Over-expression of de novo lipogenesis (DNL) genes is associated with the prognosis of various types of cancers. However, the effects of single nucleotide polymorphisms (SNPs) in these genes on recurrence and survival of non-small cell lung cancer (NSCLC) patients after surgery are still unknown. In this study, a total of 500 NSCLC patients who underwent surgery treatment were included. Eight SNPs in 3 genes (ACACA, FASN and ACLY) of the DNL pathway were examined using the Sequenom iPLEX genotyping system. Multivariate Cox proportional hazards regression and Kaplan-Meier curves were used to analyze the association of SNPs with patient survival and tumour recurrence. We found that two SNPs in the FASN gene were significantly associated with the recurrence of NSCLC. SNP rs4246444 had a significant association with lung cancer recurrence under additive model (hazard ratio [HR], 0.82; 95% confidence interval [95%CI], 0.67-1.00; p=0.05). Under the dominant model, rs4485435 exhibited a significant association with recurrence (HR, 0.75; 95%CI, 0.56-1.01; p=0.05). Additionally, SNP rs9912300 in ACLY gene was significantly associated with overall survival in lung cancer patients (HR, 1.41; 95%CI, 1.02-1.94, p=0.04) under the dominant model. Further cumulative effect analysis showed moderate dose-dependent effects of unfavorable SNPs on both survival and recurrence. Our data suggest that the SNPs in DNL genes may serve as independent prognostic markers for NSCLC patients after surgery.
Valve core shapes analysis on flux through control valves in nuclear power plants

Control valves are widely used to regulate fluid flux in nuclear power plants, and there are more than1500 control valves in the primary circuit of one nuclear power plant. With their help, the flux can beregulated to a specific level of water or steam to guarantee the energy efficiency and safety of the nuclearpower plant. The flux characteristics of the control valve mainly depend on the valve core shape. In orderto analyze the effects of valve core shapes on flux characteristics of control valves, this paper focuses onthe valve core shapes. To begin with, numerical models of different valve core shapes are established, andresults are compared with the ideal flux characteristics curve for the purpose of validation. Meanwhile,the flow fields corresponding to different valve core shapes are investigated. Moreover, relationshipsbetween the valve core opening and the outlet flux under different valve core shapes are carried out. In this paper, the decomposition formulas for radix-2, radix-4 and split radix FHT algorithm[2] are examined and modified, and fast algorithms for computing discrete Hartley transform are presented and block diagrams of one stage decomposition are given. The operation counts for the FHT algorithms of this paper are determined, the multiply counts are less than Reference[2] and addition counts are increased a little. These algorithms are in-place computing and the hardware structures are simple and attractive.

In this work, a highly sensitive method using a colorimetric probe coupled to dispersive liquid?liquid microextraction (DLLME) was developed for the quantitative determination of dopamine (DA) in serum. The DA in serum was concentrated by DLLME to increase the detection sensitivity and reduce the matrix effects. After the DLLME process, a colorimetric probe of silver triangular nanoparticles (AgTNPs) was used to detect DA, which was based on the plasma transformation of AgTNPs caused by strong interactions with melamine (MA). The results showed that DA could inhibit the aggregation of AgTNPs induced by MA, resulting in the recovery of the surface plasmon resonance (SPR) peak of AgTNPs. Thus, the DLLME method followed by colorimetric probe detection of DA can be achieved. The parameters affecting the proposed method were optimized, under the optimal conditions, a linear calibration curve was obtained over a concentration range of 5 to 250?nM with a recovery from 94.4 to 101.3%. The detection limit was 1.6 nM (at an S/N ratio of 3). The present method was successfully applied to determine DA in human serum.

Background: Many factors, including molecular ones, were demonstrated to be associated with long-term prognosis of hepatocellular carcinoma (HCC). Thus far, the expression and clinicopathologic and prognostic significance of the carboxyl terminus of Hsp70-interacting protein (CHIP) in B-type hepatitis virus (HBV)-related HCC remain unknown. Materials and Methods: CHIP expression was detected by immunohistochemical staining of surgical samples from 79 patients with HCC with HBsAg positivity. In addition, correlations with clinicopathologic parameters and patient survival were evaluated. Results: It was found that positive CHIP staining was observed in tumor, but not non-tumor, tissues. High expression of CHIP was significantly related to larger tumor size, with marginally significant associations noted for presence of portal vein invasion and higher serum a-fetoprotein level. In addition, univariate analysis showed that high CHIP expression was a powerful predictor for dismal overall and disease-free survival. However, independent prognostic implications of CHIP were not proven in multivariate Cox regression test. Conclusions: CHIP is overexpressed in HBV-related HCC and is associated with unfavorable biological behavior as well as poor prognosis. However, its prognostic role needs to be further validated.
Expression profiles of microRNAs in skeletal muscle of sheep by deep sequencing

Objective: MicroRNAs are a class of endogenous small regulatory RNAs that regulate cell proliferation, differentiation and apoptosis. Recent studies on miRNAs are mainly focused on mice, human and pig. However, the studies on miRNAs in skeletal muscle of sheep are not comprehensive. Methods: RNA-seq technology was used to perform genomic analysis of miRNAs in prenatal and postnatal skeletal muscle of sheep. Targeted genes were predicted using miRanda software and miRNA-mRNA interactions were verified by quantitative real-time polymerase chain reaction. To further investigate the function of miRNAs, candidate targeted genes were enriched for analysis using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment. Results: The results showed total of 1,086 known miRNAs and 40 new candidate miRNAs were detected in prenatal and postnatal skeletal muscle of sheep. In addition, 345 miRNAs (151 up-regulated, 94 down-regulated) were differentially expressed. Moreover, miRanda software was performed to predict targeted genes of miRNAs, resulting in a total of 2,833 predicted targets, especially miR-381 which targeted multiple muscle-related mRNAs. Furthermore, GO and KEGG pathway analysis confirmed that targeted genes of miRNAs were involved in development of skeletal muscles. Conclusion: This study supplements the miRNA database of sheep, which provides valuable information for further study of the biological function of miRNAs in sheep skeletal muscle.

The calpain I (CAPN1) gene is an important marker for meat tenderness and marbling score in the bovine, but there were no studies to determine whether the CAPN1 gene had an association with other meat quality traits. In this study, we examined the relation between genetic polymorphisms of the CAPN1 gene and some meat quality traits in Yanbian Yellow Cattle of China. By PCRSSCP and gene sequencing in 321 unrelated Yanbian yellow cattle, twenty seven single nucleotide polymorphisms (SNPs) were detected in CAPN1, two existed SNPs in exon 8 and exon 17 resulted in the change of AA at F311S and M599V, respectively, and the otherpolymorphisms were at intron 7, 8, 14, 16 and 17. There were different preponderant genotypes at the corresponding gene locus and all genotypes were not associated with tenderness but other meat traits. This is the first study of the relationship between CAPN1 and meat quality besides tenderness in Yanbian yellow cattle of China.
