Effective Cryopreservation Method by Controlling the Clump Size of Human Embryonic Stem Cells

Human embryonic stem cells (hESCs) have the potential for use in regenerative medicine and in the field of basic research. Therefore, effective cryopreservation and storage of hESCs are important for preservation of newly established cell lines for various purposes. Despite poor survival and slow recovery after thawing, the con-ventional slow freezing method is most commonly used for cryopreservation of hESCs due to its simplicity and ease of use for freezing a large number of hESCs appropriate to clinical applications. Here we controlled clump size(Group I; 400~450μm, Group II; 800~900μm, and Group III; 1500~1700μm) of hESCs at 5 days after plating using a glass pipette during cryopreservation in order to obtain a larger amount of hESCs after thawing. Attachment rates differed significantly (p<0.05) in each of the three groups and the average attachment rate of Group II was highest in SNUhES4 and H1. In particular, the attachment rate of Group II in SNUhES3 showed a significant improvement with ROCK inhibitor Y-27632. These results indicate that clump size and cell-cell adhesions of Group II are appropriate for cryopreservation compared to Group I and Group III. This method increased cell viability and reduced the recovery time leading to various experiments, and therefore has an advantage for use with hESCs like newly established in particular. We demonstrate that use of this cryopreservation method with control of the clump size of hESCs can effectively improve the attachment rate and survival of post-thaw hESCs with and without Y-27632.

The purpose of this study was to investigate the effects of sleep/wake behavior for shiftwork in air traffic controllers (ATCs) using wrist actigraph for ten workers on a continuous full-day three-team three-shift system of forward rotation including on-duty and off-duty periods. The wrist actigraph data were recorded for three days (one shift cycle) for each subject. The mean activity counts during an on-duty period progressively increased from the night, the swing, to the morning shifts. The doze length during on-duty periods showed decreases in the morning and swing shifts as compared to the night shift. Total sleep time (TST) and sleep efficiency (SE) during off-duty periods increased in the morning-1 and swing-night shift compared to the morning-2 shift. Finally, I discussed the role of doze-taking during the burden on night shift ATCs.
Fostering Attitudes and Customer Satisfaction for Sustainability by Electric Car-Sharing

Purpose: The purpose of this paper is to explore factors that affect electric car-sharing by highlighting issues of environmental sustainability and customer attitude and satisfaction. Research design, data and methodology: This study examined customers‟ perception on those issues by using electric car-sharing as usages of both electric car and car-sharing play a key role to improve sustainability. Online survey was applied to collect the data. This study also applied factor and regression analyses for data analysis. Results: The results of this study showed that effects of proposed factors including cost efficiency, emotion, safety, health, and sustainability on attitude toward electric car-sharing on attitude were significant. The results also showed that the effects on intention, satisfaction, and loyalty were significant. Conclusions: This study provides policy and managerial implications. By dealing with factors of electric car-sharing service, this study offers necessity of better strategies and policies for electric car-sharing service to electric car businesses and policy makers. This study also suggests that businesses should develop appropriate strategies for the improved usage of electric car-sharing by considering sustainability and improving relationships with customers. Further, government should consider to develop proper policies for sustainability by promoting the usages of electric car-sharing.

In this paper, we present the improvement in low moisturizing ability and stability that existing skin softeners have due to the low oil content, by developing skin softener using nanoemulsion of phospholipid liposome, based on the properties of nanoemulsion in cosmetic formulation. In this study, two types of oil; dimethicone (DC 200/6cs) or medium chain triglyceride (MCT), and two kinds of lecithin; unsaturated or saturated were respectively applied to produce nanoemulsion. In the particle size analysis of nanoemulsion, the droplet size of nanoemulsion containing DC200/6cs and unsaturated lecithin was the smallest, and all nanoemulsion showed high stability in the measurement of zeta potential. Therefore, with the smallest particle size and high stability, moisture contents and trans epidermal water loss(TEWL) were measured using the skin softener of DC200/6cs and unsaturated lecithin contained nanoemulsion, and the measurement was compared with the non-oil skin softener and the skin softener with only small amount of oil. The results showed that the moisture content of the skin softener using nanoemulsion increased greatly than other two skin softeners, showing high hydration ability and water retention capacity, and TEWL decreased greatly, therefore preventing the evaporation of moisture from the skin. As a result, the oil content and stability of the skin softener was improved by utilizing nanoemulson based of phospholipid liposome, and it is expected to be used in various ways in cosmetic industry.
