Effect of UV on Stability of Collagen with Consideration of Hydratation and Fibrillogenesis

Collagen is an abundant extracellular matrix protein that is widely used in the biomaterial, food, and medical fields. Most extracted collagen is soluble in acids and assembles into fibrils under neutral pH conditions. In order to study the effect of UV irradiation on the stability of collagen with consideration of hydratation and fibrillogenesis,UV irradiation of collagen under acid and neutral pH conditions was investigated using electrophoresis, turbidity analysis, and Fourier transform infrared spectroscopy. The relationship between the collagen structure and sensitivity to UV radiation was assessed by studying the effect of UV on collagen under different pH conditions. Results showed that structural changes in collagen caused by UV depend on the degree of hydratation. Soluble collagen completely loses fibrillogeneic ability after exposure to UV for 40 min. Fibrillogenesis of collagen can relieve the UV effect to some degree.

Henosepilachna vigintioctomaculata is one of the most serious defoliates attacking potatoes. However, studies on functional genes have greatly been limited due to the insufficiency of effective and stable endogenous references to normalize RT-qPCR data. In this report, nine housekeeping genes (RPL4, RPL6, RPL13, RPL32, RPS18, ACT, EF1α, GAPDH and α-TUB) involved in different biological processes were selected. Their expression levels under diverse experimental conditions including developmental stages, tissues, temperatures and host plants were determined using RT-qPCR technology. The tested candidate genes were comprehensively ranked based on five alternative stability analysis methods (Ct value, geNorm, NormFinder, BestKeeper and ReFinder). The results revealed that the optimal internal reference genes varied under different experimental conditions. Any gene pair among the five candidates (RPL4, RPL13, RPL32, RPS18 and EF1α) was a suitable reference gene set under different temperatures and on different host plants. A combination of RPL6 and RPL13 was recommended as the best reference gene set across different developmental stages. A pair of RPS18 and EF1α was ranked as the optimal reference gene combination within different tissues. The most suitable reference genes were RPS18 and RPL13 under four different experimental conditions. Our findings not only establish an accurate and reliable normalization of RT-qPCR data, but also lay a solid foundation for further functional gene researches in H. vigintioctomaculata

Objectives: TCR-gamma-delta+T cells (${\gamma}{\delta}$ T cells) are non-conventional T lymphocytes that can recognize and eradicate tumor cells. Our previous studies showed that infiltration and function of ${\gamma}{\delta}$ T cells were substantially attenuated in hepatocellular carcinoma (HCC). However, their prognostic value was not clarified. Methods: The association between ${\gamma}{\delta}$ T cells and the clinical outcomes was determined by immunohistochemistry (IHC) in a HCC patient cohort (n = 342). Results:Immunohistochemistry showed decreased infiltration of ${\gamma}{\delta}$ T cells in tumoral tissues compared with paired peritumoral tissues. The counts of ${\gamma}{\delta}$ T cells in peritumoral tissues were negatively correlated with tumor size (P = 0.005). Survival analysis showed that the levels of peritumoral ${\gamma}{\delta}$T cells were related to both time to recurrence (TTR) and overall survival (OS) (P = 0.010 and P = 0.036, respectively) in univariate analysis, and related to TTR in multivariate analysis (P = 0.014, H.R. [95% CI] = 0.682 [0.502-0.927]). Furthermore, the level of peritumoral ${\gamma}{\delta}$ T cells showed independent prognostic value for TTR in Barcelona Clinic Liver Cancer (BCLC) stage A patients (P = 0.038, H.R. [95% CI] = 0.727 [0.537-0.984]). However, tumoral ${\gamma}{\delta}$ T cells did not show independent prognostic value for either TTR or OS in HCC patients. Conclusions: Low counts of ${\gamma}{\delta}$ T cells in peritumoral liver tissue are related to a higher incidence of recurrence in HCC and can predict postoperative recurrence, especially in those with early-stage HCC.
An Efficient Indexing Structure for Multidimensional Categorical Range Aggregation Query

Categorical range aggregation, which is conceptually equivalent to running a range aggregation query separately on multiple datasets, returns the query result on each dataset. The challenge is when the number of dataset is as large as hundreds or thousands, it takes a lot of computation time and I/O. In previous work, only a single dimension of the range restriction has been solved, and in practice, more applications are being used to calculate multiple range restriction statistics. We proposed MCRI-Tree, an index structure designed to solve multi-dimensional categorical range aggregation queries, which can utilize main memory to maximize the efficiency of CRA queries. Specifically, the MCRI-Tree answers any query in O(nkn-1) I/Os (where n is the number of dimensions, and k denotes the maximum number of pages covered in one dimension among all the n dimensions during a query). The practical efficiency of our technique is demonstrated with extensive experiments.

The pseudorapidity distributions of charged particles produced in pp collisions at the CERN Large Hadron Collider (LHC) energies (0.9, 2.36, and 7 TeV) are studied by using the multisource thermal model. A group of extrapolated equations is used to determine various parameters. The calculated results are in agreement with the experimental data of the ALICE and CMS Collaborations at the LHC.

Thin Ga2O3 films were grown on Si (100) using trimethylgallium (TMG) and oxygen as the precursors through plasma-enhanced atomic layer deposition. The depositions were made over a temperature range of 80–250 °C with a growth per cycle of around 0.07 nm/cycle. Surface self-saturating growth was obtained with TMG pulse time ≥20 ms at a temperature of 150 °C. The root mean square surface roughness of the obtained Ga2O3 films increased from 0.1 nm to 0.3 nm with increasing the growth temperature. Moreover, the x-ray photoelectron spectroscopy analysis indicated that the obtained film was Ga-rich with the chemical oxidation states Ga3+ and Ga1+, and no carbon contamination was detected in the films after Ar+ sputtering. The electron density of films measured by x-ray reflectivity varied with the growth temperature, increasing from 4.72 to 5.80 g/cm3 . The transmittance of Ga2O3 film deposited on a quartz substrate was obtained through ultraviolet visible (UV–Vis) spectroscopy. An obvious absorption in the deep UV region was demonstrated with a wide band gap of 4.6–4.8 eV. The spectroscopic ellipsometry analysis indicated that the average refractive index of the Ga2O3 film was 1.91 at 632.8 nm and increased with the growth temperature due to the dense structure of the films. Finally, the I-V and C-V characteristics proved that the Ga2O3 films prepared in this work had a low leakage current of 7.2 × 10−11 A/cm2 at 1.0 MV/cm and a high permittivity of 11.9, suitable to be gate dielectric.
An Improved LLC Resonant Converter with Variable-Rectifier for Electric-Vehicle Charger Application

An improved LLC converter with variablerectifier (VR) is studied and evaluated for electric-vehicle charger applications with wide output voltage range. The VR can be reconfigured as center-tapped rectifier (CTR) and full-bridge rectifier (FBR) by the secondary side auxiliary switches. The output voltage range is extended by changing the configuration of rectifier, which results in reduced switching frequency range, circulating current, and conduction losses. Meanwhile, zero voltage switching are achieved for all MOSFETs. A mode transition scheme is employed to achieve smooth and fast transition between CTR and FBR modes. The operation principles are analyzed in detail. A 500W prototype with 400V input voltage and 40-160V output voltage is built and tested to verify the feasibility of the converter.
