        Mac-2 결합단백질 발현에서의 STAT3의 역할

        배경 : Mac-2 결합단백질은 인간의 몇몇 암세포들, 특히 유방암, 폐암, 위암세포의 세포액으로 분비되는 당단백질로 면역반응과 다양한 암환자 및 전염성 질환환자에서의 세포부착 활동성의 기능을 담당하고 있다. 본 연구에서는 전사단계에서의 Mac-2 결합단백질의 조절자를 동정하고자 하였다. 방법 : 위암 세포주에서 Mac-2 결합단백질 발현에 대한 EGF의 효과를 결정하기 위하여, 서로 다른 길이의 Mac-2 결합단백질 프로모터 플라스미드를 제작하고 프로모터 활성과 Mac-2 결합단백질 발현을 측정하였다. 더욱이 STAT3와 hTERT가 Mac-2 결합단백질의 음성 혹은 양성 조절자인지 그 기능을 규명하기 위하여 STAT3와 hTERT에 특이적인 siRNA를 형질전환시킨 후, Mac-2 결합단백질 수준은 정량 ELISA로 측정하였다. 결과 : HEK293 또는 위암세포주인 SNU-638, AGS에 EGF를 처리하면 Mac-2 결합단백질전사를 억제할 수 있었다. 5′-절편 프로모터 실험에서 pGL3-Mac Pro-2,377을 형질전환한 세포가 pGL3-Mac Pro-2,277을 형질전환한 세포에 비해 감소된 루시퍼레이즈 활성을 나타냈다. 또한 Mac-2 결합단백질 프로모터부위 중 -2,366/-2,356이 잠정적인 STAT3 결합부위임을 동정하였고, 양성 조절자로 알려진 siRNA hTERT에 비해 siRNA STAT3가 형질전환된 세포는 Mac-2 결합단백질이 감소되었음을 증명하였다. 결론 : EGF 신호가 Mac-2 결합단백질 발현에 중요하며, hTERT는 Mac-2 결합단백질의 양성 조절자인 반면 STAT3는 음성 조절자로 작용하였다.

      • Function of Nitric Oxide in Activation-Induced Cell Death of T Lymphocytes

        Using a murine T cell hybridoma, activation-induced cell death (AICD) was studied. As an in vitro model system for the AICD, 1 cell hybridoma expressing TCR/CD3 complex was incubated onto the immobilized purified anti-CD3 antibody. The immobilized anti-CD3 antibody induced AICD effectively up to 40%. At 1-100 $\mu$M range of SNP, an exogenous source of nitric oxide (NO), the cell proliferation was not affected, but at 1 mM SNP, cell proliferation was significantly reduced. The AICD of T cell hybridoma was inhibited by exogenous NO at non-cytotoxic concentration, In the cells undergoing AICD, the expressions of caspase-3 and FasL were detected, but not iNOS. Similar result was recognized in the apoptosis induced by dexamethasone, an apoptosis-inducing agent. However, the conversion from the inactive form of caspase-3 (32 kDa) to the active form (17 kDa) was significantly reduced in the cells in AICD induced by anti-CD3 antibody, With the result of increased PARP cleavage in the cells, we propose that another PARP cleavage pathway not involving caspase-3 may function in the anti-CD3 antibody induced AICD in the T cell hybridoma.

        Up‐regulation of Mac‐2 binding protein by hTERT in gastric cancer

        <P><B>Abstract</B></P><P>Mac‐2 binding protein (Mac‐2BP) is a secreted tumor antigen that is elevated in many cancers and implicated in tumor metastasis, as well as cell adhesion and immune functions. We focused on the human telomerase reverse transcriptase (hTERT) induced Mac‐2BP expression and the relationship between Mac‐2BP expression and the progression of gastric cancer. A cDNA expression array analysis was performed on the telomerase‐negative cell line, SW13, which was engineered to overexpress hTERT when compared with the parental SW13 cell. hTERT‐induced Mac‐2BP expression was confirmed <I>via</I> RT‐PCR and Northern blotting. ELISA and flow cytometric analyses revealed that Mac‐2BP protein was increased by 2‐ to 4‐fold in hTERT‐overexpressing cells compared with the mock control. Mac‐2BP expression was significantly reduced when the overexpressed hTERT was neutralized by the introduction of hTERT‐specific siRNA. These results suggest that Mac‐2BP expression is modulated by hTERT. Mac‐2BP levels in both gastric cancer cells and tumor tissues were determined <I>via</I> Northern blot analysis and immunohistochemistry. Mac‐2BP protein was highly expressed in most gastric cancer cell lines, and gastric tumor tissues were stained more densely than normal tissues. The intracellular and secreted Mac‐2BP levels were also evaluated <I>via</I> ELISA, indicating that Mac‐2BP was expressed and secreted more abundantly in gastric cancer patients than in healthy donors. The elevated serum Mac‐2BP level in gastric tumor patients was also significantly associated with distant metastasis (<I>p</I> = 0.05) and higher tumor stage (<I>p</I> = 0.04). Our findings suggest that Mac‐2BP is induced by hTERT, and that it may prove to be a useful prognostic marker for the detection of malignant progression of metastatic stomach cancers. © 2006 Wiley‐Liss, Inc.</P>

        Frey syndrome is characterized by sweating and flushing on the auriculotemporal region in respense to gustatory stimuli following surgery, trauma or irfection of the parotid areas. A 22-year-old man had received a surgery for sclerosis and hypertrophy of the left mandible. Two months after operation gustatory sweating occurred, whenever he eats, on the malar area without any detectable flushing or sensory impairment. Sweating starts in 30 seconds during food intake and is clearly visible with naked eyes within 2 minutes. This syndrome is encountered uncommonly to the dermatologic practitioners but more cases might be discovered if more attention could be given to this condition.

        Immortalization of Rabbit Corneal Fibroblasts by Overexpression of Simian Virus 40 Large T antigen

        Immortalization of primary corneal cells has influence on pharmacy, medical and biological fields. Especially, investigation of immortalization mechanism using viral oncoproteins is useful for medical treatments, and these cell lines will be useful materials for toxic test of medical supplies and cell biological experiments. Rabbit corneal fibroblasts in culture undergo a finite number of divisions before they reach a terminally non-proliferating state known as replicative senescence. Therefore, we attempted to induce immortalization of rabbit corneal fibroblasts with SV40 large T antigen. As a result of experiment, expression of SV40 large T antigen was confirmed, and expression of proteins related to cell cycle repressor was decreased in the transfection group compared with non-transfection group. According to the results of cell cycle phase distribution test, SV40 large T antigen-transfected cells had obtained higher proliferation rate than primary cells. It was confirmed that during induction of immortalization, SV40 large T antigen was not able to increase telomerase activity. In conclusion, we made a rabbit corneal fibroblast cell line with SV40 large T antigen. This cell line will be useful for further studies of mammalian fibroblast biology, particularly with regard to angiogenesis and malignant transformation. In addition, this cell line offers opportunity for testing potential therapeutics and can be used for toxicity tests of materials or cosmetics. In the future, our cell line can potentially be utilized in a wide range of biology related fields.

        Unilateral nevoid telangiectasia is a rare but clinica.lly distinct dermatosis first deacribed by Zeisler in 1922. It had been also called as linear telangiectasia or unilateral telangiectasia. Wilkin suggested the term unilateral dermatomal superficial telangiectasia, in 1977. There are some reports of congenital unilateral nevoid telangiectasia but it usually occurs in women during the puberty and the third decade. Clinically it is characterized by superficial telangiectatic lesions distributed along the dermatomes uniIaterally and usually on the upper part of the body. It was reported that unilateral nevoid telangiectasia was related with elevated serum estrogen level in the many cases. We present two patients, 23-year and 25-year-old male soldiers, whose leaions were consistent with unilateral nevoid telangiectasia elinically and histopathologically.

        We previously reported the result of the VDRL and TPHA tests among the general entertainers and prostitutes in the Youngdeungpo Ku, Chung Ku, Seoul and Paju area, Kyunggi province in 1979 The study resulted in an overall, VDRL reactive rate of 5 .6% in the 6, 666 subjects including general entertainers and prostitutes: 5 3% in the 5, 289 entertainers and 7.6% in the 837 prostitutes. 34.8% of VDRL reactive entertainers showed VDRL titers of 1: 8 or higher. In order to investigate the changing patterns of VDRL reactive rates in general entertainers and prostitutes, we compared the results of the present VDRL tests (1982) with those of our previous study (1979) Total 6, 808 subjects consisted of 2,931 residents who wanted to have medical certification from Jan. 4, 1982 to June 30, 1982 and 2, 562 general entertainers in the Youngdeungpo Ku and Chung Ku areas and 120 general entertainers and 1,195 prostitutes in the Paju area frorn Jan, 4, 1982 to Dec. 31, 1981.
