        Persistent Anxiety Is Associated with Higher Glycemia Post-Transition to Adult Services in Asian Young Adults with Diabetes

        Background There is little longitudinal information on psychological burden and metabolic outcomes in young adults with diabetes (YAD) in Asia. We aimed to evaluate the association between psychological status and glycemia at baseline and 2 years following transition in a cohort of YAD in Singapore. Methods Subjects with type 1 diabetes mellitus (T1DM) and type 2 diabetes mellitus (T2DM), aged 17 to 25 years, were recruited from the YAD clinic in Singapore General Hospital. The Hospital Anxiety and Depression and Problem Areas for Diabetes scales were administered at transition (baseline) and at 18 to 24 months. Glycosylated hemoglobin (HbA1c) assessed during routine visits was tracked longitudinally. Results A total of 98 T1DM (74.8%) and 33 T2DM (25.2%) subjects were recruited between January 2011 and November 2017. At baseline, mean HbA1c was 8.6%±1.7%. Only 26.0% achieved HbA1c of ≤7.5% and 16.8% achieved HbA1c of <7%. At baseline, prevalence of anxiety was 29.8%. At 24 months, 14.1% had persistent anxiety. Those with persistent anxiety had the highest mean HbA1c, particularly at 6 months (persistently anxious vs. persistently non-anxious: 9.9%±1.2% vs. 8.2%±1.9%, P=0.009). At baseline, 9.2% of subjects had depression. This group also had poorer glycemia at baseline (HbA1c of depressed vs non-depressed: 9.6%±2.1% vs. 8.5%±1.6%, P=0.04), which persisted up to 24 months. Conclusion The majority of YAD in Singapore have suboptimal glycemia. Psychological distress is a critical harbinger of poorer metabolic outcomes.

        SYNTHESIS AND FLUORESCENCE PROPERTIES OF LaF 3 :Tb 3 þ PHOSPHORS VIA ULTRASONIC- ASSISTED IONIC LIQUID METHOD

        Hexagonal phase LaF 3 :Tb 3 þnano/microcrystals were successfully synthesized via ultrasonic-assisted ionic liquid (IL) method. The ¯eld emission scanning electron microscope (FESEM)images revealed that the morphologies of the as-prepared LaF 3 :Tb 3 þsamples were graduallyconverted from nanoplates to microcylinders with increasing the concentration of Tb 3 þions. Thephotoluminescent (PL) spectrum shows that all the as-prepared La 1 ?xTbxF 3 samples show thecharacteristic emissions of Tb 3 þ 5D4 ! 7FJ (J ¼ 6-3, with 5D4 ! 7F5 green emission at 542 nmas the strongest one) transitions. The optimal Tb 3 þ-doped concentration is 20 mol.% in the LaF 3host. The photoluminescence intensity of the La 0 : 80 Tb 0 : 20 F 3 microcylinders prepared by ultra-sonic irradiation was largely improved as compared with that of the product prepared by stirring.

        Two Base Mutations of a Putative LacI-family Transcriptional Regulator, SCO7554, Reverse the Overexpression Effect on the Antibiotic Pigment Production in Streptomyces coelicolor

        The SCO7554 protein, a putative LacI-family transcriptional regulator, is important in the antibiotic pigment production in Streptomyces coelicolor M130. A single mutation at the 4th amino acid from asparagine to serine reversed the SCO7554 effect on the antibiotic pigment production in an overexpression experiment. Additional mutation at the 276th amino acid from valine to alanine intensified this effect, and the overexpression of the double mutant SCO7554 increased the production of antibiotics, whereas the overexpression of SCO7554itself decreased the antibiotic productions. This observation suggested the functional importance of the N-terminal region of a putative LacI-family transcriptional regulator, SCO7554 protein,even though it was not located on any known functional domain.

        Effects of Two Putative LacI-family Transcriptional Regulators, SCO4158 and SCO7554, on Antibiotic Pigment Production of Streptomyces coelicolor and Streptomyces lividans

        Streptomycetes produce many secondary metabolites,including antibiotics, antifungal agents, anti-parasitic drugs, and immune suppressors. Evaluation of streptomycetes, Streptomyces coelicolor and Streptomyces lividans, was carried out as research models. Overexpression of the putative LacI-family transcriptional regulator SCO4158 enhanced the production of antibiotic pigments in S. coelicolor but decreased their production in S. lividans,which serves as an example of discrepancy between two closely related strains. The comparison of biological mechanisms of the SCO4158 protein between these two strains represents a potential approach for the identification of S. coelicolor and S. lividans genes involved in antibiotic pigment production. Overexpression of the putative LacI-family transcriptional regulator SCO7554either decreased (homologous gene from the same overexpression strain) or enhanced (heterologous gene from the other strain)antibiotic pigment production by the homologous SCO7554. The SCO7554 genes in S. coelicolor and S. lividans differ by only two codon changes, neither of which alters the amino acid sequence;hence, our result suggests a role in protein synthesis for SCO7554. Codon usage by SCO7554 is, therefore, critical for its biological function on antibiotic pigment production.

        PpsA, TufA, and Glutaminase Were Modulated under Yop Secretion Conditions in Yersinia enterocolitica

        Yersinia enterocolitica expresses the Ysc type III secretion system (TTSS) for delivery of effector proteins (Yops) to host cells to manipulate immune system of the host. The study on the physiological change of Y. enterocolitica is important in determining how cells efficiently support the expression of virulence factors and adapt to the inside of hosts, which is completely different from the outside environment. Ysc TTSS also secretes Yops into medium when cells are grown in calcium-deficient medium at $37^{\circ}C$. In the present work, a phosphoprotein purification kit was used to selectively enrich proteins whose characteristics changed under the Yop production conditions. Phosphoenol pyruvate synthase (PpsA), elongation factor Tu (TufA), and glutaminase were identified by matrix-assisted laser desorption/ionization time-of-flight. These results showed that the affinity to column and/or the steady-state level of PpsA, TufA, and glutaminase are altered under the Yop secretion conditions and suggested that any protein characteristic of these three proteins was changed under the conditions for the virulence factor expression suggested that certain characteristics of these three proteins were changed. This work demonstrated that specific protein purification method can be used to identify proteins from a complex protein mixture of cell extracts with emphasis on the specific characteristics of proteins.

      • Intracellular ATP Levels Affect Secondary Metabolite Production in <i>Streptomyces</i> spp.

        <P>The addition of extracellular ATP (exATP) to four <I>Streptomyces</I> strains had similar effects: low exATP levels stimulated antibiotic production and high levels reduced it. Compared with antibiotic production, the concentrations of intracellular ATP (inATP) in the tested strains were opposite, which suggests a role of inATP in regulating secondary metabolite production. Under inactivation of the polyphosphate kinase gene (<I>ppk</I>) in <I>Streptomyces lividans</I>, we observed the same results: when the inATP level in the mutant strain was lower than in the parent strain, more antibiotic was produced. Combining all the results, a strong inverse relationship between [inATP] and the secondary metabolite production is suggested by this study.</P>

      • Analysis of Multiple Network Accessibilities and Commercial Space Use in Metro Station Areas: An Empirical Case Study of Shanghai, China

        Against the background of the rapid development of the Shanghai Metro network, this paper attempts to establish an analytical approach to evaluate the impact of multiple transport network accessibilities on commercial space use in metro station areas. Ten well-developed metro station areas in central Shanghai are selected as samples. Commercial space floor area and visitors in these areas are collected. Using ArcGIS and Spatial Design Network Analysis, the Shanghai Metro network and road network are modeled to compute diversified transport accessibilities. Evidence from land use and commercial space floor area within a 0-to-500-meter buffer zone of stations is consistent with location and land-use theory: commercial land use is concentrated closer to stations. Correlation analysis suggests that hourly visitors to the shopping mall are mainly influenced by metro network accessibility, while retail stores and restaurants are affected by both metro and pedestrian accessibility.

        한국어 학습자의 말하기 자료에 나타나는 결속 장치 사용 연구

        목적 본 연구에서는 말하기 자료에 나타나는 한국어 학습자의 결속 장치 사용 유형과 빈도를 파악하기 위하여 양적 통계와 질적인 서술로 숙달도별에 따른 특징을 살펴보고자 하였다. 방법 이를 위하여 수험자들의 한국어 말하기 시험 자료를 분석하여 한국어 결속 장치 사용 양상과 경향을 살펴보았다. 제3차 한국어능력시험 말하기 평가개발 연구에서 실시한 말하기 시험을 참석한 초,중급 학습자 60명을 대상으로 시험 문항에 대한 녹음 파일 360개를 전사 및 분석을 실시하였다. 본고는 결속 장치를 ‘지시, 접속, 어휘 반복’으로 분류하여 그 사용과 특징을 양적인 통계와 질적인 서술로 규명하였다. 결과 지시, 접속, 어휘 반복 결속 장치를 분석한 결과, 지시 표현에는 두 집단 모두 “그>이>저” 순서로 많이 사용하였으나 “그” 계열의 지시 표현을 잘못 이해하거나 감투사로 사용하는 것이 빈번히 발견되었다. 접속 표현 같은 경우에는 초급 학습자는 확대>강화>설명 순서로 사용 빈도가 높았고 중급 학습자의 경우 강화>확대>설명 순서로 자주 사용한 것으로 나타났다. 어휘 반복 표현은 세 가지 결속 장치에서 가장 높은 빈도로 나타났는데 중급의 경우 초급보다 2배 가량 빈도가 높게 나타났다. 또한 유의어나 유사한 단어, 상하위어 항목은 거의 나타나지 않았다. 결론 이상의 분석 결과를 바탕으로 한국어 학습자에게 지시 표현을 정확하게 이해를 하고 사용할 수 있도록 교육이 필요하고, 내용의 일관성을 유지하기 위해 동일어만 반복하는 것보다 유의어나 상하위어에 대한 교육을 강화하여 다양한 어휘 반복을 할 수 있도록 가르칠 필요가 있겠다. Objectives The purposes of this study were to analyzes the Korean speaking test by Korean language learners to examine discourse cohesion shown upon using cohesive device. Methods The data collected 60 beginner and intermediate learners, 360 recorded files of test questions were transcribed and analyzed. It focuses on the use of conjunction, reference and lexical reiteration devices as a cohesive device. Results As a result of analyzing conjunction, reference, lexical reiteration cohesive devices, both groups used a lot of “그>이>저”, but it was mainly found misunderstanding and use of reference expressions of the “그”. In the case of conjunction, beginner learners have mainly used ‘expansion> reinforcement> explanation, and intermediate learners frequently used reinforcement> expansion> explanation. Lexical reiteration was found to have the highest frequency in the three cohesive device, but the intermediate learners was twice as high as that of the beginner learners. In addition, there were almost no synonyms, similar words, or upper and lower words. Conclusions Through this paper, the study was able to identify the condensation systems needed for Korean language learners. Therefore, It should be taught more actively at the teaching site.
