      • The transcriptomic changes of Huipizhi Heidou (<i>Glycine max</i>), a nematode-resistant black soybean during <i>Heterodera glycines</i> race 3 infection

        <P>Glycine max (soybean) is an extremely important crop, representing a major source of oil and protein for human beings. Heterodera glycines (soybean cyst nematode, SCN) infection severely reduces soybean production; therefore, protecting soybean from SCN has become an issue for breeders. Black soybean has exhibited a different grade of resistance to SCN. However, the underlying mechanism of Huipizhi Heidou resistance against SCN remains elusive. The Huipizhi Heidou (ZDD2315) and race 3 of Heterodera glycines were chosen to study the mechanism of resistance via examination of transcriptomic changes. After 5, 10, and 15 days of SCN infection, whole roots were sampled for RNA extraction, and uninfected samples were simultaneously collected as a control. 740, 1413, and 4925 genes were isolated by padj (p-value adjusted)<0.05 after 5, 10, and 15 days of the infection, respectively, and 225 differentially expressed genes were overlapped at all the time points. We found that the differentially expressed genes (DEGs) at 5, 10, and 15 days after infection were involved in various biological function categories; in particular, induced genes were enriched in defense response, hormone mediated signaling process, and response to stress. To verify the pathways observed in the GO and KEGG enrichment results, effects of hormonal signaling in cyst-nematode infection were further examined via treatment with IAA (indo-3-acetic acid), salicylic acid (SA), gibberellic acid (GA), jasmonic acid (JA), and ethephon, a precursor of ethylene. The results indicate that five hormones led to a significant reduction of J2 number in the roots of Huipizhi Heidou and Liaodou15, representing SCN-resistant and susceptible lines, respectively. Taken together, our analyses are aimed at understanding the resistance mechanism of Huipizhi Heidou against the SCN race 3 via the dissection of transcriptomic changes upon J2 infection. The data presented here will help further research on the basis of soybean and cyst-nematode interaction.</P>

        MAPK4 silencing in gastric cancer drives liver metastasis by positive feedback between cancer cells and macrophages

        Liver metastasis is a major cause of death in gastric cancer patients, but the underlying mechanisms are poorly understood. Through a combination of in vivo screening and transcriptome profiling followed by quantitative RT-PCR and tissue array analyses, we found that mitogen-activated protein kinase 4 (MAPK4) downregulation in gastric cancer tissues from patients is significantly associated with liver metastasis and poor prognosis. The knockdown of MAPK4 in gastric cancer cells promotes liver metastasis in orthotopic mouse models. MAPK4 depletion in gastric cancer cells induces the secretion of macrophage migration inhibitory factor (MIF) to polarize tumor-associated macrophages (TAMs) in orthotopic xenograft tumors. Moreover, TAMs activate epithelial–mesenchymal transition of gastric cancer cells to suppress MAPK4 expression, which further increases MIF secretion to polarize TAMs. Taken together, our results suggest a previously undescribed positive feedback loop between cancer cells and macrophages mediated by MAPK4 silencing that facilitates gastric cancer liver metastasis.

        중국 체육학 전공 여대생의 객체화된 신체의식이 신체존중감에 미치는 영향

        The purpose of this study was to examine the relationship between objectified body consciousness and physical self-esteem of Chinese female university students majoring in physical education. To accomplish the purpose of the research, female university students who are majoring in Physical Education at three different universities(N=427) in Henan province in China were recruited via an internet based method. Collected data were analyzed by reliability test, confirmatory factor analysis and multiple linear regression analysis via Win SAS Ver. 9.4. The results of the study are as follows. First, objectified body consciousness had a negative effect on appearance esteem. Second, objectified body consciousness had a negative effect on wight esteem. Third, objectified body consciousness had a negative effect on physical function esteem. Implications of this study were discussed and a direction for future study was suggested as follows. First, the future research should include participants with various backgrounds. Second, longitudinal research is recommended to further examine the influential factors on body esteem. Lastly, more research should be conducted to with other influential factors to understand Chinese female students.

        Real-time hydrogen mud logging during the Wenchuan earthquake fault scientific drilling project (WFSD), holes 2 and 3 in SW China

        We present hydrogen gas concentration data for drilling mud extracted during the Wenchuan Earthquake Fault Scientific Drilling (WFSD; holes 2 and 3), measured on-line during drilling in SW China. Hydrogen influx into the well at depth is largely dependent on lithology and fracture density. In hole WFSD-2, the average background concentration of hydrogen is lower in granitic rocks than in sedimentary formations. More than five major hydrogen influxes were detected while drilling through the sedimentary formations, and two hydrogen-rich zones were detected in the granite (1240–1243 m and 1383.5–1405 m depth). In hole WFSD-3, mud extracted from a tectonic breccia contains high hydrogen concentrations at depths of 600–1000 m depth, and relatively low concentrations below 1000 m depth. In both holes, we observe a lack of hydrogen in the center of the fault zone and high concentrations of hydrogen in fractured zones. Hydrogen concentration in both holes displays significant vertical heterogeneity, and is positively correlated with fracture density. Hydrogen was likely sourced from interaction between water and fresh silicate minerals surfaces that were exposed during faulting, from the mantle, and from faulting events. Fracture zones provide the main channels for the migration of hydrogen gas. The variations in background hydrogen concentrations relate to changes in porosity and permeability. The two hydrogen-rich zones (642.36–676.22 m, and 1383.5–1405 m) were likely caused by earthquake activity or far-field triggering during WFSD-2 drilling. The results of this study provide gas data that can be used to model fault activity. It is important to consider the relationship between hydrogen gas and the seismic cycle, and to use such data to identify seismic precursors.

        Study on a Low-temperature Cellulose-degrading Strain: Fermentation Optimization, Straw Degradation, and the Effect of Fermentation Broth on Seed Growth

        The biodegradation of straw by cellulosedegrading strains plays an important role in the disposal of agricultural waste. In this study, a Penicillium sp. strain JiTF01 that could degrade cellulose was isolated at the low temperature (10oC), and its fermentation conditions were optimized by response surface method. Strain JiTF01 was applied to degrade rice straw at 10oC. It was found that the straw degradation ratio could reach to 46.53% on the 25th day of culture with the enzyme activity of 44.30 U/mL under optimized fermentation conditions. In addition, given that strain JiTF01 showed excellent acid resistance and the cellulase produced by strain JiTF01 exhibited higher enzymatic activity and stability under acidic conditions, the uninterrupted integration process of acid pretreatment and acid fermentation of straw was then performed. The results showed that the degradation ratio of 53.61% was observed after 25 days of the fermentation at pH 3, which increased by 7.08% than that under optimized conditions. Besides, the effect of straw fermentation broth on the growth of rice seed was also investigated, and the 10-5 and 10-6 diluted fermentation broth was found that could significantly promote the growth of rice seedlings and enhance the germination rate of rice seeds under the salt stress of 100 and 200 mmol/L. The conclusions found in this article might have important implications for straw recycling in cold regions.
