      • Expression of the Pokemon Proto-oncogene in Nasopharyngeal Carcinoma Cell Lines and Tissues

        To study the differentiated expression of the proto-oncogene Pokemon in nasopharyngeal carcinoma (NPC) cell lines and tissues, mRNA and protein expression levels of CNE1, CNE2, CNE3 and C666-1 were detected separately by reverse transcription polymerase chain reaction (RT-PCR), real-time PCR and Western-blotting. The immortalized nasopharyngeal epithelial cell line NP69 was used as a control. The Pokemon protein expression level in biopsy specimens from chronic rhinitis patients and undifferentiated non keratinizing NPC patients was determined by Western-blotting and arranged from high to low: C666-1>CNE1>CNE2> CNE3>NP69. The Pokemon mRNA expression level was also arranged from high to low: CNE1>CNE2>NP69>C666-1>CNE3. Pokemon expression of NP69 and C666-1 obviously varied from mRNA to protein. The Pokemon protein level of NPC biopsy specimens was obviously higher than in chronic rhinitis. The data suggest that high Pokemon protein expression is closely associated with undifferentiated non-keratinizing NPC and may provide useful information for NPC molecular target therapy.

        Transcriptome Analysis of Antrodia cinnamomea Mycelia from Different Wood Substrates

        Antrodia cinnamomea, an edible and medicinal fungus with significant economic value and application prospects, is rich in terpenoids, benzenoids, lignans, polysaccharides, and benzo- quinone, succinic and maleic derivatives. In this study, the transcriptome of A. cinnamomea cultured on the wood substrates of Cinnamomum glanduliferum (YZM), C. camphora (XZM), and C. kanehirae (NZM) was sequenced using the high-throughput sequencing technology Illumina HiSeq 2000, and the data were assembled by de novo strategy to obtain 78,729 Unigenes with an N50 of 4,463 bp. Compared with public databases, about 11,435, 6,947, and 5,994 Unigenes were annotated to the Non-Redundant (NR), Gene Ontology (GO), and Kyoto Encyclopedia of Genes and Genome (KEGG), respectively. The comprehensive analysis of the mycelium terpene biosynthesis-related genes in A. cinnamomea revealed that the expression of acetyl-CoA acetyltransferase (AACT), acyl-CoA dehydrogenase (MCAD), 3- hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA), mevalonate pyrophosphate decarboxylase (MVD), and isopentenyl diphosphate isomerase (IDI) was significantly higher on NZM com- pared to the other two wood substrates. Similarly, the expression of geranylgeranyltransfer- ase (GGT) was significantly higher on YZM compared to NZM and XZM, and the expression of farnesyl transferase (FTase) was significantly higher on XZM. Furthermore, the expressions of 2,3-oxidized squalene cyclase (OCS), squalene synthase (SQS), and squalene epoxidase (SE) were significantly higher on NZM. Overall, this study provides a potential approach to explore the molecular regulation mechanism of terpenoid biosynthesis in A. cinnamomea.

        Crack-tip constraint analysis of two collinear cracks under creep condition

        The higher-order asymptotic C(t) - $A_2(t)$ approach was employed to investigate the crack-tip stress of two collinear cracks in a power-law creeping material under the plane strain conditions. A comprehensive calculation was made of the single crack, collinear crack model with S/a = 0.4 and 0.8, by using the C(t) - $A_2(t)$ approach, HRR-type field and the finite element analysis; the latter two methods were used to check the constraint significance and the calculation accuracy of the C(t) - $A_2(t)$ approach, respectively. With increasing the creep time, the constraint $A_2$ was exponentially increased in the small-scale creep stage, while no discernible dependency of the constraint $A_2$ on the creep time was found at the extensive creep state. In addition, the creep time and the mechanical loads have no distinct influence on accuracy of the results obtained from the higher-order asymptotic C(t) - $A_2(t)$ approach. In comparison with the HRR-type field, the higher-order asymptotic C(t) - $A_2(t)$ solution matches well with the finite element results for the collinear crack model.

        AcuD Gene Knockout Attenuates the Virulence of Talaromyces marneffei in a Zebrafish Model

        Talaromyces marneffei is the only dimorphic species in its genus and causes a fatal systemic mycosis named talaromycosis. Our previous study indicated that knockdown of AcuD gene (encodes isocitrate lyase of glyoxylate bypass) of T. marneffei by RNA interference approach attenuated the virulence of T. marneffei, while the virulence of the AcuD knockout strains was not studied. In this study, T. marneffei-zebrafish infection model was successfully established through hindbrain microinjection with different amounts of T. marneffei yeast cells. After co-incubated at 28_C, the increasing T. marneffei inoculum doses result in greater larval mortality; and hyphae generation might be one virulence factor involved in T. marneffeizebrafish infection. Moreover, the results demonstrated that the virulence of the DAcuD was significantly attenuated in this Zebrafish infection model.

        Nitrogen and Fluorine Dual-Doped Carbon Nanosheets for High-Performance Supercapacitors

        Doping carbon materials with heteroatoms such as N, F is an effective approach to elevating the capacitive performance of supercapacitors. In this paper, nitrogen and fluorine dual-doped two-dimensional (2D) porous carbon nanosheets (PCNSs) have been fabricated by a straightforward template carbonization method, using trisodium citrate as carbon source and self-template, and ammonium fluoride as N/F dopants. The N/F-doped carbon samples are well characterized by a series of techniques and measured in a three-electrode system and two-electrode system, respectively. As a result, N/F-doped carbon has delivered large capacitance of 110 F g -1 at 1 Ag -1 and high-energy density of 3.82 W h kg -1 at the power density of 0.5 kW kg -1. It is also revealed that semi-ionic C–F bonds in PCNSs have enhanced electrical conductivity, hence, facilitating electron transport in the electrode. For comparison, ammonium chloride is used as sole dopant for producing N-doped carbon materials, whose capacitive performances are much lower than the N/F-codoped one, indicating the synergistic effect of N/F for capacitive improvement.

      • Associations of IL-10 Gene Polymorphisms with Acute Myeloid Leukemia in Hunan, China

        We investigated the possible association of interleukin-10 (IL-10) single nucleotide polymorphisms (SNPs) and susceptibility to acute myeloid leukemia (AML) in 115 patients and 137 healthy controls. Genetic analysis of IL-10 SNPs at -819 and -592 was carried out with the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) approach. The IL-10 mRNA expression of AML patients and controls with different genotype was detected by real-time quantitative polymerase chain reaction (RT-PCR). Genetic analysis of IL-10 revealed that the -819AA genotype frequencies and the -819A allele frequencies in the AML group were higher than in the controls (59.1% vs 40.9%; 75.6% vs 63.9%, respectively); there were remarkable differences in -819T/C and -592A/C gene distribution (P<0.05) and the TA haploid frequencies were higher in the AML group (75.6% vs 63.9%, P<0.05). IL-10 mRNA expression in incipient AML patients was obvious higher than the non-tumor group and the remission group ($7.78{\times}10^{-3}$ vs $2.43{\times}10^{-3}$, $3.64{\times}10^{-3}$, P<0.05).The study suggested that the haploid TA and genotype TA/TA may be associated with AML in Han people in Hunan province.The IL-10 SNPs at -819 and -592 sites were associated with AML and may affect IL-10 mRNA expression in AML patients.

        A Study of London Penetration Depth for the Non-centrosymmetric Superconductor LaNiC2

        We study the superconducting properties of the polycrystalline LaNiC2 by means of measuringthe electrical resistivity ρ(T), magnetization M(T) and London penetration depth Δλ(T). Thesuperconducting order parameter is analyzed in terms of the low temperature penetration depthΔλ(T) and the corresponding superfluid density ρs(T), and the results show strong evidence oftwo-gap Bardeen-Cooper-Schrieffer (BCS) superconductivity for LaNiC2.

        Identification and confirmation of 14-3-3 ζ as a novel target of ginsenosides in brain tissues

        Background: Ginseng can help regulate brain excitability, promote learning and memory, and resist cerebral ischemia in the central nervous system. Ginsenosides are the major effective compounds of Ginseng, but their protein targets in the brain have not been determined. Methods: We screened proteins that interact with the main components of ginseng (ginsenosides) by affinity chromatography and identified the 14-3-3 ζ protein as a potential target of ginsenosides in brain tissues. Results: Biolayer interferometry (BLI) analysis showed that 20(S)-protopanaxadiol (PPD), a ginseng saponin metabolite, exhibited the highest direct interaction to the 14-3-3 ζ protein. Subsequently, BLI kinetics analysis and isothermal titration calorimetry (ITC) assay showed that PPD specifically bound to the 14-3-3 ζ protein. The cocrystal structure of the 14-3-3 ζ protein-PPD complex showed that the main interactions occurred between the residues R56, R127, and Y128 of the 14-3-3 ζ protein and a portion of PPD. Moreover, mutating any of the above residues resulted in a significant decrease of affinity between PPD and the 14-3-3 ζ protein. Conclusion: Our results indicate the 14-3-3 ζ protein is the target of PPD, a ginsenoside metabolite. Crystallographic and mutagenesis studies suggest a direct interaction between PPD and the 14-3-3 ζ protein. This finding can help in the development of small-molecular compounds that bind to the 14-3-3 ζ protein on the basis of the structure of dammarane-type triterpenoid.

        STRUCTURE IMPROVEMENT OF TRUCK CARRIAGE WITH SEALING COVER

        SolidWorks and ANSYS Workbench were used to establish solid model and perform FEM analysis of truck with and without sealing covers respectively. In the past practical production, there has been no precedent for the modal analysis of the truck carriage with a sealing cover; the modal analysis is carried out for the first time in this paper. The vibration characteristics of the truck carriage are obtained. The first ten-order of non-rigid mode of the newly development truck carriage is obtained and the excitation frequency range of the idling state of the engine is calculated to avoid resonating with the ground. In order to avoid the resonance phenomenon during the truck running, the vibration characteristics of the new carriage model were studied, the first ten non-rigid modes of the new carriage model were extracted, and the excitation frequency range of the engine during idling was calculated. The non-rigid natural frequencies of the first 10 free models are between 6.18 ~ 26.74 Hz. The engine idling excitation frequency is 32.5 ~ 145 Hz, and the common excitation frequency caused by road roughness is 3 Hz. In theory, it does not cause resonance when driving in a normal state.
