Purpose: Notch is an important signaling pathway that regulates cell fate, stem cell maintenance and the initiation of differentiation in many tissues. It has been reported that activation of Notch-1 contributes to tumorigenesis. However, whether Notch signaling might have a role in chemoresistance of prostate cancer is unclear. This study aimed to investigate the effects of Notch-1 silencing on the sensitivity of prostate cancer cells to docetaxel treatment. Methods: siRNA against Notch-1 was transfected into PC-3 prostate cancer cells. Proliferation, apoptosis and cell cycle distribution were examined in the presence or absence of docetaxel by MTT and flow cytometry. Expression of $p21^{waf1/cip1}$ and Akt as well as activation of Akt in PC-3 cells were detected by Western blot and Real-time PCR. Results: Silencing of Notch-1 promoted docetaxel induced cell growth inhibition, apoptosis and cell cycle arrest in PC-3 cells. In addition, these effects were associated with increased $p21^{waf1/cip1}$ expression and decreased Akt expression and activation in PC-3 cells. Conclusion: Notch-1 promotes chemoresistance of prostate cancer and could be a potential therapeutic target.
Research On Optimization In NC Bending Process Of Thin-walled Tube Based on Orthogonal Experiment

In the process of research and development of tube bending forming technology, the way of improving the forming quality and establishing the optimization of forming process parameters has become an important problem that should be solved quickly. This text simulated the process of bend forming with the way of finite element, combined with the effects of geometric and physical parameters on the forming quality of bending pipe, taking the important parameters which are measurements of the pipe wall thickness variation as test indexes, made the virtual orthogonal experiment and gained the laws of the bending angle, relative bending radius, relative wall thickness, thrust, pipe friction coefficient between mode have effects on the forming quality of wall thinning rate and wall thickening. Through the comprehensive analysis and additional test, obtained the optimal parameters values, improved the accuracy of simulation and the optimization results of orthogonal test, improved the quality of tube bending forming.

Background: Role of autophagy in neutrophil function and the association of autophagy and autophagy related gene (ATG) polymorphisms with asthma susceptibility were suggested. In this study, we investigated the association of ATG5 and ATG7 polymorphisms with asthma risk, severity and neutrophilic phenotype of asthma. Methods: We recruited 408 asthma patients and 201 healthy controls (NC). Sputum neutrophil counts were determined by H&E staining. Serum IL-8 levels were measured by ELISA. ATG5 gene polymorphisms (-769T>C, -335G>A and 8830C>T) and ATG7 gene polymorphisms (-100A>G and 25108G>C) were genotyped. The functional activities of ATG5 -769T>C and -335G>A variants were investigated by luciferase reporter assays. Results: No associations of ATG5 and ATG7 gene polymorphisms with asthma susceptibility and severity were found. ATG5 -769T>C and -335G>A were in complete linkage disequilibrium. In the asthma group, GA/AA genotypes at ATG5 -335G>A were associated with higher neutrophil counts in sputum (P<0.05); CC/TT genotype at ATG5 8830C>T associated with lower % FEV1 predicted value (P<0.05). DNA fragments containing ATG5 -769T and -335G alleles had higher promoter activities compared to those with -769C and -335A in both human airway epithelial cells (A549, P<0.01) and human mast cell (HMC-1, P<0.001). GG and CC genotype at ATG7 -100A>G and 25108G>C were significantly associated with high serum levels of IL-8 (P<0.05 for both variants). Conclusions: Polymorphisms of ATG5 and ATG7 genes could contribute to neutrophilic phenotype in the pathogenesis of adult asthma.
Stochastic characteristics of reinforcement corrosion in concrete beams under sustained loads

This paper deals with the characteristics of reinforcement corrosion in concrete beams under the influence of sustained loads. The evolution and distribution laws of the reinforcement corrosion were measured periodically over time. The results show that sustained load exhibits a pronounced exacerbating effect on the reinforcement corrosion, and enlarges the nonuniformity level of corrosion as the load level increases. Accompanied with the continuous formation of the rust, the corrosion rate was also observed to be highly nonlinear and time-dependent. Moreover, to visually and quantitatively analyze the distribution of reinforcement corrosion, the 3D scanning technology combined with the probability statistics analysis was adopted, and the observed nonuniformity can be well described by the Gumbel distribution. Finally, an approach based on the three-phase spherical model was proposed to estimate the reinforcement corrosion, taking account of the effects of sustained load on the changes of concrete porosity and oxygen diffusivity.
Engineering of Self-Assembled Fibronectin Matrix Protein and Its Effects on Mesenchymal Stem Cells

<P>Fibronectin (FN) contributes to cell adhesion, proliferation, and differentiation in various cell types. To enhance the activity of fibronectin at the sites of focal adhesion, we engineered a novel recombinant fibronectin (FNIII10) fragment connected to the peptide amphiphile sequence (PA), LLLLLLCCCGGDS. In this study, the effects of FNIII10-PA on rat mesenchymal stem cells (rMSCs) were compared with those of FNIII10. FNIII10-PA showed the prominent protein adhesion activity. In addition, FNIII10-PA showed a significantly higher effect on adhesion, proliferation, and differentiation of rMSCs than FNIII10. Taken together, the FNIII10-containing self-assembled sequence enhanced rMSCs adhesion, proliferation, and differentiation.</P>
