      • A Context-Restricted Multi-Stage Korean Coreference Resolution Model

        Coreference resolution is a task of finding all word expressions that refer to the same entity in a text. Anaphors generally refer to the nearest preceding individuals with same attributes. So, position information is one of the important features in coreference resolution. In order to resolve Korean coreferences, multi-stage resolution model is developed by classifying coreference types, and then restricted the context of each processing stage differently according to its characteristics. The experimental results show that the proposed system improved 4.4 percentage point compared with the context-unrestricted system. This means that the context-restricted method plays a positive role in Korean coreference resolution.

        1,2-Dichloropropane (1,2-DCP)-Induced Angiogenesis in Dermatitis

        1,2-Dichloropropane (1,2-DCP) has been used as an industrial solvent and a chemical intermediate, as well as in soil fumigants. Human exposure may occur during its production and industrial use. The target organs of 1,2-DCP are the eyes, respiratory system, liver, kidneys, central nervous system, and skin. Repeated or prolonged contact may cause skin sensitization. In this study, 1,2-DCP was dissolved in corn oil at 0, 2.73, 5.75, and 8.75 mL/kg. The skin of mice treated with 1,2-DCP was investigated using western blotting, hematoxylin and eosin staining, and immunohistochemistry. 1,2-DCP was applied to the dorsal skin and both ears of C57BL/6J mice. The thickness of ears and the epidermis increased significantly following treatment, and the appearance of blood vessels was observed in the dorsal skin. Additionally, the expression of vascular endothelial growth factor, which is tightly associated with neovascularization, increased significantly. The levels of protein kinase-B (PKB), phosphorylated PKB, mammalian target of rapamycin (mTOR), and phosphorylated mTOR, all of which are key components of the phosphoinositide 3-kinase/PKB/mTOR signaling pathway, were also enhanced. Taken together, 1,2-DCP induced angiogenesis in dermatitis through the PI3K/PKB/mTOR pathway in the skin.

        MiR-339 attenuates LPS-induced intestinal epithelial cells inflammatory responses and apoptosis by targeting TLR4

        Background Intestinal epithelial cells are important for defending against pathogen infection. LPS is an endotoxin that is highly antigenic and cytotoxic produced by bacteria. LPS disrupts the intestine epithelium integrity and induced the intestinal epithelial cell infammation and apoptosis. Our previous study has predicted the function of exosome miRNAs through bioinformatics analysis, and we found that miR-339 had a potential function in cell infammation response. To our knowledge, no published paper has demonstrated the miR-339 function in protecting the intestine epithelium against bacterial infection. Objective The objective of this study is to evaluate the miR-339 function in regulating intestinal epithelial cells to defend against bacterial infection through biological experiments and bioinformatics analyses. Methods Through the miR-339 transfection experiment and TLR4 interfering experiment, we evaluated the function of miR339 and TLR4 in the process of infammatory responses and apoptosis. Through Bioinformatics analyses and dual-luciferase reporter experiment, we identifed the target gene of miR-339. Results miR-339 attenuates LPS-induced intestinal epithelial cells infammatory responses through the TLR4/NF-κB signaling pathway and inhibited LPS-induced apoptosis through the P53 signaling pathway. TLR4 is the target gene of miR-339. TLR4 reduced LPS-induced proinfammatory responses and apoptosis. Conclusions In conclusion, miR-339 protected the intestine epithelial cells from LPS-induced cell infammation and apoptosis through targeting TLR4. This study expanded our understanding of how miRNAs and genes work collaboratively in regulating intestinal epithelial cells to defend against bacterial infection.

        Ion-exchange coupled crystallization for the removal of calcium ions from dicyandiamide

        A new process for ion exchange coupled crystallization is introduced for the removal of calcium ions from dicyandiamide. The effects of different ion-exchange resins, temperature, reaction time, stirring rate, and treatment amount and resin dosage on the removal of calcium ions in dicyandiamide were studied. On this basis, the crystallization process of dicyandiamide was optimized by response surface methodology, together with respective investigations on the effects of cooling rate, stirring rate, seed grain size and seeding time on the removal of calcium ions in dicyandiamide. It was found that the removal efficiency of calcium ion could reach up to 98.12%, during the ion-exchange treatment, and the value increased then fell, with the rise of stirring rate and temperature; the efficiency would improve, with the accumulation of resin dosage; but it would diminish with the increase of treatment amount; and the value would first rise and then remain unchanged when the reaction time was extended. In addition, the best conditions for crystallization are also provided. When the cooling rate is at 0.3 oC/min, the stirring rate 300 rpm, the seed size 60 meshes, and the seeding time 30 minutes, seeds in uniform size with the content of Ca2+ pharmaceutically qualified would be obtained, under the optimum process conditions.

        <P>Physiological and morphological properties of the human intestine cannot be accurately mimicked in conventional culture devices such as well plates and petri dishes where intestinal epithelial cells form a monolayer with loose contacts among cells. Here, we report a novel microfluidic cell culture device (mu FCCD) that can be used to culture cells as a human intestinal model. This device enables intestinal epithelial cells (Caco-2) to grow three-dimensionally on a porous membrane coated with fibronectin between two polydimethylsiloxane (PDMS) layers. Within 3 days, Caco-2 cells cultured in the mu FCCD formed villi-and crypt-like structures with small intercellular spaces, while individual cells were tightly connected to one another through the expression of the tight junction protein occludin, and were covered with a secreted mucin, MUC-2. Caco-2 cells cultured in the mu FCCD for 3 days were less susceptible to bacterial attack than those cultured in transwell plates for 21 days. mu FCCD-cultured Caco-2 cells also displayed physiologically relevant absorption and paracellular transport properties. These results suggest that our intestinal model more accurately mimics the morphological and physiological properties of the intestine in vivo than the conventional transwell culture model.</P>

        HDAC inhibitors induce epithelial-mesenchymal transition in colon carcinoma cells.

        <P>The effects of histone deacetylase (HDAC) inhibitors on epithelial-mesenchymal transition (EMT) differ in various types of cancers. We investigated the EMT phenotype in four colon cancer cell lines when challenged with HDAC inhibitors trichostatin A (TSA) and valproic acid (VPA) with or without transforming growth factor-β1 (TGF-β1) treatment. Four colon cancer cell lines with different phenotypes in regards to tumorigenicity, microsatellite stability and DNA mutation were used. EMT phenotypes were assessed by the expression of E-cadherin and vimentin using western blot analysis, immunofluorescence, quantitative real-time RT-PCR following treatment with TSA (100 or 200 nM) or VPA (0.5 mM) with or without TGF-β1 (5 ng/ml) for 24 h. Biological EMT phenotypes were also evaluated by cell morphology, migration and invasion assays. TSA or VPA induced mesenchymal features in the colon carcinoma cells by a decrease in E-cadherin and an increase in vimentin expression at the mRNA and protein levels. Confocal microscopy revealed membranous attenuation or nuclear translocation of E-cadherin and enhanced expression of vimentin. These responses occurred after 6 h and increased until 24 h. Colon cancer cells changed from a round or rectangular shape to a spindle shape with increased migration and invasion ability following TSA or VPA treatment. The susceptibility to EMT changes induced by TSA or VPA was comparable in microsatellite stable (SW480 and HT29) and microsatellite unstable cells (DLD1 and HCT116). TSA or VPA induced a mesenchymal phenotype in the colon carcinoma cells and these effects were augmented in the presence of TGF-β1. HDAC inhibitors require careful caution before their application as new anticancer drugs for colon cancers.</P>

        한국어 ‘있다/없다’의 의미 확장 연구

        This study has a goal to analyse the direction of the meaning extension of complimental antonym words ‘iss-ta/eop-ta’. For the goal, this study applied on the cognitive theory. Complimental antonym words ‘iss-ta/eop-ta’ has five types of the meaning extension. The types of the meaning extension are as follows ① ‘object->space’ ②’object->behavior->space’, ③‘physical position-> social position-> psychological position’, ④‘content word->functional word’, ⑤‘concreteness->abstractness’. The aspects of the meaning extension are divided into two; one is symmetrical (as like as type①,②) and the other is asymmetrical (as like as type ③,④,⑤) The asymmetrical type of meaning extension made another imbalance of the productivity in the aspects of compounding lexical words, functional words and markedness. In the aspect of compounding words, ‘eop-ta’ is more productive than ‘iss-ta’, but in the side of functional words, ‘iss-ta’ is more productive. And in the aspect of markedness, ‘eop-ta’ is more productive than ‘iss-ta’, this means that the domain of the meaning of ‘iss-ta’ is wider than ‘eop-ta’.
