        Functional Characterization of aroA from Rhizobium Leguminosarum with Significant Glyphosate Tolerance in Transgenic Arabidopsis

        Glyphosate is the active component of the top-selling herbicide, the phytotoxicity of which is due to its inhibition of the shikimic acid pathway. 5-Enolpyruvylshikimate-3-phosphate synthase (EPSPS) is a key enzyme in the shikimic acid pathway. Glyphosate tolerance in plants can be achieved by the expression of a glyphosate-insensitive aroA gene (EPSPS). In this study, we used a PCR-based two-step DNA synthesis method to synthesize a new aroA gene (aroAR. leguminosarum) from Rhizobium leguminosarum. In vitro glyphosate sensitivity assays showed that aroAR. leguminosarum is glyphosate tolerant. The new gene was then expressed in E. coli and key kinetic values of the purified enzyme were determined. Furthermore, we transformed the aroA gene into Arabidopsis thaliana by the floral dip method. Transgenic Arabidopsis with the aroAR. leguminosarum gene was obtained to prove its potential use in developing glyphosate-resistant crops.

        對于泰山皮影희德個案考察

        Mountain Tai Shadow Play is kind of folk art originated in the area around Mountain Tai in Shandong Province. In June 2008, it was packed with the other 11 kinds of shadow plays, such as Beijing Shadow Play, Hejian Shadow Play and Xiuyan Shadow Play, etc. into one cultural item namely "Shadow Play" and entered the first batch of the Expanded Inventory of National Intangible Cultural Heritage. (NO, IV-91) Against the fact that this art is dying, there is very rare research about it yet. From Dec. 2007, I started to know the only inheritor of this art Mr. Fan ZhengAn and did interviews with him for a few times, which is the basis of this paper. This paer tries to demonstrate the uniqueness of this tradition through working on the following three aspects: Firstly, to define the historical evolution, the main characteristics as well as the contemporary living status of Mountain Tai Shadow Play under the local context; Secondly, to explore the training process of an apprentice to become a qualified artist; thirdly. to argue that the tradition of Mountain Tai Shadow keeps growing rather than perishing through a dynamic mechanism of innovation and the ardent efforts of the artist.

      • Differential Diagnosis of CT Images in Children with Neuroblastomas and Ganglioneuroblastomas

        Objective: To investigate the differential features of CT images in children with neuroblastomas (N) and ganglioneuroblastomas (G). Materials and Methods: Clinical data of 12 children in group G and 15 in group N undergoing CT examination and definitely diagnosed by pathology were retrospectively analyzed. The focal conditions were observed and compared in the two groups, including location, size, boundaries, morphology, enhanced degree and mode, abdominal vascular involvement, presence or absence of spanning the midline, infiltration of peripheral organs, angiography manifestations in tumors or surroundings, presence or absence of calcification and vascular tumor emboli as well as metastases of distal organs and lymph nodes. Results: In group N, the incidence of tumors in the adrenal area was conspicuously higher than in group G (P<0.05), while that of tumors with regular morphology and clear boundaries was significantly lower than in group G (P<0.01); Angiography manifestation rate and incidences of vascular embedding, lymph node metastasis, infiltration and organic metastasis in group N were all markedly higher than in group G (P<0.05). There was no statistical significance between the two groups in terms of focal size, presence or absence of calcification and spanning the midline, and enhanced degree and mode, as well as vascular tumor emboli (P>0.05). Conclusions: Mostly located in adrenal areas and with vascular embedding as a primary manifestation, the neuroblastoma extremely readily metastases to lymph nodes and other organs as well as infiltrating local tissues, with dilation on angiography frequent in or around the tumors. With vascular displacement as a primary manifestation, ganglioneuroblastoma has a regular morphology and clear boundaries.

        Experimental Study on Shear Behavior of Bolted Cement Mortar Blocks under Constant Normal Stiffness

        In this paper, the influence of bolting on shear mechanical behavior of cement mortar specimens were studied. First, a shear test apparatus which could provide CNS (constant normal stiffness) condition was designed. Then, more than 140 bolted and non-bolted cement mortar blocks (100 × 100 × 100 mm) were prepared. The strengthening effect of bolts with diameters of 0 − 10 mm on cement mortar specimens was studied under different initial normal loads ranging from 20 kN to 60 kN and constant normal stiffness of 420 kN/mm. Two simplified models of shear behavior of reinforced cement mortar blocks were proposed. The most important influence of bolt on the cement mortar blocks was to strengthen the post-peak shear strength rather than the peak shear strength, and the strengthening effect was reduced under high initial normal force. The cohesion of cement mortar blocks was reduced while internal friction angle was increased in residual sliding stage by bolts. The dilation of reinforced cement mortar specimens in postpeak shear stage was significantly reduced due to bolting, and the larger the bolt diameter, the greater the reduction in the block dilation.

        Long noncoding RNA UCA1 inhibits ischaemia/reperfusion injury induced cardiomyocytes apoptosis via suppression of endoplasmic reticulum stress

        Background Ischemia heart disease is one of the major causes of death worldwide which often associated with tissue infarction and limit the recovery of function. Multiple factors involved in the I/R-induced cardiomyocyte dysfunction which were consistent with a role of oxidative stress and altered endothelium-dependent responses. However, the pathogenic mechanisms in I/R injury remain unclear. Materials and methods The H9C2 cells were in the ischaemia/reperfusion (I/R) condition. After I/R, the cells were transfected with or without adenovirus-urothelial carcinoma associated 1(Ad-UCA1). Then qRT-PCR analysis was performed to quantify mRNA expression of different treatment groups. Cell apoptosis rate was assessed using flow cytometry and ER stress biomarker expression were measured by immunoblotting. Intracellular and mitochondrial ROS generation were assayed by fluorescence microscope after staining with the DCFDA or MitoSOX. Results I/R conditions trigger lncRNAs UCA1 expression, cellular and mitochondria ROS production, resulting in cell apoptosis through the induction of oxidative and ER stress. Overexpression of UCA1 protects H9C2 cells from I/R-induced ER stress and cell apoptosis. Moreover, UCA1 might be a potential regulator in the protective effect of I/R‑induced oxidative stress and mitochondria dysfunction. Subsequently, ER stress inhibitor attenuated the effect of siUCA1 induced injury in H9C2 cells. Conclusion The expression of UCA1 against I/R induced oxidative stress and mitochondria dysfunction via suppression of endoplasmic reticulum stress. UCA1 might be a biomarker to improved diagnosis of I/R injury.

        Genome-wide identification of the ARRs gene family in tobacco (Nicotiana tabacum)

        Background The growth of axillary buds determines the shoot branching and morphology of plants, and its initiation and development are regulated by a series of hormonal signals, such as cytokinin. Arabidopsis response regulators (ARRs) can regulate the growth and development, disease resistance and stress resistance of plants by participating in cytokinin signaling. Objective To explore the distribution and expression pattern of ARR members in tobacco. Methods The identifcation, isoelectric points, molecular weights, protein subcellular localization prediction, multiple sequence alignment, phylogenetic analysis, protein motifs and structures, chromosome distributions of deduced ARR proteins were conducted. The gene expression profling of various tissues in response to topping, low temperature and drought were analyzed by RNA-seq and qRT-PCR. Results 59 ARR genes from cultivated tobacco (Nicotiana tabacum) were identifed, namely NtARRs, including 21 type A NtARRs and 38 type B NtARRs. The 59 NtARRs were expressed mainly in all organs except the fruits. Some representative NtARRs may participate in axillary bud initiation and development, as well as in stress resistance through cytokinin signal transduction. Conclusion Understanding the roles of NtARRs in the molecular mechanisms responsible for axillary bud growth and stress tolerance could aid in targeted breeding in crops.

        Label-Free Fluorescent Detection of Hg2+ in Aqueous Media Based on N-Doped MoS2 Nanosheets

        The incorporation of dopant atoms alters the structure of MoS2, resulting in unique properties and practical applications. Herein, a facile and low-cost approach for producing N-doped molybdenum disulfide (N-MoS2) was carried out using melamine and MoS2 in a laboratory tube furnace at 400 ℃ for 2h. The N-MoS2 fluorescent probe showed sensitive response to Hg2+ in a wide concentration range of 0.4–10 μM with a low limit of 28 nM. The practical applicability of detecting Hg2+ in lake water samples may open a new way to evaluate Hg2+ in wastewater.

        Elevating Limonene Production in Oleaginous Yeast Yarrowia lipolytica via Genetic Engineering of Limonene Biosynthesis Pathway and Optimization of Medium Composition

        As an alternative terpenoid producer, nonconventional oleaginous yeast Yarrowia lipolytica was chosen for limonene production. Y. lipolytica can give high biomass yield and metabolize a broad range of substrates such as glycerol, alkanes, fatty acid, fats, and oils. As previously reported, optimization of limonene synthesis pathway and mevalonate (MVA) pathway leads to the accumulation of 112-fold higher limonene as compared to an initial strain. In this study, we introduced an additional copy of limonene synthesis gene (LS), which resulted in an increase of limonene production. This engineered strain was used to carry out further optimization study. Amongst all the carbon sources tested, the highest level of limonene production was obtained from glycerol, and citrate was selected as an auxiliary carbon source. In fed-batch fermentation with an optimized medium, the engineered strain was found to produce 165.3 mg/L limonene, which corresponds to the highest yield till date for the production of limonene in Y. lipolytica.
