        HMGB1 regulates autophagy through increasing transcriptional activities of JNK and ERK in human myeloid Leukemia cells

        HMGB1 is associated with human cancers and is an activator of autophagy which mediates chemotherapy resistance. We here show that the mRNA levels of HMGB1 are high in leukemia cells and it is involved in the progression of childhood chronic myeloid leukemia (CML). HMGB1 decreases the sensitivity of human myeloid leukemia cells K562 to anti-cancer drug induced death through up-regulating the autophagy pathway, which is confirmed by the observation with an increase in fusion of autophagosomes and autophagolysosomes. When overexpressing HMGB1, both mRNA levels of Beclin-1, VSP34 and UVRAG which are key genes involved in mammalian autophagy and protein levels of p-Bcl-2 and LC3-II are increased. Luciferase assays document that over-expression of HMGB1 increases the transcriptional activity of JNK and ERK, which may be silenced by siRNA. The results suggest that HMGB1 regulates JNK and ERK required for autophagy, which provides a potential drug target for therapeutic interventions in childhood CML. [BMB reports 2011; 44(9): 601-606]

      • 중국 유학생들의 BMI에 따른 식습관, 운동습관 및 체형인식에 관한 연구

        본 연구는 한국으로 유학을 온 중국 대학생들을 대상으로 체질량지수에 따라 식습관과 운동습관을 비교 분석하고, 저체중집단, 과체중집단 및 비만집단에 영향을 미치는 요인을 파악하여 건강생활의 습관을 개선하고 바람직한 체형을 유지하기 위하여 실시하였다. 본 연구에 참여한 중국 유학생 417명을 무작위로 선정하여 설문지를 이용하여 중국 유학생들의 식습관, 운동습관을 체질량 지수로 분류하여 체질량지수별 차이를 비교분석한 결론은 다음과 같다. 1. 중국 유학생의 식습관 요인에서 체질량지수가 높은 대학생들은 식사 속도가 빨랐고 식사량도 많았다. 그리고 간식, 외식, 청량음료, 고기나 기름 등을 자주 먹는다고 응답하였다. 남자유학생의 비만집단과 여자유학생의 과체중집단과 비만 집단이 체중조절을 위해서 식사량을 줄이는 것으로 나타났다. 2. 중국 유학생 운동습관 요인은 ``매일 30분 이상 운동 (축구, 야구, 태권도 등)을 하거나 친구와 논다``, ``일주일에 3회 이상 운동을 한다``에서는 남자유학생의 저체중집단 보다 비만 집단에서 높게 조사되었다. 남자유학생의 비만집단은 가까운 거리라도 걸어가지 않고 차를 타는 경향을 나타났다. 체질량지수가 높은 남·여 유학생들은 체중조절을 위해서 운동을 하였다. 3. 외모 및 체형의 인식 요인에서 중국 유학생의 체질량지수가 낮은 대학생들이 현재 외모에 만족도가 높고 체중과 체질량지수에 대해 남·여 학생들의 비만집단에서 관심이 높은 것으로 조사되었으며, 체중 조절에 대한 노력은 체질량지수가 높은 중국 여자유학생들이 더 적극적이었다. This study was to investigate the eating habits, exercise habits and body perception according to body mass index among Chinese international students in Korea. Total of 417 Chinese students were participated. Data were collected form March through May in 2011, by questionnaire, and analyzed by the SPSS 17.0. 1. For Chinese students, the results for imperative factor of eating habit showed that peoples with higher BMI always eating food more and always in a quick speed, in addition, it was responded that some snack, soft drinks, and meat as well as the oily food were taken usually. In order to adjust the body weight of both obese male group and overweight female group students, decreasing the food intake was a efficient way. 2. It was investigated that the factors of Chinese students` exercise habits are "Doing 30 minutes or more exercise everyday (football, baseball, taekwondo etc.) or playing with friends". In the survey for the students who exercise more than 3 times per week, the male students in obesity exercise more than the students in underweight group. Moreover, the male obese group had a tendency to rid a car instead of walking even in a short distance. In order to control the weight, the male/female students who have the high BMI should take the exercise. 3. In the survey for the factors of recognition the appearance and shape, Chinese students with lower BMI have higher satisfaction with their current appearance, moreover, it was surveyed that the male and female students in the obese group have the higher interest regarding to the body weight and BMI. The Chinese women student who has high BMI is much more proactive to control the body weight.

        Anti-Obesity and Hypolipidemic Effects of Ethanolic Extract from Alpinia officinarum Hance (Zingiberaceae) in Rats Fed High-Fat Diet

        To investigate the anti-obesity and hypolipidemic effects of Alpinia officinarum ethanolic extract (AOE) for the first time, anti-obesity models in vivo were used. Ten male Sprague-Dawley rats were fed normal control diet (NC); other groups of rats were fed a high-fat diet (HFD) with or without different proportions of AOE (AOE-1, 3%; AOE-2, 5%) for 6 weeks to examine feed intake, body and adipose tissue weight, serum total cholesterol (Total-C), triacylglycerol (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and leptin levels, alanine aminotransferase (ALT) and aspartate aminotranferase activities, hepatic Total-C and TG levels, and the pathological changes in liver and epididymal adipose tissues. Interestingly, feed intakes among the experimental groups were not significantly different. Body weight gains were significantly lowered in the AOE-1 and AOE-2 groups compared with the HFD group (P<.05) and near to the level of the NC group. AOE also improved the lipid profile in serum and the pathological changes in liver and adipose tissue and decreased the relative weights of epididymal and perirenal white adipose tissues. They improved lipid profile by lowering serum Total-C, TG, and LDL-C concentrations, leptin content, and the atherogenic index compared with the HFD group. The HDL-C concentration and the ratio of HDL-C/Total-C significantly increased compared with those of the HFD group. The serum ALT activity of the AOE-2 group was notably lower than that of the HFD group. Our data suggest that AOE can be considered as an anti-obesity agent that is effective for suppressing body weight gain and decreasing lipid profile.

        Quality Assessment of Curcuma longa L. by Gas Chromatography-Mass Spectrometry Fingerprint, Principle Components Analysis and Hierarchical Clustering Analysis

        Gas Chromatography-Mass Spectrometry (GC-MS) fingerprint analysis, Principle Components Analysis (PCA),and Hierarchical Cluster Analysis (HCA) were introduced for quality assessment of Curcuma longa L. (C. longa).The GC-MS fingerprint method was developed and validated by analyzing 33 batches of samples of C. longa from different geographic locations. 18 chromatographic peaks were selected as characteristic peaks and their relative peak areas (RPA) were calculated for quantitative expression. Two principal components (PCs) were extracted by PCA. C. longa collected from Guizhou and Fujian were separated from other samples by PC1, capturing 71.83% of variance. While, PC2 contributed for their further separation, capturing 11.13% of variance. HCA confirmed the result of PCA analysis. Therefore, GC-MS fingerprint study with chemometric techniques provides a very flexible and reliable method for quality assessment of C. longa.

        Sublethal effects of abamectin on the development, fecundity, and wing morphs of the brown planthopper Nilaparvata lugens

        The brown planthopper Nilaparvata lugens negatively affects rice yield by sucking nutrients from the rice stem and spreading viruses. In this study, the effects of sublethal concentrations of abamectin on development, fecundity, and wing morphs of N. lugens were investigated. Treatment with sublethal concentrations of abamectin showed no significant effects on the developmental period of F 0 nymphs. Although LC 10 and LC 25 treatments resulted in 10% and 25% mortality, the LC 25 treatment significantly prolonged the lifespan of macropterous and brachypterous females. Abamectin-treated brachypterous females showed significantly lower fecundity than control females. Furthermore, abamectin treatment showed no significant effects on the developmental period of F 1 nymphs. However, LC 25 treatment significantly inhibited the fecundity of brachypterous F 1 females and significantly reduced the proportion of macropterous F 1 females and brachypterous F 1 males. LC 25 treatment showed no significant effects on the fecundity of macropterous F 1 females. Considering the proportion of macropterous and brachypterous F 1 females, the relative fitness of the macropterous F 1 nymphs in the control, LC 10 , and LC 25 treatment groups was 1, 1.02, and 0.84, respectively, and that of brachypterous F 1 nymphs was 1, 0.79, and 0.93, respectively. Sublethal concentrations of abamectin inhibited N. lugens emergence. The present findings indicate the potential of abamectin for N. lugens field control.

      • Genistein Reinforces the Inhibitory Effect of Cisplatin on Liver Cancer Recurrence and Metastasis after Curative Hepatectomy

        Background: The high recurrence rate after hepatic resection in hepatocellular carcinoma (HCC) is a major obstacle to improving prognosis. The objective of the present study was to explore the function of genistein, a soy-derived isoflavone, in enhancing the inhibitory effect of cisplatin on HCC cell proliferation and on tumor recurrence and metastasis in nude mice after curative hepatectomy. Methods: Proliferation of human HCC cells (HCCLM3) was detected by 3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT) assay. Synergistic effects of genistein and cisplatin were evaluated with the median-effect formula. Nude mice bearing human HCC xenografts underwent tumour resection (hepatectomy) 10 days post implantation, then received intraperitoneal administration of genistein or cisplatin alone or the combination of the two drugs. 33 days after surgery, recurrent tumours and pulmonary metastasis were evaluated individually. MMP-2 level in recurrent tumours was detected by immunohistochemistry and real-time PCR; MMP-2 expression in HCCLM3 was detected by immunocytochemistry. Results: Genistein and cisplatin both suppressed the growth and proliferation of HCCLM3 cells. The two drugs exhibited synergistic effects even at relatively low concentrations. In vivo, mice in the combined genistein and cisplatin group had a smaller volume of liver recurrent tumors and fewer pulmonary metastatic foci compared with single drug treated groups. Cisplatin upregulated the expression of MMP-2 in both recurrent tumours and HCCLM3, while genistein abolished cisplatin-induced MMP-2 expression. Conclusions: Genistein reinforced the inhibitory effect of cisplatin on HCC cell proliferation and tumour recurrence and metastasis after curative hepatectomy in nude mice, possibly through mitigation of cisplatin-induced MMP-2 upregulation.

        <P>We present high-resolution optical tomographic images of human red blood cells (RBC) parasitized by malaria-inducing Plasmodium falciparum (Pf)-RBCs. Three-dimensional (3-D) refractive index (RI) tomograms are reconstructed by recourse to a diffraction algorithm from multiple two-dimensional holograms with various angles of illumination. These 3-D RI tomograms of Pf-RBCs show cellular and subcellular structures of host RBCs and invaded parasites in fine detail. Full asexual intraerythrocytic stages of parasite maturation (ring to trophozoite to schizont stages) are then systematically investigated using optical diffraction tomography algorithms. These analyses provide quantitative information on the structural and chemical characteristics of individual host Pf-RBCs, parasitophorous vacuole, and cytoplasm. The in situ structural evolution and chemical characteristics of subcellular hemozoin crystals are also elucidated.</P>
