        Brucella abortus 국내 분리주의 36kDa 외막단백을 암호하는 omp2b 유전자의 염기 분석

        B. abortus 국내 분리주의 분자유전학적 성상을 규명하고 분자역학적 연구에 필요한 기초자료를 얻기위해 36kDa OMP를 암호하는 omp2b 유전자를 PCR법으로 확인하고, 그리고 omp2b 유전자의 핵염기 서열과 아미노산 서열을 결정하고 이 결과를 Brucella 표준균주 및 여러 동물에서 분리된 Brucella 분리주와 상호 비교 분석하였던 바 다음과 같은 결과를 얻었다 1. B. abortus 분리주 (7주)에 대해 omp2b primers를 이용하여 PCR을 수행 한 바 공시한 참조균주와 분리균주 모두에서 1,251bp의 특이한 DNA 절편이 각각 증폭되었다 2. omp2b 유전자의 1,089bp에 대한 염기서열과 이에 대한 362개의 아미노산 서열을 결정하였다. 분리균주의 omp2b 유전자의 염기서열을 참조균주와 비교 분석한 결과 B. abortus 2308 (biovar 1) 및 B. abortus B3196 (biovar 5)과는 99.6%~l00% 및99.5%~99.9%의 매우 높은 상품성을 나타내었고, B. abortus 45/20 (biovar 1)과는 97.1%~97.4%의 상품성을 나타내었다. 또한 362개의 아미노산 서열에 대해 분석하였던 바, 분리균주들은 B. abortustus 2308 (biovar 1) 및 B. abortus B3196 (biovar 5)과는 99.2%~l00%의 매우 높은 상품성을 보였으며, B. abortus 45/20 (biovar 1)과는 96.4%~97.2%의 상품성을 보였다. 3. 본 시험에 공시된 브루셀라균주의 omp2b 유전자서열과 여러 동물에서 분리된 브루셀라 분리주의자료를 바탕으로 phylogenetic tree를 작성 한 바, 국내 분리주는 B. abortus 2308 (biovar 1), B. abortus B3196 (blovar 5), B. abortus 45/20 (biovar 1), B. suis 1330 (biovar 1), B. melitensis 16M (biovar 1), B. canis NCTC 01854, B. neotomae 5k33과 97.3%이상의 높은 근연성을 보이며 동일한 cluster에 소속되었으며, omp2b 유전자는 매우 잘 보존됨을 알 수 있었다. omp2b gene of Brucella abortus isolates encoding 36kDa outer membrane protein (OW) was amplified by polymerase chain reaction (PCR) and the sequences of nucleotide and amino acid were determined and analyzed to obtain the basic data for molecular epidemiological features of brucellosis in Korea. In PCR using omp2b primers for the gene of 36kDa OMP, the specific signals at 1,251bp were demonstrated, The nucleotide sequences of omp2b gene of 1,089bp were sequenced by dye terminator cycle sequencing method, and 362 amino acid sequences were predicted. As compared the nucleotide sequences with those of B. abortus reference strains, the isolates showed the very high homology to the reference strains ranging 97.1%-100%. The similar results were also obtained in the analysis of the amino acid sequences. In the phylogenetic tree, the isolates clustered with the reference strains and the majority of strains originated from pig, dog, wood rat and cattle.

      • Pore Flow Model에 의한 투과증발기구의 해석에 관한 연구

        A study was carried out on the separation of water-ethanol mixtures by pervaporation using cellulose acetate membrance. The experimental data were further analyzed by using a transport theory developed on the basis of the pore flow mechanism. The experimental result for pure liquid is well coresponded to calculated one by pore flow mechanism. But due to interactions and coupling effects between permeats and membrance, difference is existed between experimental and calculated results for mixture. In pervaporation of single component, the total permeate flux decreased with an increase of permeate pressure and increased with an increase of temperature. The experimental permeate flux with permeate pressure was well fitted with calculated one by pore flow model. The activation was 11.714 KJ/mol for water, 1,508 KJ/mol for ethanol. For the mixture, the total permeate flux increased with an increase of temperature and decreased with an increase of feed ethanol mole fraction. However, there are considerable difference between experimental and calculated one due to interactions and coupling.

        계면활성제의 친수·소수성 그룹의 부분전하와 CMC 특성 (양이온 및 양쪽성 계면활성제)

        On the basis of theory of Bratsch's electronegativity equalization, the electronegativity equalization, the group electronegativities and the group partial charges for cationic and amphoteric surfactants could be calculated using Pauling's electronegativity parameters. From calculated output, we have investigated relationships between CMC(critical micelle concentration) and partial charge and group electronegativity of hydrophilic and hydrophobic groups, structural stability of micelle for cationic and amphoteric surfactants. As a result, CMC depends upon partial charge and electronegativity of hydrophilic and hydrophobic groups for surfactants. In case of cationic surfactant, as the increment of carbon number in hydrophilic group, partial charge of hydrophilic group is increased, but CMC and electronegativity of hydrophilic group is decreased. With increasing the carbon number of hydrophilic group for cationic surfactant, its partial charge is increased, but CMC and its electronegativity are decreased. With increasing the carbon number of hydrophobic group for cationic and amphoteric surfactant, its partial charge is increased, but CMC and its electronegativity are decreased

      • 피르빅산 탈수소 효소 복합체 구성성분의 혈청학적 분석

        One of multienzyme complexes involved in oxidative decarboxylation of 2-oxo acids, pyruvate dehydrogenase complex(PDHC) has an important role in cellular energy metabolism. PDHC was isolated from bovine kidney and the anti PDHC anti sera was raised in rabbit. The antisera were inmunoreacted with all the subunits(E2, E3, protein-X, Ela and E1β ) of PDHC. An attempt is being made to identify PDHC subunits serologically in human leukocyte using the anti-PDHC antisera.

        It has been observed that Pt(IV) in Hydrochloric acid solution, when treated with Tin(II) chloride give a orange color and the color that is due to chloroplatin-ous acid, is stable reproducible and quantitative. Thus it is possible to determine platinum by use of spectrophotometer. This method saves time in determination of Pt and is applied to estimation of small amounts of Pt in its ores and alloys.

      • 수직 안진을 동반한 뇌간뇌염 1례

        It is known that meningoencephalitis is the disease usually characterized by a hstory of headache, high fever and disturbance of consciousness. Its cause is usually virus. In most cases, inflammatory reaction involve the brain diffusely, but occasionally involve brain locally. In those cases in whom inflammatory reaction involved brain locally, specific symptoms appear according to the location involved. Clinically, we can suspect the site of brain where inflammatory reaction involved, according to clinical symptoms. We experienced a case of meningoencephalitis with downbeat nystagmus implying that the inflammatory lesion involve pontomedullary junction. It was confirmed by MRI. So, we report it with review of literatures.

      • BRS 생체정보 분석방법(分析方法)의 정확도(正確度) 확인 연구

        BRS는 인간의 잠재능력을 적극적으로 계발 · 활용하여 임의의 시료에 대한 미지 (未知)의 생체정보 (Bio-Information)를 편리하게 분석할 수 있는 효과적이고 경제적인 분석장비이다. 그러나 BRS와 관련된 분석방법 자체의 정확도에 대한 연구는 충분하게 이루어지지 않고 있다. 그래서 BRS 분석방법의 정확도를 과학적으로 확인해보기 위해서 본 연구를 진행했다 실험결과, BRS에 의한 생체정보 분석방법 의 정확도는 측정자의 숙련정도에 따라 다르지만 고급 측정자의 경우에는 95% 이상의 높은 확률로 표준시료의 생체정보를 정확하게 분석할 수 있으며, 측정자의 분석능력이 떨어지는 경우에는 여러 명의 측 정자를 대상으로 비교검토(Cross-Check) 방식으로 실험을 보완하면 90% 이상의 정확도를 확보할 수 있었다. BRS is a sort of analysis-instrument which help to estimate some Bio-Information for a sample conveniently and effectively But, for lack of the study to verify the confidence-degree of the Bio-Information analysis method related to BRS, the availability of the method is not known clearly yet, It was the reason that made authors perform this study And It was possible to arrive at the following two conclusions I though the confidence-degree of the Bio-Information analysis method ny BRS was different according to the degree of the expertness of a person using the instrument in the case of the very skillful expert, the confidence-degree for the standard sample used in this study was reached to 95% up 2 In the general case, to improve of the confidence-degree of the Bio-Information analysis method by BRS, the cross-check method by the several measuring person can be introduced.

      • 소 卵胞의 體外培養에 의한 卵子의 體外成熟과 體外受精에 관한 硏究

        This study was to investigate the effects of the follicular culture from which the oocytes origenate on their subsequent in vitro maturation. The follicular oocytes were cultured in TCM-199 medium containing FCS, hormones, BFF for 24-30 hrs in CO^2 incubator with 5% CO^2 in air at 38.5℃ and then matured oocytes were again 18-20 hrs with motile capacitated spermatozoa in the BO solution containing 100 ㎍/㎖ of heparin. The results obtained in these experiments were summarized as follows : 1.Ovarian follicles were isolated and cultured according to size 1-2 ㎜, 2-4 ㎜, 4-6 ㎜ and 6-8 ㎜ for 24-30 hrs. The rates of GVBD in each group were 80.0%(40/50), 84.4%(38/45), 77.8%(35/45) and 84.0%(42/50), but oocytes maturation were supressed at anaphaes-Ⅰ or telephase-Ⅰ stage. 2. When oocytes cultured for 4 hrs at 35℃ in medium with BFF and hormones after follicular culture for 24-30 hrs, the maturation completed to metphase-Ⅱ stage. However, rates of GVBD in oocytes from follicular culture were higher than oocytes cultured in medium. 3. When oocytes from follicle cultured for 4 hrs at various temperature and culture time were cultured for 24-30hrs the rates of oocytes maturation from follicle cultured at 20℃(43.6%, 24/55), 30℃(47.3%, 26/55) and 35℃(52.7%, 29/55) were significant higher than group cultured at 4℃(24.4%, 11/45). 4. When oocytes from follicles cultured for hrs at 35℃, the rates of oocytes maturation and fertilization rates were 67.3%(101/150), 29.3%(44/150) and oocytes cultured for 24-30hrs at 35℃, thr rates of oocytes maturation and in vitro fertilization were 86.5%(122/141), 53.9%(76/141), respectively.
