      • Quniolone 화합물의 물, 옥탄올계에서의 전이에 대한 열역학적 함수의 고찰

        Norfloxacin, ofloxacin, enoxacin, ciprofloxacin, KR 10755의 5가지 quinolone 화합물을 대상으로 온도를 달리하면서 물에서 n-octanol로의 분배계수를 측정한 결과 아래와 같은 결론을 얻었다. 1. Norfloxacin, ofloxacin, enoxacin, ciprofloxacin 등의 화합물은 온도상승에 따라 점차 분배계수 K_D값이 증가하였으나 KR 10755는 온도상승에 따라 K_D값이 일정하게 변화하지 않았다. 2. 이 실험에 사용한 모든 quinolone 화합물들은 1.0 이하의 K_D값을 나타냈고, KR 10755< ofloxacin < enoxacin < norfloxacin < ciprofloxacin 순서로 증가하였다. 3. 산출된 열역학적 함수, ??D, ??H, ??S의 결과를 종합해 보면, 1) 양의 ??G의 값은 본 실험에서 사용한 모든 quinolone 화합물들이 물에서 n-octanol로의 이행이 용이하지 않다는 것을 의미한다. 2) 모든 quinolone 화합물들이 양의 ??S 값을 나타내는 것으로 보아 화합물들이 n-octanol에서 더욱 무질서해지는 것 같다. 3) 모든 quinolone 화합물들의 두 용매 사이에서의 평형분배반응은 흡열반응이다. The partition coefficients of quinolones-norfloxacin, ofloxacin, enoxacin, ciprofloxacin, KR 10755 from water to n-octanol have been measured as a function of temperature. These data are used to evaluate the thermodynamic parameters ??G, ??H, ??S for the solute transfer process. The partition coefficients increased in order of ciprofloxacin < norfloxacin < enoxacin < ofloxacin < KR 10755. All transfer processes from water to n-octanol of quinolones using in this study are not favoured and the positive entropy changes of all quinolones suggest that all compounds become disorder in n-octanol. All transfer processes were endothermic.

      • 핵전이에 의한 Aspergillus속과 Penicillium 속의 잡종으로부터 생산된 esterase특성

        핵전이에 의해 Aspergillus oryzae var oryzae와 MPV 31 사이의 잡종을 획득하였고, 그특성과 esterase 활성 및 동위효소 양상을 조사한 결과는 다음과 같다. UV와 MNNG의 이중처리에 의해 A. oryzae var oryzae와 P. verruculosum F-3의 영양요구성 돌연변이를 각각 3주, 6주를 획득하였으며, 핵전이를 위한 원형질체 형성의 최적조건은 PH 5.8, 1.0% Novozym 234, 0.6 M KCl 그리고 효소의 처리시간은 2 시간으로 하였다. 또한 원형질체 재생을 위한 삼투안정제도 역시 0.6 M KCl이었으며, 재생율은 각각 56.4%, 52.3%로 나타났다. 핵전이에 의한 형질전환율은 3.0 x 10<윗첨자>-5</윗첨자>~1.0 x 1<윗첨자>-5</윗첨자>으로 나타났으며, 형질전환체의 유전적 안정성과 conidial size, 핵염색, 그리고 DNA 함량의 측정결과핵형은 aneuploid로 추정되었고 효소 활성의 측정결과 형질전환체가 모균주에 비해 1.3~2.2배 가량 증가하였으며, 동위효소 양상은 형질전환체와 모균주에서 유사하게 나타났으나 효소활성에 차이가 있음을 알수 있었다. Intergeneric hybrids formed between Aspergillus oryzae var oryzae and Penicillium verruculosum F-3 were obtained by nuclear transfer technique. Several auxotrophic mutants isolated from conidiospores of the two strains mutagenized with ultraviolet and N-methyl-N-nitrosoguani-dine. Optimal conditions for formation of intergeneric hybrids were investigated. Frequencies of hybrid formation by nuclear transfer were 1 x 10<윗첨자>-5</윗첨자>~3 x 10<윗첨자>-5</윗첨자>. From observation of genetic stability, conidial size, DNA content, nuclear stain, it was suggested that their karyptypes are aneuploid. The hybrid posses the 1.2~l.8 fold higher esterase activities than those of parental strains. It was also revealed that some hybrids had different isozyme patterns compared with those of parental strains by esterase activity assays.

        전통한과류에 대한 소비자선호 및 구매 후 만족에 관한 연구 - 주부소비자를 대상으로 -

        본 연구는 주부소비자를 대상으로 첫째, 한과류에 대한 소비자의 선호와 구매 후 만족을 설명하고, 둘째, 한과류에 대한 소비자선호와 구매 후 소비자만족에 영향을 주는 관련요인을 파악하였다. 연구결과 47%의 응답자가 한과류를 선호하는 것으로 나타났으며, 약 47%의 소비자가 구매우 만족한 것으로 나타났다. 다음으로 소비자선호와 구매 후 만족의 결정요인 분석결과를 살펴보면 첫째, 소비자선호에 유의하게 영향을 미치는 요인으로는 연령, 학력, 소득, 가격부담인지, 안전식품인지로 나타났다. 나이가 많을수록, 교육수준이 높을수록, 소득수준이 높을수록, 한과류를 안전한 식품으로 인지할수록 한과류를 선호할 확률은 증가하는 것으로 나타났다. 반면, 한과류의 가격이 비싸다고 인지할 경우 한과류를 선호할 확률은 감소하는 것으로 나타났다. 둘째. 한과류에 대한 구매 후 소비자만족여부는 구매비용과 외부정보원이 유의한 요인으로 나타났다. 구매비용이 클수록, 정보탐색시 외부 정보원을 활용할 경우 구매 후 만족할 확률은 증가하는 것으로 타타났다. The purposes of this study were to explain consumer preference and satisfaction associated with Han-gwa and to explore the influencing factors on them. The results of this study were as follows: 1) Forty seven percent of respondents preferred Han-gwa. and about 47% of them were satisfied from purchasing Han-gwa. 2) The consumer preference was found to be significantly influenced by age. education. income. price and food safety. If consumers were old. had higher education, had higher income and recognized Han-gwa as a safe food. they were likely to prefer Han-gwa. But if consumers recognized Han-gwa as an expensive food. they were less likely to prefer Han-gwa. In addition. the consumer satisfaction was found to be significantly influenced by purchasing expenses and external information sources such as relatives. friends and salesmen.

        Enhancement of SOX-2 expression and ROS accumulation by culture of A172 glioblastoma cells under non-adherent culture conditions.

        <P>More efficient isolation and identification of cancer stem cells (CSCs) would help in determining their fundamental roles in tumor biology. The classical tool for this purpose is anchorage-independent tumorsphere culture. We compared the effects of differently textured culture plates and serum deprivation on the acquisition of CSC properties of A172 glioblastoma cells. Cells were cultured on standard polystyrene-treated plates, ultra-low attachment, poly (2-hydroxyethyl methacrylate)-coated plates, and 1% agar-coated plates with 10% serum or in serum-free glioblastoma sphere medium (GBM). Based on mitochondrial reductase activity and subG1 proportions, non-adherent conditions had a greater impact on A172 cell viability than serum deprivation. Among the stemness-related genes, SOX-2 expression was significantly upregulated by serum deprivation under non-adherent conditions, while several epithelial-to-mesenchymal transition (EMT)-related genes were less dependent on serum. In addition, reactive oxygen species (ROS) accumulation in A172 cells was significantly increased in GBM under non-adherent conditions. Despite the correlation between SOX-2 induction and ROS accumulation, treatment with the ROS scavenger N-acetyl-l-cysteine did not prevent SOX-2 expression, suggesting that ROS accumulation is not an essential requirement for induction of SOX-2. Our results suggested that cultivation of cancer cells under conditions of serum deprivation in an anchorage-independent manner may enrich SOX-2-expressing CSC-like cells in vitro.</P>

        Mycobacterium avium paratuberculosis in Wild Boars in Korea

        <P>Mycobacterium avium subsp. paratuberculosis (MAP) causes chronic infectious enteritis in various domestic and wild mammals and is widely distributed globally. Interspecies transmission has been frequently reported. We investigated the presence of MAP from December 2010 to March 2011 in blood and feces collected from 222 hunter-killed wild boars. We collected 197 serum and 180 fecal samples and examined them by culture, PCR, and enzyme-linked immunosorbent assay. We investigated the status of MAP infection and the MAP genotypes in the wild boar population of Korea by using IS900 PCR and IS1311-restriction endonuclease analysis typing. Of the 180 fecal samples cultured, MAP colonies were recovered from two. By PCR, 18 animals were positive for MAP and one serum sample had a strong humoral response to MAP. The PCR-positive DNA samples from the colonies and the feces samples were genotyped as 'cattle type' and 'bison type,' which are major MAP genotypes infecting domestic species in Korea. Our study provides new information on mycobacterial infection among wild boars, and suggests that a more effective program should be developed to monitor mycobacterial infections in wild animal populations in Korea.</P>

        The herd-level prevalence of antibodies against Mycobacterium avium subspecies paratuberculosis in the Gyeonggi and Chungbuk of Korea, as detected by bulk tank milk ELISA

        Mycobacterium avium subspecies paratuberculosis (MAP) causes Johne`s disease (JD) in ruminants. This is the first large scale report to estimate the herd-level prevalence of antibodies against MAP by using an ELISA to detect antibodies in bulk tank milk (BTM) samples. The samples were collected from January 2011 to November 2011, from 636 herds of the dairy farms in the Gyeonggi and Chungbuk areas of Korea. The overall apparent prevalence of MAP antibody-positive herds was 8.5%, and regional prevalence were 32/440 (7.3%) and 22/196 (11.2%) of dairy farms in the Gyeonggi and Chungbuk areas, respectively. The results did not differ significantly by region. While we have determined the prevalence rate of MAP in the Gyenoggi and Chungbuk areas in this study, there is a continuing need for well-designed studies to calculate the prevalence of MAP in dairy herds based on culture and molecular findings.

        High fat diet-induced obesity leads to proinflammatory response associated with higher expression of NOD2 protein

        Obesity has been reported to be associated with low grade inflammatory status. In this study, we investigated the inflammatory response as well as associated signaling molecules in immune cells from diet-induced obese mice. Four-week-old C57BL mice were fed diets containing 5% fat (control) or 20% fat and 1% cholesterol (HFD) for 24 weeks. Splenocytes (1 × 107 cells) were stimulated with 10 μg/mL of lipopolysaccharide (LPS) for 6 or 24 hrs. Production of interleukin (IL)-1β, IL-6, and TNF-α as well as protein expression levels of nucleotide-binding oligomerization domain (NOD)2, signal transducer and activator of transcription (STAT)3, and pSTAT3 were determined. Mice fed HFD gained significantly more body weight compared to mice fed control diet (28.2 ± 0.6 g in HFD and 15.4 ± 0.8 g in control). After stimulation with LPS for 6 hrs, production of IL-1β was significantly higher (P = 0.001) and production of tumor necrosis factor (TNF)-α tended to be higher (P < 0.064) in the HFD group. After 24 hrs of LPS stimulation, splenocytes from the HFD group produced significantly higher levels of IL-6 (10.02 ± 0.66 ng/mL in HFD and 7.33 ± 0.56 ng/mL in control, P = 0.005) and IL-1β (121.34 ± 12.72 pg/mL in HFD and 49.74 ± 6.58 pg/mL in control, P < 0.001). There were no significant differences in the expression levels of STAT3 and pSTAT3 between the HFD and the control groups. However, the expression level of NOD2 protein as determined by Western blot analysis was 60% higher in the HFD group compared with the control group. NOD2 contributes to the induction of inflammation by activation of nuclear factor κB. These findings suggest that diet-induced obesity is associated with increased inflammatory response of immune cells, and higher expression of NOD2 may contribute to these changes.
