Modification of nucleocytoplasmic proteins with O-GlcNAc regulates a wide variety of cellular processes and has been linked to human diseases. The enzymes O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA) add and remove O-GlcNAc, but the mechanisms regulating their expression remain unclear. Here, we demonstrate that retention of the fourth intron of OGT is regulated in response to O-GlcNAc levels. We further define a conserved intronic splicing silencer (ISS) that is necessary for OGT intron retention. Deletion of the ISS in colon cancer cells leads to increases in OGT, but O-GlcNAc homeostasis is maintained by concomitant increases in OGA protein. However, the ISS-deleted cells are hypersensitive to OGA inhibition in culture and in soft agar. Moreover, growth of xenograft tumors from ISS-deleted cells is compromised in mice treated with an OGA inhibitor. Thus, ISS-mediated regulation of OGT intron retention is a key component in OGT expression and maintaining O-GlcNAc homeostasis.

An apparatus is made for the purpose of thermoluminezcence dosimetry. It has the functions of reading out and recording the thermoluminescence emitted from a heated TL phosphor. The dark current of the PM-tube used can be lowered down to the level of 10^-8 amperes at heater temperature 300℃. CaSO_4 : Dy is adopted as a TL phosphor and it is irradiated by the γ-ray from 258.5mCi of ^137Cs. The γ-ray exposure and the heating rate on the TL phosphor are changeable, and the various glow-curves can be drawn for a given exposure and heating rate. Even though the TL phosphor irradiated to the same exposure, the height and width of the glow-curve can be varied by the operating heating rate. In this experiment, three different heating rates namely, 1.4℃/S, 2.7℃/S and 3.4℃/S were examined. The last one, 3.4℃/S, was turned out to be the most appropriate for reading out thermoluminescence in our particular case of study in which CaSO_4 : Dy phosphors were used.

Background : Peroxiredoxins(Prx) play an important role in regulating cellular differentiation and proliferation in several types of mammalian cells. One mechanism for this action involves modulation of hydrogen peroxide(H2O2)-mediated cellular responses. This report examines the expression of Prx I and Prx II in thyroid cells and their roles in eliminating H2O2 produced in response to TSH. Methods : The expression of Prx-I and Prx-II were quantiated in FRTL-5 after stimulation with Thyroid stimulating hormone (TSH), Forskolin (FSK), Methimazole (MMI) and hydrogen peroxide (H2O2). Transient transfections were carried out with FRTL-5 cells at 80% confluency and 20?g of pCRprx I and pCRprx II or equivalent molar amounts of the pCR3.1TM basic vector. Transient transfection used an electroporation technique. Intracellular H2O2 was assayed in FRTL-5 cells with a fluorescent dye, 2', 7'-dichlorofluoresceindiacetate(DCFH-DA). Apoptosis of cells were evaluated by using an detection kit (Promega, Inc., Madison, WI). Results : Prx I and Prx II are constitutively expressed in FRTL-5 thyroid cells. Prx I expression, but not Prx II expression, is stimulated by exposure to TSH and H2O2. In addition, methimazole(MMI) induces a high level of Prx I mRNA and protein in these cells. Overexpression of Prx I and Prx II enhance the elimination of H2O2 produced by TSH in FRTL-5 cells. Treatment with 500?M H2O2 causes apoptosis in FRTL-5 cells as evidenced by standard assays of apoptosis(i.e., terminal deoxynucleotidyl transferase deoxyuridine triphosphate-biotin nick end-labeling(TUNEL), BAX expression and PARP cleavage. Overexpression of Prx I and Prx II reduces the amount of H2O2-induced apoptosis measured by these assays. Conclusion : These results suggest that Prx I and Prx II are involved in the removal of H2O2 in thyroid cells, and can protect these cells from undergoing apoptosis. These proteins are likely to be involved in the normal physiological response to TSH-induced production of H2O2 in thyroid cells(J Kor Soc Endocrinol15:55-69, 2000).

Soil microarthropods play crucial role in decomposition of organic matters and nutrient cycling in soil ecosystem, and contribute to the energy flow. Herbicide is one of the disturbance factor in soil ecosystem specially in agricultural soil. Non-target effect of the herbicide, Pendimethalin, application on soil microarthropods were studied. Pendimethalin application showed mixed effects on the abundance of soil microarthropods. Short-term reduction of the density was shown in managed agricultural soil system where the natural vegetation density was low. However, in heavily vegetated plots in Pyeongchang, there was no short-term change after treatment. In the latter case, soil microarthropod density was lower from the samples of one year after the treatment than in control plot. Collemboans and predatory mesostigmatid mites were abundant in the study system.

The purpose of this study is to reveal coaches’ leadership types and their positive factors for team culture and improved performance, and provide important information to extend coaches’ understanding of leadership behavior types and players in handball players registered for the Korea Handball Association. This study will contribute to build up the foundation to improve and supplement coaching methods. For the purpose, 400 handball players in national middle and high schools were sampled using cluster random sampling. Unreliable questionnaires which included dual responses and no response were excluded from data analysis. Finally, 384 samples were used for the final analysis via data clarification such as a search for entry error. A questionnaire survey was used as an data collection instrument. The questionnaire whose reliability and validity of a questionnaire was tested in previous researches at home and aborad was modified and added for the purpose of the study. The questionnaire consisted of a total of 46 items: demographic characteristics(5 items), leadership(23 items), team culture(hierarchy culture, adhocracy culture, and rational culture)(12 items), and performance(game influence and game operation) (6 items). This researcher and an assistant researcher visited handball matches to closely investigate the relationship between coach leadership perceived by players and team culture and performance. The purpose and administration way of the questionnaire were explained to subjects. Self administration was used to complete the questionnaire. Unreliable responses and data which included a dual answer or were not answered were first excluded from data analysis. Reliable data were individually inputted. Data processing was made for the purpose of analysis, using a SPSS/PC 18.0 statistics program. Detailed data processing methods were as follows. First, standard multiple regression was conducted to examine the effects of handball coaches’ leadership on team culture. Second, standard multiple regression was conducted to examine the effects of handball coaches’ leadership on performance. Third, correlation analysis was conducted to examine the relationship among handball coaches’ leadership, team culture, and performance. The effects of handball coaches’ leadership type on team culture and performance were analyzed through the above methodology and procedures. The results were as follows. First, handball coaches’ leadership types had a significant effect on team culture. Second, handball coaches’ leadership types had a significant effect on performance. Third, all factors were positively correlated among handball coaches’ leadership, team culture, and performance.
A Study of the Inorganic Scintillator Properties for a Phoswich Detector

CsI(Tl), $CdWO_4(CWO),\;Bi_4Ge_3O_{12}(BGO)\;and\;Gd_2SiO_5:Ce(GSO)$ scintillators were studied to manufacture a phoswich detector. The maximum wavelengths of the CsI(Tl), CWO, BGO and GSO scintillators are 550 nm, 475 nm, 490 nm and 440 nm for the radioluminescence, and the absolute light outputs of the CsI(Tl), CWO, BGO and GSO scintillators are 54890 phonon/MeV, 17762 phonon/MeV, 8322 phonon/MeV and 8932 phonon/MeV with a neutral filter, and the decay time of the CsI(Tl), CWO, BGO and GSO scintillators is $1.3{\mu}s,\;8.17{\mu}s$, 213 ns and 37 ns by a single photon method. The phoswich detector which was manufactured with plastic and CsI(Tl) scintillators could separate the ${\beta}$ particle and ${\gamma}$ ray. The phoswich detector could also measure the pulse height spectra of the ${\beta}$ particle and ${\gamma}$ ray by a PSD method. Phoswich 검출기를 제작하기 위하여 무기 섬광체인 CsI(Tl), $CdWO_4(CWO),\;Bi_4Ge_3O_{12}(BGO)$와 $Gd_2SiO_5:Ce(GSO)$의 특성을 연구하였다. CsI(Tl), CWO, BGO 및 GSO 섬광체의 radioluminescence 중심파장은 550 nm, 475 nm, 490 nm 및 440 nm이였고, neutral filter를 사용하여 측정한 CsI(Tl), CWO, BGO 및 GSO 섬광체의 절대광량은 각각 54890 phonon/MeV, 17762 phonon/MeV, 8322 phonon/MeV 및 8932 phonon/MeV이였으며, single photon method로 측정한 형광감쇠시간은 각각 $1.3{\mu}s,\;8.17{\mu}s$, 213 ns 및 37 ns이였다. 플라스틱 섬광체와 CsI(Tl) 섬광체를 사용하여 phoswich 검출기를 제작하였고 PSD(pulse shape discriminator) 방법으로 ${\beta}$ 입자와 ${\gamma}$ 선을 구별하며 각각의 방사선에 대한 파고 스펙트럼을 측정하였다.
中寒冷環境에서 艾灸刺戟 期間 및 形態가 CD4 T세포에 미치는 영향

Purpose; This Study was aimed to Investigate the different effects according to the form and term of moxibustion therapy on the depression of the immune response induced by cold stress in rats. Methods, The control group and the Sample groups were also exposed to cold stress and they were redivided into 1, 2, 3, 4 day-group by total term of time exposed to cold stress. The sample group were classified into sample I (direct moxibustion therapy), sample I (Bongrae-moxibustion therapy) according to the form of moxibustion, To all of sample groups, each moxibustion therapy were peformed 2 times a day on the loci corresponding Bi-su(BL20) of Human body and Blank loci. Experimental studies frere performed for the observation of the No. of RBC, WBC and Lymphocyte in blood CD4+ T helper cell count, spleen CD4? T helper cell count, axillary lymph node CD4? T helper cell count, and Thymus CD4+T helper cell count in rats exposed to cold stress. Results ; 1. The CD4? T ce11 count in blood was increased statistically significant in all of the sample I (1, 2, 3, 4-day group) as compared with the control group. And in the sample II only 2-day group gained the statistical significance as compared with the control group. 2. The CD4? T cell count in axillyary lymph node was increased statistically significant in all of the sample I (1, 2, 3, 4-day group) compared with the control group. But all of the sample II were increased without the statistical significance compared with the control group. 3. The CD4? cell count in Spleen was increased with the statistical nonsignificance in all of the sample I (1, 2, 3, 4-day group) as compared with the control group. And in the sampleII only 1-day group gained the statistical significance as compared with the control group. all of the sample I (1, 2, 3, 4-day group) as compared with the control group. And in the sample II only 1-day group gained the statistical significance as compared with the control group. 4. The CD4? T cell count in Thymus was increased with the statistical non significance in the Conclusions ; On the depression of the immune response induced by cold stress in rats, direct moxibustion therapy is more effective than Bongrae-moxibustion therapy for the recovery of immune response.
Screening of Oriental Herbal Medicines for Antibacterial Activities

The water extracts of oriental herbal medicines which have been clinically used to treat bacterial infections in Korea were screened for in vitro antibacterial activity by the paper disc assay method. Two Gram positive bacteria, Staphylococcus aureus SG511, Bacillus subtilis ATCC 6633 and two Gram negative bacteria, Escherichia coli 055, Pseudomonas aeruginosa 9027 were used as test organisms. Among 83 of the extracts tested, 25 were active against Staphylococcus aureus SG 511, 9 were active against Bacillus subtilis ATCC 6633, while none showed inhibitory activity against Eschelichia coli 055 and Pseudomonas aeruginosa 9027. Among them, Hwangyonhaedoktang plus hwangyon, Chongwisan, and Ssangbaksan showed remarkably potent antibacterial activity.
