NGAL (neutrophil gelatinase-associated lipocalin) is a novel cancer-related protein involves multiple functions in many cancers and other diseases. We previously overexpressed NGAL to analyze its role in esophageal squamous cell carcinoma (ESCC). In this study, a protein-protein interaction (PPI) was constructed and the shortest paths from NGAL to transcription factors in the network were analyzed. We found 28 shortest paths from NGAL to RELA, most of them obeying the principle of extracellular to cytoplasm, then nucleus. These shortest paths were also prioritized according to their normalized intensity from the microarray by the order of interaction cascades. A systems approach was developed in this study by linking differentially expressed genes with publicly available PPI data, Gene Ontology and subcellular localizaton for the integrated analyses. These shortest paths from NGAL to DEG transcription factors or other transcription factors in the PPI network provide important clues for future experimental identification of new pathways.
Structure Analysis of 16S rDNA Sequences from Strains of Acidithiobacillus ferrooxidans

Four strains of Acidithiobacillus ferrooxidans with different iron oxidation capacity were isolated from different mine drainage stations. The 16S rRNA gene of these strains were cloned and sequenced. Based on our sequences analysis on the four strain and the data on the other strains deposited in Genbank, all A. ferrooxidans may be classified into three phylogenetic groups. The analysis data showed that nucleotide variables (signature sites) were detected in 21 positions, and most of them were found in the first 800bp from 5' terminal except position 970 and 1375. Interestingly, the first 13 signature sites were located in two main regions:the first region (position 175-234) located in V2 while the second region (position 390-439) were detected in constant region between V2 and V3. Furthermore, the secondary structure and minimal free energy were determined in two regions among strains of three groups. These results may be useful in characterizing the microevolutionary mechanisms of species formation and monitoring in biohydrometallurgical application.

BACKGROUND: Reliable in vitro cellular models are needed to study the phenotypic modulation of smooth muscle cells (SMCs) in health and disease. The aim of this study was to optimize gelatin methacrylate (GelMA)/alginate hydrogels for bioprinting three-dimensional (3D) SMC constructs. METHODS: Four different hydrogel groups were prepared by mixing different concentrations (% w/v) of GelMA and alginate: G1 (5/1.5), G2 (5/3), G3 (7.5/1.5), and G4 (7.5/3). GelMA 10% was used as control (G5). A circular structure containing human bladder SMCs was fabricated by using an extrusion-based bioprinter. The effects of the mixing ratios on printability, viability, proliferation, and differentiation of the cells were investigated. RESULTS: Rheological analysis showed that the addition of alginate significantly stabilized the change in mechanical properties with temperature variations. The group with the highest GelMA and alginate concentrations (G4) exhibited the highest viscosity, resulting in better stability of the 3D construct after crosslinking. Compared to other hydrogel compositions, cells in G4 maintained high viability ([80%), exhibited spindle-shaped morphology, and showed a significantly higher proliferation rate within an 8-day period. More importantly, G4 provided an optimal environment for the induction of a SMC contractile phenotype, as evidenced by significant changes in the expression of marker proteins and morphological parameters. CONCLUSION: Adjusting the composition of GelMA/alginate hydrogels is an effective means of controlling the SMC phenotype. These hydrogels support bioprinting of 3D models to study phenotypic smooth muscle adaptation, with the prospect of using the constructs in the study of therapies for the treatment of urethral strictures.

The primary task of machine fault diagnosis is to judge whether the current state is normal or damaged, so it is a typical binary classification problem with mutual exclusion. Mutually exclusive events and out-of-domain detection have one thing in common: there are two types of data and no intersection. We proposed a fusion model method to improve the accuracy of machine fault diagnosis, which is based on the mutual exclusivity of events and the commonality of out-of-distribution detection, and finally generalized to all binary classification problems. It is reported that the performance of a convolutional neural network (CNN) will decrease as the recognition type increases, so the variational auto-encoder (VAE) is used as the primary model. Two VAE models are used to train the machine's normal and fault sound data. Two reconstruction probabilities will be obtained during the test. The smaller value is transformed into a correction value of another value according to the mutually exclusive characteristics. Finally, the classification result is obtained according to the fusion algorithm. Filtering normal data features from fault data features is proposed, which shields the interference and makes the fault features more prominent. We confirm that good performance improvements have been achieved in the machine fault detection data set, and the results are better than most mainstream models.

To enhance the friction and wear resistance of GCr15/PTFE sliding contact surfaces, the synergistic lubrication effect of micro-textures and solid lubricants were studied in this paper. Micro-textures were processed on GCr15 samples by Nd:YAG laser, which were filled with MoS 2 . Tribological performance tests were carried out on a Rtec-MFT 5000 tester. Tribological properties were optimal with the textures density of 20 % and depth of 9 μm. A continuous composite solid lubricating film was formed by PTFE and MoS 2 . As load increases, friction coefficient of the sample shows a rapid decline and then stabilizes. With the increase of reciprocating frequency, friction curve of the sample shows a trend of slight fluctuation. When frequency is 2.7 Hz, friction coefficient is the lowest. Tribological properties of PTFE/GCr15 contact surfaces could be significantly optimized by micro-textures and MoS 2 , which performs favorable prospect of engineering application such as guide rails, bearings, machinery seal, etc.
A Joint Transform Correlator Encryption System Based on Binary Encoding for Grayscale Images

A binary encoding method for grayscale images is proposed to address their unsatisfactory decryption results from joint transform correlator (JTC) encryption systems. The method converts the encryption and decryption of grayscale images into that of binary images, and effectively improves decrypted-image quality. In the simulation, we replaced unencoded grayscale images with their binary encoded counterparts in the JTC encryption and decryption processes, then adopted a median filter to suppress saturation noise while keeping other settings unchanged. Accordingly, decrypted-image quality was clearly enhanced as the correlation coefficient (CC) between a decrypted image and its original rose from 0.8237 to 0.9473 initially, and then further to 0.9937, following the above two steps respectively. Finally, optical experimental results confirmed that the proposed encryption system works correctly.

The charged poly(vinylidene fluoride) (PVDF) membranes were prepared by composite thermallyinduced phase separation (C-TIPS) method with perfluorosulfonic acid (PFSA) as additive. The resultsverify that the porosity, surface hydrophilicity and pure water permeation flux (JW) increase with theaddition of PFSA for the membranes. The highest JW of 507.2 1.2 L/(M2 H bar) is obtained for themembrane cast at 90 8C. Much higher casting temperature isn’t advantageous to improve JW value for thePVDF membranes. All membranes exhibit negative apparent zeta potential (zapp). The crystallinity in b phaseincreases as casting temperature increases for all PVDF membranes.

A study of 426 rabbits from 3 cities in Jilin province (Changchun City and Jilin City) and Liaoning province (Shenyang City) was conducted between May and June 2015. The overall prevalence of E. bieneusi in rabbits was 0.94% (4/426), with 0% (0/116), 1.72% (3/174), and 0.74% (1/136) in Jilin, Changchun, and Shenyang City, respectively. Only 3 farms (farm 1 and farm 3 in Changchun City, farm 8 in Shenyang City) were PCR-positive for E. bieneusi. Moreover, rabbits of more than 6 months (1.72%) had the highest E. bieneusi prevalence, followed by rabbits of 4-6 months (1.26%), 2-3 months (0.58%), and less than 1 month (0%). Analysis of ITS gene of E. bieneusi suggested that all 4 E. bieneusi isolates were genotype D, and were classified as group 1a. The present results first demonstrated the existence of zoonotic E. bieneusi in domestic rabbits in China. Effective control measures should be implemented to prevent E. bieneusi infection in domestic rabbits, other animals, and humans.

Sheath blight disease (ShB) severely afects rice production; however, the details of defense against ShB remain unclear. To understand the rice defense mechanism against ShB, an RNA sequencing analysis was performed using Rhizoctonia solani inoculated rice leaves after 48 h of inoculation. Among them, 3417 genes were upregulated and 2532 were downregulated when compared with the control group (>twofold or<1/2). In addition, the diferentially expressed genes were classifed via Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), and MapMan analyses. Fifty-nine GO terms and seven KEGG pathways were signifcantly enriched. A MapMan analysis demonstrated that the phytohormone and metabolic pathways were signifcantly altered. Interestingly, the expression levels of 359 transcription factors, including WRKY, MYB, and NAC family members, as well as 239 transporter genes, including ABC, MFS, and SWEET, were signifcantly changed in response to R. solani AG1-IA inoculation. Additionally, OsWRKY53 and OsAKT1 negatively regulate the defense response in rice against R. solani via gain of function study for OsWRKY53 and loss of function study for OsAKT1, respectively. Furthermore, several diferentially expressed genes contain R. solani-responsive cis acting regulatory elements in their promoter regions. Taken together, our analyses provide valuable information for the additional study of the defense mechanisms against ShB, and the candidate genes identifed in this study will be useful resource for future breeding to enhance resistance against ShB.
