        Toll-like Receptor3-mediated Induction of Chemokines in Salivary Epithelial Cells

        Toll-like receptors (TLRs) functionally expressed in salivary epithelial cells, but their roles remain elusive. Among TLRs family, TLR3 is activated by dsRNA, a byproduct of viral infection. The aim of this study was to investigate the role of TLR3 in the inflammatory immune responses using HSG cells. Reverse transcriptase-polymerase chain reaction (RT-PCR), real-time PCR and ELISA were performed to identify expression of TLRs and TLR3-mediated chemokine inductions. The chemotaxis assay of activated T lymphocytes was also performed. Treatment of HSG cells with polyinosinic: polycytidylic acid (poly(I:C)) significantly increased interferon-γ-inducible protein 10 (IP-10), interferon- inducible T-cell α chemoattractant (I-TAC), and regulated on activation, normal T-cells expressed and secreted (RANTES) gene expressions in a concentration-dependent manner. Anti-TLR3 antibody blocked the increases of IP-10 and I-TAC genes. Poly(I:C)-induced increases of IP-10 and I-TAC were also confirmed at protein levels from cell lysates, but their release into extracellular medium was detected only in IP-10. We found that the culture media from HSG cells stimulated with poly(I:C) significantly increases T lymphocyte migration. Our results suggest that TLR3 plays an important role in chemokine induction, particularly IP-10, in salivary epithelial cells.

        A Novel Recognition Algorithm Based on Holder Coefficient Theory and Interval Gray Relation Classifier

        The traditional feature extraction algorithms for recognition of communication signals can hardly realize the balance between computational complexity and signals` interclass gathered degrees. They can hardly achieve high recognition rate at low SNR conditions. To solve this problem, a novel feature extraction algorithm based on Holder coefficient was proposed, which has the advantages of low computational complexity and good interclass gathered degree even at low SNR conditions. In this research, the selection methods of parameters and distribution properties of the extracted features regarding Holder coefficient theory were firstly explored, and then interval gray relation algorithm with improved adaptive weight was adopted to verify the effectiveness of the extracted features. Compared with traditional algorithms, the proposed algorithm can more accurately recognize signals at low SNR conditions. Simulation results show that Holder coefficient based features are stable and have good interclass gathered degree, and interval gray relation classifier with adaptive weight can achieve the recognition rate up to 87% even at the SNR of -5dB.

        A New Robust Signal Recognition Approach Based on Holder Cloud Features under Varying SNR Environment

        The unstable characteristic values of communication signals along with the varying SNR (Signal Noise Ratio) environment make it difficult to identify the modulations of signals. Most of relevant literature revolves around signal recognition under stable SNR, and not applicable for signal recognition at varying SNR. To solve the problem, this research developed a novel communication signal recognition algorithm based on Holder coefficient and cloud theory. In this algorithm, the two-dimensional (2D) Holder coefficient characteristics of communication signals were firstly calculated, and then according to the distribution characteristics of Holder coefficient under varying SNR environment, the digital characteristics of cloud model such as expectation, entropy, and hyper entropy are calculated to constitute the three-dimensional (3D) digital cloud characteristics of Holder coefficient value, which aims to improve the recognition rate of the communication signals. Compared with traditional algorithms, the developed algorithm can describe the signals` features more accurately under varying SNR environment. The results from the numerical simulation show that the developed 3D feature extraction algorithm based on Holder coefficient cloud features performs better anti-noise ability, and the classifier based on interval gray relation theory can achieve a recognition rate up to 84.0%, even when the SNR varies from -17dB to -12dB.

        중국의 코로나19 대응은 성공적인가? : 국내와 국제적 대응 관점에서

        2019년 발생한 코로나 19는 동아시아는 물론 유럽 전역에까지 일순간에 확산되었고, 인류가 관여하는 거의 모든 분야에서 심각한 피해를 입었다. 미국과 중국 간의 패권전쟁으로 긴장감이 날로 높아지고 있다. 코로나19 발생의 진원지 문제는 미국과 중국의 갈등을 증폭시키는 계기가 되었고, 백신의 공공재화 문제는 백신 외교전으로 까지 확산되었다. 중국의 코로나19 방역 대응은 단순히 바이러스의 전파를 막기 위한 행위가 아니라 국내적으로는 체제안정과 국외적으로 중국의 부정적 이미지 개선과 리더십 회복을 위한 적극적 대응 전략으로 봐야 할 것이다. 본 연구는 중국의 코로나19 방역에서 대응전략 및 효과와 문제점을 알아보고자 하는 것이다. 국내적 대응으로 중국의 중앙과 지방정부의 대처, 대중 참여를 알아본다. 국제적 대응으로 국제사회의 코로나19 진원지 문제와 백신개발 문제를 알아보고 평가한다. Corona 19, which occurred in 2019, spread in an instant to not only East Asia but also to all of Europe, and suffered serious damage in almost every field in which mankind is involved. Tensions are rising day by day due to the war for hegemony between the United States and China. The issue of the origin of the outbreak of COVID-19 served as an opportunity to amplify the conflict between the United States and China and the issue of public goods of vaccines has spread to the vaccine diplomacy war. China’s COVID-19 quarantine response is not just an act to prevent the spread of the virus, but domestic system stability and abroad, it should be viewed as an active response strategy for improving China’s negative image and restoring leadership. The purpose of this study is to investigate the countermeasures, effects, and problems in the prevention and control of COVID-19 in China. As a domestic response, it examines the response of China’s central and local governments and public participation. As an international response, the international community examines and evaluates the issue of the origin of COVID-19 and the issue of vaccine development.

      • Expression of the Na<sup>+</sup>-HCO<sub>3</sub><sup>−</sup>cotransporter and its role in pH<sub>i</sub>regulation in guinea pig salivary glands

        <P>Patterns of salivary HCO3<SUP>−</SUP>secretion vary and depend on species and gland types. However, the identities of the transporters involved in HCO3<SUP>−</SUP>transport and the underlying mechanism of intracellular pH (pHi) regulation in salivary glands still remain unclear. In this study, we examined the expression of the Na<SUP>+</SUP>-HCO3<SUP>−</SUP>cotransporter (NBC) and its role in pHiregulation in guinea pig salivary glands, which can serve as an experimental model to study HCO3<SUP>−</SUP>transport in human salivary glands. RT-PCR, immunohistochemistry, and pHimeasurements from BCECF-AM-loaded cells were performed. The amiloride-sensitive Na<SUP>+</SUP>/H<SUP>+</SUP>exchanger (NHE) played a putative role in pHiregulation in salivary acinar cells and also appeared to be involved in regulation in salivary ducts. In addition to NHE, NBC also played a role in pHiregulation in both acini and ducts. In the parotid gland, NBC1 was functionally expressed in the basolateral membrane (BLM) of acinar cells and the luminal membrane (LM) of ducts. In the submandibular gland, NBC1 was expressed only in the BLM of ducts. NBC1 expressed in these two types of salivary glands takes up HCO3<SUP>−</SUP>and is involved in pHiregulation. Although NBC3 immunoreactivity was also detected in submandibular gland acinar cells and in the ducts of both glands, it is unlikely that NBC3 plays any role in pHiregulation. We conclude that NBC1 is functionally expressed and plays a role in pHiregulation in guinea pig salivary glands but that its localization and role are different depending on the type of salivary glands.</P>

        Ca2+-activated K+ Currents of Pancreatic Duct Cells in Guinea-pig

        There are numerous studies on transepithelial transports in duct cells including Cl- and/or HCO3-. However, studies on transepithelial K+ transport of normal duct cells in exocrine glands are scarce. In the present study, we examined the characteristics of K+ currents in single duct cells isolated from guinea pig pancreas, using a whole-cell patch clamp technique. Both Cl- and K+ conductance were found with KCl rich pipette solutions. When the bath solution was changed to low Cl-, reversal potentials shifted to the negative side, -75±4 mV, suggesting that this current is dominantly selective to K+. We then characterized this outward rectifying K+ current and examined its Ca2+ dependency. The K+ currents were activated by intracellular Ca2+. 100 nM or 500 nM Ca2+ in pipette significantly (P<0.05) increased outward currents (currents were normalized, 76.8±7.9 pA, n=4 or 107.9±35.5 pA, n=6) at +100 mV membrane potential, compared to those with 0 nM Ca2+ in pipette (27.8±3.7 pA, n=6). We next examined whether this K+ current, recorded with 100 nM Ca2+ in pipette, was inhibited by various inhibitors, including Ba2+, TEA and iberiotoxin. The currents were inhibited by 40.4±% (n=3), 87.0±% (n=5) and 82.5±% (n=9) by 1 mM Ba2+, 5 mM TEA and 100 nM iberiotoxin, respectively. Particularly, an almost complete inhibition of the current by 100 nM iberiotoxin further confirmed that this current was activated by intracellular Ca2+. The K+ current may play a role in secretory process, since recycling of K+ is critical for the initiation and sustaining of Cl- or HCO3- secretion in these cells.
