        Overexpressed CYP450 mediated apoptosis evaluates cytotoxicity and teratotoxicity of Cucumis callosus

        Fruits of Cucumis callosus (Rottl.) Cogn. (Family: Cucurbitaceae) plant, are commonly known as “bitter cucumber” (English) and “Kachri” (Hindi) in India and have been traditionally used for antioxidant, rich source of vitamin C, antidiabetic and anticancer actions. Tribal peoples of Odissa and West-Bengal are using these fruits during worship and as a vegetable. In vitro cytotoxicity of methanolic seed extract of C. callosus (MSCC) at different concentrations (25, 50, 100, 200, 400) μg/ ml and methanolic pericarp extract of C. callosus (MPCC) at (30, 60, 120, 240, 360) μg/ml on Ehrlich Ascites Carcinoma (EAC) cell line, has been evaluated by using the MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay and Trypan blue dye exclusion assay for a period of 3 h treatment. Acute toxicity and in vivo teratotoxicity of both extracts were also evaluated using the Zebrafish embryo developmental assay. The MSCC and MPCC showed the direct cytotoxic effect on EAC cells in a dose-dependent manner with IG50 value 273.17 and 235.08 μg/ml respectively. The MPCC in 240 and 360 μg/ml showed a strong teratotoxic effect on Zebrafish embryo in a dose-dependent manner. The cytotoxicity and teratotoxicity of MPCC were due to overexpressed CYP450 mediated apoptosis in μthe tissue.

      • Assessment of diffusion tensor image quality across sites and vendors using the American College of Radiology head phantom

        <P>The purpose of this study was to explore the feasibility of assessing quality of diffusion tensor imaging (DTI) from multiple sites and vendors using American College of Radiology (ACR) phantom. Participating sites (Siemens (n=2), GE (n=2), and Philips (n=4)) reached consensus on parameters for DTI and used the widely available ACR phantom. Tensor data were processed at one site. <SUB>B0</SUB> and eddy current distortions were assessed using grid line displacement on phantom Slice 5; signal‐to‐noise ratio (SNR) was measured at the center and periphery of the b=0 image; fractional anisotropy (FA) and mean diffusivity (MD) were assessed using phantom Slice 7. Variations of acquisition parameters and deviations from specified sequence parameters were recorded. Nonlinear grid line distortion was higher with linear shimming and could be corrected using the 2nd order shimming. Following image registration, eddy current distortion was consistently smaller than acquisition voxel size. SNR was consistently higher in the image periphery than center by a factor of 1.3–2.0. ROI‐based FA ranged from 0.007 to 0.024. ROI‐based MD ranged from 1.90×<SUP>10−3</SUP> to 2.33×<SUP>10−3</SUP><SUP>mm2</SUP>/s(median=2.04×<SUP>10−3</SUP><SUP>mm2</SUP>/s). Two sites had image void artifacts. The ACR phantom can be used to compare key quality measures of diffusion images acquired from multiple vendors at multiple sites.</P><P>PACS number(s): 87.57.‐s, 87.19.lf</P>

        RICHARDSON EXTRAPOLATION OF ITERATED DISCRETE COLLOCATION METHOD FOR EIGENVALUE PROBLEM OF A TWO DIMENSIONAL COMPACT INTEGRAL OPERATOR

        In this paper, we consider approximation of eigenelements of a two dimensional compact integral operator with a smooth kernel by discrete collocation and iterated discrete collocation methods. By choosing numerical quadrature appropriately, we obtain convergence rates for gap between the spectral subspaces, and also we obtain superconvergence rates for eigenvalues and iterated eigenvectors. We then apply Richardson extrapolation to obtain further improved error bounds for the eigenvalues. Numerical examples are presented to illustrate theoretical estimates.

        Inner harbour wave agitation using boussinesq wave model

        Short crested waves play an important role for planning and design of harbours. In this context a numerical simulation is carried out to evaluate wave tranquility inside a real harbour located in east coast of India. The annual offshore wave climate proximity to harbour site is established using Wave Model (WAM) hindcast wave data. The deep water waves are transformed to harbour front using a Near Shore spectral Wave model (NSW). A directional analysis is carried out to determine the probable incident wave directions towards the harbour. Most critical threshold wave height and wave period is chosen for normal operating conditions using exceedence probability analysis. Irregular random waves from various directions are generated confirming to Pierson Moskowitz spectrum at 20m water depth. Wave incident into inner harbor through harbor entrance is performed using Boussinesq Wave model (BW). Wave disturbance experienced inside the harbour and at various berths are analysed. The paper discusses the progresses took place in short wave modeling and it demonstrates application of wave climate for the evaluation of harbor tranquility using various types of wave models.

        SOME SUBORDINATION PROPERTIES OF THE LINEAR OPERATOR

        In this paper, subordination results of analytic function $f{\in}{\mathcal{A}}_p$ involving linear operator ${\mathcal{K}}^{{\delta},{\lambda}}_{c,p}$ are obtained. By applying the differential subordination method, results are derived under some sufficient subordination conditions. On using some hypergeometric identities, corollaries of the main results are derived. Furthermore, convolution preserving properties for a class of multivalent analytic function associated with the operator ${\mathcal{K}}^{{\delta},{\lambda}}_{c,p}$ are investigated.

        Migrating foreign body in an adult bronchus: An aspirated denture

        As a safety measure, dentures are routinely removed before surgery. Aspiration of a denture could be catastrophic, with medicolegal implications. Foreign body aspiration is uncommon in adults; however, aspirations may remain asymptomatic and undiagnosed for long periods of time. We report an adult male who presented with a cough for more than 6 months. On radiography, a foreign body was found migrating within the tracheobronchial tree from one mainstem bronchus to the other, at different time points. The foreign body was later found to be a portion of his denture. The aspiration may have occurred at the time of a surgical procedure.

        COEFFICIENT BOUNDS FOR CERTAIN SUBCLASSES OF MEROMORPHIC AND BI-UNIVALENT FUNCTIONS

        In the present investigation, the author introduces two interesting subclasses of normalized meromorphic univalent functions $w=f(z)$ defined on $\tilde{\Delta}:=\{z{\in}\mathbb{C}:1<{\mid}z{\mid}<{\infty}\}$ whose inverse $f^{-1}(w)$ is also univalent meromorphic in $\tilde{\Delta}$. Estimates for the initial coefficients are obtained for the functions in these new subclasses.

        Expression profiling of cultured podocytes exposed to nephrotic plasma reveals intrinsic molecular signatures of nephrotic syndrome

        Background: Nephrotic syndrome (NS) is a common renal disorder in children attributed to podocyte injury. However, children with the same diagnosis have markedly variable treatment responses, clinical courses, and outcomes, suggesting molecular heterogeneity. Purpose: This study aimed to explore the molecular responses of podocytes to nephrotic plasma to identify specific genes and signaling pathways differentiating various clinical NS groups as well as biological processes that drive injury in normal podocytes. Methods: Transcriptome profiles from immortalized human podocyte cell line exposed to the plasma of 8 subjects (steroid-sensitive nephrotic syndrome [SSNS], n=4; steroid-resistant nephrotic syndrome [SRNS], n=2; and healthy adult individuals [control], n=2) were generated using microarray analysis. Results: Unsupervised hierarchical clustering of global gene expression data was broadly correlated with the clinical classification of NS. Differential gene expression (DGE) analysis of diseased groups (SSNS or SRNS) versus healthy controls identified 105 genes (58 up-regulated, 47 down-regulated) in SSNS and 139 genes (78 up-regulated, 61 down-regulated) in SRNS with 55 common to SSNS and SRNS, while the rest were unique (50 in SSNS, 84 genes in SRNS). Pathway analysis of the significant (P≤0.05, -1≤ log2 FC ≥1) differentially expressed genes identified the transforming growth factor-β and Janus kinase-signal transducer and activator of transcription pathways to be involved in both SSNS and SRNS. DGE analysis of SSNS versus SRNS identified 2,350 genes with values of P≤0.05, and a heatmap of corresponding expression values of these genes in each subject showed clear differences in SSNS and SRNS. Conclusion: Our study observations indicate that, although podocyte injury follows similar pathways in different clinical subgroups, the pathways are modulated differently as evidenced by the heatmap. Such transcriptome profiling with a larger cohort can stratify patients into intrinsic subtypes and provide insight into the molecular mechanisms of podocyte injury.

        Effect of Pre-partum Feeding of Crossbred Cows on Growth Performance, Metabolic Profile and Immune Status of Calves

        The effects of pre-partum feeding management in terms of birth weight, growth, metabolic profile and immunity of calves were studied using 24 crossbred (Bos taurus${\times}$Bos indicus) cows, divided into three equal groups. The dietary treatments included feeding of either 3.0 kg concentrate/head/d throughout the 60 d pre-partum (T$_1$), or 3.0 kg concentrate during 60-22 d pre-partum and thereafter at an increased allowance at 0.25 kg/d during the next 21 d till it reached 1% of live weight (T$_2$). The third group of cows was fed similar to T$_2$, except that the concentrate feeding during 60-22 d pre-partum was reduced to 2.0 kg (T$_3$). All the groups had access to ad libitum green fodder throughout. The results revealed that the mean daily dry matter (DM) intake by the cows was similar (p>0.05) among the three groups during the 60 days of the pre-partum but T2 animals tended to gain more live weight (41.25 kg) than T$_1$ (38.12 kg) and T$_3$ (36.25 kg). The body condition score of the cows did not change appreciably over the experimental period. The mean birth weight of the calves was 24.00${\pm}$1.10, 24.63${\pm}$1.17 and 23.25${\pm}$1.19 kg for the three groups, respectively, with the corresponding average daily gain of 154.2, 155.0 and 169.7 g during the subsequent 60 days; both these parameters did not vary significantly ascribable to prepartum feeding regimens of their dams. The total immunoglobulin (Ig) concentration in the colostrum was 6.31${\pm}$0.34, 5.80${\pm}$0.21 and 6.13${\pm}$0.30 g/dl for the three groups, respectively, showing no influence of dietary treatments. The mean serum Ig levels (T$_1$ 2.10${\pm}$0.09, T$_2$ 2.05${\pm}$0.09 and T$_3$ 2.10${\pm}$0.12 g/dl) of calves at 5 d of age were similar among the dietary groups as was the case with various serum biochemical constituents. It is concluded that the variations in pre-partum dietary management elicited no significant influence on the calf performance including the immune status.

        Design of tensegrity structures using artificial neural networks

        This paper focuses on the application of artificial neural networks (ANN) for optimal design of tensegrity grid as light-weight roof structures. A tensegrity grid, 2 m ${\times}$ 2 m in size, is fabricated by integrating four single tensegrity modules based on half-cuboctahedron configuration, using galvanised iron (GI) pipes as struts and high tensile stranded cables as tensile elements. The structure is subjected to destructive load test during which continuous monitoring of the prestress levels, key deflections and strains in the struts and the cables is carried out. The monitored structure is analyzed using finite element method (FEM) and the numerical model verified and updated with the experimental observations. The paper then explores the possibility of applying ANN based on multilayered feed forward back propagation algorithm for designing the tensegrity grid structure. The network is trained using the data generated from a finite element model of the structure validated through the physical test. After training, the network output is compared with the target and reasonable agreement is found between the two. The results demonstrate the feasibility of applying the ANNs for design of the tensegrity structures.
