      • Acremonium chrysogenum에 의한 Cephalosporin C 생산 및 모델링

        The production of cephalosporin C and its biosynthetic modeling were studied. Cultivation experiments were carried out in an agitated and aerated fermentor using Acremonium chrysogenum. Corn steep liquor was used as a major carbon source. Intracellular and extracellular concentrations of metabolites and enzymes were measured. Based on enzymatic kinetics the biosynthetic modeling of cephalosporin C was developed and used to compare the experimental data with simulation results. In the simulation the effect of dissolved oxygen concentraction on cephalosporin C production was investigated. For the high production yield of cephalosporin C the dissolved oxygen concentration should be kept over 40 % during the cultivation. The biosynthetic modeling can help further to understand the biosynthetic machinery of the microorganism to produce cephalosporin C.

      • 연쇄중합효소반응을 이용한 태아성별진단

        For sex determination by the PCR method, oligoprimers to Y-chromosome gene, DYZ1, SRY, and AMGL were synthesized and genomic DNA was extracted from male and female placenta for the control use. DYZ1 represented 154 bp single band to 0.001pg/ml male genomic DNA but did not represent 154 bp band in female genomic DNA, SRY represented 341 bp band to 1 pg/ml male genomic DNA but did not represent 341 bp band in female genomic DNA, and AMGL represented 977 and 788 bp double band to 1 pg/ml male genomic DNA and 977 bp single band to 1 pg/ml female genomic DNA in 2% agarose gel electrophoresis stained with ethidium bromide. DYZ1 was 1,000-fold sensitive than SRY and AMGL. DYZ1 and SRY could not identify the PCR failure from female but AMGL identified. To increase the sensitivity, the dual amplification of AMGL was performed and the sensitivity increased to 1,000-fold. During the dual amplification of female genomic DNA mixed with male genomic DNA, 0.00125pg/l, 1:400 part male genomic DNA contamination represented double bands as male. In 2 cases of 46, XY female, DYZ1 and AMGL amplification represented male band but SRY amplification did not represent male band. It was suggested that SRY gene was deleted in two 46, XY female cases. for fetal sex determination, PCR with DYZ1, SRY, and AMGL was performed in 10 cases of chorionic villi and 15 cases of amnionic cells. By the comparison with karyotyping result, fetal sex determination was achieved successfully in all 23 samples using PCR of SRY and AMGL but false result was detected in 3 cases(13%) using DYZ1. According to our results, it was concluded that DYZ1 was 1,000-fold sensitive than SRY and AMGL but could not be used because of its false results, and AMGL and SRY must be used concomitantly for precise sex determination.

        사회복지조직의 실제채용기준을 반영할 수 있는 채용공고에 대한 탐색적 연구 : 서울·대전지역을 중심으로

        본 연구의 목적은 사회복지조직의 공개 채용공고가 실제채용기준을 얼마나 반영하고 있는지 파악하여 채용공고의 변화가능성을 탐색하기 위함이다. 이러한 목적을 달성하기 위해 2008년 한국사회복지사협회의 채용공고 중 210건의 조직을 표집하여 기초분석을 통해 정보내용을 파악하였고, 서울과 대전의 23개 조직의 기관장을 대상으로 1:1면접을 통해 사례연구를 진행하였다. 분석결과, 실제채용기준은 자원봉사와 실습을 통한 현장경험, 사회복지사 1급 자격증, 지원자와 조직의 비전 일치, 사회복지 마인드, 대인관계 등으로 드러났다. 그러나 이는 채용공고에 제시되지 않거나 질적인 차이를 보였다. The purpose of this study is to understand how an open employment notice has reflected the actual criterions of the recruitment, and to discover a potential for any changes. To achieve this purpose, a systematic random sample of 210 open employment notices was selected from Korea Association of Social Workers in 2008, and a descriptive analysis was conducted. The qualitative research was carried out for the case study with 23 interviewees from social welfare organizations in Seoul and Daejeon. Consequently, the actual criterions of the recruitment included the following elements: social work field practicum and volunteer work, the level one social welfare license, degree of harmony with the vision of the organization, dept of social welfare-oriented mind, and human networks. However, these criterions may not be presented in open employment notices or show that there are qualitative differences between open employment notices and the actual employment criterions.

      • 잠재적으로 기능을 갖는 마우스 칼리크레인 유전자(mGK-21) 클로닝과 그 특성에 관한 연구

        Mouse glandular kallikreins are a highly homologous subfamily of serine proteases encoded by 25 genes, all located on chromosome 7^((8)). By protein and cDNA sequence analysis, it was demonstrated that prorenin converting enzyme(PRECE) in the ICR mouse submandibular gland is identical to the epidermal growth factor-binding protein(EGF-BP) type B, the mouse Grandular Kallikrein-13(mGK-13) gene product identified Balb/c^((10, 11)). To examine whether the mGK-21 which cleaves the mouse pro-mouse glandular kallikrein-21(mGK-21) to its mature form is involved in the processing of prorenin, We have cloned the cDNAs of the mGK-21 from a library of male ICR mouse submandibular gland(SMG) and determined the sequences of the cDNA of mGK-21. And then Chinese Hamster Ovary(CHO) cells were transfected with the mGK-21 expression plasmids, and stable cell lines expressing a high level of the mGK-21 precursor were obtained. The conditioned medium from CHO/GK21 cells was then treated with trypsin, since trypsin is known to activate prokallikrein. Therefore, We treated the conditioned medium of CHO/GK21 cells with trypsin, which has been demonstrated to effectively convert the pro mGK-21 to enzymematically active form. Then, the radiolabeled-conditioned medium of CHO/MRB cells, which stablely express mature Ren-2 prorenin, was incubated with the trypsin-treated CHO/GK21. In vitro translation, the product of mGK-21 cDNA was detected and in the northern blotting, mGK-21 were expressed in submandibular gland. In the present study, even though the recombinant mGK-21 do not show a prorenin converting activity, it is apparent that they may possess a serine protease activity.

      • CSVT법에 의해 증착시킨 CdS 박막의 전기적 특성

        CSVT법에 의해 증착시킨 CdS 박막의 전기적 특성 및 열전기 특성을 조사하기 위하여 시료의 X-선 회절, 직류 전기전도도, Hall 이동도 그리고 TSC를 측정하였다. 그 결과 CSVT 법에 의해 제작된 CdS 박막이 보통중착법에 의한 것에 비하여 결정성장이 향상되고 Hall 이동도가 크며 저항이 작다는 것을 알았다. 또한 TSC 측정결과, 광조사 조건에 따라 TSC 피크가 다르게 나타나며 이러한 피크 특성은 photochemical reaction model로서 설명될 수 있고, 그리고 제작조건이 다른 두 CdS 박막에서 나타나는 TSC 피크의 비교로부터 CSVT법에 의해 제작된 시료가 보통증착법에 의한 것보다 상온 이상에서의 photochemical reaction 효과 적음을 알 수 있다. To investigate electrical property and thermally stimulated current (TSC) for the CdS films which were grown by close-spaced vapor transport (CSVT), the measurements of X-ray diffraction pattern (XRD), d.c. conductivity, Hall mobility and TSC were carried out. Form the result of the measurements, CSVT-CdS films are found to be better oriented and to be less resistive with better mobility as compared with the films evaporated by the conventional method. TSC spectra under illumination conditions show different TSC peaks. These can be explained by photochemical reaction model. The result suggests that the effect of photochemical reaction above room temperature is to be less than conventional evaporated films in the case of CSVT-CdS films.

      • Rhizopus oryzae KCTC 11970을 이용한 푸마르산 생산조건의 최적화

        This work was developed in several steps to find the optimum conditions of fumaric acid production by Rhizopus oryzae KCTC 11970 in flask culture. Six different carbon sources and eleven nitrogen sources were tested. CaCO_3 was used as a neutralizing agent to control pH of medium. The optimum carbon source and nitrogen source were glucose 70 g/L and ammonium sulfate 0.4 g/L, respectively. At this optimum condition, the fumaric acid concentration, yield, and volumetric productivity were 19.33 g/L, 52.8%, and 0.32 g/L/h, respectivity, after fermentation time of 60 hr.

      • 결핵성 수막염 환자에서 결핵균 30 kDa 및 32 kDa 단백항원에 대한 항체반응의 비교

        The 30 and 32 kDa antigens from Mycobacterium tuberculosis H37Rv culture filtrates, identified as biologically important proteins in the immune responses against mycobacterial infection, were purified and used in enzyme linked ummunosorbent assay (ELISA) for the determination in specific IgG and IgM levels in cerebrospinal fluids of 15 patients with tuberculous meningitis and 17 controls with nontuberculous diseases. High reactivity to both antigens was observed in tuberculous meningitis. Mean IgG and IgM antibody levels differed significantly (P < 0.001) between patients and con trols. Mean IgG antibody levels were also higher than IgM levels in tuberculous meningitis. A comparison of the antibody levels against the 30 and 32 kDa antigen within the tuberculous meningitis patients showed higher IgG reactivity to the 30 kDa than to the 32 kDa antigen, suggesting that the antibody response of these patients is directed predominantly against the 30 kDa protein. However, the patterns of reactivity to 30/32 kDa, 30 kDa and 32 kDa antigen in individual subjects were similar. By the cut-off value adding 2 stsndard deviation to the mean absorbance of controls, the sensitivity and specificity of the IgG antibody to the 30 kDa antigen were 100% and 94.4%, respectively. These values were higher than those obtained by 30/ 32 kDa and 32 kDa antigen. From the above results, it is suggested that the 30 kDa antigen may be dominant antigen and more valuable in the diagnosis of tuberculous meningitis than 32 kDa antigen.
