        Structural covariance of neostriatal and limbic regions in patients with obsessive-compulsive disorder.

        <P>Frontostriatal and frontoamygdalar connectivity alterations in patients with obsessive-compulsive disorder (OCD) have been typically described in functional neuroimaging studies. However, structural covariance, or volumetric correlations across distant brain regions, also provides network-level information. Altered structural covariance has been described in patients with different psychiatric disorders, including OCD, but to our knowledge, alterations within frontostriatal and frontoamygdalar circuits have not been explored.</P>

        Human Adipose-Derived Stromal Cells as a Feeder Layer to Improve Keratinocyte Expansion for Clinical Applications

        The aim of this work is to propose a keratinocytes (KC) culture method for clinical practice with irradiated adipose-derived mesenchymal stromal cells (ASCs) as human feeder layer, avoiding murine immortalized fibroblasts, commonly request for producing skin substitutes. ASCs were isolated, expanded, irradiated, and co-cultured with autologous or allogeneic KC. All experiments were performed using murine fibroblasts as control. Cell counts, flow cytometric analysis and ELISA were carried out, in order to define cell yield, viability and cytokine secretion. Results indicate that the optimal X-ray dose for ASCs is 120 Gy and the optimal seeding density is 625 cells/cm2; moreover, flow cytometric analysis shows that the percentage of feeder layer cells reaches values lower than 1%, within 8 days of co-culture. KC reach confluence in 6.9 days on ASCs substrate and, after confluence, the number of live cells increases again in a multilayered structure. Moreover, results show higher levels of interleukin (IL)-1α in co-culture with ASCs compared with 3T3, while no differences were observed for IL-6 and IL-8. Therefore, human ASCs enable to obtain effectively in vitro expanded KC and represent a viable alternative to murine fibroblasts for the production of clinical use skin substitutes.

        Cross-sectional Evaluation of the Sarcopenia Quality of Life (SarQoL) Questionnaire: Translation and Validation of its Psychometric Properties

        Background: The SarQoL, a quality-of-life questionnaire specific to sarcopenia, was developed in 2015 and has since been translated into a number of other languages. The main reason to introduce this new Ukrainian version of the questionnaire was to measure sarcopenic individuals’ perceptions regarding their positions in life in the context of their culture and value systems. Methods. The questionnaire was translated using a forward-backward approach with a pre-test. A total of 49 subjects were recruited for the validation study. Sarcopenia was diagnosed according to the Ishii test. The validation analyses included discriminative power, internal consistency, floor and ceiling effects, construct validity, and test-retest reliability. We compared the SarQoL questionnaire to the Short-Form 36 and the EuroQoL-5 Dimensions. Results: A total of 28 subjects out of 49 were categorized as probably sarcopenic. They had a significantly lower quality of life (overall score 58.43±17.13 vs. 69.89±13.31; p=0.014). The internal consistency was excellent (α=0.898), with none of the domains showing a disproportionate influence on the homogeneity of the questionnaire. Convergent construct validity was also confirmed. The results indicated a near-perfect degree of test-retest reliability. Conclusions: The first Ukrainian version of the questionnaire is equivalent to the available original English version. We developed a valid, reliable, and consistent new Ukrainian version of the SarQoL that may be used with confidence for clinical and research purposes.

        Intra-regional reform in East Asia and the new Hague Principles on Choice of Law in International Commercial Contracts

        지난 20년, 동아시아 역내 국가들은 국가 차원에서의 국제사법 개혁에 대해 점차 개방적인 모습을 보여 주었다. 한국은 2001년 시행된 『국제사법』의 개정을 통해 개혁의 선구자가 되었고, 일본은 2007년 발효된 『法の適用に?する通則法』을 통해 동일한 개혁의 발자취를 따랐다. 이후 중국도 상당한 진전을 보여주고 있다. 중국의 국제사법인 『涉外民事?系法律适用法』을 2011년 제정하였고, 최근 2013년 1월 7일부로 시행된 최고인민법원의 사법해석(1)에 의해 동법을 보충하였다. 이러한 발전의 관점에서, 학자들은 역내 및 국제적 관여를 촉진하기 위한 보다 많은 대화와 개혁을 요청해 왔다. 최근 한 학자는 동아시아 역내의 공통된 가치와 헤이그국제사법회의의 다양한 국제협약(international instruments)의 정신을 반영한 국제사법을 발전시켜야 함을 강조하였다. 이 통찰력 넘치는 견해는 헤이그국제사법회의가 동아시아 지역과 공유하는 오랫동안 지속된 유대관계를 반영하고 있다. 일본은 1957년 6월 헤이그국제사법회의의 회원이 되었고, 중국은 1987년 7월, 한국은 1997년 8월에 헤이그국제사법회의 의 회원이 되었다. 헤이그국제사법회의는 동아시아를 넘어 2012년 홍콩에 새로운 아태평양지역사무소를 개설하고 유라시아의 터키를 포함하여 싱가폴을 9번째 아시아의 헤이그국제사법회의 회원국으로 받아들일 준비를 함으로써 이 지역에서 그 존재감을 넓혀가고 있다. 아시아 국가들이 헤이그국제사법회에 많이 참여함으로써 아시아 지역의 필요에 보다 잘 대응하는 새로운 국제협약의 발전을 촉진시킬 것으로 기대한다. 이 논문은 그러한 국제협약의 하나로서 곧 승인될 예정인 새로운 국제상사계약의 준거법 결정에 관한 헤이그원칙(이하, “헤이그원칙”)을 검토한다. 헤이그원칙이 추구하는 바는 국제계약의 준거법 결정에 있어 당사자자치의 승인 및 제한과 관련한 현행 모범실무(best practice)의 국제적 규준(code)이 될 수 있도록 하는 데 있다. 이 논문은 헤이그원칙의 전개, 형식 및 범위와 헤이그원칙의 주석서에 대해 상설하고, 다방면의 모범실무 규정과 혁신적 규정을 소개한다. 또한 이 글은 한국, 중국 및 일본의 해당 국제사법 규정을 면밀히 검토한 후 헤이그원칙이 개별 국가 내지 동아시아 역내 차원의 개혁을 위한 유용한 도구가 될 수 있을 것인가에 대한 문제를 제기한다. The last two decades have seen East Asian States evince an increasing openness to private international law reform at the national level. The Republic of Korea was the harbinger in this regard with Law Number 6465 of the Act amending the Conflict of Laws Act taking effect in 2001. Japan followed suit with the Act on General Rules for Application of Laws taking effect in 2007. The People’s Republic of China has since made significant advances. China’s Law on the Application of Law to Foreign-related Civil Relations entered into force in 2011 and has recently been supplemented by the Supreme People’s Court Interpretation I on the Foreign-related Civil Relations Law, which entered into force on 7 January 2013. In light of these developments, commentators have called for further dialogue and reform with a view to facilitating increased intra-regional and international engagement. One commentator recently emphasised the importance of developing private international law rules reflecting common values in the region and doing so in parallel with various instruments of the Hague Conference on Private International Law. This prescient observation reflects the longstanding, enduring bond that the Hague Conference shares with the East Asian region. Japan became a Member of the Hague Conference in June 1957; China in July 1987 and the Republic of Korea in August 1997. Beyond East Asia, the Hague Conference has a growing presence in the Asia Pacific, highlighted by the opening of its new regional office in Hong Kong in 2012 and the recent membership of Singapore as the 9th Asian member of the Hague Conference (including Eurasian Turkey). It is hoped that an increased participation of Asian States in the Hague Conference will facilitate the development of new instruments that are better adapted to the needs of the region. This article will examine one such instrument, the new Hague Principles on Choice of Law in International Commercial Contracts (“the Hague Principles”), the endorsement of which is expected shortly. The Hague Principles seek to serve as an international code of current best practice with respect to the recognition and limits of party autonomy in choice of law for international contracts. The article will detail the development, form and scope of the Hague Principles and their accompanying Commentary before exploring various best practice provisions and innovative provisions. It will offer a high-level comparison with the conflict of laws rules applicable in the Republic of Korea, China and Japan and, in doing so, raise the question of whether the Hague Principles may be a useful instrument for reform at a national or regional level within East Asia.

        Potential for colonization of O111:H25 atypical enteropathogenic E. coli

        Using clonal phylogenetic methods, it has been demonstrated that O111:H25 atypical enteropathogenic E. coli (aEPEC) strains belong to distinct clones, suggesting the possibility that their ability to interact with different hosts and abiotic surfaces can vary from one clone to another. Accordingly, the ability of O111:H25 aEPEC strains derived from human, cat and dogs to adhere to epithelial cells has been investigated, along with their ability to interact with macrophages and to form biofilms on polystyrene, a polymer used to make biomedical devices. The results demonstrated that all the strains analyzed were able to adhere to, and to form pedestals on, epithelial cells, mechanisms used by E. coli to become strongly attached to the host. The strains also show a Localized-Adherence- Like (LAL) pattern of adhesion on HEp-2 cells, a behavior associated with acute infantile diarrhea. In addition, the O111:H25 aEPEC strains derived either from human or domestic animals were able to form long filaments, a phenomenon used by some bacteria to avoid phagocytosis. O111:H25 aEPEC strains were also encountered inside vacuoles, a characteristic described for several bacterial strains as a way of protecting themselves against the environment. They were also able to induce TNF-α release via two routes, one dependent on TLR-4 and the other dependent on binding of Type I fimbriae. These O111:H25 strains were also able to form biofilms on polystyrene. In summary the results suggest that, regardless of their source (i.e. linked to human origin or otherwise), O111:H25 aEPEC strains carry the potential to cause human disease.

        Heart Rate Variability and Somatization in Adolescents With Irritable Bowel Syndrome

        Background/AimsChanges in autonomic regulation and psychological distress play an important role in the pathobiology of irritable bowel syndrome (IBS). The aim of the current study is to evaluate the autonomic function and to link it to the levels of somatization in adolescents with IBS. MethodsWe enrolled 30 adolescents with various types of IBS and 35 healthy controls. Time and frequency domain indexes of heart rate variability (HRV) were measured in supine (baseline) and standing (orthostasis) positions using short-term electrocardiographic recordings. The somatic symptoms index was assessed with the modified Screening for Somatoform Symptoms questionnaire. ResultsAdolescents with IBS showed no differences of HRV parameters in the supine position compared to healthy control. In orthostasis, a decrease in the standard deviation of normal RR intervals as well as main spectral index total power (TP) were observed. The reduction of TP was attributed to the reduced activities of the high- and low frequency components. Increased somatic symptoms index in IBS patients negatively correlated with TP in orthostasis (r = –0.485, P = 0.007). A subgroup analysis revealed that adolescents with IBS with TP values either < 2500 msec2 or > 5500 msec2 in the supine position demonstrated significantly reduced activity of the low frequency component. ConclusionsAdolescents with IBS showed signs of autonomic dysfunction only during the orthostatic test, which were associated with increased somatization scores. Further research is needed to establish the links between emotional wellbeing and autonomic function in this population.

        Identification of IRF8, TMEM39A, and IKZF3-ZPBP2 as Susceptibility Loci for Systemic Lupus Erythematosus in a Large-Scale Multiracial Replication Study

        Systemic lupus erythematosus (SLE) is a chronic heterogeneous autoimmune disorder characterized by the loss of tolerance to self-antigens and dysregulated interferon responses. The etiology of SLE is complex, involving both heritable and environmental factors. Candidate-gene studies and genome-wide association (GWA) scans have been successful in identifying new loci that contribute to disease susceptibility; however, much of the heritable risk has yet to be identified. In this study, we sought to replicate 1,580 variants showing suggestive association with SLE in a previously published GWA scan of European Americans; we tested a multiethnic population consisting of 7,998 SLE cases and 7,492 controls of European, African American, Asian, Hispanic, Gullah, and Amerindian ancestry to find association with the disease. Several genes relevant to immunological pathways showed association with SLE. Three loci exceeded the genome-wide significance threshold: interferon regulatory factor 8 (IRF8; rs11644034; p<SUB>meta-Euro</SUB> = 2.08 x 10<SUP>-10</SUP>), transmembrane protein 39A (TMEM39A; rs1132200; p<SUB>meta-all</SUB> = 8.62 x 10<SUP>-9</SUP>), and 17q21 (rs1453560; p<SUB>meta-all</SUB> = 3.48 x 10<SUP>-10</SUP>) between IKAROS family of zinc finger 3 (AIOLOS; IKZF3) and zona pellucida binding protein 2 (ZPBP2). Fine mapping, resequencing, imputation, and haplotype analysis of IRF8 indicated that three independent effects tagged by rs8046526, rs450443, and rs4843869, respectively, were required for risk in individuals of European ancestry. Eleven additional replicated effects (5 x 10<SUP>-8</SUP> < p<SUB>meta-Euro</SUB> < 9.99 x 10<SUP>-5</SUP>) were observed with CFHR1, CADM2, LOC730109/IL12A, LPP, LOC63920, SLU7, ADAMTSL1, C10orf64, OR8D4, FAM19A2, and STXBP6. The results of this study increase the number of confirmed SLE risk loci and identify others warranting further investigation.

        Novel biocidal N-halamine plastic based on poly(vinyl chloride): Preparation and characteristics

        Preparation method of new biocidal plastics based on plasticized PVC has been elaborated. Thederivative of poly(4-vinylbenzyl chloride) containing N-halamine substituents has been mixed with PVCgranules and processed by dry blend extrusion and injection moulding. The properties of obtainedmaterial have been tested using SEM/EDX and thermogravimetry. The biocidal efficacy of modified PVCresin has been tested against two strains of bacteria: Escherichia coli (Gram-negative) and Staphylococcusaureus (Gram-positive). It was found that even small amount of biocidal component yields to efficient biocidal activity of thewhole plastic. Chemical composition and other properties of PVC remain almost unchanged
