        Ribosomal Protein S6, a Target of Rapamycin, Is Involved in the Regulation of rRNA Genes by Possible Epigenetic Changes in <i>Arabidopsis</i>

        <P>The target of rapamycin (TOR) kinase pathway regulates various biological processes, including translation, synthesis of ribosomal proteins, and transcription of rRNA. The ribosomal protein S6 (RPS6) is one of the well known downstream components of the TOR pathway. Ribosomal proteins have been known to have diverse functions in regulating cellular metabolism as well as protein synthesis. So far, however, little is known about other possible role(s) of RPS6 in plants, besides being a component of the 40 S ribosomal subunit and acting as a target of TOR. Here, we report that RPS6 may have a novel function via interaction with histone deacetylase 2B (AtHD2B) that belongs to the plant-specific histone deacetylase HD2 family. RPS6 and AtHD2B were localized to the nucleolus. Co-expression of RPS6 and AtHD2B caused a change in the location of both RPS6 and AtHD2B to one or several nucleolar spots. ChIP analysis suggests that RPS6 directly interacts with the rRNA gene promoter. Protoplasts overexpressing both <I>AtHD2B</I> and <I>RPS6</I> exhibited down-regulation of pre-18 S rRNA synthesis with a concomitant decrease in transcription of some of the ribosomal proteins, suggesting their direct role in ribosome biogenesis and plant development. This is consistent with the mutation in <I>rps6b</I> that results in reduction in 18 S rRNA transcription and decreased root growth. We propose that the interaction between RPS6 and AtHD2B brings about a change in the chromatin structure of rDNA and thus plays an important role in linking TOR signaling to rDNA transcription and ribosome biogenesis in plants.</P>

        To reduce the exessive noise from the engine of the farm machinery, the precise measuring equipments and analyzing apparatus are required. The exhaust noise of an engine varies continuously between several ranges, it can not be, therefore, measured accurately by the noise indicator commercially available. In this study, acoustic measuring and analyzing system with a high reproducibility of the exhaust noise were developed to investigate desingn paramenters of a muffler and to manufacture an effective silencer of the small farm diesel engine. The system consisted of a microcomputer, sound recognition board, A/D converter, FFT and PSD analysis programs, etc. The results of the experiments were summarized as follows; 1. At the experimental apparatus were produced with the resonance and abnormal sound induced at the rear of the speaker were absorbed, and playbacks of the recorded sound were set up as the original sound resources. The sound resources recorded on the engine removed muffer and were input and analyzed at the microcomputer. 2. The developed system with high sensitivity microphone and microcomputer could be applied as the range as 1㎑, and proved to be useful in the noise measurement and analysis for small diesel engines, and the result of statistical analysis showed a high significance below 1% between original and reproduced sound conditions.

        Figaro type gas sensor was fabricated with sensing materials in which mixed with SnO_(2) , Al_(2)O_(3) binder and palladium catalyst and, developed electronic parts. The SnO_(2) powders obtained by sol-gel method had a mean particle size of 0.93 p m and specific surface area of 32.5 m^(2)/g. And the γ-Al_(2)O_(3)with surface area of 180 m^(2)/g and a solution of 1M. PdCl_(2) were used for the synthesis of the sensing material. Mullite tubes of 0.8mm diamrter, Au pad electrodes of 0.3mm clearance, Au-Pd alloy lead wire of 80μm dia, heating element coiled with Kanthal wire of 60 μm dia were developed to use the gas sensor. The sensitivity of the sensors was excellent for the detection of methane, propane and carbon monoxide at about 490℃, 440℃ and 140℃, respectively.

        Optimization of Ni-zirconia based anode support for robust and high-performance 5 x 5 cm<sup>2</sup> sized SOFC via tape-casting/co-firing technique and nano-structured anode

        High-performance Ni-zirconia based anode supported cells were developed via a cost-effective tape-casting/co-firing technique and a nano structured anode. The fundamental properties for conventional NiO-YSZ anode supports - such as porosity, shrinkage, electrical conductivity and mechanical strength - were measured as a function of the proportion of NiO and YSZ (coarse and fine powders). Electrical conductivity, shrinkage, porosity, strength were found to be 1200 S/cm, 22%, 45% and 55 MPa, respectively, for a composition of NiO:YSZ (60:40 wt%) and coarse:fine YSZ (50:50 wt%). However, warping of the cell and delamination was frequently observed between the anode and the electrolyte after the co-firing step. The NiO/YSZ-ScSZ (40/30-30 wt%) nano-composite anode was synthesized to increase the connectivity of Ni phase, the sinter-ability of YSZ phases and to match the shrinkage with ScSZ electrolyte. It displayed strength of 95 MPa, an electrical conductivity of 1400 S/cm with thermal stability after cycling 10 times, 50% porosity, and 28% shrinkage; the latter being particularly similar to the ScSZ electrolyte. Moreover, the 5 x 5 cm<SUP>2</SUP> sized single cell consisting of the NiO/YSZ-ScSZ anode, ScSZ electrolyte and an LSM-YSZ cathode showed 19 @?/5 cm of flatness and a power of over 13.3 W (0.83 W/cm<SUP>2</SUP>) with hydrogen at 700 <SUP>o</SUP>C.

        한우와 홀스타인 비육우의 근육 위성세포 Cloning 및 배양

        The objectives of this study were to exam optimum culture condition and to compare growth rate of muscle satellite cells derived from Hanwoo and Holstein cattle. Animal growth depends on the proliferation and differentiation of muscle satellite cells. Muscle satellite cells were separated by pronase digestion from 250g muscle and cloned pure cell lines. The optimum cell culture condition was tested with three different culture medium (DME / F12, DMEM, McCoy's 5A) and three different source of serums (fetal calf serum, chicken serum, horse serum). The number of nuclei per unit area was lower in DME/F12 (68%) and McCoy's 5A (64%) than that of DMEM (100%) (P$lt;0.05). Also cell growth rate was lower in CS(37.3%) and HS (10.2 %) than that in FCS (P$lt;0.05). Therefore. the optimum muscle satellite cell culture condition was DMEM + 10 % FCS in both Hanwoo and Holstein cattle.

      • Impact of Education and Culture on Entrepreneurship

        This paper explored the impact of secondary education level and national culture on the entrepreneurial activities. Specifically, we examine how higher education and one dimension of national culture (individualist-collectivist orientation) are related to the ratio of opportunity-entrepreneurship in a country. The research model is developed as: high education and individualism are positive determinants of the entrepreneurial activity, but only when the motivation for starting new ventures is opportunity or improvement and not when people do it for necessity. Our variables were developed with GEM data, World Economic Forum's Global Competitiveness Report and Hofstede's cultural determinants for 57 countries for the years 2009, 2010 and 2011, and we test our hypothesis empirically. In addition to our testing independent variables, we added 3more control variables, economic freedom, and perception of corruption and property rights, in order to specify our findings. Our analysis proves that higher education and individualism are determinants for superior opportunity entrepreneurial activity. Education levels and individualism, however, do not show the interaction effect. Our results show that, after controlling for other entrepreneurship determinants variables borrowed from the institutional theory of entrepreneurship, education at the tertiary level i.e., university, college, etc., positively affects the choice of individuals to pursue opportunity driven entrepreneurship. Moreover, individualism is also found to positively affect entrepreneurs' choice. We hope this research contribute in expanding our understanding on global entrepreneurship activities.

        As we use computer commonly and can get information easily through network, the sharing information on the web has become generalized trend in manufacturing industry field. And so, the sharing information among cooperating companies as well as internal departments is also indispensible requirements. In order to cope with the information society we implemented a universal Internet Simulation on the Web at low cost. To implement three dimension Simulation of a pair of standard spur gears via browser on the web, we have used Pentium MMX 200㎒ CPU, 64MB RAM, 4x CD-ROM Drive Speed, Windows NT 4.0, and Internet Explorer 4.0. And we also used Visual C^++ Language Program, Open GL Graphic Library. ActiveX Control and HTML, which enable this gear simulation to be seen on the WEB. We set standard spur gears data according to KS B1414 regulations. The purpose of implementation of Gear Simulation System on the Web is as follows. First, we can decrease cost in implementing simulation of three dimensions because this system can be implemented on the personal computer without any special device or software. Second, the utilization of world wide web enables us to communicate freely and implement the design of simultaneous engineering escaping from the limitation of time and space. Third, we suggest the possibility of implementation of simulation of three dimensions on the web of three dimensions in order to design products quickly and to connect computer application systems automatically.

      • D.L-α-Tocopherol 投與에 依한 臟器內 Transaminase 活性變動에 關한 硏究

        In order to observe some change in protein metabolism by feeding vitamin E enriched diet, G.O.T. and G.P.T. activities in the liver, spleen and serum of experimental animals were measured. The animals, female albino rats from a pure strain, weighing 226-265g were maintained by fasting or feeding different quality of diets with or without enriching V-E. The animals were divided into six groups: the control group(S group,), the fasting group(A group), the high carbohydrate diet group(B group), the high carbohydrate with V-E(added 25㎎ per day) diet group(BE group), the high protein diet group(C group) and the high protein with V-E(added 25㎎ per day) diet group(CE group). The animals in the A group were sacrified on the 1st 2nd 4th and 8th day after fasting; in the B and BE groups, sacrified on the 10th 20th and 30th day after feeding respective diet; and in the C and CE groups, sacrified on the 5th 10th 20th and 30th, day after feeding. The results obtained are summarized as follows: 1. The change of G. O. T. and G. P. T. activities in the liver or spleen showed opposed tendency when the animals were maintained with the V-E enriched high carbohydrate diet. 2. The G.O.T. activities in the liver of the B and BE groups were increased during first 10 days compared with those of the S group, but the activities of the BE group returned to the control level during 20-30 days. The activities in the liver of the B group were maintained for 20-30 days in the increased state. 3. The change of G.O.T. and G.P.T. activities in the spleen between the B group and the BE group showed different tendencies, but the change of the activities between the C group and CE group showed simillar tendencies. 4. The G.O.T. and G.P.T. activities in the blood were not changed much by feeding different diets. 5. The G.O.T. and G.P.T. activities in the liver were not changed much at the early stage of the fasting, but the activities were significantly increased at the late stage(4-8 days' fasting) of the fasting. 6. The G.O.T. and G.P.T. activities in the liver of the A and B groups were increased compared with those of the S group.

      • Conserved sequences of thrombospondin‐related adhesive protein gene of <i>Plasmodium vivax</i> in clinical isolates from Korea

        <P><B>Summary</B></P><P>Thrombospondin‐related adhesive protein (TRAP) from <I>Plasmodium vivax</I> (<I>P.?vivax</I>) became one of the important vaccine candidates for malaria, because <I>P.?vivax</I> TRAP (PvTRAP) is responsible for the sporozoite–host interactions. PvTRAP polymorphisms in the isolates from Republic of Korea (ROK) were analysed, setting the valuable baseline data for the future vaccine developments and clinical trials with PvTRAP, as a strong vaccine candidate. A total of 54 isolates were collected in 2010. PvTRAP genes from above isolates were amplified and sequenced, and the results were analysed and compared against Sal‐1 strain. Sequencing analysis of 1424‐bp‐size PvTRAP PCR products revealed one major allelic type with six non‐synonymous substitutions, where S81T, E95D, I121V and T127R substitutions were found in region II, and K371N and A425E substitutions from region IV. The ROK isolates revealed the limited sequence polymorphisms in PvTRAP in comparison with the reported isolates from other nations.</P>

        INFLUENCE OF ENDOGENOUS SOMATOSTATIN ON LACTATION IN RATS

        The role of endogenous somatostatin on lactation in rats was examined by passive immuno-neutralization of Wistar rats. In one study, the rats were given either immunoglobulin raised in sheep against somatostatin, or non-specific sheep immunoglobulins by daily s.c. injection from parturition through the first two weeks of lactation. The growth of the pups was recorded by weighting every second day, and the milk yield calculated from the pup weight and weight gain. Immunoneutralization of maternal somatostatin during pregnancy had a slight effect (p < 0.05) on the mean birth weight of the pups but no subsequent effect on postnatal growth rate of the pups or milk yield ($25.32{\pm}0.88g/day$) compared with young control rats given normal sheep serum ($25.55{\pm}1.04g/day$). Similarly, passive immunization against somatostatin during lactation ($21.96{\pm}1.57g/day$) also did not affect milk yields compared with controls ($24.85{\pm}1.03g/day$). These data do not support a significant role for endogenous somatostatin in regulating milk production in lactating rats.
