Angelica 속 식물의 종판별을 위한 연구현황 및 전망

약용 식물 자원은 천연 화장품 소재, 제약 산업 그리고 인간의 건강을 위한 기능성 식품의 개발로 확대 된 이후 중요한 자산이 되고 있다. 그러나 세계적으로 약용 식물의 명칭을 각각 다르게 표기하고 있으며 계통 발생학적 기원이 명확하지 않다는 단점을 지니고 있다. 이 때문에 소비자들은 매우 큰 혼란을 겪고 있으며 특히 형태학적으로 유사한 식물의 말린 뿌리로 유통될 때에는 전문가들도 그들의 기원을 구별하기가 매우 어렵다. 이러한 이유때문에 이처럼 광범위하고 다양한 작물의 기원을 식별하기 위해 분자표지 기법을 적용하여 활용되고 있다. 이 리뷰에서 본 연구자들은 Angelica종의 분화에 관한 적합한 '기원정립'을 위한 현재의 연구 성과들을 정리했다. 결론적으로 Angelica종의 식별을 위해 개발된 분자적 연구에 대해 설명하고 약용작물의 유전체 정보를 활용하여 그들의 기원정립 및 판별에 대해 논의할 것이다. Medicinal plants resources are becoming important assets since their usages have been expanded to the development of functional foods for human health, natural cosmetics, and pharmaceutical industries. However, names are different from each country and their phylogenetic origins are not clear. These lead consumers to be confused. In particular, when they are morphologically similar and distributed as dried roots, it is extremely difficult to differentiate their origins even by specialists. Recently, molecular markers have been extensively applied to identify the origin of many crops. In this review, we tried to overview the current research achievements for the development of suitable 'origin identification' regarding to the differentiation of Angelica species. Furthermore, more advanced techniques including amplification genome based marker analyses are also discussed for their practical applications in the authentication of particular medicinal plant in Angelica species.
해당화 열매 성숙단계에 따른 생리활성 및 기능성 물질 변화 분석

열매가 성숙하는 과정은 유전학적으로 프로그램화 되어 있는 과정으로 다른 여러 유전자 발현과 효소의 작용에 의한 생화학적, 생리적 조절의 결과물이다. 이런 일련의 과정에서 과실과 채소에 화학적 조성과 활성 물질에 변화가 나타나는 것을 당연한 결과일 것이다. 해당화의 열매가 성숙하는 과정에서 이들 활성 물질의 변화를 측정하기 위해 우리는 성숙단계 별로 채집 된 열매 시료 추출물에서의 항산화 활성의 차이, 항 elastase 활성 및 polyphenol 화합물의 함량을 분석하였다. 결과적으로 미성숙 단계의 열매 추출물에서 높은 phenolic 화합물과 플라보노이드 함량이 검출되었으며, 그에 따라 높은 radical 소거능, 환원력 및 ORAC 활성을 나타냈다. 또한 elastase 저해 활성에서도 다른 성숙 단계의 추출물에 비해 미성숙 단계의 열매 추출물에서 높은 활성을 나타내는 것으로 조사되었다. 더불어 성숙과정에서 플라보노이드 생합성 과정에 관련된 유전자의 발현이 감소하는 것을 확인하였으며, 이를 통해 플라보노이드의 함량이 감소하는 것을 설명할 수 있었다. HPLC분석을 통해 다른 성숙 단계의 추출물과 비교하여 미성숙 열매 추출물에서 높은 함량의 myricetin, caffeic acid, chlorogenic acid, syringic acid, р-coumaric acid 등이 검출되었다. 구조 기반의 molecular doking을 사용하며 이들 화합물과 elastase의 결합부위와 패턴을 분석하였으며 이를 통해 chlorogenic acid와 elastase가 강하게 결합하는 것을 확인하였다. 따라서, 본 연구를 통해서 해당화 열매의 성숙 정도가 이 식물의 약리학적 가치에 영향을 미치는 것을 규명하였고, 미성숙한 해당화 열매는 향후 활성산소 생성에 의해 유발되는 피부 노화 억제 효과뿐 아니라 피부의 탄력성 강화를 위한 기능성 천연소재로서의 가능성을 입증하였다. Fruit ripening is a genetically programmed process involving a number of biochemical and physiological processes assisted by variations in gene expression and enzyme activities. This process generally affects the phytochemical profile and the bioactive principles in fruits and vegetables. To appraise the variation in bioactive principles of fruits from Rosa rugosa during its ripening process, we analyzed the changes in antioxidant and anti-elastase activities and polyphenolic compounds during the four ripening stages of fruits. Overall, an extract of unripe fruits contained the highest levels of total phenolic and flavonoid contents, radical scavenging activity, reducing power, oxygen radical antioxidant capacity, and elastase inhibitory activity, compared with the extracts of fruits at other stages of ripening. Additionally, we found that the reduction of flavonoid content occurs because of decreased transcriptional levels of genes involved in flavonoid biosynthesis pathway during the ripening process. Based on HPLC analysis, we found that the extract of unripe fruits contained the highest amount of myricetin, caffeic acid, chlorogenic acid, syringic acid, and p-coumaric acid and suggested that the antioxidant and anti-elastase activities of the extract obtained from stage 1, should be mediated by the presence of these compounds. Additionally, we analyzed the interaction sites and patterns between these compounds and elastase using the structure-based molecular docking approach, and suggested that chlorogenic acid strongly interacted with elastase. Together, these findings suggest that the maturity of fruits has profound effects on the pharmaceutical value of R. rugosa.
