        Background: Superior colliculus is a part of midbrain, and participates in the visual reflexes, It receives afferent fibers from optic nerve, visual cortex, and spinotectal tract. After optic deprivation, the microscopic structure of the superior colliculus changed. Calcium-binding proteins (CBPs) Play an important role in the neuronal protection, differentiation and reorganization of the central nervous system, Objectives and Methods: The effects of neonatal retinal deafferentation on a CBPs, calbindm D-28k (CB), Parvalbumin (PB) and calretimn (CR), and the existence of colocalization between the CBPs were examined immunohistochemically in the rat superior colliculus. Results: On the experimental (contralateral to enucleation) side of superior colliculus, the number of CB-immunoreactive (IR) cells was reduced (77.4% compared to control), but not fibers. The number of PB-IR neurons and fibers was also reduced on the experimental side (88.5% compared to control), In the other hand, the CR-IR cells were dramatically increased (642% compared to control), but CR-IR fibers were markedly decreased on the experimental side. The colocalization between CB-CR and PV-CR was rarely observed in the superior colliculus Conclusion: These results suggest that the changes of retinotectal projection may alter the expressional pattern of CBPs in different manners; relatively stable in CB- and PV-IR neurons and plastic in CR-IR neurons.

        The response of eukaryotic cells to double-strand breaks in genomic DNA includes the sequestration of many factors into nuclear foci. Recently it has been reported that a member of the histone H2A family, H2AX, becomes extensively phosphorylated within 10-30 minutes of DNA damage and forms foci at break sites. Histone H2AX has a role in suppressing genomic instability and cancer. Phosphorylated H2AX (gamma-HZAX) is essential to the efficient recognition and (or) repair of DNA double strand breaks (DSBs), and many molecules, often thousands, of H2AX become rapidly phosphorylated at the site of each nascent DSB. gamma-H2AX foci formation is a sensitive biological dosimeter and presents new and exciting opportunities to understand important biological processes, human diseases, and individual variations in radiation sensitivity. These potentialities demonstrate the importance of understanding the parameters and functions of gamma-H2AX formation.

        Chest wall perforator flaps for partial breast reconstruction: Surgical outcomes from a multicenter study

        Background Perforator artery flaps based on the branches of intercostal arteries and lateral thoracic artery can be used for reconstruction after breast-conserving surgery (BCS). Although described more than a decade ago, these have not been adopted widely in clinical practice. We report on short-term and long-term surgical outcomes of partial breast reconstruction using chest wall perforator flaps from a prospective multicenter audit. Methods All patients operated for BCS and partial breast reconstruction using intercostal artery perforator or lateral thoracic artery perforator flaps from January 2015 to October 2018 were included in the analysis. Oncoplastic breast surgeons with appropriate level of training performed all tumor excisions and reconstructions as a single-stage procedure. Patient characteristics, treatment details and surgical outcomes were noted. Specific outcomes recorded were margin re-excision and complication rates. Results One hundred and twelve patients underwent the procedure in the given study period. The median age was 54 years. Median specimen weight was 62.5 g and median volume of excision was 121.4 mL. Fifteen patients (13.39%) underwent a margin re-excision for close or positive margins without additional morbidity. One patient required a completion mastectomy. Eight patients (7.14%) had an early complication. None of the patients required a contralateral symmetrization procedure. The results were comparable across the participating centers. Conclusions Chest wall artery perforator-based flaps are an excellent option for lateral and inferior quadrant partial breast reconstructions. The short and long-term surgical outcomes are comparable across sites and can be performed with minimal morbidity. Patient-reported outcome measures need to be studied.

        Identifification of microRNAs from transcriptome data in gurmar (Gymnema sylvestre)

        MicroRNAs (miRNAs) are single-stranded, non-coding, small (~ 22 nt) RNAs that regulate mRNA targets in plants andanimals. Gymnema sylvestre (Retz.) is an important medicinal plant that lacks genomic as well as transcriptomic information. Here, to identify homologous miRNAs, we screened 6028 unique known plant miRNAs against 272,161 unigenes of G. sylvestre (Retz.) generated by paired-end deep transcriptome sequencing. We utilized 76 aligned unigenes for extracting theprecursor sequences in G. sylvestre (Retz.) and identifi ed 16 potential candidate miRNAs belonging to 12 miRNA families. The mean minimal folding free energy index (MFEI) value of these 16 predicted miRNAs was − 0.966, ranging from − 0.653to − 2.37, while the G + C% content varied between 27.3 and 61.9%. Nucleotide composition analysis revealed that cytosinewas the dominant nucleotide in mature miRNAs (26.3%), while uracil was the next most prevalent nucleotide (25.7%), followedby guanine (24.3%) and adenine (23.7%). In most cases (81%), the fi rst position of the 5′ end was occupied by uracilin the identifi ed mature G. sylvestre (Retz.) miRNAs. Diff erential expression of target unigenes revealed that, as comparedto leaf tissue, a total 16 and 12 target unigenes were diff erentially expressed in fl ower and fruit tissues, respectively. Amongthe diff erentially expressed target unigenes were Auxin Response Factors 17-like, Copia-type polyprotein Phytoene synthase2, as well as several encoding unnamed, predicted, uncharacterized, and/or hypothetical proteins. After detailed predictionof miRNAs and their corresponding target unigenes, we report for the fi rst time a total of 13 miRNA and 213 correspondingtarget unigenes in G. sylvestre (Retz.).

        Acute Pancreatitis: A Rare Post-Colonoscopy Sequela

        Abdominal pain is a common but benign symptom after colonoscopy. We report a case of acute pancreatitis that occurred just after anelective screening colonoscopy; this is a rare event with very few reported cases. A healthy, asymptomatic male underwent screeningcolonoscopy at our center and developed abdominal pain and emesis after the procedure. An abdominal X-ray ruled out perforation but laboratory tests revealed elevated levels of amylase and lipase. The patient had no etiological risk factors for pancreatitis. Thepresumed mechanism of pancreatitis in this case is mechanical and pressure trauma from excessive insuffation, external abdominal pressure, and repeated withdrawal of the colonoscope due to tight angulation of the splenic flexure, a structure that is in close proximityto the pancreatic tail. Acute pancreatitis should be considered in the differential diagnosis of patients who present with abdominal painafter colonoscopy once more common etiologies have been excluded.

        Normal Values of High-resolution Anorectal Manometry of Healthy Indians

        Background/AimsHigh-resolution anorectal manometry (HRAM) measures anal sphincter function and anorectal co-ordination. This study aims to provide normal data for HRAM and evaluate the effect of gender, age, and body mass index (BMI) on anorectal functions in healthy Indian subjects. MethodsHRAM was performed on 93 healthy volunteers using a 20-channel, water-perfused catheter. We evaluated anorectal pressures, rectal sensation, and balloon expulsion time. Measurements were recorded during rest, squeeze, and simulated defecation (push). ResultsMedian anal resting pressure (88 mmHg vs 94 mmHg, P = NS), anal squeeze pressure (165 mmHg vs 147 mmHg, P = NS) were not significantly different between males and females. Rectal pressure (70 mmHg vs 54 mmHg, P = 0.024) and anal pressure (82 mmHg vs 63 mmHg, P = 0.008) during simulated evacuation without rectal distention, were higher in males. The threshold for the first sensation was lower in females (40 mL vs 30 mL, P = 0.021) but desire to defecate (105 mL vs 90 mL, P = NS) and maximum tolerable volume (160 mL vs 140 mL, P = NS) were not significantly different in males and females. Anal residual pressure (median mmHg 83 vs 71 mmHg, P = 0.025) was higher in subjects < 40 years of age. Maximum anal squeeze pressure (185 mmHg vs 165 mmHg, P = 0.024) and maximum rectal pressure (75 mmHg vs 62 mmHg, P = 0.032) during push higher in BMI < 23 kg/m2. ConclusionsThe present study provides normal data for the Indian population that can be used for comparison and further work. Age, gender, and BMI affect anorectal parameters in HRAM and should be considered while reporting.
