The genus Diadegma is a well known parasitoid group in various lepidopteran insect such as Plutella xylostella andsome are known to have symbiotic virus. Previously we reported a novel ichnovirus as named Diadegma fenestrale Ichnovirus(DfIV, 62 segments, 247,238bp) from D. fenestrale which parasitized in the P. xylostella. Here we report another ichnovirus,DsIV from Taiwan strain of D. semiclausum which parasitized in the P. xylostella. Total 64 genomic segments were sequenced,185,053bp from DsIV. Two Ichnovirus showed high similarity in 99 regions in most of segments. We already confirmedintegration of DfIV genome segments into the chromosome from the P. xylostella via genome sequencing of the lepidopteranhost. Therefore some of recombination could be passible inside of the lepidopteran genome.

The habitat of the cockroach varies by species. The German cockroach (Blattella germanica) lives in human dwellings, while the Japanese field roach (Blattella nipponica) lives in a mountainous region. Based on phylogenetic analysis of mtCO I, the two species are closely related to each other and B. germanica is divergent from wild species such as B. nipponica. Their habitats and walking speed differ even though the two species have similar morphology. We hypothesized that habitats might influence walking speed via changes to appendage morphology and enzyme-based physiological differences. We found that phenotype such as appendage length and esterase isozyme expression were clearly different between the two species. These differences might be responsible for the observed difference in walking speed.
Molecular diagnosis of insecticide resistance and their application

Increasing numbers of insecticide resistant insect is one of the main problem in current agriculture. To manage the insecticide resistance, IRAC suggest some tips for farmers : First, sequence or alteration of insecticide based on MOA. Second, choosing the effective insecticide. In this point raised following question : How to distinguish the effective or non-effective insecticide. Until now, bioassay is the right answer. However that is time-consuming, labor-intensive and costly process. So we have been steadily advancing molecular based insecticide resistance diagnosis for supporting and/or substitution of classical bioassay. It can summarized following steps : First, identification of insecticide resistance mechanism. Second, marker development. Third, development molecular diagnosis such as PASA, LAMP etc. Forth, application on field collected insects. Here we discuss some case of these study particular several major pests in highland agriculture.
Potato Virus Y detection in single aphid by one step RT-PCR with boiling technique

Potato Virus Y (PVY) (Potyviridae: potyvirus) is one of the serious emerging virus of seed potato world-wide. It affects the seed potato by transmitting non-persistently via aphids. Here, we developed a simple PVY detection method which used the boiling technique for releasing of the viral RNA from aphid such as stylet and amplification by PVY specific primers located in the viral coat protein gene which suitable for various strains. This simplified method could save the time compared to earlier detection method due to the simplified RNA extraction step. Following this procedure, we tested this one step RT-PCR based PVY detection method by using three PVY vectoring aphid species (M. persicae, A. gossypii and M. euphorbiae) as well as other sucking type insect such as thrips (F. occidentalis). This PVY detection method is rapid, easy-to-use and suitable for large-scale testing in laboratories of seed potato.

It can summarized following steps : 1. Identification of insecticide resistance mechanism 2. Marker development 3. Molecular diagnosis 4. Selection of non-effective insecticide. Here we focused on the development of insecticide resistance marker. Point mutation in target site of insecticide is a good example. Up or down-regulation of specific gene and/or existence of chimeric gene is another example from Helicoverpa armigera.
도시설계기법 유출저감 효과분석을 위한 FFC2Q와 XP-SWMM 비교 연구

인적ㆍ물적 자원이 고밀화된 도시에서 발생하는 홍수 또는 침수는 도시의 기능을 마비시키거나 많은 인명피해를 발생시킬 수 있다. 본 연구에서는 유역을 재해발생지점, 재해직접영향권, 재해간접영향권으로 구분하고 각 구역별로 적합한 재해저감 도시설계기법을 적용하여 피해를 최소화하고자 한다. 이를 위해서는 우선적으로 도시설계기법을 적용하여 시뮬레이션하고 정량적으로 효과를 검증할 필요가 있다. 따라서 본 논문에서는 국내에서 사용되고 있는 도시유출모형을 활용하여 재해저감 도시설계기법의 효과 분석을 위한 적용성을 검토하였다. 검토결과로써 효과분석을 위한 FFC2Q모형, XP-SWMM과 같은 집중형 모형의 한계점을 제시하였다. The flood or inundation that occur in high-density city can paralyze urban functions and cause a lot of casualties. In this study, to minimize the damage, the disaster mitigating urban design techniques for the divided basin as disaster occurring point, disaster vulnerable site, urban responding region are applied. First of all, to do this, it is necessary to verify the effectiveness of urban design techniques by simulating them. Therefore, in this paper, the applicability of urban runoff models used in domestic disaster reduction study was investigated to analyze the outflow decrease efficiency of urban design techniques. As the reviewing results, the limitations of the lumped models such as FFC2Q and XP-SWMM are presented.
