        대학생의 사고력을 높일 수 있는 쓰기와 토의 활동의 전공 연계 학습 모델

        최근에 대학 글쓰기 교과 기능이 기초글쓰기 또는 계열별 글쓰기에서 전공지식과 내용을 주제로 한 전공연계 글쓰기(WAC)로 전환하는 시점에 있다. 이러한 맥락에서 본고에서는 글쓰기의 선행 경험이 매우 부족한 전문대학생들을 대상으로 1차적으로 기초글쓰기를 지속적으로 습관화할 수 있는 전공글쓰기 프로그램인 <학습 활동지>작성 운영과정과 결과를 제시하였다. 또 본격적인 전공지식을 주제로 글쓰기를 하는 <프로젝트 전문세미나>교과목의 운영과정과 결과를 제시하였다. 두 개의 과정을 통해 학생들이 글쓰기에 대한 기초력을 다지고 지식의 구조를 체계화하는 논리성고 문장력도 강화하였다. 향후 4차산업 혁명이 주도하는 미래사회의 변화 속에서 창의융합형 인재에 대한 수요를 충족하기 위해서는 전공연계 글쓰기와 같은 생각하는 수업 모델이 개발되어야 할 것이다. Recently, university writing curriculum functions are at the point of transition from basic writing or writing by division to major-linked writing (WAC) based on major knowledge and content. In this context, this paper presents the operation process and results of < Learning Activity Sheet >, a major writing program that allows students to continue to habituate basic writing primarily for vocational college students who have very little prior experience in writing. In addition, the operation process and results of the < Project Seminar >, where writing on the subject of full-scale major knowledge, was presented. Through the two courses, students have strengthened their basic skills in writing and strengthened their logic and writing skills to systematize the structure of their knowledge. In order to meet the demand for creative and convergent talent amid the changes of the future society led by the 4th Industrial Revolution, a thinking class model, such as major-linked writing, should be developed.

        Functional Identification of a Nuclear Localization Signal of MYB2 Protein in Giardia lamblia

        MYB2 protein was identified as a transcription factor that showed encystation-induced expression in Giardia lamblia. Although nuclear import is essential for the functioning of a transcription factor, an evident nuclear localization signal (NLS) of G. lamblia MYB2 (GlMYB2) has not been defined. Based on putative GlMYB2 NLSs predicted by 2 programs, a series of plasmids expressing hemagglutinin (HA)-tagged GlMYB2 from the promoter of G. lamblia glutamate dehydrogenase were constructed and transfected into Giardia trophozoites. Immunofluorescence assays using anti-HA antibodies indicated that GlMYB2 amino acid sequence #507–#530 was required for the nuclear localization of GlMYB2, and this sequence was named as NLS<SUB>GlMYB2</SUB>. We further verified this finding by demonstrating the nuclear location of a protein obtained by the fusion of NLS<SUB>GlMYB2</SUB> and G. lamblia glyceraldehyde 3-phosphate dehydrogenase, a non-nuclear protein. Our data on GlMYB2 will expand our understanding on NLSs functioning in G. lamblia.

        Evaluation of α-Tubulin as an Antigenic and Molecular Probe to Detect Giardia lamblia

        The α/β-tubulin heterodimer is the basic subunit of microtubules in eukaryotes. Polyclonal antibodies specific to recombinant α-tubulin of Giardia lamblia were made, and found effective as a probe to specifically detect G. lamblia by immunofluorescence assays. Nucleotide sequences of α-tubulin genes were compared between G. lamblia WB and GS strains, prototypes of assemblage A and assemblage B, respectively. A set of primers was designed and used to amplify a portion of the α-tubulin gene from G. lamblia. PCR-RFLP analysis of this α-tubulin PCR product successfully differentiated G. lamblia into 2 distinct groups, assemblages A and B. The results indicate that α-tubulin can be used as a molecular probe to detect G. lamblia.

        The Early Trauma Inventory Self Report-Short Form: Psychometric Properties of the Korean Version

        Objective-Experiencing traumatic events in childhood is related to various psychiatric problems in adulthood, and a comprehensive tool for measuring childhood trauma is necessary in this field. This study aimed to examine the psychometric properties, and factor structure of the Korean version of the Early Trauma Inventory Self Report-Short Form (ETISR-SF). ETISR-SF measures the childhood trauma, including physical, and emotional sexual abuse, as well as general traumas. Methods-A clinical and nonclinical samples comprising of 97 subjects from a local community, and 207 patients with the ETISR-SF, were assessed. Other tools, including the Childhood Trauma Questionnaire-Short Form (CTQ-SF), the Beck Depression Inventory (BDI), and the Beck Anxiety Inventory (BAI) were used to assess clinical symptoms. Additional data from 69 college students was used to examine the test-retest reliability. Results-The original four-factor model was supported by the confirmatory factor analysis scale [χ2 (351, n=304)=3374.025, p<0.001, TLI=0.969, CFI=0.972, RMSEA=0.030]. The ETISR-SF was found to be a reliable instrument (Cronbach’s α=0.869). Comparison of the ETISR-SF scores discriminated the clinical group from that of the control group. The measure showed good convergent and divergent validity, in that the scores were correlated higher with the scores on the CTQ-SF (0.691) than with the scores on the BDI or BAI (0.424, 0.397 respectively). The ETISR-SF was found to be temporally stable, showing the moderate to high correlation (0.844). Conclusion-These findings suggest that the Korean version of the ETISR-SF appears to be a reliable and valid instrument for the measurement of reported childhood trauma.

      • Use of Natural and Cultural Heritage in Tourism Marketing. Case of Latvia

        The strategic resources of tourism in Latvia are tangible and intangible cultural and natural heritage, which manifests itself not only in the diversity of species, but also in the landscape created by long-lasting interaction between man and nature and other resources. In the last decade, increasing attention has been devoted to the concept of "ecosystem services" whose economic and social value has not been fully appreciated. The ecosystem services are most often associated with the use of nature, but the use of cultural tourism resources is also closely linked to the ecosystem services. The main objective of the research (2017-2018) was to ascertain: 1) the ways tourism entrepreneurs and tourism information centres of municipalities use the cultural and natural heritage (including ecosystem services) as a tool for tourism product and place marketing; 2) the role of the internationally recognized brand UNESCO in marketing.

        Chronic Stress Induces Neurotrophin-3 in Rat Submandibular Gland

        Purpose: Plasma neurotrophin-3 (NT-3) levels are associated with several neural disorders. We previously reported that neurotrophins were released from salivary glands following acute immobilization stress. While the salivary glands were the source of plasma neurotrophins in that situation, the association between the expression of neurotrophins and the salivary gland under chronic stress conditions is not well understood. In the present study, we investigated whether NT-3 levels in the salivary gland and plasma were influenced by chronic stress. Materials and Methods: Expressions of NT-3 mRNA and protein were characterized, using real-time polymerase chain reactions, enzyme-linked immunosorbent assay, and immunohistochemistry,in the submandibular glands of male rats exposed to chronic stress (12 h daily for 22 days). Results: Plasma NT-3 levels were significantly increased by chronic stress (p<0.05), and remained elevated in bilaterally sialoadenectomized rats under the same condition. Since chronic stress increases plasma NT-3 levels in the sialoadenectomized rat model, plasma NT-3 levels were not exclusively dependent on salivary glands. Conclusion: While the salivary gland was identified in our previous study as the source of plasma neurotrophins during acute stress, the exposure to long-term stress likely affects a variety of organs capable of releasing NT-3 into the bloodstream. In addition, the elevation of plasma NT-3 levels may play important roles in homeostasis under stress conditions.

        A 6 Gb/s Low Power Transimpedance Amplifier with Inductor Peaking and Gain Control for 4-channel Passive Optical Network in 0.13 ㎛ CMOS

        This paper presents a 6 Gb/s 4-channel arrayed transimpedance amplifiers (TIA) with the gain control for 4-channel passive optical network in 0.13 ㎜ complementary metal oxide semiconductor (CMOS) technology. A regulated cascode input stage and inductive-series peaking are proposed in order to increase the bandwidth. Also, a variable gain control is implemented to provide flexibility to the overall system. The TIA has a maximum 98.1 ㏈Ω gain and an input current noise level of about 37.8 ㎀/㎐. The die area of the fabricated TIA is 1.9 ㎜ x 2.2 ㎜ for 4-channel. The power dissipation is 47.64 ㎽/1ch.

        Hepatitis C Virus Core Protein Activates p53 to Inhibit E6-associated Protein Expression via Promoter Hypermethylation

        E6AP (E6-associated protein)는 C형 간염바이러스(hepatitis C virus, HCV)의 코어 단백질 유비퀴틴화와 프로테오좀 분해를 유도하여 캡시드 조립을 저해함으로써 HCV 복제를 억제하는 것으로 알려져 있다. 반면에 HCV코어 단백질은 숙주의 항바이러스 방어계에 대항하고 자신의 유비퀴틴-의존적 프로테아좀 분해를 막기 위하여 DNA 메틸화를 통하여 E6AP 발현을 저해하는 전략을 진화과정에서 획득하였다. 본 연구에서는 HCV 코어 단백질이 E6AP 발현을 저해하는 기전을 밝혀내고자 하였다. HCV 코어 단백질은 HepG2 세포에서 DNA 메틸화 효소들인 DNMT1, 3a 및 3b의 단백질 수준과 효소 활성을 증가시켜 프로모터 과메틸화를 통하여 E6AP 발현을 저해하였지만 p53를 발현하지 않는 Hep3B 세포에서는 이러한 효과들이 관찰되지 않았다. 흥미롭게도 Hep3B 세포에 p53만 과발현시키면 HCV 코어 단백질이 없더라도 DNMT가 활성화되고 프로모터 과메틸화를 통하여 E6AP 발현이 저해되었다. 또한 p53 녹다운 및 과발현 실험을 통하여 p53 활성화가 HCV 코어 단백질의 효과에 필수적임을 알 수 있었다. 이로 인하여 Hep3B 보다 HepG2 세포에서 낮은 수준의 유비퀴틴화된 HCV 코어 단백질이 검출되었다. 따라서 HCV 코어 단백질은 p53-의존적으로 자신의 유비퀴틴-매개성 프로테아좀 분해를 저해한다. The E6-associated protein (E6AP) is known to induce the ubiquitination and proteasomal degradation of HCV core protein and thereby directly impair capsid assembly, resulting in a decline in HCV replication. To counteract this anti-viral host defense system, HCV core protein has evolved a strategy to inhibit E6AP expression via DNA methylation. In the present study, we further explored the mechanism by which HCV core protein inhibits E6AP expression. HCV core protein upregulated both the protein levels and enzyme activities of DNA methyltransferase 1 (DNMT1), DNMT3a, and DNMT3b to inhibit E6AP expression via promoter hypermethylation in HepG2 cells but not in Hep3B cells, which do not express p53. Interestingly, p53 overexpression alone in Hep3B cells was sufficient to activate DNMTs in the absence of HCV core protein and thereby inhibit E6AP expression via promoter hypermethylation. In addition, upregulation of p53 was absolutely required for the HCV core protein to inhibit E6AP expression via promoter hypermethylation, as evidenced by both p53 knockdown and ectopic expression experiments. Accordingly, levels of the ubiquitinated forms of HCV core protein were lower in HepG2 cells than in Hep3B cells. Based on these observations, we conclude that HCV core protein evades ubiquitin-dependent proteasomal degradation in a p53-dependent manner.
