      • Silica coated gold nanorods for imaging and photo-thermal therapy of cancer cells.

Mallick, Sudipta,Sun, In-Cheol,Kim, Kwangmeyung,Yil, Dong Kee American Scientific Publishers 2013 Journal of Nanoscience and Nanotechnology Vol.13 No.5

<P>Due to their efficient conversion of absorbed light energy to heat gold nanorods have been proved to be an amazing tool for minimally invasive photo-thermal cancer therapy. The present in vitro study demonstrates the ability of silica coated Au nanorods to function as a dual probe for cancer-cell therapy and imaging without any toxic side-effects. HeLa cells were incubated with silica coated Au nanorods and imaged inside the cell just after 1 hour of incubation by a dark field set up due to strong surface enhanced Raman scattering. To induce hyperthermia, silica coated Au nanorod incubated HeLa cells were illuminated with a diode laser (671 nm, 200 mW, 10 min). Cell destruction was observed even at a very low dose of nanorods, whereas none was observed in the absence of nanorods. Silica coated Au nanorods thus offer a promising, novel class of selective photo-thermal agents for cancer therapy and diagnosis.</P>

      • Radiation Induced Cystitis and Proctitis - Prediction, Assessment and Management

Mallick, Supriya,Madan, Renu,Julka, Pramod K,Rath, Goura K Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.14

Cystitis and proctitis are defined as inflammation of bladder and rectum respectively. Haemorrhagic cystitis is the most severe clinical manifestation of radiation and chemical cystitis. Radiation proctitis and cystitis are major complications following radiotherapy. Prevention of radiation-induced haemorrhagic cystitis has been investigated using various oral agents with minimal benefit. Bladder irrigation remains the most frequently adopted modality followed by intra-vesical instillation of alum or formalin. In intractable cases, surgical intervention is required in the form of diversion ureterostomy or cystectomy. Proctitis is more common in even low dose ranges but is self-limiting and improves on treatment interruption. However, treatment of radiation proctitis is broadly non-invasive or invasive. Non-invasive treatment consists of non-steroid anti-inflammatory drugs (NSAIDs), anti-oxidants, sucralfate, short chain fatty acids and hyperbaric oxygen. Invasive treatment consists of ablative procedures like formalin application, endoscopic YAG laser coagulation or argon plasma coagulation and surgery as a last resort.

      • Quality assessment and forecast sensitivity of global remote sensing observations

        Mallick, S.,Dutta, D.,Min, K. H. Springer Science + Business Media 2017 Advances in atmospheric sciences Vol.34 No.3

<P>The satellite-derived wind from cloud and moisture features of geostationary satellites is an important data source for numerical weather prediction (NWP) models. These datasets and global positioning system radio occultation (GPSRO) satellite radiances are assimilated in the four-dimensional variational atmospheric data assimilation system of the UKMO Unified Model in India. This study focuses on the importance of these data in the NWP system and their impact on short-term 24-h forecasts. The quality of the wind observations is compared to the short-range forecast from the model background. The observation increments (observation minus background) are computed as the satellite-derived wind minus the model forecast with a 6-h lead time. The results show the model background has a large easterly wind component compared to satellite observations. The importance of each observation in the analysis is studied using an adjoint-based forecast sensitivity to observation method. The results show that at least around 50% of all types of satellite observations are beneficial. In terms of individual contribution, METEOSAT-7 shows a higher percentage of impact (nearly 50%), as compared to GEOS, MTSAT-2 and METEOSAT-10, all of which have a less than 25% impact. In addition, the impact of GPSRO, infrared atmospheric sounding interferometer (IASI) and atmospheric infrared sounder (AIRS) data is calculated. The GPSRO observations have beneficial impacts up to 50 km. Over the Southern Hemisphere, the high spectral radiances from IASI and AIRS show a greater impact than over the Northern Hemisphere. The results in this study can be used for further improvements in the use of new and existing satellite observations.</P>

Handling Of Sensitive Data With The Use Of 3G In Vehicular Ad-Hoc Networks

Mallick, Manish,Shakya, Subarna,Shrestha, Surendra,Shrestha, Bhanu,Cho, Seongsoo The Institute of Internet 2016 International Journal of Internet, Broadcasting an Vol.8 No.2

Data delivery is very challenging in VANETs because of its unique characteristics, such as fast topology change, frequent disruptions, and rare contact opportunities. This paper tries to explore the scope of 3G-assisted data delivery in a VANET within a budget constraint of 3G traffic. It is started from the simple S_Random (Srand) and finally reached the 3GSDD, i.e., the proposed algorithm. The performance evaluation of different algorithms is done through the two metrics delivery ratio and average delay. A third function utility is created to reflect the above two metrics and is used to find out the best algorithm. A packet can either be delivered via multihop transmissions in the VANET or via 3G. The main challenge is to decide which set of packets should be selected for 3G transmissions and when to deliver them via 3G. The aim is to select and send those packets through 3G that are most sensitive and requiring immediate attention. Through appropriate communication mechanism, these sensitive information are delivered via VANET for 3G transmissions. This way the sensitive information which could not be transmitted through normal VANET will certainly find its destination through 3G transmission unconditionally and with top priority. The delivery ratio of the packets can also be maximized by this system.

      • A Best Effort Classification Model For Sars-Cov-2 Carriers Using Random Forest

Mallick, Shrabani,Verma, Ashish Kumar,Kushwaha, Dharmender Singh International Journal of Computer ScienceNetwork S 2021 International journal of computer science and netw Vol.21 No.1

The whole world now is dealing with Coronavirus, and it has turned to be one of the most widespread and long-lived pandemics of our times. Reports reveal that the infectious disease has taken toll of the almost 80% of the world's population. Amidst a lot of research going on with regards to the prediction on growth and transmission through Symptomatic carriers of the virus, it can't be ignored that pre-symptomatic and asymptomatic carriers also play a crucial role in spreading the reach of the virus. Classification Algorithm has been widely used to classify different types of COVID-19 carriers ranging from simple feature-based classification to Convolutional Neural Networks (CNNs). This research paper aims to present a novel technique using a Random Forest Machine learning algorithm with hyper-parameter tuning to classify different types COVID-19-carriers such that these carriers can be accurately characterized and hence dealt timely to contain the spread of the virus. The main idea for selecting Random Forest is that it works on the powerful concept of "the wisdom of crowd" which produces ensemble prediction. The results are quite convincing and the model records an accuracy score of 99.72 %. The results have been compared with the same dataset being subjected to K-Nearest Neighbour, logistic regression, support vector machine (SVM), and Decision Tree algorithms where the accuracy score has been recorded as 78.58%, 70.11%, 70.385,99% respectively, thus establishing the concreteness and suitability of our approach.

      • Liposomes containing cholesterol and mitochondria-penetrating peptide (MPP) for targeted delivery of antimycin A to A549 cells

        Mallick, Sudipta,Thuy, Le Thi,Lee, Seulgi,Park, Jong-II,Choi, Joon Sig Elsevier 2018 Colloids and surfaces. B, Biointerfaces Vol.161 No.-

<P><B>Abstract</B></P> <P> <UL> <LI> Mitochondria are exclusively employed to produce energy required for the vital metabolic functions of the cell. However, mitochondria also play a key role in mammalian cell death. Dissipation in the mitochondria membrane potential causes cell death. Therefore, in cancer therapy, mitochondria are a novel target. Herein, we developed a nano-formulation of Antimycin A specifically targeted towards mitochondria and lung cancer; A549 cell. The liposomes were prepared using cholesterol and a mitochondria-penetrating peptide (MPP) having a phenylalanine-arginine-phenylalanine-lysine (FRFK) peptide sequence. The FRFK peptide was synthesized using solid phase peptide synthesis (SPPS) and contained cholesterol in the N-terminal end of the phenylalanine (Chol-FRFK). The synthesized material was confirmed using 1H NMR, Fourier transform infrared spectroscopy (FT-IR) and matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI TOF/MS). 1,2-Dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE) and Chol-FRFK, at a molar ratio of 1:1, were used for liposomal (Chol-FRFK/D) formulations; the sizes of the liposomes were confirmed using dynamic light scattering (DLS). Cytotoxicity was evaluated in A549 cells. Cellular uptake and mitochondria targeting were confirmed by flow cytometry and confocal microscopy, respectively. Antimycin A, a hydrophobic and mitochondrial electron transporter inhibitor was encapsulated into the Chol-FRFK/D liposomes. Our results indicate that Chol-FRFK/D liposomes may potentially be used for the nano-formulation of cytotoxic drugs and enhancing their bioavailability in cancer therapy. </LI> </UL> </P> <P><B>Highlights</B></P> <P> <UL> <LI> Idea of mitochondria targeting carrier to deliver mitochondria-specific drug, is proposed. </LI> <LI> Chol-FRFK was synthesized as a liposomal component. </LI> <LI> Chol-FRFK/D liposomes were prepared for delivery of Antimycin A to the mitochondria. </LI> <LI> Nano-formulation of Antimycin A showed higher toxicity in A549 cells. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Effect of Neem (Azadirachta indica) oil on the progressive growth of a spontaneous T cell lymphoma

Mallick, Sanjaya Kumar,Gupta, Vivekanand,Singh, Mahendra Pal,Vishvakarma, Naveen Kumar,Singh, Nisha,Singh, Sukh Mahendra Kyung Hee Oriental Medicine Research Center 2008 Oriental pharmacy and experimental medicine Vol.7 No.5

The present study was undertaken to investigate the effect of in vivo administration of neem oil intra-peritoneally (i.p.) to mice bearing a progressively growing transplantable T cell lymphoma of spontaneous origin, designated as Daltons lymphoma (DL), on the tumor growth. Mice were administered various doses of neem oil mixed in groundnut oil, which was used as a diluting vehicle or for administration to control DL-bearing mice. Administration of neem oil resulted in an acceleration of tumor growth along with a reduction in the survival time of the tumor-bearing host. Neem oil administered DL-bearing mice showed an augmented apoptosis in splenocytes, bone marrow cells and thymocytes along with an inhibition in the anti-tumor functions of tumor-associated macrophages. Thus this study gives an altogether a novel information that neem oil instead of the popular belief of being anti-tumor and immunoaugmentary may in some tumor-bearing conditions, behave in an opposite way leading to an accelarated tumor progression along with a collapse of the host's anti-tumor machinery. These observations will thus have long lasting clinical significance, suggesting caution in use of neem oil for treatment of cancer.

Liposomes: Versatile and Biocompatible Nanovesicles for Efficient Biomolecules Delivery

Mallick, S.,Choi, J.S. American Scientific Publishers 2014 Journal of nanoscience and nanotechnology Vol.14 No.1

Since the revolutionary discovery that phospholipids can form closed bilayered structures in aqueous systems, liposomes have become a very interesting topic of research. Because of their versatility and amazing biocompatibility, the use of liposomes has been widely accepted in many scientific disciplines. Their applications, especially in medicine, have yielded breakthroughs with anticancer-drug carriers over the past few decades. Specifically, their easy preparation and various structural aspects have given rise to a broadly usable way to internalize biomolecules such as drugs, DNA, RNA and even imaging probes. This review article reports recent developments in liposomal drug delivery and gene delivery, and thoroughly covers the synthesis and different kinds of liposomal surface modification techniques that have resulted in higher stability and efficiency with respect to the use of liposomes in tumor cell targeting, site-specific release, and extending blood retention times.
