        <P><B>Abstract</B></P><P>Synthesis, radiolabelling, and <I>in vitro</I> evaluation of a new <SUP>125</SUP>I‐labelled iodouracil hexitol nucleoside analogue are reported. The target compound was successfully synthesized by an iodination–destannylation method and then purified by reverse phase HPLC. The radiochemical purity of the product was >99% with decay‐corrected yields of 48±3%. <I>In vitro</I> cellular uptake testing was carried out using MCA and MCA‐tk cell lines for comparison of compound 1 with [<SUP>18</SUP>F]FHBG. The newly synthesized compound 1 showed higher accumulation in herpex simplex virus type 1 thymidine kinase (HSV1‐tk) gene expression cell line (MCA‐tk cell line) than in the wild type MCA cell line compared with [<SUP>18</SUP>F]FHBG. The MCA‐tk to MCA cellular uptake ratio for compound 1 was higher than that of [<SUP>18</SUP>F]FHBG from 2 h after incubation. The radioiodine‐labelled compound 1 (I‐125, <I>t</I><SUB>1/2</SUB>=59.37 days) has a longer physical half‐life than F‐18‐(<I>t</I><SUB>1/2</SUB>=110 min) labelled FHBG. Radioiodine‐labelled compound 1 could be used for monitoring gene expression for a long time. The selectivity for MCA‐tk cell line makes compound 1 a promising imaging agent for HSV1‐tk expression.       • M/G/I Queue 에 관한 硏究

        本 論文에서는 M/G/I Queue에 對한 몇 가지 것들을 알아본다. 먼저 Arriving 'Customer's Districution, Departing Customer's Distribution, Outside Observer's Distribution과 이들 사이의 關係를 알아본다. 그 다음 Little 의 定理를 證明하고 이것들 이외에 M/G/I Queue를 分析하는 데 必要한 것들을 몇 가지 더 알아 본 다음 M/G/I Queue의 mean Queue Length와 Mean Waiting Time 을 구해 본다. Inbedded Markov Chain을 證明하고 이를 利用하여 M/G/I Queue Length 에 對한 Probability Generating Function과 Waiting Time 에 對한 Moment Generating Function을 誘導한다. This study is intended to investigate a few things about M/G/I queue. First of all, the researcher intends to know the relationship among Arriving Customer's Distribution, Departing Customer's Distribution, and Outside Observer's Distribution. Then the researcher tries to prove a theorem of Little and to find out a few more things needed to analyze the M/G/I queue additionally. Moreover the researcher intends to find out the mean queue length and the men waiting time of M/G/I queue. Rastly the researcher tries to induce the moment generating function about waiting time and the probability generating function about M/G/I queue length, by explaining and using the Imbedded Markov Chain.

        Identification and characterization of a carboxypeptidase N1 from red lip mullet (<i>Liza haematocheila</i>); revealing its immune relevance

        <P><B>Abstract</B></P> <P>Complement system orchestrates the innate and adaptive immunity <I>via</I> the activation, recruitment, and regulation of immune molecules to destroy pathogens. However, regulation of the complement is essential to avoid injuries to the autologous tissues. The present study unveils the characteristic features of an important complement component, anaphylatoxin inactivator from red lip mullet at its molecular and functional level. Mullet carboxypeptidase N1 (MuCPN1) cDNA sequence possessed an open reading frame of 1347 bp, which encoded a protein of 449 amino acids with a predicted molecular weight of 51 kDa. <I>In silico</I> analysis discovered two domains of PM14-Zn carboxypeptidase and a C-terminal domain of M14 N/E carboxypeptidase, two zinc-binding signature motifs, and an N-glycosylation site in the MuCPN1 sequence. Homology analysis revealed that most of the residues in the sequence are conserved among the other selected homologs. Phylogeny analysis showed that MuCPN1 closely cladded with the <I>Maylandia zebra</I> CPN1 and clustered together with the teleostean counterparts. A challenge experiment showed modulated expression of MuCPN1 upon polyinosinic:polycytidylic acid and <I>Lactococcus garviae</I> in head kidney, spleen, gill, and liver tissues. The highest upregulation of MuCPN1 was observed 24 h post infection against poly I:C in each tissue. Moreover, the highest relative expressions upon <I>L. garviae</I> challenge were observed at 24 h post infection in head kidney tissue and 48 h post infection in spleen, gill, and liver tissues. MuCPN1 transfected cells triggered a 2.2-fold increase of nitric oxide (NO) production upon LPS stimulation compared to the un-transfected controls suggesting that MuCPN1 is an active protease which releases arginine from complement C3a, C4a, and C5a. These results have driven certain way towards enhancing the understanding of immune role of MuCPN1 in the complement defense mechanism of red lip mullet.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Carboxypeptidase N1 complement component was identified from the red lip mullet. </LI> <LI> Ubiquitous expression of MuCPN1 was observed in healthy mullet tissues. </LI> <LI> Modulated transcriptions of MuCPN1 revealed the importance in the immune responses. </LI> <LI> MuCPN1 was enhanced the nitric oxide production at an inflammatory condition. </LI> </UL> </P>

      • Identification of thioredoxin domain-containing protein 17 from big-belly seahorse <i>Hippocampus abdominalis</i>: Molecular insights, immune responses, and functional characterization

        <P><B>Abstract</B></P> <P>Thioredoxin domain-containing protein 17 (TXNDC17) is a small protein (∼14 kDa) involved in maintaining cellular redox homeostasis via a thiol-disulfide reductase activity. In this study, TXNDC17 was identified and characterized from <I>Hippocampus abdominalis</I>. The open reading frame (ORF) consisted of 369 bp and 123 amino acids. Similar to the other thioredoxins, TXNDC17 contained a conserved WCXXC functional motif. The highest spatial mRNA expressions of <I>HaTXNDC17</I> were observed in the muscle, brain, and intestine. Interestingly, the mRNA expression of <I>HaTXNDC17</I> in blood showed significant upregulation at 48 h against all the pathogen associated molecular patterns (PAMPs) and bacteria. Further, <I>HaTXNDC17</I> transcripts in the trunk kidney were significantly upregulated at 24–48 h by bacterial endotoxin lipopolysaccharides (LPS), viral mimic polyinosinic: polycytidylic acid (poly I:C), and gram-negative bacteria (<I>Edwardsiella tarda</I>). The DPPH assay showed that the radical scavenging activity varies in a concentration-dependent manner. The insulin reduction assay demonstrated a significant logarithmic relationship with the concentration of rHaTXNDC17. Moreover, FHM cells treated with recombinant HaTXNDC17 significantly enhanced cellular viability under oxidative stress. Together, these results show that HaTXNDC17 function is important for maintaining cellular redox homeostasis and that it is also involved in the immune mechanism in seahorses.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Big-belly seahorse thioredoxin domain containing protein 17 maintain redox homeostasis. </LI> <LI> TXNDC17 is ubiquitously found in cytosol and extracellular space. </LI> <LI> Thiol active CXXC conserved motif consists in HaTXNDC17. </LI> <LI> Spatial and temporal mRNA expression was evaluated. </LI> <LI> Radical scavenging ability, antioxidant activity and cellular viability were measured. </LI> </UL> </P>

        Complement factor D homolog involved in the alternative complement pathway of rock bream (Oplegnathus fasciatus): Molecular and functional characterization and immune responsive mRNA expression analysis

        <P>The complement system serves conventional role in the innate defense against common invading pathogens. Complement factor D (CfD) is vital to alternative complement pathway activation in cleaving complement factor B. This catalytic reaction forms the alternative C3 convertase that is crucial for complement-mediated pathogenesis. In this study, rock bream (Oplegnathus fasciatus) CfD (OfCfD) was characterized and OfCfD mRNA expression was investigated. OfCfD encodes 277 amino acids (aa) for a 30-kDa polypeptide. A domain analysis of the deduced OfCfD aa sequence showed a single serine protease trypsin superfamily domain, a serine active region, three active sites, and three substrate-binding sites. Pairwise sequence comparisons indicated that OfCfD has the highest identity (84.5%) with Oreochromis niloticus CID. The phylogenetic tree revealed a common ancestral origin of CfD members, with fish CfD distinct from other vertebrate orthologs. The structural arrangement of the OfCfD gene (2451 bp) contained five exons interrupted by four introns. A spatial transcriptional analysis indicated that OfCfD transcripts constitutively expressed in all of the examined rock bream tissues, and that they were highest in the spleen and liver. In addition, OfCfD transcripts were immunologically upregulated by lipopolysaccharide (LPS) (12 h p.i.), Streptococcus iniae (12 h p.i.), rock bream iridovirus (RBIV) (6-12 h p.i.), and poly I:C (6 h p.i.) in spleen tissue. OfCfD is a trypsin protease and its recombinant protein showed strong protease activity similar to that of trypsin, indicating its catalytic function in the alternative pathway.         Cellular uptake mechanism and intracellular fate of hydrophobically modified glycol chitosan nanoparticles

        Polymeric nanoparticle-based carriers are promising agents for the targeted delivery of therapeutics to the intracellular site of action. To optimize the efficacy in delivery, often the tuning of physicochemical properties (i.e., particle size, shape, surface charge, lipophilicity, etc.) is necessary, in a manner specific to each type of nanoparticle. Recent studies showed an efficient tumor targeting by hydrophobically modified glycol chitosan (HGC) nanoparticles through the enhanced permeability and retention (EPR) effect. As a continued effort, here the investigations on the cellular uptake mechanism and the intracellular fate of the HGC nanoparticles are reported. The HGC nanoparticle, prepared by a partial derivatization of the free amino groups of glycol chitosan (GC) with 5β-cholanic acid, had a globular shape with the average diameter of 359 nm and the zeta potential of ca. 22 mV. Interestingly, these nanoparticles showed an enhanced distribution in the whole cells, compared to the parent hydrophilic GC polymers. In vitro experiments with endocytic inhibitors suggested that several distinct uptake pathways (e.g., clathrin-mediated endocytosis, caveolae-mediated endocytosis, and macropinocytosis) are involved in the internalization of HGC. Some HGC nanoparticles were found entrapped in the lysosomes upon entry, as determined by TEM and colocalization studies. Given such favorable properties including low toxicity, biocompatibility, and fast uptake by several nondestructive endocytic pathways, our HGC nanoparticles may serve as a versatile carrier for the intracellular delivery of therapeutic agents.

        Synergistic effect of MWNT/fly ash incorporation on the EMI shielding/absorbing characteristics of cementitious materials

        The present study aims to develop electromagnetic interference (EMI) shielding materials of which the EM wave shielding/absorbing capabilities were enhanced by a synergistic effect of multi-wall carbon nanotube (MWNT) and fly ash (FA) in terms of EM characteristics. The MWNT/FA-added cementitious materials were fabricated with various content ratios of FA and silica fume (SF). SF was incorporated as a dispersion agent of MWNT and EMI shielding effectiveness (SE) of the cementitious materials was assessed in an effort to determine optimum content ratios of FA and SF. The best EMI SE was achieved by the specimen comprised of MWNT 0.6%, SF 20%, and FA 75% (C-FA75-SF20) as exhibiting -8.0~-57.1dB at a frequency range from 1 to 18GHz, indicating a synergistic effect of MWNT and FA. The superior EMI SE of the C-FA75-SF20 was assessed by three different tests, i.e., SEM observation, energy-dispersive X-ray spectroscopy analysis, and electrical & electromagnetic properties evaluation. Furthermore, a computational simulation work demonstrated that the experimental EMI SE results of the C-FA75-SF20 correspond to the simulation results.

      • Electrochemical behavior of CrN coating for polymer electrolyte membrane fuel cell

        <P>CrN films on a bipolar plate in polymer electrolyte membrane fuel cells have several advantages owing to their excellent corrosion resistance and mechanical properties. Three CrN samples deposited at various radio frequency (RF) powers by RF magnetron sputtering were evaluated under potentiodynamic, potentiostatic and electrochemical impedance spectroscopy conditions. The electrochemical impedance spectroscopy data were monitored for 168 h in a corrosive environment at 70 °C to determine the coating performance at +600 mV<SUB>SCE</SUB> under simulated cathodic conditions in a polymer electrolyte membrane fuel cell. The electrochemical behavior of the coatings increased with decreasing RF power. CrN films on the AISI 316 stainless steel substrate showed high protective efficiency and charge transfer resistance, i.e. increasing corrosion resistance with decreasing RF power. X-ray diffraction confirmed the formation of a CrN(200) preferred orientation at low RF power.</P>

        In order to find out the current tendency of medication practiced for 8 years, 1980-1987, at the Oriental Medicine Hospital, Kyung-Hee University Medical Centre, the published data on the usage of herbal preparations and crude material medica were further analysed. In practice, four doctrines of prescriptions are currently in use in Korean traditional medicine, namely,(i) "sang-han" (originated from "Sang-Han-Lon") : (ii) "hu-se" (from so-called the late Chinese traditional medicine) : (iii) "sa-sang"(form Lee Chemas(李濟馬), Korean constitutional medicine) and (iv) oriental medical doctor's own empirical prescription. Some of important findings were as fellows : 1.The ratio between the use of two forms of medications packted herbs and herbal preparations(pills or liquid etc) was 59% and 41%. 2.Of 1,104 kinds of prescriptions available, numbers of prescriptions lead to consumption of more than 10,000 packets during any one year were 252, which occupied 23% of the total number of proscriptions issued. These 252 kinds of prescriptions were based on the following four doctrines perceived in Korean traditional medicine : "hu-se"(173 kinds or 69%) : "sa-sang"(52 or 20%) : doctor's own empirical(24 or 10%) and "sang-han"(3 or 1%). 3.The usage of 76 popular herbal preparations according to classification of disorders was : for general purpose, e.g., tonicum-21 kinds or 28% : gastroin-testinal disorders-17 or 22% ; neuro-psychiatric disorders-6 or 8% ; diseases of circulatory system-6 or 8% ; gynaecological disorders-5 or 7% : musculo-ske-letal disorders-4 or 5% ; paediatric disorders-4 or 5% etc etc in order. 4.The kind of prcscriptions which was ranked at the top in the consumption was "bo-hyul-an-shin-tang(補血安神湯)". A total of 313.914 packets was used for eight years. 5.The amount of the usage of crude herbs was highest for Fraxini cortex(陳皮). followed by Atractylodes rhizoma alba(白□), Angelicae gigantis(當歸), clycyrrhizae radix(甘草) and Dioscoreae rhizoma(山□). 6.The numbers of species of orudc herbs which were consumed more than 100kg during any one year were 131. Among them, diuretica comprising 11 material was ranked at the top.
