Identification of pathways and genes associated with cerebral palsy

Cerebral palsy (CP) is a non-progressive neurological disease, of which susceptibility is linked to genetic and environmental risk factors. More and more studies have shown that CP might be caused by multiple genetic factors, similar to other neurodevelopmental disorders. Due to the high genetic heterogeneity of CP, we focused on investigating related molecular pathways. Ten children with CP were collected for whole-exome sequencing by next-generation sequencing (NGS) technology. Customized processes were used to identify potential pathogenic pathways and variants. Three pathways (axon guidance, transmission across chemical synapses, protein–protein interactions at synapses) with twenty-three genes were identified to be highly correlated with CP. This study showed that the three pathways associated with CP might be the molecular mechanism of pathogenesis. These findings could provide useful clues for developing pathway-based pharmacotherapies. Further studies are required to confirm potential roles for these pathways in the pathogenesis of CP.
Hybrid ceramics-based cancer theranostics

Cancer is a major threat to human lives. Early detection and precisely targeted therapy/therapies for cancer is the most eff ective way to reduce the diffi culties (e.g., side eff ects, low survival rate, etc.) in treating cancer. To enable eff ective can- cer detection and treatment, ceramic biomaterials have been intensively and extensively investigated owing to their good biocompatibility, high bioactivity, suitable biodegradability and other distinctive properties that are required for medical devices in oncology. Through hybridization with other materials and loading of imaging agents and therapeutic agents, nanobioceramics can form multifunctional nanodevices to simultaneously provide diagnostic and therapeutic functions for cancer patients, and these nanodevices are known as hybrid ceramics-based cancer theranostics. In this review, the recent developments of hybrid ceramics-based cancer theranostics, which include the key aspects such as their preparation, bio- logical evaluation and applications, are summarized and discussed. The challenges and future perspectives for the clinical translation of hybrid ceramics-based cancer theranostics are also discussed. It is believed that the potential of hybrid ceramic nanoparticles as cancer theranostics is high and that the future of these theranostics is bright despite the diffi culties along the way for their clinical translation.
Parameter importance ranking for SBLOCA of CPR1000 with moment-independent sensitivity analysis

The phenomenon identification and ranking table (PIRT) is an important basis in the nuclear power plant (NPP) thermal-hydraulic analysis. This study focuses on the importance ranking of the input parameters when lacking the PIRT, and the target scenario is the small break loss of coolant accident (SBLOCA) in a pressurized water reactor (PWR) CPR1000. A total of 54 input parameters which might have influence on the figure of merit (FOM) were identified, and the sensitivity measure of each input on the FOM was calculated through an optimized moment-independent global sensitivity analysis method. The importance ranking orders of the parameters were transformed into the Savage scores, and the parameters were categorized based on the Savage scores. A parameter importance ranking table for the SBLOCA scenario of the CPR1000 reactor was obtained, and the influences of some important parameters at different break sizes and different accident stages were analyzed.

<P>ABSTRACT</P><P>The masses of the central supermassive black holes (BHs) and the Eddington ratios for a sample of 65 young radio galaxies [27 gigahertz-peaked spectrum (GPS) and 38 compact steep-spectrum (CSS) sources] are estimated by various methods. We find that the average BH mass of these young radio galaxies is 〈log <I>M</I><SUB>BH</SUB>〉≃ 8.3, which is less than that of radio loud quasi-stellar objects (QSOs) and low-redshift radio galaxies (〈log <I>M</I><SUB>BH</SUB>〉≃ 9.0). The CSS/GPS sources have relatively high Eddington ratios with average ratio 〈log <I>L</I><SUB>bol</SUB>/<I>L</I><SUB>Edd</SUB>〉=−0.56, which are similar to those of narrow-line Seyfert 1 galaxies (NLS1s). It suggests that the CSS/GPS sources may not only be in the early stage of radio activities, but also in the early stage of their accretion activities.</P><P>We find that the young radio galaxies as a class deviate systematically from the <I>M</I><SUB>BH</SUB>–σ<SUB>*</SUB> relation defined by nearby inactive galaxies, when using σ<SUB>[O <SMALL>III</SMALL></SUB> as a surrogate for the stellar velocity dispersion σ<SUB>*</SUB>. There is no significant correlation between the deviation of the [O <SMALL>III</SMALL>] emission line width, Δσ≡σ<SUB>[O <SMALL>III</SMALL>]</SUB>−σ<SUB>[pred]</SUB> and the jet/accretion power, where σ<SUB>[pred]</SUB> are calculated from the Tremaine et al. relation using the estimated BH masses. However, we find that the deviation Δσ in young radio galaxies is well correlated with the Eddington ratio, and this correlation is found to be similar to that of radio quiet active galactic nuclei (AGN) (QSOs/NLS1s) where the radio jet is absent or weak. We suggest that the accretion activities may still play an important role in shaping the kinematics of [O <SMALL>III</SMALL>] narrow line in these young radio galaxies.</P>

Objective: This study aimed to identify differentially expressed genes (DEGs) by transcriptome analysis to elucidate a potential mechanism by which governor vessel electroacupuncture (GV-EA) promotes neuronal survival, axonal regeneration, and functional recovery after complete transection spinal cord injury (SCI). Methods: Sham, control, or GV-EA group adult female Sprague Dawley rats underwent a complete transection SCI protocol. SCI area RNA-seq investigated the DEGs of coding and noncoding RNAs 7 days post-SCI. Gene ontology (GO) and kyoto encyclopedia of genes and genomes (KEGG) enrichment analyses were used to classify DEGs functions, to explain a possible molecular mechanism. Immunofluorescence and BBB (Basso, Beattie, and Bresnahan) score were used to verify a GV-EA treatment effect following SCI. Results: GV-EA treatment could regulate the expression of 173 mRNA, 260 lncRNA, and 153 circRNA genes among these DEGs resulted by SCI. GO enrichment analysis showed that the DEGs were most enriched in membrane, actin binding, and regulation of Toll-like receptor signaling pathway. KEGG pathway analysis showed enriched pathways (e.g. , Tolllike receptors, MAPK, Hippo signaling). According to the ceRNA network, miR-144-3p played a regulatory role by interacting with lncRNA and circRNA. GV-EA also promoted the injured spinal cord neuron survival, axonal regeneration, and functional improvement of hind limb locomotion. Conclusion: Results of our RNA-seq suggest that post-SCI GV-EA may regulate characteristic changes in transcriptome gene expression, potential critical genes, and signaling pathways, providing clear directions for further investigation into the mechanism of GV-EA in subacute SCI treatment. Moreover, we found that GV-EA promotes neuronal survival, nerve fiber extension, and motor function recovery in subacute SCI.
