        Identification of growth trait related genes in a Yorkshire purebred pig population by genome-wide association studies

        Objective: The aim of this study is to identify genomic regions or genes controlling growth traits in pigs. Methods: Using a panel of 54,148 single nucleotide polymorphisms (SNPs), we performed a genome-wide Association (GWA) study in 562 pure Yorshire pigs with four growth traits: average daily gain from 30 kg to 100 kg or 115 kg, and days to 100 kg or 115 kg. Fixed and random model Circulating Probability Unification method was used to identify the associations between 54,148 SNPs and these four traits. SNP annotations were performed through the Sus scrofa data set from Ensembl. Bioinformatics analysis, including gene ontology analysis, pathway analysis and network analysis, was used to identify the candidate genes. Results: We detected 6 significant and 12 suggestive SNPs, and identified 9 candidate genes in close proximity to them (suppressor of glucose by autophagy [SOGA1], R-Spondin 2 [RSPO2], mitogen activated protein kinase kinase 6 [MAP2K6], phospholipase C beta 1 [PLCB1], rho GTPASE activating protein 24 [ARHGAP24], cytoplasmic polyadenylation element binding protein 4 [CPEB4], GLI family zinc finger 2 [GLI2], neuronal tyrosine-phosphorylated phosphoinositide-3-kinase adaptor 2 [NYAP2], and zinc finger protein multitype 2 [ZFPM2]). Gene ontology analysis and literature mining indicated that the candidate genes are involved in bone, muscle, fat, and lung development. Pathway analysis revealed that PLCB1 and MAP2K6 participate in the gonadotropin signaling pathway and suggests that these two genes contribute to growth at the onset of puberty. Conclusion: Our results provide new clues for understanding the genetic mechanisms underlying growth traits, and may help improve these traits in future breeding programs.

        Experimental and Numerical Study on Fatigue Behavior of RC Beams Strengthened with U-Shaped Steel Casing

        To achieve a rapid and great enhancement in the fl exural performance of reinforced concrete beams, this study proposes a new strengthening method using U-shaped steel casing and focuses on studying the fatigue performance of strengthened beams. In this paper, ten specimens were tested under fatigue loads. Testing results demonstrated that no obvious damage was observed from the strengthened beam after 2 million fatigue cycles compared with the reference beam which failed at 58.97 × 10 4 fatigue cycles, indicating the good joint performance of the U-shaped steel casing and the RC beam. The strengthened beams generally failed by the sudden rupture of the bottom steel plate accompanied by concrete crushing. The mid-span defl ection and material strains all increased rapidly at the beginning, then steadily developed during nearly 80% of the fatigue life, and grew rapidly again after the weld toe cracking until the strengthened beam failed. The fatigue life was signifi cantly extended with the increase of either the height of the steel box or the thickness of the bottom plate, while it was dramatically shortened with the increase in the fatigue load amplitude. To facilitate the use of the outcomes, a simplifi ed formula for predicting the concrete residual strain of strengthened beams was derived. The predicted results matched the experimental ones well. The derived formula is theoretically only applicable to strengthened beams with dimensions same as the specimens in the experimental tests under similar loading conditions. Reasonably good agreements were found between the fi nite element analyses and experimental results. The developed fi nite element model can well simulate the failure mode under static load and the damage evolution under fatigue load. Based on the stress analysis results, the fatigue lives of the strengthened beams were predicted relatively accurately using the Fe-safe software, with the deviations mostly less than 15%.

      • Unified SVPWM Strategy for Post-fault Three-level NPC Voltage Source Inverters

        Eight switch three-phase inverters (ESTPIs) are the reconfigured topologies of the three-level neutralpoint-clamped (NPC) inverter with a failure leg. This failure leads to different topologies of ESTPIs, which can be controlled by different space vector pulse width modulation (SVPWM) strategies. By analyzing the relationship of three SVPWM strategies for different ESTPIs, a unified coordinate transformation is proposed, which can make one of the three SVPWM strategies be a unified control strategy of ESTPIs. And the unified coordinate transformation is suited for two different SVPWM strategies for ESTPIs. Consequently, the unified strategy not only simplifies the control strategy, but also makes the fault tolerant control strategy more flexible. Simulation and hardware-in-the-loop (HIL) test results have shown the validity and feasibility of the proposed unified SVPWM strategies.

        Compressive performances of concrete filled Square CFRP-Steel Tubes (S-CFRP-CFST)

        Sixteen concrete filled square CFRP-steel tubular (S-CFRP-CFST) stub columns under axial compression were experimentally investigated. The experimental results showed that the failure mode of the specimens is strength loss of the materials, and the confined concrete has good plasticity due to confinement of the CFRP-steel composite tube. The steel tube and CFRP can work concurrently. The load versus longitudinal strain curves of the specimens can be divided into 3 stages, i.e., elastic stage, elasto-plastic stage and softening stage. Analysis based on finite element method showed that the longitudinal stress of the steel tube keeps almost constant along axial direction, and the transverse stress at the corner of the concrete is the maximum. The confinement effect of the outer tube to the concrete is mainly focused on the corner. The confinements along the side of the cross-section and the height of the specimen are both non-uniform. The adhesive strength has little effect both on the load versus longitudinal strain curves and on the confinement force versus longitudinal strain curves. With the increasing of the initial stress in the steel tube, the load carrying capacity, the stiffness and the peak value of the average confinement force are all reduced. Equation for calculating the load carrying capacity of the composite stub columns is presented, and the estimated results agree well with the experimental results.

        Interleukin-17 Indirectly Promotes M2 Macrophage Differentiation through Stimulation of COX-2/PGE2 Pathway in the Cancer Cells

        PurposeInterleukin-17 (IL-17) is a proinflammatory cytokine that plays important roles ininflammation, autoimmunity, and cancer. The purpose of this study was to determineif IL-17 indirectly regulates macrophage differentiation through up-regulation ofcyclooxygenase-2 (COX-2) expression in the cancer cell lines. Materials and MethodsHuman cervical cancer HeLa, human lung cancer A549, and mouse prostate cancerMyc-CaP/CR cell lines were treated with recombinant IL-17; Western blot analysis,enzyme-linked immunosorbent assay, and quantitative real-time polymerase chainreaction analysis were utilized to examine the cellular responses. ResultsIL-17 up-regulated expression of COX-2 mRNA and protein in HeLa, A549, and Myc-CaP/CR cell lines. IL-17’s effects were mediated through nuclear factor-κB andERK1/2 signaling pathways as the inhibitors of these pathways could inhibit IL-17-induced COX-2 expression. The conditional medium obtained from the cancer cellscontained prostaglandin E2, the levels of which were increased by IL-17 treatment. When treated with the conditional medium, particularly with the IL-17-induced conditionalmedium, mouse RAW264.7 macrophages and human THP-1 monocytes expressedhigher levels of IL-10 (a marker of M2 macrophages) than inducible nitricoxide synthase or tumor necrosis factor α (markers of M1 macrophages). In contrast,when RAW264.7 and THP-1 cells were treated directly with IL-17, expression of thesemarker genes was not markedly changed. ConclusionThe results of this study suggest that IL-17 indirectly promotes M2 macrophagedifferentiation through stimulation of the COX-2/PGE2 pathway in the cancer cells,thus IL-17 plays an indirect role in regulating the tumor immune microenvironment.

        Upregulating miR-637 aggravates endoplasmic reticulum stress-induced apoptosis in gastric cancer cells by suppressing Calreticulin

        Gastric cancer is a leading cause of cancer death worldwide. Endoplasmic reticulum (ER) stressinduced apoptosis has been confirmed to be important in the treatment of gastric cancer. MiR- 637 has recently been found to exert inhibitory effects on gastric cancer, and this study aimed to investigate whether miR-637 could regulate apoptosis through ER stress. The results showed that tunicamycin (TM) induced downregulation of miR-637 in gastric cancer cells (AGS) and increase of apoptosis and ER stress. Overexpression of miR-637 promoted TM-induced apoptosis and expression of ER stress associated proteins (GRP78 and CHOP), but inhibited expression of Calreticulin. MiR-637 could bind with the 3ʹ-UTR of CALR, and negatively regulated the expression of CALR. The co-transfection of miR-637 and CALR in AGS cells show that, CALR overexpression could reverse the pro-apoptosis effects of miR-637 in TM-treated cells. In conclusion, the present study suggests that miR-637 participates in ER stress-induced apoptosis in gastric cancer cells by suppressing CALR expression. miR-637 or CALR may be a future potential target for gastric cancer treatment.

      • ENVIRONMENTAL CONDITIONS OF DUST EVENTS IN NORTHERN CHINA DURING 2000-2001

        To investigate the large-scale environmental conditions of dost storms occurring in Northern China, a synoptic situation of ten dust events in Northern China during 2000-2001 was composed and its dynamic features were identified. The composite synoptic situation showed that the occurrence of the ten dust events was associated with the eastward movement of a strong cold high from Mongolia, and the development of a cyclone in Northeast China, which caused the pressure gradient to intensify considerably in Northern China. The upper level circulation revealed that a 500 hPa trough developed rapidly above Northern China, and diagnosis showed that composite ascending motion was pronounced above the Siberian district. The change of vertical motion in the middle and low troposphere above Mongolia was also related to the occurrence of the ten dust events. When the descending current, 12 hr before the occurrence, was completely replaced by the ascending current, the instable atmospheric stratification of the lower troposphere enabled dry dust to be transported from the surface to high altitudes toward Northern China.

        Analysis of Bearing Capacity of Circular Concrete Filled CFRP-Steel Tubular Beam-Column

        In practice, it is common for circular concrete-filled CFRP-steel tube (C-CF-CFRP-ST) member severing under eccentric load, but there are few related investigations on its mechanical behavior currently. In this study, experimental study and finite element analysis are employed to reveal the static behavior of circular concrete filled CFRP-steel tube under eccentric compression. Total 32 C-CF-CFRP-ST specimens, taking the slenderness ratio (λ) and eccentric ratio (e) as principal parameters, were designed and tested firstly, obtaining the failure modes, N-um curve and the strains in the steel tube and CFRP. It was found that the damage of concrete became more serious with the decrease of the slenderness ratio but got mitigated with the increase of eccentricity. Additionally, it was indicated that the N-um curve of C-CF-CFRP-ST consisted of three stages, namely elastic stage, elastic-plastic stage and plastic stage. In the elastic-plastic stage, the damage is gradually accumulated in CFRP and concrete, and the specimens finally exhibited ductile failure in the end of plastic stage. In the entire loading process, CFRP has almost same strains compared to the steel tube. Following the experimental study, finite element analysis was conducted to investigate the static behavior of C-CF-CFRP-ST comprehensively. The proposed finite element model reproduced the compressive behavior of C-CF-CFRP-ST, in terms of failure modes and N-um curves. The influence of material strength, layers of CFRP and steel ratio on the static behavior of C-CF-CFRP-ST specimens were then studied through numerical parametric study. In the end, equations for predicting the bearing capacity of C-CF-CFRP-ST are proposed, and the predicted static strengths of the specimens are basically consistent with the experimental results.
