      • Integration of Design of Experiments into Quality Function Deployment

        Quality Function Deployment (QFD) is a useful planing process in new product development. It achieves the maximum custom satisfaction through translating Voice of the Customer (VOC) into voice of the engineer in the House of Quality (HOQ), and ensures the customer focus from house to house translation. In QFD, the relationship matrix and correlation matrix are important parts, which will influence the final priorities of the technical response. According to the traditional methods, these matrices are determined by the people's experience and filled with ordinal numbers. It would inevitably result in inaccuracy and curtness. Design of Experiments (DOE) is employed to determine the effect of variables on the process. In this paper, the author puts forward the idea of integrating DOE into the process of QFD. This integration helps to produce scientific decision and robust product design. A framework on QFD/DOE is presented and explanations are also made.

        Investigation of Proteomic Responses of Streptomyces lydicus to Pitching Ratios for Improving Streptolydigin Production

        Streptomyces lydicus has been reported to produce antibiotic streptolydigin. Pitching ratios play crucial roles in primary and secondary metabolism of Streptomyces bacteria. The higher pitching ratio (30%, v/v)significantly enhanced the levels of streptolydigin products in S. lydicus. Proteome analysis revealed that betaglucosidase and UTP-glucose-1-phosphate uridylyltransferase were up-regulated to accelerate the starch hydrolyzation at the high pitching ratios. Enhancement in the levels of UDPN-acetylmuramoylalanyl-D-glutamate-2, 6-diaminopimelate ligase and glycine cleavage system aminomethyltransferase were involved in the conversion of amino acids into secondary metabolites. Additionally, the expression levels of PfkA2, PfkA3, Zwf2, SucD, GalE1, GatB, TktA1 and ThcA, associated with glycolysis, pentose phosphate pathway, TCA cycle and amino acid metabolism, were dramatically elevated at high pitching ratios, which play important roles in the enhanced streptolydigin production in S. lydicus E9. Interestingly, the levels of proteins (glutamine synthetase I, glutamate synthase subunit beta and glutamine synthetase) were down-regulated with the increases of pitching ratios and fermentation progress,revealing that pitching ratio altered the glutamine synthetase levels and consequently regulated the streptolydigin production of S. lydicus E9. The up-regulation of proteins (eg, aldehyde dehydrogenase and alkyl hydroperoxide reductase) was involved in the redox-based regulation network triggered by an imbalance of the intracellular cell redox homeostasis and by crosstalk with secondary metabolism at the higher pitching ratio. These results settle new insights into physiological facts of S. lydicus E9 in responses to pitching ratios and will eventually improve the antibiotic production schemes in industry.

        Functional Characterization of aroA from Rhizobium Leguminosarum with Significant Glyphosate Tolerance in Transgenic Arabidopsis

        Glyphosate is the active component of the top-selling herbicide, the phytotoxicity of which is due to its inhibition of the shikimic acid pathway. 5-Enolpyruvylshikimate-3-phosphate synthase (EPSPS) is a key enzyme in the shikimic acid pathway. Glyphosate tolerance in plants can be achieved by the expression of a glyphosate-insensitive aroA gene (EPSPS). In this study, we used a PCR-based two-step DNA synthesis method to synthesize a new aroA gene (aroAR. leguminosarum) from Rhizobium leguminosarum. In vitro glyphosate sensitivity assays showed that aroAR. leguminosarum is glyphosate tolerant. The new gene was then expressed in E. coli and key kinetic values of the purified enzyme were determined. Furthermore, we transformed the aroA gene into Arabidopsis thaliana by the floral dip method. Transgenic Arabidopsis with the aroAR. leguminosarum gene was obtained to prove its potential use in developing glyphosate-resistant crops.

        In this research paper, it will identify whether the Heart Vein Opening Drink, a solution to cardiovascular diseases that is widely spreaded among the community, have the cardioprotective properties as the community said. Cardiovascular diseases is now one of the most threatening diseases to mankind, thus if there is a prevention or cure to it, research should be done to identify it and to find the reason behind this. However, the results in this research are conflicting and demands for further development of research.

        The Gender-Sensitive Social Risk Factors for Internet Addiction in College Undergraduate Students

        Objective The current study aims to explore precipitating and social risk factors for internet addiction (IA) in university undergraduate students, and to provide evidence for interventions and the early prevention of IA in different genders. Methods Four thousand eight hundred and fifty-eight college sophomores completed an online survey on their internet use-related behaviours and social risk factors. Results We found that more male (8.3%) than female students (5.4%) had moderate and severe IA. The main online activity in the moderate and severe IA groups was online gaming in males and online streaming in females. Roommates engaging in similar internetbased entertainment was a risk factor of IA only for males, while not being in a romantic relationship was a risk factor of IA for females only. Infatuation with the internet before college and adjustment problems for college life were shared risk factors for both genders in the mild and moderate IA groups. Conclusion IA was a common phenomenon in college students with shared and unique precipitating and social risk factors in males and females. The gender-sensitive risk factors for IA warranted earlier and individualized intervention and prevention strategies for IA in this population.

        Effect of all-trans retinoic acid on casein and fatty acid synthesis in MAC-T cells

        Objective: Caseins and fatty acids of milk are synthesized and secreted by the epithelial cells of the mammary gland. All-trans retinoic acid (ATRA), an active metabolite of vitamin A, has been shown to promote mammary development. This study was conducted to determine the effect of ATRA on casein synthesis and fatty acid composition in MAC-T cells. Methods: MAC-T cells were allowed to differentiate for 4 d, treated with ATRA (0, 1.0, 1.5, and 2.0 μM), and incubated for 3 d. We analyzed the fatty acid composition, the mRNA expression of casein and fatty acid synthesis-related genes, and the phosphorylation of casein synthesis-related proteins of MAC-T cells by gas chromatography, quantitative polymerase chain reaction, and western blotting, respectively. Results: In MAC-T cells, ATRA increased the mRNA levels of α<sub>S1</sub>-casein and β-casein, janus kinase 2 (JAK2) and E74-like factor 5 of the signal transducer and activator of transcription 5 β (STAT5-β) pathway, ribosomal protein S6 kinase beta-1 (S6K1) and eukaryotic translation initiation factor 4E binding protein 1 of the mammalian target of rapamycin (mTOR) pathway, inhibited the mRNA expression of phosphoinositide 3-kinase and eukaryotic initiation factor 4E of the mTOR pathway, and promoted the phosphorylation of STAT5-β and S6K1 proteins. Additionally, ATRA increased the de novo synthesis of fatty acids, reduced the content of long-chain fatty acids, the ratio of monounsaturated fatty acids to saturated fatty acids (SFA), the ratio of polyunsaturated fatty acids (PUFA) to SFA, and the ratio of ω-6 to ω-3 PUFA. The mRNA levels of acetyl-CoA carboxylase 1, fatty acid synthase, lipoprotein lipase, stearoyl-CoA desaturase, peroxisome proliferator-activated receptor gamma, and sterol regulatory element-binding protein 1 (SREBP1) were enhanced by ATRA. Conclusion: ATRA promotes the synthesis of casein by regulating JAK2/STAT5 pathway and downstream mTOR signaling pathway, and it improves the fatty acid composition of MAC-T cells by regulating SREBP1-related genes.

        An automated control system for concrete temperature development in construction

        PLC and its expansion module, electric ball valve and cooling pipe, electric heating steel plate and various components of the system, which is used to control test and process data. By automatically adjusting the opening of the valve, the system makes the top temperature and cooling speed develop along the ideal temperature diachronic curve. Moreover, the system enables the temperature difference between inside and surface of test block limited in a given range by automatically controlling the surface board heating. The method of physical simulation test by sandbox with built-in cooling water pipe and heating rod is adopted. On the premise of a given standard value, the operation of the system is checked under different working conditions. Further, an extension of this system is proposed, which enables its application to obtain some thermal parameters when cooperating with numerical simulation.

        Calculation on the Ion Flow Field under HVDC Transmission Lines Considering Wind Effects

        Based on Deutsch assumption, a calculation method on the electric field over the ground surface under HVDC transmission lines in the wind is proposed. Analyzing the wind effects on the electric field and the space charge density the existing method based on Deutsch assumption is improved through adding the wind speed to the ion flow field equations. The programming details are illustrated. The calculation results at zero wind speed are compared with available data to validate the code program. Then the ionized fields which resulted from corona of ±800kV HVDC lines are analyzed. Both the electric field and the current density on the ground level are computed under different wind direction and speed. The computation results are in good agreement with measurements. The presented method and code program can be used to rapidly predict and evaluate the wind effects in HVDC transmission engineering.

        Cytisine, a Partial Agonist of α4β2 Nicotinic Acetylcholine Receptors, Reduced Unpredictable Chronic Mild Stress-Induced Depression-Like Behaviors

        Cytisine (CYT), a partial agonist of α4β2-nicotinic receptors, has been used for antidepressant efficacy in several tests. Nicotinic receptors have been shown to be closely associated with depression. However, little is known about the effects of CYT on the depression. In the present study, a mouse model of depression, the unpredictable chronic mild stress (UCMS), was used to evaluate the activities of CYT. UCMS caused significant depression-like behaviors, as shown by the decrease of total distances in open field test, and the prolonged duration of immobility in tail suspension test and forced swimming test. Treatment with CYT for two weeks notably relieved the depression-like behaviors in the UCMS mice. Next, proteins related to depressive disorder in the brain region of hippocampus and amygdala were analyzed to elucidate the underlying mechanisms of CYT. CYT significantly reversed the decreases of 5-HT1A, BDNF, and mTOR levels in the hippocampus and amygdala. These results imply that CYT may act as a potential anti-depressant in the animals under chronic stress.
