Effect of welding defects on mechanical properties of welded joints subjected to temperature

Welding defects negatively affect the safety of steel structures. In fire, the steel connections with welding defects would fracture prematurely that cause the structure lose their fire resistance. However, the knowledge of effects of welding defects on welding connections at elevated temperature are still limit. This paper conducted steady-state tensile tests on butt welded specimens with artificial defects to investigate the effect of defects on the mechanical properties of butt welding at high temperatures. These effects on the evolution law of stress and strain, fracture strengths and extension abilities were discussed. The results show that the stress concentration caused by the defects in the welding zone reduced the yield strengths of the weldments at high-temperature, and the stress concentration induced brittle fracture occurring at the welding zone at high temperature with low levels and produced different failure modes. The extension abilities of the weldments at different temperatures were influenced by the defect levels significantly.

In the hospitality and tourism sectors, service robots have become increasingly adopted by companies to facilitate frontline service to reduce human labor, improve efficiency and provide better customer experience. In addition, the COVID-19 pandemic has intensified consumers’ demand for service robots to reduce human contact. To better understand the impact of service robots in the service sector, researchers have been examining user behavior of different technologies. While prior empirical studies have mainly focused on utilitarian-based acceptance models, other perspectives, such as the interpersonal relations, have been largely overlooked. With the ubiquitous of social media, people become connected but alone, leading to negative effects on interpersonal relations.

In the present experimental study, cecal ligation and puncture significantly increased the myocardial injury assessed in terms of excess release of creative kinase-MB (CK-MB), cardiac troponin I (cTnI), interleukin (IL)-6 and decrease of IL-10 in the blood following 12 h of laparotomy procedure as compared to normal control. Also, a significant increase in protein expression levels of high-mobility group box 1 (HMGB1) and decreased phosphorylation of glycogen synthase kinase-3β (GSK-3β) was observed in the myocardial tissue as compared to normal control. A single independent administration of telmisartan (2 and 4 mg/kg) and AR-A014418 (1 and 2 mg/kg) substantially reduced sepsis-induced myocardial injury in terms of decrease levels of CK-MB, cTnI and IL-6, HMGB1, GSK-3β and increase in IL-10 and p-GSK-3β in the blood in sepsis- subjected rats. The effects of telmisartan at dose 4 mg/kg and AR-A014418 at a dose of 2 mg/kg were significantly higher than the telmisartan at a dose of 2 mg/kg and AR-A014418 1 mg/kg respectively. Further, no significant effects on different parameters were observed in the sham control group in comparison to normal. Therefore it is plausible to suggest that sepsis may increase the levels of angiotensin II to trigger GSK-3β-dependent signaling to activate the HMGB1/receptors for advanced glycation end products, which may promote inflammation and myocardial injury in sepsis-subjected rats.

The oligonucleotide microarray, a high-throughput polymorphism detection technology, holds great promise for the characterization of complex genetic variance. To achieve greater sensitivity and specificity for it to be an effective platform technology we present results and discuss some of the factors influencing signal intensities and single-mismatch discrimination in array-based mutation/SNP detection. Probes with a series of concentrations were spotted onto the slide in order to find the optimal concentration with the identifiable satisfying signals and the stable ratios between matched and mismatched probes. It was found that under our experimental conditions, when the initial probe concentration is higher than the maximum immobilization capability of the slide (7.5 µM), the hybridization signal will be saturated and the ratio between matched and mismatched probes will be more stable than at a lower probe concen-tration. Considering the cost of probes and the systematic stability, a constant spotting cone n-tration of 10 µM was selected. The stability of different types of mismatched oligo-DNA du-plexes on the glass surface was also confirmed. The results show that the order of stability of mismatched oligo-DNA duplexes on a glass surface is in general agreement with previous reports conducted using liquid and polyacryl-amide gel pads. This suggests that the influence of the mismatched base pair on the stability of the duplex in a solid hybridization system is similar to that in the solution hybridization environment.

Double stranded targets on the cDNA microarray contain representatives of both the coding and noncoding strands, which will introduce hybridization competition with probes. Here, the effect of double and single strands of targets on the signal intensity and the ratios of Cy5/Cy3 within the same slide were compared. The results show that single stranded targets can increase the hybridization efficiency without changing the Cy5/Cy3 ratio. Based on these results, a new strategy was established by generating cDNA targets with asymmetric PCR, instead of conventional PCR, to increase the sensitivity of the cDNA microarray. Furthermore, the feasibility of this approach was validated. The results indicate that the cDNA microarray system based on asymmetric PCR is more sensitive, with no decrease in the reliability and reproducibility as compared with that based on conventional symmetric PCR.
