Analysis of the Bacterial Community in a Full-scale Printing and Dyeing Wastewater Treatment System Based on T-RFLP and 454 Pyrosequencing

        Qingxiang Yang,Jia Wang,Xinkuan Han,Yuanyuan Xu,Dong Liu,Hongxin Hao,Xuemei L,Yuhui Guo,Tianqi Niu,Shiyue Qi 한국생물공학회 2014 Biotechnology and Bioprocess Engineering Vol.19 No.1

In this study, the bacterial dynamics andstructure compositions in the two-stage biological processof a full-scale printing and dyeing wastewater (PDW)treatment system were traced and analyzed by terminalrestriction fragment length polymorphism (T-RFLP) and454 pyrosequencing techniques. T-RFLP analysis showedthat the microbial communities experienced significantvariation in the process of seed sludge adaptation to thePDW environments and were in constant evolution duringthe whole running period of the system, despite theconstant COD and color removal effects. Pyrosequencingresults indicated that the two-stage biological systemharbored rather diverse bacteria, with Proteobacteria beingthe predominant phylum during the steady running period, although its microbial compositions differed. The firststageaerobic tank was dominated by α-Proteobacteria(89.05% of Proteobacteria), whereas in the second-stageaerobic tank, β- and γ-Proteobacteria, besides α-Proteobacteria,were the dominant bacterial populations.

Effect of Replacing Corn and Wheat Bran With Soyhulls in Lactation Cow Diets on In Situ Digestion Characteristics of Dietary Dry Matter and Fiber and Lactation Performance

        Meng, Qingxiang,Lu, Lin,Min, Xiaomei,McKinnon, P.J.,Xiong, Yiqiang Asian Australasian Association of Animal Productio 2000 Animal Bioscience Vol.13 No.12

An in situ digestion trial (Experiment 1) and a lactation trial (Experiment 2) were conducted to determine the effects of replacing corn and wheat bran with soyhulls (SH) in lactating dairy cow diets on the extent and kinetics of digestion of DM and NDF, and lactation performance. In experiment 1, five mixed feeds consisting of mixed concentrate and roughages (50:50 on a DM basis) were formulated on isonitrogenous and isoenergetic bases to produce five levels (0, 25, 50, 75 and 100%) of SH replacement for corn and wheat bran. SH had high in situ digestion (92 and 89% for potentially digestible DM and NDF) and fairly fast digestion rate (7.2 and 6.3 %/h for DM and NDF). Increasing level of SH replacement resulted in increased NDF digestibility (linear, p=0.001-0.04) and similar DM digestibility (beyond 12 h incubation, p=0.10-0.41). As level of SH replacement increased, percentage of slowly digestible fraction (b) of DM increased (linear, p=0.03), percentage of rapidly digestible fraction (a) of DM tended to decrease (linear, p=0.14), and DM digestion lag time tended to be longer (linear, p=0.13). Percentage of potentially digestible fraction (a+b) and digestion rate (c) of slowly digestible fraction of dietary DM remained unaltered (p=0.36-0.90) with increasing SH in the diet. Increasing level of SH for replacing corn and wheat bran in the diet resulted in increases in percentages of b (quadratic, p<0.001), a (linear, p=0.08), a+b (quadratic, p=0.001) and a tendency to increase in c for NDF (linear, p<0.19). It was also observed that there was a satisfactory fit of a non-linear regression model to NDF digestion data ($R^2=0.986-0.998$), but a relatively poor fit of the model to DM digestion data ($R^2=0.915-0.968$). In experiment 2, 42 lactating Holstein cows were used in a randomized complete block design. SH replaced corn and wheat bran in mixed concentrates at 0, 25, and 50%, respectively. These mixed concentrates were mixed with roughages and fed ad libitum as complete diets. Replacing corn and wheat bran with SH at 0, 25 and 50% levels did not influence (p=0.56-0.95) DM intakes (18.4, 18.6, and 18.5 kg/d), milk yields (27.7, 28.4 and 27.6 kg/d), 4% fat-corrected-milk (FCM) yields (26.2, 27.6, and 27.3 kg/d) and percentages of milk protein (3.12, 3.17 and 3.18%), milk lactose (4.69, 4.76 and 4.68%) and SNF (8.50, 8.64, and 8.54%). On the other hand, milk fat percentges linearly increased (3.63, 3.85 and 3.90% for SH replacement rates of 0, 25 and 50% in the diet, p=0.08), while feed costs per kg FCM production were reduced.

        勸導技術与小學生教育類App的融合設計硏究

        주경상(Zhu, Qingxiang),이진욱(Lee, Jin Wook) 한국전시산업융합연구원 2020 한국과학예술융합학회 Vol.38 No.2

Educational Apps have been widely used by elementary school students, but the design of them needs to be improved. The purpose of this paper is to study the convergence design of persuasive technology and elementary school students" educational App. Through the persuasive design, to alleviate the elementary school students" weariness, improve their learning enthusiasm and initiative. Firstly, the learning behavior of elementary school students is studied by means of participatory observation, and it is concluded that the learning behavior can be divided into four stages: the stage of learning intention, the stage of learning preparation, the stage of learning execution, and the stage of learning completion. On this basis, the factors affecting the weariness of elementary school students at each stage were obtained by means of participatory observation and interview. Then, by analyzing the influence factors of weariness in each stage, persuasive strategies are proposed from the three dimensions of motivation, ability and trigger, and the persuasive design strategies of elementary school students" educational Apps are summarized. Finally, the results of this study were applied in an App design project, and the effectiveness of these strategies was verified by a controlled experiment, which can help primary school students to improve their enthusiasm and initiative in learning and can effectively reduce the weariness of elementary school students. The results of this study can provide reference for the persuasive design of elementary school students" educational Apps.

Weighted Moore--Penrose inverses of adjointable operators on indefinite inner-product spaces

        Mengjie Qin,Qingxiang Xu,Ali Zamani 대한수학회 2020 대한수학회지 Vol.57 No.3

Necessary and sufficient conditions are provided under which the weighted Moore--Penrose inverse $A^\dag_{MN}$ exists, where $A$ is an adjointable operator between Hilbert $C^*$-modules, and the weights $M$ and $N$ are only self-adjoint and invertible. Relationship between weighted Moore--Penrose inverses $A^\dag_{MN}$ is clarified when $A$ is fixed, whereas $M$ and $N$ are variable. Perturbation analysis for the weighted Moore--Penrose inverse is also provided.

Effects of Nitrate Addition on Rumen Fermentation, Bacterial Biodiversity and Abundance

        Liping Zhao,Qingxiang Meng,Liping Ren,Wei Liu,Xinzhuang Zhang,Yunlong Huo,Zhenming Zhou 아세아·태평양축산학회 2015 Animal Bioscience Vol.28 No.10

This study examined changes of rumen fermentation, ruminal bacteria biodiversity and abundance caused by nitrate addition with Ion Torrent sequencing and real-time polymerase chain reaction. Three rumen-fistulated steers were fed diets supplemented with 0%, 1%, and 2% nitrate (dry matter %) in succession. Nitrate supplementation linearly increased total volatile fatty acids and acetate concentration obviously (p = 0.02; p = 0.02; p<0.01), butyrate and isovalerate concentration numerically (p = 0.07). The alpha (p>0.05) and beta biodiversityof ruminal bacteria were not affected by nitrate. Nitrate increased typical efficient cellulolytic bacteria species (Ruminococcus flavefaciens, Ruminococcus ablus, and Fibrobacter succinogenes) (p<0.01; p = 0.06; p = 0.02). Ruminobactr, Sphaerochaeta, CF231, and BF311 genus were increased by 1% nitrate. Campylobacter fetus, Selenomonas ruminantium, and Mannheimia succiniciproducens were core nitrate reducing bacteria in steers and their abundance increased linearly along with nitrate addition level (p<0.01; p = 0.02; p = 0.04). Potential nitrate reducers in the rumen, Campylobacter genus and Cyanobacteria phyla were significantly increased by nitrate (p<0.01; p = 0.01).To the best of our knowledge, this was the first detailed view of changes in ruminal microbiota by nitrate. This finding would provide useful information on nitrate utilization and nitrate reducer exploration in the rumen.

Commutability Assessment of Processed Human Plasma Samples for Normetanephrine and Metanephrine Measurements Based on the Candidate Reference Measurement Procedure

        Deng Yuhang,Liu Qingxiang,Liu Zhenni,Zhao Haijian,Zhou Weiyan,Zhang Chuanbao 대한진단검사의학회 2022 Annals of Laboratory Medicine Vol.42 No.5

Background: To identify candidate external quality assessment (EQA) materials for normetanephrine and metanephrine measurements, we assessed the commutability of eight processed human plasma samples. The agreement between routine assays and the candidate reference measurement procedure (cRMP) was also evaluated. Methods: Fifty-three clinical samples and eight processed plasma samples were prepared. The processed samples included pooled and individual plasma samples spiked with pure normetanephrine and metanephrine and non-spiked pooled and individual plasma samples. The clinical and processed samples were subjected to four routine isotope dilution tandem mass spectrometry assays and cRMP. Commutability was assessed based on two approaches recommended by the CLSI and International Federation of Clinical Chemistry (IFCC). Passing–Bablok regression and Bland–Altman analysis were used to evaluate the agreement between the routine assays and cRMP. Results: The commutability results of the CLSI approach were better than those of the IFCC approach. For the CLSI approach, spiked individual plasma samples and spiked high-concentration pooled plasma samples were commutable for all routine assays for both analytes. The non-spiked pooled plasma sample was commutable for two out of four routine assays for metanephrine and three out of four routine assays for normetanephrine. The agreement between the routine assays and the cRMP was satisfactory, except for one routine assay showing significant bias. Conclusions: High-concentration spiked pooled plasma samples and spiked individual plasma samples are candidate EQA materials for normetanephrine and metanephrine measurements.
