Optimization of a Synthesis Process for Octenylsuccinic Anhydride Modified Gum Arabic

Gum arabic (GA) was esterified using octenylsuccinic anhydride (OSA) in an aqueous solution. HPLC was used to analyze the bound OS content (%OS) after esterification, and total OS content (%OStotal) results from HPLC were in agreement with titration results. The effects of temperature, reaction time, pH value, and GA concentration on the %OS value were determined using single factor experiments. Temperature, time, and pH were used for further optimization. Process parameters for preparation of octenylsuccinic anhydride modified gum arabic (OS-GA) were optimized using response surface methodology (RSM). The theoretical optimum conditions were a temperature of 37.9oC, a reaction time of 116.4 min, and a pH of the reaction system=7.99. Under optimum conditions, the %OS value of OS-GA was 2.07%.
A SPACE-TIME DISCONTINUOUS GALERKIN METHOD FOR FIRST ORDER HYPERBOLIC SYSTEMS

We present a new space-time discontinuous Galerkin (DG) method for solving the time dependent, positive symmetric hyperbolic systems. The main feature of this DG method is that the discrete equa- tions can be solved semi-explicitly, layer by layer, in time direction. For the partition made of triangle or rectangular meshes, we give the stability analysis of this DG method and derive the optimal error estimates in the DG-norm which is stronger than the L₂-norm. As application, the wave equation is considered and some numerical experiments are provided to illustrate the validity of this DG method.

In order to develop the application of the natural polysaccharide lentinan (LNT) and decrease the side effects of doxorubicin, we successfully synthesized a novel folate-modified maleilated lentinan-doxorubicin (FA-M.LNT-DOX) polymer and used it for tumor-targeted drug delivery. The release efficiency and cytotoxicity of the prodrugs were evaluated in vitro. Although DOX release from FA-M.LNT-DOX was quite slow in a neutral buffer, it was particularly fast in an acidic solution with a pH of 5.0. Compared with DOX, FA-M.LNT-DOX showed higher cytotoxicity in HeLa cells and significantly lower cytotoxicity in normal cells. These results suggested that FA-M.LNT-DOX could be considered as a potential drug delivery candidate for folate receptor-positive cancer therapy.
