        Bioaccumulation of Arsenic in Recombinant Escherichia coli Expressing Human Metallothionein

        The recombinant Escherichia coli(E.coli) expressing human hepatic metallothionein_IA (hMT_IA) was constructed for bioaccumulation of Arsenic (As). The gene sequence of hMT_IA was modified for codon preference of E.coli and synthesized using chemical method. The vector of pGEX_4T_1 was used=and hMT_IA was expressed as the fusion protein with glutathione S-transferase (GST) tag. The bioaccumulation capability of arsenite compounds As(III) of the recombinant E.coli increased more than 3-fold from 76.3 to 319.6 μg/g dry cells compared with the control. The conditions of 50 μM of As(III) and low pHs were optimal for As(III) bioaccumulation. The heavy metals of Cd, Hg, and Zn inhibited As(III) bioaccumulation. The bioaccumulation reached 70% of the saturated value within 1 h. The recombinant E.coli will be useful in bioremediation of arsenic or other kinds of heavy metal contaminated water.       • Relationship Between GSTT1 Gene Polymorphism and Hepatocellular Carcinoma in Patients from China

        Objective: The results from studies on associations of the glutathione S-transferase T1 (GSTT1) gene polymorphism and hepatocellular carcinoma (HCC) risk in Chinese populations are still conflicting. This meta-analysis was performed to evaluate the relationship in detail. Methods: Eligible reports were recruited into this meta-analysis from the databases of PubMed, Embase, Cochrane Library and CBM-disc (China Biological Medicine Database). Results were expressed with odds ratios (OR) for dichotomous data, and 95% confidence intervals (CI) were also calculated. Results: Eighteen investigations were identified for the analysis of association between polymorphic deletion of GSTT1 and HCC, consisting of 2,693 patients with HCC and 4,696 controls. Null genotype of GSTT1 was associated with HCC susceptibility in Chinese (OR=1.53, 95%CI: 1.28-1.82; P<0.00001). Conclusion: The GSTT1 null genotype is associated with HCC susceptibility in Chinese.

        An improved interval analysis method for uncertain structures

        Based on the improved first order Taylor interval expansion, a new interval analysis method for the static or dynamic response of the structures with interval parameters is presented. In the improved first order Taylor interval expansion, the first order derivative terms of the function are also considered to be intervals. Combining the improved first order Taylor series expansion and the interval extension of function, the new interval analysis method is derived. The present method is implemented for a continuous beam and a frame structure. The numerical results show that the method is more accurate than the one based on the conventional first order Taylor expansion.

        Molecular characterization of the sans fille gene in Antheraea pernyi: A highly conserved gene during the evolution of animals

        Sans-fille (SNF) is the Drosophila homologue ofmammalian general splicing factors U1A and U2B″, and plays an important role in sex determination in Drosophilamelanogaster. In this study, the snf gene fromAntheraea pernyi (Lepidoptera: Saturniidae), an economically important insect, was isolated and characterized. The obtained 925 bp cDNA sequence contains an open reading frame of 669 bp encoding a polypeptide of 222 amino acids,showing 78% sequence identity to that from D. melanogaster. A database search revealed that SNF protein homologs are present in many animals, including invertebrates and vertebrates, with more than 70% amino acid sequence identities, suggesting that they were highly conserved during the evolution of animals. Phylogenetic analysis revealed that A. pernyi SNF was closely related to Bombyx mori SNF. Quantitative real-time PCR (qRT-PCR) analysis showed that the A. pernyi snf gene was transcribed during five larval developmental stages,and in six tested tissues (ovaries, testes, silk glands, fat body, integument, and hemolymph),with the most abundance determined in the gonads (ovaries or testes). Investigation of expression changes throughout embryonic development indicated that A. pernyi snfmRNAwas expressed at a lowlevel fromdays 0 to 4, and reached amaximum level at day 10, but decreased to a low level before hatching. These results suggest that the product of the snf gene may play important roles in the development of A. pernyi.

        Electric Stimulation Hyperthermia Relieves Inflammation via the Suppressor of Cytokine Signaling 3-Toll Like Receptor 4 Pathway in a Prostatitis Rat Model

        Purpose: Chronic prostatitis (CP), including chronic pelvic pain syndrome (CPPS), is the most commonly encountered manifestation of prostatitis. The aim of this study was to evaluate the effect of electric stimulation hyperthermia treatment (ESHT) on CP/CPPS and to explore the underlying mechanism. Materials and Methods: RWPE-2 cells with lipopolysaccharide-induced inflammation and a prostatitis rat model induced by 17β-estradiol and dihydrotestosterone underwent sham, electric stimulation, or ESHT treatment. Four weeks later, cells, supernatants, and rat prostates were collected for analysis using immunohistochemistry, Western blots, and enzyme-linked immunosorbent assays. Results: We found that ESHT improved prostatitis in vivo and attenuated inflammation in vitro. ESHT significantly induced suppressor of cytokine signaling 3 (SOCS3) expression and subsequently promoted HSP70. It attenuated inflammation through decreased expression of toll-like receptor 4 (TLR4), nuclear factor kappa B, and subsequent inflammatory cytokines. ESHT also inhibited apoptosis and released growth factor in tissue affected by prostatitis. Conclusions: ESHT improved CP/CPPS and reversed pathologic changes of prostatitis by inhibiting the SOCS3-TLR4 pathway.

      • EMG-Based Continuous and Simultaneous Estimation of Arm Kinematics in Able-Bodied Individuals and Stroke Survivors

        <P>Among the potential biological signals for human-machine interactions (brain, nerve, and muscle signals), electromyography (EMG) widely used in clinical setting can be obtained non-invasively as motor commands to control movements. The aim of this study was to develop a model for continuous and simultaneous decoding of multi-joint dynamic arm movements based on multi-channel surface EMG signals crossing the joints, leading to application of myoelectrically controlled exoskeleton robots for upper-limb rehabilitation. Twenty subjects were recruited for this study including 10 stroke subjects and 10 able-bodied subjects. The subjects performed free arm reaching movements in the horizontal plane with an exoskeleton robot. The shoulder, elbow and wrist movements and surface EMG signals from six muscles crossing the three joints were recorded. A non-linear autoregressive exogenous (NARX) model was developed to continuously decode the shoulder, elbow and wrist movements based solely on the EMG signals. The shoulder, elbow and wrist movements were decoded accurately based only on the EMG inputs in all the subjects, with the variance accounted for (VAF) > 98% for all three joints. The proposed approach is capable of simultaneously and continuously decoding multi-joint movements of the human arm by taking into account the non-linear mappings between the muscle EMGs and joint movements, which may provide less effortful control of robotic exoskeletons for rehabilitation training of individuals with neurological disorders and arm impairment.</P>

        Composition and Antioxidant Activity of Water-Soluble Polysaccharides from Tuber indicum

        Crude water-soluble Chinese truffle Tuber indicum polysaccharide (TIP) was extracted from the fruiting bodies with water and then successively purified by DEAE–cellulose 52 and Sephadex G-100 column chromatography,yielding two major polysaccharide fractions: TIP1-1 and TIP2-1. High-performance gel permeation chromatography analysis showed that the average molecular sizes of TIP1-1 and TIP2-1 were approximately 1.75×10⁴Da and 5.73×10³ Da, respectively. Monosaccharide component analysis by gas chromatography indicated that TIP1-1 was composed of mannose,glucose, galactose, and rhamannose in the respective molar ratio of 3.93:1.24:0.75:1.26 and that TIP2-1 contained mannose,glucose, and arabinose in the respective molar ratio of 5.27:1.44:0.43. The antioxidant activity analyses revealed that TIP1-1and TIP2-1 possessed considerable antioxidant activity. Compared with TIP1-1, which has a higher molecular weight and contains no uronic acid, TIP2-1 exhibited a protective effect on PC12 cells injured by H₂O₂and a higher scavenging activity against free radicals. The relative effects of the lower molecular size, the presence of uronic acid, and the antioxidant activity of TIP2-1 appear to be significant. Accordingly, the Chinese truffle T. indicum might serve as an effective antioxidative healthcare food and source of natural antioxidants.

        Serotype-Independent Protection against Pneumococcal Infections Elicited by Intranasal Immunization with Ethanol-Killed Pneumococcal Strain, SPY1

        The 23-valent polysaccharide vaccine and the 7-valent pneumococcalconjugate vaccine are licensed vaccines that protectagainst pneumococcal infections worldwide. However,the incidence of pneumococcal diseases remains high in lowincomecountries. Whole-cell vaccines with high safety andstrong immunogenicity may be a favorable choice. We previouslyobtained a capsule-deficient Streptococcus pneumoniaemutant named SPY1 derived from strain D39. As anattenuated live pneumococcal vaccine, intranasal immunizationwith SPY1 elicits broad serotype-independent protectionagainst pneumococcal infection. In this study, forsafety consideration, we inactivated SPY1 with 70% ethanoland intranasally immunized BALB/c mice with killed SPY1plus cholera toxin adjuvant for four times. Results showedthat intranasal immunization with inactivated SPY1 inducedstrong humoral and cellular immune responses. Intranasalimmunization with inactivated SPY1 plus cholera toxin adjuvantelicited effective serotype-independent protection againstthe colonization of pneumococcal strains 19F and 4 as well aslethal infection of pneumococcal serotypes 2, 3, 14, and 6B. The protection rates provided by inactivated SPY1 againstlethal pneumococcal infection were comparable to those ofcurrently used polysaccharide vaccines. In addition, vaccinespecificB-cell and T-cell immune responses mediated theprotection elicited by SPY1. In conclusion, the 70% ethanolinactivatedpneumococcal whole-cell vaccine SPY1 is a potentiallysafe and less complex vaccine strategy that offersbroad protection against S. pneumoniae.
