This study aimed to evaluate the effects of Limosilactobacillus fermentum and Lactiplantibacillus plantarum isolated from human feces coordinating with inulin on the composition of gut microbiota and metabolic profiles in db/db mice. These supplements were administered to db/db mice for 12 weeks. The results showed that the Lactobacillaceae coordinating with inulin group (LI) exhibited lower fasting blood glucose levels than the model control group (MC). Additionally, LI was found to enhance colon tissue and increase the levels of short-chain fatty acids. 16S rRNA sequencing revealed that the abundance of Corynebacterium and Proteus, which were significantly increased in the MC group compared with NC group, were significantly decreased by the treatment of LI that also restored the key genera of the Lachnospiraceae_NK4A136_group, Lachnoclostridium, Ruminococcus_ gnavus_group, Desulfovibrio, and Lachnospiraceae_UCG-006. Untargeted metabolomics analysis showed that lotaustralin, 5-hydroxyindoleacetic acid, and 13(S)-HpODE were increased while L-phenylalanine and L-tryptophan were decreased in the MC group compared with the NC group. However, the intervention of LI reversed the levels of these metabolites in the intestine. Correlation analysis revealed that Lachnoclostridium and Ruminococcus_gnavus_group were negatively correlated with 5-hydroxyindoleacetic acid and 13(S)-HpODE, but positively correlated with L-tryptophan. 13(S)-HpODE was involved in the "linoleic acid metabolism". L-tryptophan and 5-hydroxyindoleacetic acid were involved in "tryptophan metabolism" and "serotonergic synapse". These findings suggest that LI may alleviate type 2 diabetes symptoms by modulating the abundance of Ruminococcus_gnavus_group and Lachnoclostridium to regulate the pathways of "linoleic acid metabolism", "serotonergic synapse", and" tryptophan metabolism". Our results provide new insights into prevention and treatment of type 2 diabetes.

<P>Arabidopsis protein NCA1 interacts with catalases in the cytosol and increases catalase activity through maintaining catalase folding state, which is required for stress responses.</P><P>Catalases are key regulators of reactive oxygen species homeostasis in plant cells. However, the regulation of catalase activity is not well understood. In this study, we isolated an <I>Arabidopsis thaliana</I> mutant, <I>no catalase activity1-3</I> (<I>nca1-3</I>) that is hypersensitive to many abiotic stress treatments. The mutated gene was identified by map-based cloning as NCA1, which encodes a protein containing an N-terminal RING-finger domain and a C-terminal tetratricopeptide repeat-like helical domain. NCA1 interacts with and increases catalase activity maximally in a 240-kD complex in planta. In vitro, NCA1 interacts with CATALASE2 (CAT2) in a 1:1 molar ratio, and the NCA1 C terminus is essential for this interaction. CAT2 activity increased 10-fold in the presence of NCA1, and zinc ion binding of the NCA1 N terminus is required for this increase. NCA1 has chaperone protein activity that may maintain the folding of catalase in a functional state. NCA1 is a cytosol-located protein. Expression of <I>NCA1</I> in the mitochondrion of the <I>nca1-3</I> mutant does not rescue the abiotic stress phenotypes of the mutant, while expression in the cytosol or peroxisome does. Our results suggest that NCA1 is essential for catalase activity.</P>
Berberine protects against palmitate induced beta cell injury via promoting mitophagy

Background: Destruction of pancreatic beta cells is the most typical characteristic of diabetes. Objective: We aimed to evaluate the effect of berberine (BBR), a bioactive isoquinoline derivative alkaloid, on beta cell injury. Methods: Rodent pancreatic beta cell line INS-1 was treated with 0.5 mM palmitate (PA) for 24 h to establish an in vitro beta cell injury model. Results: BBR at 5 µM promoted cell viability, inhibited cell apoptosis and enhanced insulin secretion in PA-induced INS-1 cells. BBR treatment also suppressed PA-induced oxidative stress in INS-1 cells, as evidenced by the decreased ROS production and increased activities of antioxidant enzymes. In addition, suppressed ATP production and reduced mitochondrial membrane potential were restored by BBR in PA-treated INS-1 cells. It was further determined that BBR affected the expressions of mitophagy-associated proteins, suggesting that BBR promoted mitophagy in PA-exposed INS-1 cells. Meanwhile, we found that BBR facilitated nuclear expression and DNA-binding activity of Nrf2, an antioxidative protein that can regulate mitophagy. Finally, a rescue experiment was performed and the results demonstrated that the effect of BBR on cell viability, apoptosis and mitochondrial function in PA-induced INS-1 cells were cancelled by PINK1 knockdown. Conclusions: BBR protects islet β cells from PA-induced injury, and this protective effect may be achieved by regulating mitophagy. The present study may provide a novel therapeutic strategy for β cell injury in diabetes mellitus.

This paper traces W. B. Yeats’s bearings on the Australian playwright Louis Esson. The branching out of Yeats’s influence in discussion can be traced back to one single play, Cathleen Ni Houlihan (1902). Cathleen Ni Houlihan is structured by the twin formal factors, the diegetic-space centeredness, and the mimesis of the protagonist’s embodied reorientation, which together constitute what I call “the technique of spatialization.” What Esson takes from this play, by reconstruction, is not the perpetuation of Irish folklores, but the staging form of spatialization. This particular staging technique has been extensively adapted by Esson to offer the spectators the imagination of the hostile space of Australia in three one-act plays, including Dead Timber, The Sacred Place, and The Drovers. This essay looks at the global Yeats in these plays in the context of early twentieth-century Anglophone modernism and offers a way to explain how the (re)mapping of space is at stake in the playwriting for cultural nationalism.
Research on Microstructure and Properties of TiN, (Ti, Al)N and TiN/(Ti, Al)N Multilayer Coatings

Magnetron sputtered TiN, (Ti, Al)N and TiN/(Ti, Al)N multilayer coatings grown on cemented carbide substrates have been characterized by using electron probe microanalysis (EPMA), X-ray diffraction (XRD), scanning electron spectroscopy (SEM), nanoindentation, scratcher and cutting tests. Results show that TiN coating is bell mouth columnar structures, (Ti, Al)N coating is straight columnar structures and the modulation structure has been formed in the TiN/(Ti, Al)N multilayer coating. TiN/(Ti, Al)N multilayer coating exhibited higher hardness, better adhesion with substrate and excellent cutting performance compared with TiN and (Ti, Al)N coating.
An Emerging Identity of Capital Owners in Charles Dickens’ Great Expectations

This paper discusses the relationship of capital, things, and selfimprovement in Charles Dickens’ Great Expectations (1860-1) and focuses on the character of Abel Magwitch to delineate a positive attitude toward the possession of capital shown in the novel. Dickens reproduces the chill-and-kill culture in the early 19th century brought on by the punitive financial prohibition measures of capital punishment, long-term imprisonment or transportation out to colonies, while highlighting the optimistic anticipation of profit making by personal participation in capital investment in the 1850s after the financial reforms. Meanwhile, Dickens also avoids a swing to a total embrace of things without any assertion of a person’s agency. Great Expectations offers readers a transit for entering a status of coexistence with things, from an unformulated fear of owning things to techniques of detaching the self from the dominance of things over subjectivities. By this study I wish to contribute to the understanding of high-Victorian novels as complex reactions to the classical idea of homo oeconomicus, which underpins the political economy initiated by Adam Smith and others.

Matrine, a main active component extracted from dry roots of Sophora flavecens, has been reported to exert antitumor effects on A549 human non-small lung cancer cells, but its mechanisms of action remain unclear. To determine effects of matrine on proliferation of A549 cells and assess possible mechanisms, MTT assays were employed to detect cytotoxicity, along with o flow cytometric analysis of DNA content of nuclei of cells following staining with propidium iodide to analyze cell cycle distribution. Western blotting was performed to determined expression levels of Bax, Bcl-2, VEGF and HDAC1, while a microarray was used to assessed changes of miRNA profiles. In the MTT assay, matrine suppressed growth of human lung cancer cell A549 in a dose- and timedependent manner at doses of 0.25-2.5 mg/ml for 24h, 48h or 72h. Matrine induced cell cycle arrest in G0/G1 phase and decreased the G2/M phase, while down-regulating the expression of Bcl2 protein, leading to a reduction in the Bcl-2/Bax ratio. In addition, matrine down regulated the expression level of VEGF and HDAC1 of A549 cells. Microarray analysis demonstrated that matrine altered the expression level of miRNAs compared with untreated control A549 cells. In conclusion, matrine could inhibit proliferation of A549 cells, providing useful information for understanding anticancer mechanisms.
Optical Properties of K9 Glass Waveguides Fabricated by Using Carbon-Ion Implantation

K9 glass is a material with promising properties that make it attractive for optical devices. Ion implantation is a powerful technique to form waveguides with controllable depth and refractive index profile. In this work, optical planar waveguide structures were fabricated in K9 glasses by using 6.0-MeV C3+-ion implantation with a fluence of 1.0 × 1015 ions/cm2. The effective refractive indices of the guided modes were measured by using a prism-coupling system. The refractive index change in the ion-irradiated region was simulated by using the intensity calculation method. The modal intensity profile of the waveguide was calculated and measured by using the finite difference beam propagation method and the end-face coupling technique, respectively. The transmission spectra before and after the implantation showed that the main absorption band was not influenced by the low fluence dopants. The optical properties of the carbon-implanted K9 glass waveguides show promise for use as integrated photonic devices.
Conceptual design study on Plutonium-238 production in a multi-purpose high flux reactor

Plutonium-238 has always been considered as the one of the promising radioisotopes for space nuclear power supply, which has long half-life, low radiation protection level, high power density, and stable fuel form at high temperatures. The industrial-scale production of 238Pu mainly depends on irradiating solid 237NpO2 target in high flux reactors, however the production process faces problems such as large fission loss and high requirements for product quality control. In this paper, a conceptual design study of producing 238Pu in a multipurpose high flux reactor was evaluated and analyzed, which includes a sensitivity analysis on 238Pu production and a further study on the irradiation scheme. It demonstrated that the target structure and its location in the reactor, as well as the operation scheme has an impact on 238Pu amount and product quality. Furthermore, the production efficiency could be improved by optimizing target material concentration, target locations in the core and reflector. This work provides technical support for irradiation production of 238Pu in high flux reactors.
