        Overexpression of SIRT2 contributes tumor cell growth in hepatocellular carcinomas

        The mammalian homolog of yeast Sir2protein is the sirtuin family of histone deacetylases (HDACs), a NAD+-dependent protein deacetylase in humans. Accumulating evidence suggests that sirtuin 2 (SIRT2) co-localizes with the microtubule network and deacetylates α-tubulin, and is involved in various cellular processes including calorie restriction-dependent life span extension, mitotic cell cycle regulation,cellular apoptosis, DNA damage repair, and genomic silencing. However, the underlying mechanisms of action remains poorly understood, especially in hepatocarcinogenesis. Hence in this study, to determine the association between the aberrant expression of SIRT2and liver cancer development and progression, SIRT2expression was investigated in ten selected hepatocellular carcinoma (HCC) tissues and matched normal liver tissues, using RT-PCR and Western blot analysis. Next, SIRT2 was disrupted by siRNA-mediated protein knockdown method to investigate the biological role of SIRT2 in hepatocarcinogenesis in Hep3B cells. As a result, we identified that SIRT2 expression was significantly up-regulated in HCC tissues compared to corresponding normal liver tissues. In addition, suppression of SIRT2 caused regression of tumor cell growth and proliferation. We also found that SIRT2could interact with α-tubulin and regulates the acetylation status of α-tubulin in Hep3B cells. In conclusion,we suggest that SIRT2 is aberrantly regulated in HCCs which may contribute to the mitogenic potential of tumor cells during the development and progression of HCC, and could be a novel molecular target for therapeutic intervention in liver cancer.

        Cadmium induces apoptosis in primary rat osteoblasts through caspase and mitogen-activated protein kinase pathways

        Exposure to cadmium (Cd) induces apoptosis in osteoblasts (OBs); however, little information is available regarding the specific mechanismsof Cd-induced primary rat OB apoptosis. In this study, Cd reduced cell viability, damaged cell membranes and induced apoptosis in OBs. We observed decreased mitochondrial transmembrane potentials, ultrastructure collapse, enhanced caspase-3 activity, and increasedconcentrations of cleaved PARP, cleaved caspase-9 and cleaved caspase-3 following Cd treatment. Cd also increased the phosphorylationof p38-mitogen-activated protein kinase (MAPK), extracellular signal-regulated kinases (ERK)1/2 and c-jun N-terminal kinase (JNK) in OBs. Pretreatment with the caspase inhibitor, N-benzyloxycarbonyl-Val-Ala-Asp-fluoromethylketone, ERK1/2 inhibitor (U0126), p38 inhibitor(SB203580) and JNK inhibitor (SP600125) abrogated Cd-induced cell apoptosis. Furthermore, Cd-treated OBs exhibited signs of oxidativestress protection, including increased antioxidant enzymes superoxide dismutase and glutathione reductase levels and decreased formationof reactive oxygen species. Taken together, the results of our study clarified that Cd has direct cytotoxic effects on OBs, which are mediatedby caspase- and MAPK pathways in Cd-induced apoptosis of OBs.

      • Research on the Influencing Factors and Transformation Path of Logistics Enterprise Low-Carbon Performance

        Purpose - This study expounds that logistics enterprises should make use of an information-sharing mechanism, benefit distribution mechanism, and government guarantee mechanism to optimize and balance resource allocation with cooperative participants. Design/Methodology/Approach - This study developed a theoretical research model with interface management analysis, interface resource allocation, and interface management measures as independent variables; an interface connection mechanism and collaborative innovation capability as intermediary variables; and the low-carbon performance of enterprises as a dependent variable based on the structural equation model (SEM) and the empirical test, by selecting a certain number of logistics enterprises nationwide as research objects. Findings - This study discloses that elements such as interface management analysis, interface resource allocation, interface management measures, interface connection mechanism, and collaborative innovation capability contribute proportionally to the low-carbon performance of logistics enterprises. Meanwhile, the interface connection mechanism plays an intermediary role in the impact of interface management analysis and interface management measures on the low-carbon performance of enterprises. Research Implications - This study proposes that the collaborative innovation capability of enterprises should be improved through various interface management measures, and that R&D and the application of low-carbon technology and equipment must be accelerated, thus promoting the improvement of the low-carbon performance of enterprises, and enabling a low-carbon transformation in logistics enterprises, and even the whole logistics industry, so as to help achieve the carbon peak and move toward carbon neutrality as soon as possible.

        An optical fibre monitoring system for evaluating the performance of a soil nailed slope

        Conventional geotechnical instrumentation techniques available for monitoring of slopes, especially soil-nailed slopes have limitations such as electromagnetic interference, low accuracy, poor longterm reliability and difficulty in mounting a series of strain sensors on a soil nail bar with a small-diameter. This paper presents a slope monitoring system based on fibre Bragg grating (FBG) sensing technology. This monitoring system is designed to perform long-term monitoring of slope movements, strains along soil nails, and other slope reinforcement elements. All these FBG sensors are fabricated and calibrated in laboratory and a trial of this monitoring system has been successfully conducted on a roadside slope in Hong Kong. As part of the slope stability improvement works, soil nails and a toe support soldier-pile wall were constructed. During the slope works, more than 100 FBG sensors were installed on a soil nail, a soldier pile, and an in- place inclinometer. The paper presents the layout and arrangement of the instruments as well as the installation procedures adopted. Monitoring data have been collected since March 2008. This trial has demonstrated the great potential of the optical fibre monitoring system for long-term monitoring of slope performance. The advantages of the slope monitoring system and experience gained in the field implementation are also discussed in the paper.

        CircularRNA_104670 plays a critical role in intervertebral disc degeneration by functioning as a ceRNA

        This study was carried out to explore the roles of circular RNAs (circRNAs) in nucleus pulposus (NP) tissues in intervertebral disc degeneration (IDD). Differentially expressed circRNAs in IDD and normal NP tissues were identified based on the results of microarray analysis. Bioinformatics techniques were employed to predict the direct interactions of selected circRNAs, microRNAs (miR), and mRNAs. CircRNA_104670 was selected as the target circRNA due to its large multiplier expression in IDD tissues. After luciferase reporter and EGFP/RFP reporter assays, we confirmed that circRNA_104670 directly bound to miR-17-3p, while MMP-2 was the direct target of miR-17-3p. The receiver-operating characteristic (ROC) curve showed that circRNA_104670 and miR-17-3p had good diagnostic significance for IDD (AUC circRNA_104670 = 0.96; AUC miRNA-17-3p = 0.91). A significant correlation was detected between the Pfirrmann grade and expression of circRNA_104670 (r = 0.63; p = 0.00) and miR-17-3p (r = −0.62; p = 0.00). Flow-cytometric analysis and the MTT assay showed that interfering with circRNA_104670 using small interfering RNA (siRNA) inhibited NP cell apoptosis (p < 0.01), and this inhibition was reduced by interfering with miR-17-3p. Interfering with circRNA_104670 suppressed MMP-2 expression and increased extracellular matrix (ECM) formation, which were also reduced by interfering with miR-17-3p. Finally, an MRI evaluation showed that circRNA_104670 inhibition mice had a lower IDD grade compared with control mice (p < 0.01), whereas circRNA_104670 and miRNA-17-3p inhibition mice had a higher IDD grade compared with circRNA_104670 inhibition mice (p < 0.05). CircRNA_104670 is highly expressed in the NP tissues of IDD and acts as a ceRNA during NP degradation.

        Ellipsometric Study of the Optical Properties of Silver Oxide Prepared by Reactive Magnetron Sputtering

        A series of AgOx thin films were prepared by the DC-magnetron reactive-sputtering method at room temperature under different sputtering power and oxygen to argon ratio conditions. The optical properties of the AgOx films were studied by analysis of the spectroscopic ellipsometry data with the Tauc-Lorentz multi-oscillator model in the 1.5 4.5-eV photon-energy range. The spectra of absorption, XPS and PL were measured. The results indicate that the AgOx film changes from metallic to dielectric structure with increasing oxygen flux. Under the saturated oxidation condition after annealing, the dominating component will be Ag2O in the film to show indirect interband transitions occurring at about 0.57 eV and 2.43 eV, respectively, that are in agreement with the results of PL measurement and Lorentz analysis.

        Harnack estimates for nonlinear backward heat equations with potentials along the Ricci-Bourguignon flow

        In this paper, we derive various differential Harnack estimates for positive solutions to the nonlinear backward heat type equations on closed manifolds coupled with the Ricci-Bourguignon flow, which was done for the Ricci flow by J.-Y. Wu \cite{[WJY12]}. The proof follows exactly the one given by X.-D. Cao \cite{[CXD08]} for the linear backward heat type equations coupled with the Ricci flow.

        A Coumarin-based Fluorescent Sensor for Selective Detection of Copper (II)

        Cu (II) detection is of great importance owing to its significant function in various biological processes. In this report, we developed a novel coumarin-based chemosensor bearing the salicylaldimine unit (2) for Cu2+ selective detection. The results from fluorescence spectra demonstrated that the sensor could selectively recognize Cu2+ over other metal cations and the detection limit is as low as 0.2 μM. Moreover, the confocal fluorescence imaging in HepG2 cells illustrated its potential for biological applications.

      • Improved Optimization for Data Disaster Recovery System over Low-Bandwidth Networks

        Data generated by various fields are increasing exponentially and thus results in challenges for data performances in both scales of diversity and complexity. The problem how to solve the bottlenecks of low -bandwidth networks has been of fatal significance for all kinds of network status. We present a new approach on improved optimization for data disaster recovery system (DDRS) over low-bandwidth networks that not only aims to improve the defects and deficiencies of mainstream DDRS but also helps ensure the reliable network resources for operators to conduct multi-services. A novel bandwidth self-adaptive approach (BSAA) for data packing replication was essentially established to make contribution to the integral performance improvement. A Hidden Markov Model (HMM) for predicting network status was also built to ensure system availability and stability. Experiments showed that the DDRS over low-bandwidth networks named InfoDr can effectively optimize the workload with better performance and better application self-adaptability for multi-services.
