        AGS 인체위암세포에서 발효된 아가콩 추출물에 의한 apoptosis 유도

        본 연구에서는 대두(FS)와 아가콩의 발효추출물(FYA)의 항암활성 기전을 확인하기 위해 AGS 인체위암세포의 증식에 미치는 영향을 조사하였다. AGS 세포에서 FS 및 FYA 처리로 인하여 암세포의 증식이 처리 농도 의존적으로 강하게 억제하였고, apoptosis 유발을 의미하는 세포의 전반적인 형태 및 핵의 변형 또한 동반하였다. 또한 세포주기 분석을 통하여 이 현상이 apoptosis 유도에 의한 것임을 확인하였다. AGS 세포에 처리된 FS 및 FYA는 pro-apoptotic factor인 Bax의 발현 증가를 통한 intrinsic pathway나, death receptor 관련 유전자의 발현 증가를 통한 extrinsic pathway를 활성화시키며, 더 나아가서 IAP family인자의 발현 억제 및 caspases의 활성 증가를 일으켜 apoptosis를 유발시키는 것을 유추할 수 있었는데, 이러한 효과들은 FS보다 FYA에서 더욱더 탁월하였다. 이는 향후 아가콩 발효추출물이 항암치료를 위한 적용 가능성이 매우 우수함을 제시하여 주는 결과이다. Extracts of soybeans fermented by Bacillus subtilis have a wide variety of functions, such as enhancing the body's immune function, fibrinolysis activity, anti-inflammation, anti-cancer, estrogen function and anti-infection effects. Recently, it was reported that the extracts of fermented beans exhibit strong anti- inflammatory and anti-cancer properties by suppressing the transcription of pro-inflammatory cytokine genes and induction of apoptosis, respectively. However, the mechanisms of their cytotoxicity in human gastric cancer cells are poorly understood. In the present study, we investigated the effects of ethyl alcohol extracts from fermented soybean (FS) and yellow agabean (FYA) on cell growth and apoptosis in AGS human gastric cancer cells. A treatment of FS and FYA inhibited the growth of AGS cells in a concentration-dependent manner by inducing apoptosis. FS- and FYA-induced apoptosis were associated with down-regulation of XIAP and cIAP-2, and up-regulation of pro-apoptotic Bax expression. Moreover, a treatment of FS and FYA not only triggered an increase in the levels of death receptor (DR)4, DR5, Fas and FasL, but also induced the activation of casepase-3, -8 and -9. These findings illustrate that FS and FYA may have a therapeutic potential in human gastric AGS cells and as a functional food.

        Comparative analysis of fat and muscle proteins in fenofibrate-fed type II diabetic OLETF rats: the fenofibrate-dependent expression of PEBP or C11orf59 protein

        Fenofibrate, an agonist of PPARα, plays an important role in activating many proteins catalyzing lipid metabolism, and it also has a considerable effect on improvement of insulin sensitivity in the diabetic condition. To investigate fenofibrate- dependent expression of peripheral tissue proteins in diabetes, we analyzed whole muscle or fat proteins of fenofibrate-fed OLETF rats, an animal model of type II diabetes, using 2-dimensional gel electrophoresis. We found that many proteins were specifically expressed in a fenofibrate-dependent manner in these diabetic rats. In particular, a functionally unknown C11orf59 protein was differentially expressed in the muscle tissues (about 5-fold increase) in fenofibrate-fed OLETF rats as compared to control rats. Additionally, the signal proteins phosphatidylethanolamine binding protein and IkB interacting protein were differentially regulated in the fenofibrate-treated adipose tissues. We suggest here that these proteins might be involved in controlling lipid or carbohydrate metabolism in diabetes via PPARα activation. [BMB reports 2010; 43(5): 337-343]

        Bacillus sp.에 의하여 발효된 신품종 아가콩 발효 산물의 혈전 용해 활성 및 항산화 효과

        본 연구에서는 혈전용해능이 우수한 Bacillus sp. sm26을 선별하고 신품종 기능성 콩인 노란 아가콩(YA)와 검정 아가콩(BA)을 발효시킨 발효콩(FYA, FBA)의 혈전용해 활성과 70% ethanol 추출(FYAE, FBAE)에 대한 항산화 효능을 조사하였다. FYA와 FBA의 혈전용해 활성은 각각 6.38±0.5와 6.83±0.5 U/ml로 대두 발효물(FSB)보다 각각 1.3배와 1.4배 높은 것으로 나타났다. 총 페놀 함량은 FYAE와 FBAE가 각각 3.40±0.44 ㎎/g, 2.45±0.20 ㎎/g으로 나타나, 대두 발효 추출물(FSBE)에 비하여 약 2배 이상의 높은 페놀 함량을 나타내었다. FYAE의 총 단백질과 총 당 함량은 각각 0.56±0.11 ㎎/g과 2.41±0.48 ㎎/g이었으며 FYBA는 각각 0.39±0.12 ㎎/g과 2.72±0.63 ㎎/g으로 나타나, FYAE가 FSBE보다 각각 4.7배와 1.7배 높은 총 단백질과 총 당 함량을 지닌 것으로 나타났다. DPPH radical 소거능은 FBAE가 1 ㎎/ml에서 79 %로 가장 높게 나타났으며 2배 높은 활성을 나타내었다. 환원력은 FBAE가 1 ㎎/ml의 농도에서 가장 높은 환원력을 나타내었으며 FSBE에 비해 2배 높은 활성을 보였다. 지질 과산화능은 1 ㎎/ml일 때 FBAE는 93%로 FSBE의 약 3배 높은 활성을 나타내었고 FYAE 또한 80%로 2.5배 높은 활성을 보였다. 과산화 수소에 대한 소거능은 1 ㎎/ml 농도에서는 FBAE가 82%로 FSBE에 비해 4배 높은 효과를 나타내고 FYAE의 경우 2.5배 높은 효과를 나타낸다. 이상의 연구 결과에서, 기능성 콩인 아가콩 발효물의 혈전용해 활성과 항산화 효능은 일반 콩에 높게 나타났으며 이는 앞으로 새로운 천연 기능성 식품으로서의 이용 가능성을 시사하고 있다. In this study, fibrinolytic activities of fermented yellow agabean (FYA) and black agabean (FBA), and the antioxidation efficiencies of 70% ethanol extract of fermented yellow agabean (FYAE) and black agabean (FBAE) were investigated by selecting Bacillus sp. sm26 strain. Fibrinolytic activities of FYA and FBA were 6.38±0.5 and 6.83±0.5 U/ml, which were 1.3 and 1.4 times higher than that of FSB, respectively. With regard to total phenolic contents, FYAE and FBAE were 3.40±0.44 ㎎/g and 2.45±0.20 ㎎/g respectively, suggesting that their contents were about twice as high as that of fermented soybean extract (FSBE) used as a control. In comparison with FSBE, total protein and sugar contents of FYAE were 0.56±0.11 and 2.41±0.48 ㎎/g, respectively, and those of FBAE were 0.39±0.12 and 2.72±0.63 ㎎/g, respectively. This result suggests that FYAE was 4.7 and 1.7 times higher than FSBE, respectively. The DPPH radical scavenging activity of FBAE was 79% at 1 ㎎/ml, which was highest among the fermented bean extracts, and was twice as high as FSBE in regards to activity. In addition, FBAE exhibited the highest reducing power at 1 ㎎/ml, which was higher than FSBE by two-fold. With regard to lipid peroxidation, FBAE and FYAE were 93% and 80% at 1 ㎎/ml, which were 3 and 2.5 times higher than FSBE, respectively. Of note, the hydrogen peroxide scavenging activities of FBAE and FYAE were 82% and 54% at 1 ㎎/ml, offering activity that was 4 and 2.5 times higher than FSBE, respectively. Based on these results, the fibrinolytic activity and antioxidation efficiency of the fermented agabeans were significantly higher than other soybeans. Therefore, these studies may suggest that the functional agabeans can be a potential candidate for a natural functional food.

        항공스포츠의 활성화 방안

        Paragliding was introduced 15 years ago in the country. But Paragliding has not been popularized nowadays because of several problems. After `88 Olympic Games, such leisure sports alike watersurface and under water sport events have developed in accordance with a local characteristic. The other hand, air-sports are in a stalemate now because of the nonchalance of the general public, and the idleness of the persons concerned. Therefore, the purpose of this study is to investigate the present condition of domestic paragliding and to promote the solution about them. Paragliding has not been popularized for the matter of the cultivation of leaders, the matter of the settlement of accidents, and matters of policy and facility. The followings are suggested to activate paragliding event in the country. First, We should try to lead the general public to participate in paragliding activity. The essential prerequisite is the amplification of paragliding space and facility for enjoying the event. Second, To widespread paragliding, it should be also strengthened to establish the policy to organize and support paragliding clubs. Third, All local paraglidng tournament should be developed through packageing various events, so as to make it local festival.

        수상스포츠 참가자의 스포츠 사회화 요인에 관한 연구

        The purpose of this study is to find out the participating frequency by analyzing the effect of the characteristics of participant, sport socialization subject, and situational features on the water sports participating frequency. To achieve this purpose, it was used that questionnaires to 207 people who reside in the cities of Kangrung, Taonghae, Samchuk, Sokcho in the east sea and participate in the water sports at least for 1 year. the results are as follows. 1. As to the characteristics of the participants, the age, marriage status, education level do not affect much on the water sports participating frequency. 2. As to the characteristics of socialization subject, the financial support from spouse and parents does not affect much on the water sports participating frequency, but the stronger the colleges` canvassing and encouragement and the better the instructor`s personal relationship is the higher the participating frequency becomes. 3. As to the characteristics of sports situation, the higher the satisfaction with the amount of program time and supporting facilities, the higher the participation frequency becomes but the satisfaction with the instructor does not affect much. In conclusion in the water sports participating frequency, the personal characteristics did not affect much and the characteristics of socialization subject-the financial support from spouse and parents did nut affect either. It is because of the speciality of this sea. As this area is nearby the sea and supporting facilities are avaliable, people can easily access to the water sports. As to the characteristics of sports situation, instructor`s teaching methods or contents did not affect much on the water sports participating frequency. It is because that after beginner`s period, people do not feel the need of help form other professionals. Therefore educational plan and system for professional instructors must be established.

        RKIP phosphorylation–dependent ERK1 activation stimulates adipogenic lipid accumulation in 3T3-L1 preadipocytes overexpressing LC3

        <P><B>Abstract</B></P> <P>3T3-L1 preadipocytes undergo adipogenesis in response to treatment with dexamethaxone, 1-methyl-3-isobutylxanthine, and insulin (DMI) through activation of several adipogenic transcription factors. Many autophagy-related proteins are also highly activated in the earlier stages of adipogenesis, and the LC3 conjugation system is required for formation of lipid droplets. Here, we investigated the effect of overexpression of green fluorescent protein (GFP)-LC3 fusion protein on adipogenesis. Overexpression of GFP-LC3 in 3T3-L1 preadipocytes using poly-<SMALL>L</SMALL>-lysine-assisted adenoviral GFP-LC3 transduction was sufficient to produce intracellular lipid droplets. Indeed, GFP-LC3 overexpression stimulated expression of some adipogenic transcription factors (e.g., C/EBPα or β, PPARγ, SREBP2). In particular, SREBP2 was highly activated in preadipocytes transfected with adenoviral GFP-LC3. Also, phosphorylation of Raf kinase inhibitory protein (RKIP) at serine 153, consequently stimulating extracellular-signal regulated kinase (ERK)1 activity, was significantly increased during adipogenesis induced by either poly-<SMALL>L</SMALL>-lysine-assisted adenoviral GFP-LC3 transduction or culture in the presence of dexamethasone, 1-methyl-3-isobutylxanthine, and insulin. Furthermore, RKIP knockdown promoted ERK1 and PPARγ activation, and significantly increased the intracellular accumulation of triacylglycerides in DMI-induced adipogenesis.
