥10 mm, 65 (30%) had a positive T-SPOT.<I>TB</I> test, and 25 (12%) had an indeterminate T-SPOT.<I>TB</I> test. The induration size of TST was significantly associated with a high positivity rate on T-SPOT.<I>TB</I> (<I>P</I><0.001). Agreement between T-SPOT.<I>TB</I> test and TST≥10 mm was fair (<I>k</I>=0.24, 95% confidence interval 0.11–0.36). However, neither univariate nor multivariate analysis showed any association between the clinical risk for LTBI and positivity on T-SPOT.<I>TB</I> or TST.</P><P>Conclusion</P><P>T-SPOT.<I>TB</I> test was more frequently positive than TST in renal transplant candidates. However, further longitudinal studies are awaited to determine whether the ability of T-SPOT.<I>TB</I> assay to detect LTBI in renal transplant recipients can better predict the development of TB than can TST after transplantation.</P>
