        The In Vitro and In Vivo Efficacy of Hen IgY Against Vibrio parahaemolyticus and Vibrio vulnificus

        The inhibitory effect of IgY against Vibrio parahaemolyticus and Vibrio vulnificus responsible for seafood-borne diseases was investigated in this study. Water-soluble fractions (WSF) of protein containing IgYs were isolated from the egg yolk of hens initially immunized with formalininactivated V. parahaemolyticus or V. vulnificus. Protein, total and specific IgY contents of the WSF were determined. The inhibitory and protective effects of IgYs on the growth of V. parahaemolyticus and V. vulnificus were assayed in liquid medium and in mice. IgYs showed high affinity to their corresponding antigens with high titer from day 28 onwards. Protein contents and total IgY concentrations remained stable throughout the immunization period, whereas specific IgY concentrations increased steadily and reached a plateau at day 49. Specific IgY powder (150 mg/ml) significantly inhibited further multiplication of both V. parahaemolyticus and V. vulnificus in liquid medium as compared with the control IgY. The bacteria count in mice feces was lower in mice pretreated with specific IgYs than in those pretreated with PBS or control IgY. Higher survival of mice was observed in the experimental groups pretreated with either anti-V. parahaemolyticus (75% survival) or anti-V. vulnificus (87% survival) IgYs, compared with those in the control groups pretreated with PBS or nonspecific IgY. All mice in the control groups died within three days after bacteria inoculation; hence, the protective effect of specific IgYs against infection caused by V. parahaemolyticus and V. vulnificus was demonstrated.

        Production of Mouse Anti-Quail IgY and Subsequent Labeling with Horseradish Peroxidase Using Cyanuric Chloride

        Polyclonal antibodies labeled with a tracer have been commonly used as secondary antibodies in immunochemical assays to quantify the concentration of antibody-antigen complexes. The majority of these antibodies conjugated with a tracer are commercially available, with the exception of few untouched targets. This study focused on the production and application of mouse anti-quail IgY as an intermediate antibody to link between quail egg yolk IgY and goat anti-mouse IgG-HRP as primary and secondary antibodies, respectively. Subsequently, the produced mouse anti-quail IgY was labeled with horseradish peroxidase (HRP) and its efficiency on enzyme linked immunosorbent assay (ELISA) was compared with that of commercial rabbit anti-chicken IgY-HRP. As an intermediate antibody, mouse anti-quail IgY was successfully produced with good affinity and sensitivity (1:10,000) to the primary and secondary antibodies. Subsequently, mouse anti-quail IgY was effectively conjugated with HRP enzyme, resulting in a secondary antibody with good sensitivity (1:10,000) to quail anti-V. parahaemolyticus and V. vulnificus IgY. The detection limit was 105 CFU/ml for both V. parahaemolyticus and V. vulnificus. The efficiency of the produced conjugate to detect quail IgY on ELISA was comparable to that of the commercial rabbit anti-chicken IgY-HRP, and hence the produced and labeled mouse anti-quail IgY-HRP can be used as a secondary antibody to detect any antibody produced in quail.

        THE EFFECTS OF PROTEIN LEVELS ON THE TOTAL SULPHUR AMINO ACID REQUIREMENTS OF BROILERS DURING TWO GROWTH PERIODS

        Studies on the effects of protein levels on the total sulphur amino acid (TSAA) requirements of chickens were conducted on the starter broilers by feeding four levels of crude protein(16, 18, 20 and 23%) at three levels of TSAA (0.83, 0.93 and 1.03%) and on the grower broilers by feeding three levels of crude protein (16, 18 and 20%) at three levels of TSAA(0.72, 0.79 and 0.86%). The metabolisable energy of the diets was maintained constant at 3,200 kcal/kg and the experiments were carried out for two growing periods: starter (0-3 wk) and grower (3-6 wk). The results showed that there were significant differences in body weight gain, feed intake and feed:gain ratio under different protein levels of the starters. Crude protein, ME and TSAA intake were significantly affected by increasing the CP levels. The TSAA requirement of the starter broilers is recommended at 0.93% and it is not influenced by different protein levels used in the experiment. For the grower period, body weight gain and feed:gain ratio improved significantly at higher protein diets. Birds fed higher protein diet consumed greater quantities of protein. Responses to TSAA supplementation for body weight gain, feed intake and feed:gain ratio were not significant. The present results showed that the TSAA level of 0.79 to 0.86% was required for grower diets and that the protein levels of the diet did not influence the TSAA requirement.

        THE EFFECTS OF DIETARY ENERGY ON THE TOTAL SULPHUR AMINO ACID REQUIREMENTS OF BROILERS DURING TWO GROWTH PERIODS

        There levels of dietary ME (3,000, 3,200 and 3,400 kcal/kg) and four levels of Total Sulphur Amino Acid (TSAA) (0.73, 0.83, 0.93 and 1.03%) were studied in the starter period (0-3 wks) of the broilers. Three levels of dietary ME (3,000, 3,200 and 3,400 kcal/kg) combined with four levels of TSAA (0.65, 0.72, 0.79 and 0.86%) were studied in the grower period (3-6 wks). The crude protein content of the diet of the starter period was 23% while the diet of the grower period was 20%. The performance data of the starter broilers indicated that the dietary energy levels had no significant effects on body weight gain, feed intake and feed:gain ratio. However, TSAA levels had a significant influence on the growth and feed parameters. The response pattern for the grower period was similar to the starter period. The present experiment showed that in the tropics the TSAA requirement for the starter period was between 0.83 to 0.93% which is similar to the values recommended by NRC while for the grower period the TSAA requirement was between 0.79 to 0.86% at all the three energy levels which is higher than the values recommended by NRC.

      • Validity and Reliability of the Fagerstrom Test for Cigarette Dependence in a Sample of Arabic Speaking UK-Resident Yemeni Khat Chewers

        Background: The Fagerstrom Test for Cigarette Dependence (FTCD) (formally FTND) is widely used for measuring physical dependence on nicotine. Objective: To explore the cross cultural validity and reliability of FTCD amongst Arabic speaker cigarette consumers who chew khat leaf, a stimulant green leaf. Methods: The psychometric properties of the FTCD were assessed in a subsample (91regular cigarette smokers) of purposively selected 204 UK-resident Yemeni khat chewers recruited during random visits to khat sale outlets. Data were collected via a structured face-to-face interview. Data analyses included descriptive tests and factor analysis. Results: Two factors were obtained by a principle axis factor analysis and these were termed as urgency of restoring the level of nicotine after abstinence during sleeping and maintaining the level of nicotine during waking. The internal reliability (Cronbach's alpha coefficient) of the whole FTCD is low (.68) as well as for the two subscales (.60) and (.62) respectively. Conclusion: The psychometric properties of the Arabic version of the FTCD scale in this sample of Yemeni khat chewers who smoked regularly confirmed what has been established in other cultural settings. The findings of this study have yet to be cross validated amongst other appropriately representative sample of Arabic speakers.

        THE INFLUENCE OF COPPER ON THE TOTAL SULPHUR AMINO ACIDS REQUIREMENT OF BROILERS DURING TWO GROWING PERIODS

        A study was conducted to assess the influence of copper on the total sulphur amino acid requirements of broiler chickens reared under two growing periods. The TSAA levels used were 0.73, 0.83, 0.93% with copper levels of 0, 125, 250 and 375 mg/kg for the starter period and the TSAA levels of 0.72, 0.79 and 0.86% with copper levels of 0, 125, 250 and 375 mg/kg for the grower period. Total feed consumption, body weight gain and feed L gain ratio were used as the parameters for the assessment. The results showed that adding copper at 250 mg/kg to the diets improved feed : gain ratio of the starter broilers and resulted in small improvement of body weight gain and feed : gain ratio of the grower broilers. Growth was depressed in relation to the reduction of feed intake on the chicks fed diet containing 375 mg/kg copper. There was a significant interaction between dietary TSAA and copper levels for feed intake, hence, indicating that the supplementation of copper at the level of 375 mg/kg increased the TSAA requirement of the starter broilers, although no interference with the requirement of grower broilers.

        THE EFFECTS OF DIETARY ENERGY LEVELS ON THE CARCASS COMPOSITION OF THE BROILERS

        A study was conducted to determine the carcass composition of broilers when fed with three varying levels of dietary energy (3,000, 3,200 and 3,400 kcal/kg ME) at 20% crude protein and 0.79% Total Sulphur Amino Acid. The results showed that there was a significant (p < 0.05) increase in the ME intake of the chickens when the ME of the diet increased. Other factors like the protein intake, dressing percentage, weight gain and feed intake were not significantly affected. On the carcass, the increasing dietary ME levels resulted in a significant (p < 0.05) increase in the abdominal fat percentage and carcass fat percentage and a significant (p < 0.05) decrease in the carcass protein percentage. Similarly, the dietary ME produced a significant (p < 0.05) increase in the amount of and fat percentages of breast meat, thigh meat, and drumstick meat with a significant reduction in the protein percentage. There seemed to be an inverse relationship between the percentages of protein and fat. An increase in fat percentages always resulted in similar reduction in the protein content of the meat. These results also showed major differences between the white and red meat of the chicken.

        Influence of Adaptor on the Calibration of Inductance Standards

        Influence of the adaptors on the calibration of $100{\mu}H$ inductance standard was studied as a function of torque, applied when tightening the standard inductor terminal with the adaptor. Two different homemade adaptors of BPO gold-plated brass (BPO-Au) and banana-copper (BN-Cu) were made for the connection between the LCR meter and the inductance standard. The measured inductance (L) of the standard inductor and the contact resistance ($R_C$) between the adaptor and the standard inductor terminal showed exponential decreases against the torque increase from $25cN{\cdot}m$ to $150cN{\cdot}m$. The measured L and the calculated equivalence series resistance ($R_S$) were dependent on the adaptor type as well as on the $R_C$. The results of the adaptor analysis imply that the BPO-Au adaptor with the lower $R_C$ is more suitable for the inductance calibration. The calculated inductance of $99.956{\mu}H$ corrected by subtraction of the adaptor inductance and the contact resistance contributions from the measured value using the BPO-Au adaptor agreed well with the certificate ($99.948{\mu}H$) of the PTB within the measurement uncertainty of $140{\mu}H/H$.

        EFFECTS OF ASCORBIC ACID (VITAMIN C) SUPPLEMENTATION IN LAYER AND BROILER DIETS IN THE TROPICS

        An experiment was conducted to study the effects of Ascorbic Acid (AA) supplementations in the layer and broiler diets kept in the natural hot humid tropical climate ($20-35^{\circ}C$). The layers and the broilers were fed on normal commercial diet as control while supplementation of 400 and 600 mg/kg Ascorbic Acid made up the experimental diets. The results showed that AA supplementation in the layers significantly reduced egg weight and increased Haugh unit values of the eggs, but produced no significant effects on feed intake, body weight, egg production, respiratory rate and body temperature. The shell thickness was slightly improved, though not significantly, with AA supplementation. AA supplementation in broilers improved body weight gain and FCR and reduced the effect of heat stress as shown by lower body temperature and respiratory rates.

        A preliminary study of T-2 and HT-2 toxins in cereals sold in traditional market in South Korea

        Fusarium species are responsible for the production of harmful trichothecenes mycotoxins in cereals. These mycotoxins are cytotoxic, potentially immunosuppressive and potent fast-acting inhibitors of protein and nucleic acid synthesis. This study validated a HPLC method for simultaneous detection of T-2 and HT-2 toxins. The method was then used for the detection of T-2 and HT-2 toxins in cereals sold in traditional markets in Gyeongnam Province, South Korea. Seventy five samples analyzed, out of which 13 and 25 samples were found to be contaminated with T-2 (35.2-431.0 ng g<SUP>-1</SUP>) and HT-2 (21.1-442.7 ng g<SUP>-1</SUP>) toxins, respectively and 4 samples were found to be contaminated with both toxins. This study provides data on the contamination level of T-2 and HT-2 toxins in cereals in traditional market in Gyeongnam province, South Korea.
