        Identification of Genes Modulated by High Extracellular Calcium in Coculture of Mouse Osteoblasts and Bone Marrow Cells by Oligo Chip Assay

        Calcium concentration in the bone resorption lacunae is high and is in the mM concentration range. Both osteoblast and osteoclast have calcium sensing receptor in the cell surface, suggesting the regulatory role of high extracellular calcium in bone merabolism. In vitro, high extracellular calcium stimulated osteoclastogenesis in coculture of mouse osteoblasts and bone marrow cells. Therefore we examined the genes that were commonly regulated by both high extracellular calcium and 1,25(OH)_(2)vitaminD_(3)(VD3) by using mouse oligo 11 K gene chip. In the presence of 10 mM[Ca^(2+)]e or 10 nM VD3, mouse calvarial osteoblasts and bone marrow cells were co-cultured for 4 days when tartrate resistant acid phosphatase-positive multinucleated cells start to appear. Of 11,000 genes examined, the genes commonly regulated both by high extracellular calcium and by VD3 were as follows; 1) the expressions of genes which were osteoclast differentiation markers or were associated with osteoclastogenesis were up-regulated both by high extracellular calcium and by VD3; trap, mmp9, car2, ctsk, ckb, atp6b2, tm7sf4, rab7, 2) several chemokine and chemokine receptor genes such as sdf1, scya2, scyb5, scya6, scya8, scya9, and ccr1 were up-regulated both by high ectracellular calcium and by VD3, 3) the genes such as mmp1b, mmp3 and c3 which possibly stimulate bone resorption by osteoclast, were commonly up-regulated, 4) the gene such as c1q and msr2 which were related with macrophage function, were commonly down-regulated, 5) the genes which possibly stimulate osteoblast differentiation and/or mineralization of extracellular matrix, were commonly down-regulated;slc8a1, admr, plod2, lox, fosb, 6) the genes which possibly suppress osteoblast differentiation and/or mineralization of extracellular matrix, were commonly up-regulated;s100a4, npr3, mme, 7) the genes such as calponin 1 and tgfbi which possibly suppress osteoblast differentiation and/or mineralization of extracellular matrix, were up-regulated by high extracelluar calcium but were down-regulated by VD3. These results suggest that in coculture condition, both high extracellular calcium and VD3 commonly induce osteoclastogenesis but suppress osteoblast differentiation/mineralization by regulating the expression of related genes.

        황남대총 98호분 출토 유리의 과학적 분석

        황남대총 98호분 북분과 남분에서 출토된 유리시료 40점에 대한 과학적인 분석을 실시하였다. 유리의 성분조성은 주사전자현미경-에너지분산형분광기(SEM-EDS)를 사용하여 정량분석 하였고 다변량해석법을 통하여 시료를 분류하였다. 그 결과 시료 40점 모두 Na₂O를 약 20%정도 함유한 소다-석회(Na₂O CaO-SiO₂)유리임을 확인하였으며, 다시 5개 주성분(SiO₂, AI₂O₃, Na₂O, CaO, K₂O)으로 다변량해석[주성분분석(PCA)]을 실시한 결과 2개의 群으로 분류되었다. I 群에 포함된 시료는 Al₂O₃의 농도가 9.7%로 높고 CaO의 농도는 2.2%인데 비하여 Ⅱ群에서는 각각 3.2%,4.9%의 범위로 나타났다. 특히 시료 No.12의 노란 색으로 편석된 부분을 미소부위 XRD로 분석한 결과 PbSnO₃임을 국내 최초로 확인할 수 있었다. 鉛을 함유한 시료 중 No.12, 17은 열이온화질량분석기(TIMS)로 납동위원소비를 측정하였고 선형판별식 분석법(SLDA)을 이용하여 납의 산지를 추정하였다. 그 결과 각각 중국 남부, 한국 남부의 납광석을 사용한 것으로 나타났다. Elemental analysis of 40 glass samples from the Northern Tomb and the Southern Tomb of Hwangnam-daechong No. 98 was performed. Fourteen compositions of each sample were analyzed quantitatively by SEM-EDS and glass samples were classified by multivariate analysis such as PCA. All of 40 samples were confirmed to be Na₂O CaO-SiO₂ system with about 20% of Na₂O. Samples were classified into two groups by doing PCA on concentrations of 5 major elements (SiO₂, AI₂O₃, Na₂O, CaO, K₂O). Samples included in group I showed the concentration of AI₂O₃ is about 9.7% and that of CaO, about 2.2%. In group II, concentration of Al₂O₃ is about 3.2% and that of CaO, about 4.9%. Especially yellow grains embedded in sample No.12 were shown to be PbSnO₃ by micro XRD, which was the first coloring material ever found in Korea. Lead isotope ratios of samples No.12 and No.17 which contained lead were measured by TIMS. The origin of lead was traced by means of multivariate analysis such as SLDA. The result showed that lead from southern China and southern Korea had been used for making glass.

        중앙아시아 벽화 보존처리(1) : 벽화(본4074, 본4096)의 상태조사

        국립중앙박물관은 용산 새 박물관의 전시 유물로 선정된 중앙아시아 벽화의 연차적인 보존처리를 위해 “독립행정법인 일본 도쿄문화재연구소’’와 공동연구를 모색하게 되었다. 2001년에 본4074, 본4096 작은 벽화편 2점의 보존처리를 시작하면서 벽화의 균열 박락 등 손상상태, 구조와 구성재료, 이전 보존처리에서 적용된 재료와 방법에 대한 기초조사를 실시하였다. 조사결과 본4074, 본4096 2점의 벽화는 흙과 지푸라기를 반죽하여 만든 벽체에 석고를 발라 바탕으로 하고 그 위에 채색을 한 것으로 나타났으며 벽체 속에 포함된 지푸라기의 방사선 탄소연대측정 결과 본 벽화는 10세기 말에서 13세기 초에 만들어진 것으로 나타났다. 또한 X-선회절 분석 결과 화면의 흰색 바탕은 gypsum[Ca(SO₄)·2H₂O]과 CaSO₄, Calcite(CaCO₃)가 적색계통은 연단(鉛丹:Pb₃O₄)과 led arsenate[Pb(As₂O₆)], 녹색계통은 Cuprite(Cu₂O)와 arsenolite(As₂O₃), arsenic oxide(As₂O₄) 등이 사용된 것으로 조사되었다. This article is about a joint project carried out by the National Museum of Korea and the Tokyo Cultural Properties Research Institute for the conservation of central Asia Wall painting that has been selected for the exhibition at the new Seoul National Museum of Korea at Yongsan. The investigation of the wall painting revealed very useful information. This includes the condition of the object, and the identification of evident damage, such as cracks, loss of pigment, plus materials and methods employed during the object's creation, as well as previous conservation treatment. The object was mainly made by applying plaster to the body (wall) that consisted of a mixture of soils and rice straws. Then, on the surface of the wall-painting, pigments were used to draw and to colour it. As a part of the investigation, radiocarbon dating was conducted using straw samples taken from the object. The result indicates that the object is probably dated form between the end of the 10th Century and the beginning of the 13th Century. The result of X-ray diffraction also revealed the composition of the pigments used on the surface. These are 1. gypsom[Ca(SO₄)·2H₂O], CaSO₄ and Calcite(CaCO₃) and Calcite(CaCO₃) that were used for the white background. 2. Pb₃O₄ and led Arsenate [Pb(As₂O₆) that were used for the red colouring. 3. Cuprite(Cu₂O), Arsenolite(As₂O₃) and Arsenic Oxide(As₂O₄) that were used for the green colouring.

      • 쌀의 취반 중 Phenthoate 농약 잔류분의 제거

        인위적으로 phenthoate 농약성분을 부착시킨 쌀의 취반(炊飯), 즉 수세 및 가열조리시 농약잔류분의 제거율을 실험한 결과는 다음과 같다. 가정에서의 세척방법에 준하여 3회 반복 수세한 쌀에서 phenthoate는 49%가 잔존하였으므로 수세에 의해 51%가 제거되었다. 수세과정에서는 첫번째 세척액(뜨물)에서 37.3%(여과액 7.8%, 잔사 29.5%), 두번째 세척액에서 14.3%(여과액 6.2%, 잔사 8.1%), 세번째 세척액에서 8.9%(여과액 5.8%, 잔사 3.1%)가 제거되었다. 세척액 중 phenthoate 잔류분은 뜨물 여과액보다 뜨물 잔사에서 더 많이 발견되었다. 전기밥솥에서 가열조리한 쌀밥에서 phenthoate 잔존율은 41%이었으므로 수세 및 가열조리에 의해 오염된 phenthoate의 59%가 제거되었다. Phenthoate 성분은 열에 안정해 취반시에도 분해되지 않는 것으로 생각된다. 결론적으로 쌀 중에 잔류하는 phenthoate 성분은 취반과정시 주로 수세과정에 의하여 제거되므로 가열조리전 수세과정을 반드시 3회 이상 거치는 것이 바람직하다. 본 연구결과는 쌀 중 phenthoate의 잔류 허용기준 설정시 감소계수(0.4)로 활용될 수 있을 것이다. This study was undertaken in order to elucidate the elimination of phenthoate residues by washing and cooking processes of rice which is the most important food crop in Korea. When contaminated rice was washed with distilled water three times, the removal rate of total phenthoate was 51%. The removal rate in the successive washings was 37.3%(wash filtrate 7.8%, wash sediment 29.5%)in the first, 14.3%(wash filtrate 6.2%, wash sediment 8.1%)in the second and 8.9%(wash filtrate 5.8%, wash sediment 3.1%)in the third washings. More than half of the residue was removed by the first washing and most residues were foumd in the sediment rather than in the filtrate of the rice washings. The residue rate of phenthoate after cooking by an electric rice cooker was 41%, indicating that the removal rate after cooking was 59%, because phenthoate is thermally stable at the cooking temperature. In conclusion, phenthoate residues contaminated in rice grains are greatly removed in the washing process and it is desirable to wash the grains before cooking in order to decrease the hazards from pesticide residues such as phenthoate. Reduction factor of phenthoate in rice cooking is proposed to be 0.4.

      • Ciprofloxacin의 돼지 호흡기 감염증 및 설사증에 대한 치료효과

        To evaluate the clinical efficacy of ciprofloxacin (CFX) on treatment of the pigs with respiratory diseases or diarrhea. CFX at various dosages were administered intramuscularly to the diseased pigs selected from the pig farms located in Chungnam province. One hundreds and fourteen heads of pigs with respiratory diseases were divided into five groups and CFX at 0.25, 0.5, 1.0 (= 2.5mg CFX/Kg B.W.) and 2.0 doses were administered i.m. for 5 days. The recovery rates were found 31.8%, 73.9%, 91.3% and 92.0%, respectively, being higher than that of the untreated control group (19.0%). The mortality of the treated groups were 0%, that is lower than that of the control (9.5%). Seven kinds of bacteria were isolated from the respiratory organs of 86 pigs, and 34 (39.5%) of 86 pigs showed the complicated infection. In analyzing by the pigs infected with a single pathogen as P. multocida; A. pleuropneumonia. Staphyloccus spp. or Mymplasma spp., the recovery rates were 0∼66.6% at 0.25 dose, 66.6∼100% at 0.5 dose and 100% in all of 4 pathogens at 1.0 to 2.0 doses. At 0.25 dosage the pigs infected with A. pleuropneumonia showed the lowest recovery rate. One hundreds and twenty two heads of pigs with diarrhea were divided into five groups and CFX at 0.25, 0.5, 1.0 (= 2.5mg CFX/Kg B.W.) and 2.0 doses were administered i.m. for 5 days. The recovery rates were 29.1 %, 40.0%, 80.0% and 88.0%, respectively. The mortality of the treated groups were 0∼8.3%, being much lower than that of the control. Four kinds of bacteria were isolated from the fecal specimens of 94 pigs, and 84 (89.0%) of 94 pigs showed the complicated infection.

      • Monocrotaline으로 유발시킨 흰쥐 폐고혈압에 대한 Flunarizine의 약리작용

        Monocrotaline treatment (60mg/kg, i.p.) in normotensive rats resulted the indications and related toxicities of pulmonary hypertension, including the increase of hematocrit value, right ventricle hypertropy, pulmonary edema and lethality, after 4 weeks later. However, daily administrtion of flunarizine (5mg/kg, p.o.) for 4 weeks seemed to decrease the above responxes, and also there was no significant change on the systemic mean arterial blood pressure and heart rate. In the anesthetized rats, hypotensive effect induced by isoproterenol bolus injection was signficantly inhibited in flunarizine-treated pulmonary hypertensive rats, comparing to the untreated rats, even if those by acetylcholine, adenosine and ATP were not. In the isolated pulmonary artery experiments, vasorelaxation induced by isoproterenol, but not acetylcholine, adenosine or ATP, were also selectively inhibited in flunarizine treated rats, comparing to the control rats. Also vasoconstriction induced by norepinephrine were increased in flunarizine treated rats comparing to the control rats, but those by KCI were not. However, pretreatment of flunarizine did not significantly affected on the vasodilation induced by acetylcholine, isoproterenol, adenosine or ATP and on the vasoconstriction induced by norepinephrine in isolated pulmonary artery of the normal rats. These results suggest that flunarizine will decrease the pathogenesis of pulmonary hypertension through the change of α- and β-adrenergic receptor activity in pulmonary artery due to the up-regulation of voltage dependent Ca++ channels.

      • 이공의 형태, 크기 및 위치에 관한 X-선학적 연구

        The purpose of this study was to evaluate the size, shape and position of mental foramen in intraoral radiographs. For this study, periapical radiographs of right and left premolar areas were obtained from the 117 adults with normal occlusion. And then the author evaluated visualization or failure, shape and size of the radiographic mental foramen, horizontal position and vertical position of mental foramen relative to the apex of the adjacent teeth. The results were as follows : 1. The mental foramen was visualized on the radiographs 77.4% of the samples. 2. Reasons for failure to visualize the radiographic mental foramen were undetectable(69.8%), multiple radiolucencies( 17.0%), masked by adjacent anatomical structures(13.2%) in descending order of frequency. 3. Shapes of mental foramen were observed elliptical(56.9%), round or oval(43.1%) in descending order of frequency. 4. The average radiographic size of the mental foramen was 4.7mm horizontally and 3.1mm vertically. 5. Horizontal position of mental foramen was observed at the 2nd premolar area(60.2%), the area between the 1st and 2nd premolar (33.1%),the area between the 2nd premolar and the 1st molar(3.9%), the 1st premolar area(2.2%), the area between the canine, the 1st premolar area(0.6%) in descending order of frequency. 6. Vertical position of mental foramen was most frequently observed at radiographic apex of the 2nd premolar(68.5%), at apex(23.8%), overlap with apex(6.1%), superior to apex( 1.7%) in descending order of frequency. 7. Vertical dimension between the mental foramen and the apex was obtained minimum 0.5mm, maximum 5.2mm and average 2.8mm.

        고등학생들의 시험불안과 대처전략

        Objects : The purpose of this study was to see the relationship between the level of test anxiety and coping strategy in high school students. Methods : The subjects were 952 high school students(411 boys and 541 girls) in Kwangju. A self reporting questionnaire, test anxiety scales and coping strategy scales were handed out to the students on April 1994. Result : 1) Test anxiety scores in gradeⅠ and Ⅱ were higher than those in grade Ⅲ. Test anxiety scores were correlated with difficulty in attention, depressive mood and suicidal ideas. 2) The high test anxiety group used more coping strategies than the low test anxiety group and coping strategies were varied among high test anxiety groups according to the grade. 3) Test anxiety correlated positively with coping strategy. 4) Predicting factors of test anxiety were grade, attention difficulty, depressive mood, suicidal idea, withdrawal or disengagement, life evert, internal suppression of emotion and distance. Conclusion : The above results suggest that test anxiety correlated positively with coping strategy and the high test anxiety group used much more coping strategies in order to overcome stress using their potential inner resource than the low test anxiety group.

      • 파라캇에 의한 대뇌 피질 손상 : 증례 보고

        Paraquat is widely used as a universal herbicide. Paraquat induces brain damage in case of fatal poisoning. It is well documented that paraquat exposure is associated with Parkinson's disease. However, many reports suggest paraquat is not selective for the dopaminergic nigro-strial system. Preferably, there are reports that cortical neurons were found to be more sensitive to paraquat. A39-year-old woman committed suicide by drinking 20 ml of a 20% solution of Paraquat. After 5 days, she had a convulsive seizure and lost consciousness. Brain MRI showed diffuse cerebral cortical edema of bilateral frontal and temporal lobes. She died 2 days after progressive pulmonary fibrosis. Here, we report a patient with paraquat induced cortical lesion. 파라캇은 세계적으로 널리 쓰이는 제초제이다. 파라캇은 중독되었을 경우 뇌손상을 일으키기도 한다. 파라캇과 파킨슨병과의 관련성은 비교적 잘 알려져 있다. 하지만, 파라캇이 도파민 줄무늬흑체 계통(dopaminergic nifro-striatal system)에 선택적이지 않다는 많은 보고들이 있다. 오히려, 대뇌피질이 파라캇 중독에 더욱 민감하다는 일부 보고들도 있다. 39세 여자환자가 자살할 목적으로 20% 농도의 파라캇 20 ml를 마시고 본원에 내원하였다. 환자는 입원 5일째 갑자기 전신 발작 증상과 함께 의식 저하 소견을 보였다. 다음날 시행한 뇌자기공명영상 촬영상 양쪽 측두엽과 전두엽에 대칭성 세포 도겅부종 소견을 보이고 있었다. 그녀는 2일 후 폐섬유화증에 의한 호흡부전으로 사망하였다. 저자들은 파라캇 중독 환자에서 대뇌 피질 손상을 확인한 1예를 경험하였기에 보고하는 바이다.
