        Construction of graphene/AuNPs based amperometric immunosensor for detecting bladder cancer biomarker apolipoprotein A1

        This paper presents the construction and characterization of an amperometric immunosensor based on the graphene/gold nanoparticles (AuNPs/GO) nanocomposite for the detection of the bladder cancer biomarker, apolipoprotein A1 (Apo-A1). The morphological analysis of the AuNPs/GO nanocomposite demonstrated an almost spherical shape of AuNPs and the successful coverage of their surface by graphene oxide. An increased G peak and decreased D peak after the association of AuNPs with GO, implied a reduction in graphene defects. The X-ray photoelectron spectroscopy (XPS) indicated a significant decrease in the quantity of oxygen-containing functional groups in the AuNPs/GO nanocomposite, as compared to the original GO. Furthermore, the developed sensor demonstrated commendable sensitivity and selectivity, with a wide linear range for Apo-A1 detection. Importantly, the immunosensor exhibited remarkable stability over a period of 14 days, signifying its potential for practical applications.

        Quantile regression for robust inference on varying coefficient partially nonlinear models

        In this paper, we propose a robust statistical inference approach for the varying coefficient partially nonlinear models based on quantile regression. A three-stage estimation procedure is developed to estimate the parameter and coefficient functions involved in the model. Under some mild regularity conditions, the asymptotic properties of the resulted estimators are established. Some simulation studies are conducted to evaluate the finite performance as well as the robustness of our proposed quantile regression method versus the well known profile least squares estimation procedure. Moreover, the Boston housing price data is given to further illustrate the application of the new method.

      • Accessibility Assessment Via Workspace Estimation

        The process of evaluating a built environment for accessibility is known as “accessibility assessment.” Determining accessibility is closely related to the problem of determining possible motions of a specific kinematic structure – given an environment and a mobile device, how much of the environment is accessible? Given these similarities, here the accessibility assessment process is reformulated as a motion planning problem. Rather than treating each of the degrees of freedom ‘equally’ while planning, we explore a hierarchical characteristic of all of the degrees of freedom when constructing the roadmap. The approach is demonstrated on simulated environments as well as on a student residence at York University.

      • Lateral Control for Vehicles’ Automatic Steering with Front Sensor and GPS

        This paper proposes an automated steering control system for passenger cars. The control strategy used is a look-down reference system which uses a sensor at the front bumper to measure the lateral displacement and a GPS to measure the heading orientation. Firstly, the road curvature estimator is designed based on the steering angle. Secondly, the steering angles can be estimated by using the sensor and the GPS information. The estimation algorithm employed is an observer for a new extended system model, in which the steering angle and its derivative are viewed as two state variables. Then, the lateral displacements estimation and the control algorithm are investigated. An accurate and real-time estimation of the lateral displacements with respect to the road can be accomplished in such a control system. The closed loop controller is used as a compensator to control the lateral dynamics of the vehicle. The vehicle dynamic and control system are simulated with Matlab Simulink. The proposed estimation and control algorithms are validated by simulation results. The results show that the system provides a good and robust performance for vehicle to follow or track a reference path.

        Taurocholic acid promotes hepatic stellate cell activation via S1PR2/p38 MAPK/YAP signaling under cholestatic conditions

        Background/Aims: Disrupted bile acid regulation and accumulation in the liver can contribute to progressive liver damage and fibrosis. However, the effects of bile acids on the activation of hepatic stellate cells (HSCs) remain unclear. This study investigated the effects of bile acids on HSC activation during liver fibrosis, and examined the underlying mechanisms. Methods: The immortalized HSCs, LX-2 and JS-1cells were used for the in vitro study. in vitro, the adeno-associated viruses adeno-associated virus-sh-S1PR2 and JTE-013 were used to pharmacologically inhibit the activity of S1PR2 in a murine model of fibrosis induced by a 3,5-diethoxycarbonyl-1,4-dihydrocollidine (DDC) diet. Histological and biochemical analyses were performed to study the involvement of S1PR2 in the regulation of fibrogenic factors as well as the activation properties of HSCs. Results: S1PR2 was the predominant S1PR expressed in HSCs and was upregulated during taurocholic acid (TCA) stimulation and in cholestatic liver fibrosis mice. TCA-induced HSC proliferation, migration and contraction and extracellular matrix protein secretion were inhibited by JTE-013 and a specific shRNA targeting S1PR2 in LX-2 and JS-1 cells. Meanwhile, treatment with JTE-013 or S1PR2 deficiency significantly attenuated liver histopathological injury, collagen accumulation, and the expression of fibrogenesis-associated genes in mice fed a DDC diet. Furthermore, TCAmediated activation of HSCs through S1PR2 was closely related to the yes-associated protein (YAP) signaling pathway via p38 mitogen-activated protein kinase (p38 MAPK). Conclusions: TCA-induced activation of the S1PR2/p38 MAPK/YAP signaling pathways plays a vital role in regulating HSC activation, which might be therapeutically relevant for targeting cholestatic liver fibrosis.

        Genetic variation and population structure of the oriental fruit moth Grapholita molesta in Shanxi, a major pome fruits growing region in North China

        Grapholitamolesta, commonly known as the oriental fruit moth, is one of the most destructive fruit pests both in China and in other regions of the world. We examined the genetic diversity and population structures of G. molesta populations from 11 locations in a major pome fruit production region (Shanxi province) in North China using seven microsatellite markers and sequence variations in two mitochondrial genes. We found that G.molesta in the studied region exhibited high levels of genetic diversity. Among 271 individual insects examined using microsatellite markers, mean number of alleles ranged from 5.86 to 9.00 with average 8.01. Population structure analysis revealed that populations from the eleven sampling sites formed two clusters without apparent distance isolation. Cluster 1 consisted of eight populations distributed in five regions of the sampling sites, whereas Cluster 2 consisted of three populations distributed in two distant locations of the sampling sites. Genetic exchange analysis revealed that about half of the haplotypes were shared among over 90% individual insects from different regions. Our results indicated that the population structure and genetic exchange of G. molesta in the studied region might have been affected by limited dispersal ability of the insect, environmental condition, colonization history and human-mediated dispersal. These findings should provide useful information for the implementation of area-wide pest management and for further characterization of genetic variation and population structure of this economically important pest.

        AKA-PLA: Enhanced AKA Based on Physical Layer Authentication

        Existing authentication mechanisms in cellular mobile communication networks are realized in the upper layer by employing cryptographic techniques. Authentication data are broadcasted over the air in plaintext, enabling attackers to completely eavesdrop on the authentication and get some information about the shared secret key between legitimate nodes. Therefore, reusing the same secret key to authenticate several times results in the secret key`s information leakage and high attacking rate. In this paper, we consider the most representative authentication mechanism, Authentication and Key Agreement (AKA), in cellular communication networks and propose an enhanced AKA scheme based on Physical Layer Authentication (AKA-PLA). Authentication responses generated by AKA are no longer transmitted in plaintext but masked by wireless channel characteristics, which are not available to adversaries, to generate physical layer authentication responses by a fault-tolerant hash method. The authenticator sets the threshold according to the authentication requirement and channel condition, further verifies the identity of the requester based on the matching result of the physical layer authentication responses. The performance analyses show that the proposed scheme can achieve lower false alarm rate and missing rate, which are a pair of contradictions, than traditional AKA. Besides, it is well compatible with AKA.

        Barrier Lyapunov Functions-based Output Feedback Control for a Class of Nonlinear Cascade Systems With Time-varying Output Constraints

        In this study, a class of nonlinear systems with integral input to state stability (iISS) inverse dynamics and unknown control direction are examined for the issue of time-varying asymmetric output constraints of adaptive output feedback controller. To deal with unmeasured state variables and unknown directions, the state observer is constructed using a Rickati matrix differential equation with time variation. A backstepping-based method is recommended for establishing the dynamic output feedback control law. By ensuring boundedness for the timedependent barrier Lyapunov function (BLF) in the closed loop, we may not only maintain the boundedness and stability of other signals, but also avoid breaking the time-varying asymmetric constraint of the output. Finally, simulation analyses are used to confirm the scheme’s efficacy.
