      • Fluorescence Enhancement of a Microbial Rhodopsin via Electronic Reprogramming

        <P>The engineering of microbial rhodopsins with enhanced fluorescence is of great importance in the expanding field of optogenetics. Here we report the discovery of two mutants (W76S/Y179F and L83Q) of a sensory rhodopsin from the cyanobacterium <I>Anabaena</I> PCC7120 with opposite fluorescence behavior. In fact, while W76S/Y179F displays, with respect to the wild-type protein, a nearly 10-fold increase in red-light emission, the second is not emissive. Thus, the W76S/Y179F, L83Q pair offers an unprecedented opportunity for the investigation of fluorescence enhancement in microbial rhodopsins, which is pursued by combining transient absorption spectroscopy and multiconfigurational quantum chemistry. The results of such an investigation point to an isomerization-blocking electronic effect as the direct cause of instantaneous (subpicosecond) fluorescence enhancement.</P> [FIG OMISSION]</BR>

        Estudiamos en este trabajo la sintaxix del verbo haber como existencial. Para ello, nos basamos tanto en los ejemplos en los que este verbo concuerda con su complemento nominal como en los que no existe tal concordanci´a (los normativos). Discutimos la hipo´tesis de que este verbo sea un verbo inacusativo y la rechazamos segu´n criterios sinta´cticos como la posicio´n del argumento. Planteamos despune´s la hipo´tesis de que este verbo impersonal existencial en realidad proyecta dos argumentos, uno LOCATIVO y otro nominal. Aunque ha habido autores que han defendido el CASO PARTITIVO para este complemento, nosotros creemos que es ACUSATIVO, incluso en las ocasiones en que se produce su concordancia con el verbo, ya que se producen en espan~ol casos simiares, como con el verbo ser en oraciones como son las dos. Por u´ltimo, intentamos extender nuestra hipo´tesis a todos los verbos impersonales del espan~ol, y en esta clase incluimos dos subcategori´as, los impersonales le´xicos, como haber, estar, ser, existir, etc, y los impersinales sinta´cticos estructuras que se convierten en impersonales so´lo si aparece un argumento espacio-temporal. Aunque parece que no es necesario en todos los casos que el sintagma espacio-temporal este´ en una posicio´n argumental, si´ es necesario que estev presinte o, al menos, sea interpretable contextualmente.

        Is there a characteristic pattern of ambulatory blood pressure profile in type 1 diabetic children and adolescents?

        Purpose: To examine the characteristics of ambulatory blood pressure (ABP) including blood pressure variability (BPV) and its association with albuminuria in type 1 diabetic (T1D) children and to identify potential predictors of high-normal albuminuria and microalbuminuria. Methods: ABP monitoring was performed in 201 T1D children and adolescents (mean age, 14.7±3.8 years) with T1D duration over 1 year. The level of albuminuria was assessed as the albumin/creatinine ratio (ACR) and patients were further classified as low-normal, high-normal or microalbuminuria. Results: Fifteen T1D children (7.5%) were hypertensive using office blood pressure (BP) and 10 (5%) according to ABP. T1D subjects had elevated 24-hour systolic BP (SBP) and diastolic BP (DBP) (+0.2 and + 0.3 standard deviation score [SDS]) and nighttime SBP and DBP (+0.6 and +0.8 SDS) compared to reference values. Patients with microalbuminuria had significantly higher 24-hour, daytime and nighttime DBP compared to normoalbuminuric subjects. There was a high percentage of nondippers (74.1%). Nighttime diastolic BPV was significantly higher in subjects with high-normal compared to low-normal albuminuria (P=0.01). A weak correlation was found between ACR and daytime DBP SDS (r=0.29, P<0.001 and nighttime DBP SDS (r=0.21, P=0.003). Age and nighttime diastolic BPV were predictors of high-normal albuminuria while nighttime DBP was a strong predictor for microalbuminuria. Conclusion: T1D children have impaired BP regulation although most of them do not fulfill the criteria for sustained hypertension. There is an association between diastolic ABP and diastolic BPV with rising levels of albuminuria pointing to a clear connection between BP and incipient diabetic nephropathy.

        Pre- and post-operative cardiac evaluation of dogs undergoing lobectomy and pneumonectomy

        This study aimed to assess the influence of lobectomy and pneumonectomy on cardiac rhythm and on the dimensions and function of the right-side of the heart. Twelve dogs undergoing lobectomy and eight dogs undergoing pneumonectomy were evaluated preoperatively and one month postoperatively with electrocardiography and Doppler echocardiography at rest. Pulmonary artery systolic pressure (PASP) was estimated by the tricuspid regurgitation jet (TRJ) via the pulse wave Doppler velocity method. Systemic inflammatory response syndrome criteria (SIRS) were also evaluated based on the clinical and hematological findings in response to lobectomy and pneumonectomy. Following lobectomy and pneumonectomy, we predominantly detected atrial fibrillation and varying degrees of atrioventricular block (AVB). Dogs that died within seven days of the lobectomy (n = 2) or pneumonectomy (n = 1) had complete AVB. Preoperative right atrial, right ventricular, and pulmonary artery dimensions increased gradually during the 30 days (p<0.05) following pneumonectomy, but did not undergo significant changes during that same period after lobectomy. Mean PASP was 56.0 ± 4.5 mmHg in dogs having significant TRJ after pneumonectomy. Pneumonectomy, but not lobectomy, could lead to increases (p<0.01) in the SIRS score within the first day post-surgery. In brief, it is important to conduct pre- and postoperative cardiac evaluation of dogs undergoing lung resections because cardiac problems are a common postoperative complication after such surgeries. In particular, complete AVB should be considered a lifethreatening complication after pneumonectomy and lobectomy. In addition, pneumonectomy appears to increase the likelihood of pulmonary hypertension development in dogs.

        Implementation of Fair Trade Practices in Peruvian Artisanal Organizations: Analysis through Case Studies

        In Peru handicraft production is an activity complementary to agriculture which transmits cultural traditions across the generations. This article presents an analysis of the Fair Trade practices inside two organizations which support the production and marketing of Peruvian artisanal handicrafts. First, basic information is presented concerning the Fair Trade, its definition,practices, and products. Next, information regarding Peruvian artisanal products is presented as along with Fair Trade handcrafts export data. Two handicrafts producer organizations are then analyzed as case studies of Fair Trade practices carried out for producer communities in the Peruvian Andes where conditions of extreme poverty predominate. Through these case studies the article provides a detailed description of how the implementation of Fair Trade practices in the selected Peruvian handicrafts producer organizations has helped them enhance their markets.

        Electromyographic and Strength Analyses of Activation Patterns of the Wrist Flexor Muscles after Acupuncture

        This study analyzed the electromyographic and strength responses of the flexor muscles of the wrist following stimulation of acupuncture points. A total of 52 participants were randomly divided into four groups: local (heart 3, HT3), distant (heart 4, HT4), control (bladder 60, BL60), and naı ¨ve control groups. To obtain the root mean square electromyo- graphic activity, we placed surface electrodes over the wrist flexors. To obtain kilogram force (kgf) values, we attached a force transducer to the floor and to the hands of par- ticipants. Both values were recorded over three repetitions of maximal isometric wrist flexion contractions. Data were analyzed using one-way analyses of variance, followed by Dunnett’s post-hoc tests. We found reductions in electromyographic activity contralat- eral to the stimulated point in the distant group 10 minutes after removal of the needles (F 3,48 = 3.25; p < 0.05). Regarding muscle strength, ipsilateral and contralateral stimu- lation in the distant group produced kgf levels prior to and 10 minute and 20 minutes after withdrawal of the acupuncture needle that were lower than that obtained prior to inser- tion of the needle (F 3,48= 5.82; p < 0.05). Thus, stimulation of the acupuncture points distant from the wrist flexors reduced ipsilateral and contralateral muscle strength and decreased the root mean square values contralateral to the site of stimulation.

        Agave Fructans: Their Effect on Mineral Absorption and Bone Mineral Content

        In this study we investigate the effect that Agave fructans as new prebiotics have on mineral absorption improvement. Forty-eight 12-week-old C57BL/6J mice were used in this study. Forty mice were ovariectomized and eight were sham-operated controls. Mice were fed standard diets or diets supplemented with 10% Agave fructans or 10% inulin fructans. Calcium and magnesium were evaluated as well as their excretion in feces. Osteocalcin levels were also measured; femur structure was studied by scanning electron microscopy. Other parameters, such as food intake, body weight, glucose, and short-chain fatty acid content, were recorded. Calcium in plasma and bone increased in Agave fructan groups (from 53.1 to 56 and 85 mg/L and from 0.402 to 0.474 and 0.478 g/g, respectively) and osteocalcin increased in all fructan groups (> 50%). Scanning electron microscopy showed that fructans were able to mitigate bone loss. In conclusion, we demonstrated that supplementation with Agave fructans prevents bone loss and improves bone formation.

        Cytotoxic Activity of the Methanolic Extract of Turnera diffusa Willd on Breast Cancer Cells

        Turnera diffusa Willd, commonly known as Damiana, is employed in traditional medicine as a stimulant, aphrodisiac, and diuretic. Its leaves and stems are used for flavoring and infusion. Damiana is considered to be safe for medicinal use by the FDA. Pharmacological studies have established the hypoglycemic, antiaromatase, prosexual, estrogenic, antibacterial, and antioxidant activity of T. diffusa. The aim of the present study was to evaluate the possible cytotoxic effect of extracts and organic fractions of this plant on five tumor cell lines (SiHa, C-33, Hep G2, MDA-MB-231, and T-47D) and normal human fibroblasts. The results show that the methanolic extract (TdM) displayed greater activity on MDA-MB-231 breast cancer cells (with an IC50 of 30.67 μg/mL) than on the other cancer cell lines. Four organic fractions of this extract exhibited activity on this cancer cell line. In the most active fraction (F4), two active compounds were isolated, arbutin (1) and apigenin (2). This is the first report of a cytotoxic effect by T. diffusa on cancer cells. The IC50 values suggest that the methanolic extract of T. diffusa has potential as an anticancer therapy

        Intelligent Network Data Analytics Function in 5G Cellular Networks using Machine Learning

        5G cellular networks come with many new features compared to the legacy cellular networks, such as network data analytics function (NWDAF), which enables the network operatorsto either implement their own machine learning (ML) based dataanalytics methodologies or integrate third-party solutions to theirnetworks. In this paper, the structure and the protocols of NWDAFthat are defined in the 3rd Generation Partnership Project (3GPP)standard documents are first described. Then, cell-based syntheticdata set for 5G networks based on the fields defined by the 3GPPspecifications is generated. Further, some anomalies are added tothis data set (e.g., suddenly increasing traffic in a particular cell),and then these anomalies within each cell, subscriber category,and user equipment are classified. Afterward, three ML models,namely, linear regression, long-short term memory, and recursiveneural networks are implemented to study behaviour informationestimation (e.g., anomalies in the network traffic) and network loadprediction capabilities of NWDAF. For the prediction of networkload, three different models are used to minimize the mean absoluteerror, which is calculated by subtracting the actual generated datafrom the model prediction value. For the classification of anomalies, two ML models are used to increase the area under the receiver operating characteristics curve, namely, logistic regressionand extreme gradient boosting. According to the simulation results, neural network algorithms outperform linear regression innetwork load prediction, whereas the tree-based gradient boosting algorithm outperforms logistic regression in anomaly detection. These estimations are expected to increase the performance of the5G network through NWDAF.
