Bio diesel is one of the clean alternative fuels, and is known to produce less air pollutants than petroleum-based diesel. Bio diesel blended with petroleum diesel can be used. In this paper, exhaust emissions were investigated at various bio diesel mixing rations. Chassis dynamometer testing in connection with high-resolution FTIR spectroscopy was used to measure exhaust emissions including formaldehyde(HCHO) and greenhouse gases such as nitrous oxide(N₂O) and methane(CH₄). Exhaust emissions were collected for CVS-75 driving mode. The results showed that THC and PM decreased but CO, NOx, CH₄, N₂O and HCHO increased gradually, as the mixing ratio increases.

Most of the therapeutic enzymes, which is used for treatment of lysosomal storage diseases, require the glycans containing mannose-6-phosphate (M-6-P). It has been shown that the increased content of M-6-P glycan enhances lysosomal targeting and, therefore, the efficacy of therapeutic enzymes [1]. We have constructed the glyco-engineered yeast harboring very high content of mannosylphosphorylated N-glycans, which can be converted to M-6-P glycan through an uncapping process [2]. In this study, cell wall mannoproteins of the glyco-engineered yeast were extracted by using hot citrate buffer and then digested with pronase, which generated mannosylphosphorylated glycans as attached to asparagine amino acid (Asn). Through the mild acid hydrolysis for uncapping and α(1,2)-mannosidase treatment for trimming, these Asn-linked glycans were converted to Asn-linked M-6-P glycans, which were specifically purified by titanium dioxide resin. The purified Asn-linked M-6-P glycans could be easily conjugated with the crosslinkers containing an amine-reactive group such as a N- hydroxysuccinimide esters which attacks amine group of Asn. Using a chemical group (maleimide, azide or dibenzo-bicyclo-octyne) at the opposite site of the attached cross-linkers, M-6-P glycans were successfully conjugated to target proteins. Proper conjugation of M-6-P glycan to target protein was confirmed by increase of molecular weight in SDS-PAGE and lectin blot using domain 9 of cation-independent M-6-P receptor. Further, M-6-P glycan-conjugated protein was shown to be internalized and targeted to lysosomes of cells.

Chassis dynamometer testing in connection with high-resolution FTIR spectroscopy was used to measure the non-regulated emissions such as methane(CH₄), nitrous oxide(N₂O) and ammonia(NH₃) as well as the regulated emissions such as carbon monoxide(CO), total hydrocarbon(THC) and nitrogen oxides(NOx). A LPLi type passenger car and a LPGi type passenger car were used. As C3 content of LPG fuel increases, CO and THC emissions were decreased but NOx was increased in LPLi passenger car. In a LPGi type passenger car, however, the opposite trend was observed in exhaust emissions.

Chassis dynamometer testing in connection with high-resolution FTIR spectroscopy was used to measure exhaust emissions of nitrous oxide(N₂O) and methane(CH₄) from a representative fleet of several vehicles. Exhaust emissions were collected for three driving cycles: CVS-75, NEDC(ECE+EUDC), KIER city mode. The average N₂O and CH₄ emissions are increased in driving cycle with higher average speed and aggressive driving condition, as expected. The previously reported conversion factors of CH₄/THC and N₂O/NOx could not be supported in LPG-powered and diesel vehicles with DPF.

Chassis dynamometer testing in connection with high-resolution FTIR spectroscopy was used to measure exhaust emissions of nitrous oxide(N₂O) from a in-use fleet of 20 passenger cars. Exhaust emissions were collected for KIER city and KIER highway driving mode. In-use vehicles investigated, including LPG powered passenger cars, were recruited for various emissions testing requirements during 1999 and 2007. A high correlation between N₂O and nitrogen oxides was observed.
