DNA methylation and mRNA expression of COL6A3 in antler mesenchyme of female and male reindeer

Backgroud Reindeer is the only deer species that both male and female produce antlers, which provides a particularly interesting case in studying the differences between antlers of the two sexes. Alpha 3(VI) Collagen Gene (COL6A3), forms a microfibrillar network associated with the structural integrity and biomechanical properties, has been found to be one of the differentially expressed genes in antler mesenchyme of female and male reindeer. Objective and Methods The promoter sequence of reindeer COL6A3 gene was obtained using the cloning technology and analyzed by the bioinformatics methods. Bisulfite sequencing PCR (BSP) was used to detect the methylation status of the COL6A3 promoter in reindeer antler mesenchyme. Real-time quantitative PCR was used to detect COL6A3 expression in the antler mesenchyme of female and male reindeer. Results Sequence analysis revealed that the reindeer COL6A3 partial promoter sequence was 983 bp including the possible promoter region at + 105 bp to + 155 bp. Homology and phylogenetic analysis indicated that the COL6A3 promoter of reindeer had the closest genetic distance with Bos taurus, Capra hircus and Ovis aries. BSP results indicated that the methylation level of COL6A3 promoter in the female reindeer antler mesenchyme was significantly higher than in the male. Correlating with increased methylation status, we also found that COL6A3 mRNA expression in female reindeer antler mesenchyme was significantly lower than in the male. Conclusion The higher methylation level of the COL6A3 gene in female reindeer antler mesenchyme coincides with decreased COL6A3 mRNA expression, thereby affecting the transposon silencing mechanism and possibly contributing to apparent differences of antlers in female and male reindeer.
Impact of geometrical parameters on SGEMP responses in cylinder model

This paper is aimed to find out the impact of the geometrical parameters, mainly the radius and the height of a cylinder, on the SGEMP response including the famous scaling law in the classical cylinder model using a homemade PIC code UNIPIC-3D. We computed the electric fields at the center and at the edge on the emission head face with different radii and heights under normal X-rays incidence. The results show that the electric field will increase with the radius but decrease with the height. We analyze the scaling law that links the electric field product and fluence product, and whereafter an irreconcilable contradiction raises when the radius is changeable, which limits the application range of the scaling law. Moreover, the field-height-radius relation is found and described by a combination of logarithmic and minus one-quarter numerical fitting law firstly. Particle and magnetic field distributions are used to explain all the behaviors of the fields reasonably. All the findings will assist the evaluation of SGEMP response in spacecraft protection.
Shape Recognition and Classification Based on Poisson Equation- Fourier-Mellin Moment Descriptor

In this paper, we present a new shape descriptor, which is named Poisson equation-Fourier-Mellin moment Descriptor. We solve the Poisson equation in the shape area, and use the solution to get feature function, which are then integrated using Fourier-Mellin moment to represent the shape. This method develops the Poisson equation-geometric moment Descriptor proposed by Lena Gorelick, and keeps both advantages of Poisson equation-geometric moment and Fourier-Mellin moment. It is proved better than Poisson equation-geometric moment Descriptor in shape recognition and classification experiments.

To develop a new method for producing aspartame (APM), a simple and efficient strategy for the isolation of certain microorganisms producing APM from soil samples was designed. A newly strain with secreting certain specific dipeptidase to directly synthesize APM from the substrates of L-aspartic acid (L-Asp) and Lphenylalanine methyl ester (L-PM) without protection for amino acid side chains was screened from soil samples. APM concentration in reaction mixture was quantified by high performance liquid chromatography (HPLC), yield reached 0.015 g/L. Examination of the general morphological characteristics and data from the sequence analysis of the rDNA-ITS gene led to identification of the isolate as a strain of fungal endophyte spp. The newly isolated strain had a high potential for application in industrial processes for APM production. In particular, this new method was low cost for synthesis of APM during the reaction due to avoiding protection for amino acid side chains and optical resolution of the mixtures. As we known it, this is first report that a newly strain with a high potential for selective synthesis of the APM was isolated from soil.
Tetrazolium Violet Induced Apoptosis and Cell Cycle Arrest in Human Lung Cancer A549 Cells

Tetrazolium violet is a tetrazolium salt and has been proposed as an antitumor agent. In this study, we reported for the fl rst time that tetrazolium violet not only inhibited human lung cancer A549 cell proliferation but also induced apoptosis and blocked cell cycle progression in the G1 phase. The results showed that tetrazolium violet signifl cantly decreased the viability of A549 cells at 5-15 M. Tetrazolium violet -induced apoptosis in A549 cells was confl rmed by H33258 staining assay. In A549, tetrazolium violet blocked the progression of the cell cycle at G1 phase by inducing p53 expression and further up-regulating p21/WAF1 expression. In addition, an enhancement in Fas/APO-1 and its two forms of ligands, membrane-bound Fas ligand (mFasL) and soluble Fas ligand (sFasL), as well as caspase, were responsible for the apoptotic effect induced by tetrazolium violet. The conclusion of this study is that tetrazolium violet induced p53 expression which caused cell cycle arrest and apoptosis. These fl ndings suggest that tetrazolium violet has strong potential for development as an agent for treatment lung cancer.

Effects of controlled- and uncontrolled-pH operations on phenylalanine ammonia lyase (PAL) production by a recombinant Escherichia coli strain were investigated at uncontrolled-pH (pHUC) and controlled-pH (pHC) of 5.5, 6.0, 6.5, 7.0, 7.5, 8.0, and 8.5 in bioreactor systems. The results showed that the recombinant PAL activity was improved significantly by controlled pH strategy. Among the pHC operations, the highest PAL activities were obtained under pHC 7.5 strategy where cell mass (OD600 ㎚) and PAL activity was 1.3 and 1.8 fold higher than those of pHUC, respectively. The maximum PAL activity reached 123 U/g. The pHC 7.5 strategy made recombinant plasmid more stable and therefore allowed easier expression of PAL recombinant plasmid, which increased PAL production. It was indicated that the new approach (controlled-pH strategy) obtained in this work possessed a high potential for the industrial production of PAL, especially in the biosynthesis of Lphenylalanine.

Purpose: To assess the compliance of Chinese urologists with China’s benign prostatic hyperplasia (BPH) clinical practice guideline and to explore the diagnosis and therapy modalities for geriatric patients with BPH. Methods: A cross-sectional survey study was carried out in 33 medical centers in 11 different cities in China. A total of 190 urologists participated in a survey to record their preferences for diagnostic tests and treatment options for BPH outpatients. Diagnostic test results as well as health care demands were collected by surveying 2,027 outpatients aged 60 years and older. Results: The survey response rate was 97.4%. The respondents generally used the diagnostic tests recommended in China’s BPH clinical practice guideline at varying rates. The used rates for medical history, ultrasonography, and urinalysis were above 90.0%; that for uroflowmetry was 31.2%. In addition, the rate of use of recommended tests was higher among doctors in the north than among those in the south. Combination therapy with α-adrenoceptor antagonists and 5α-reductase inhibitors was the preferred treatment option for BPH, and was increasingly used with worsening lower urinary tract symptoms. Finasteride was the most prescribed medication (48.0%), followed by tamsulosin (22.7%). Conclusions: This study assessed the preferences of urologists in the diagnosis and treatment of BPH, which will serve as an important reference for updating and improving China’s current BPH clinical practice guideline.
Multi-mode Radar Signal Sorting by Means of Spatial Data Mining

Formulti-mode radar signals in complex electromagneticenvironment, different modes of one emitter tend to be deinterleavedinto several emitters, called as “extension”, when processingreceived signals by use of existing sorting methods. The “extension”problem inevitably deteriorates the sorting performance ofmulti-mode radar signals. In this paper, a novel method based onspatial data mining is presented to address above challenge. Basedon theories of data field, we describe the distribution information offeature parameters using potential field, and makes partition clusteringof parameter samples according to revealed distribution features. Additionally, an evaluation criterion based on cloud modelmembership is established to measure the relevance between differentcluster-classes, which provides important spatial knowledgefor the solution of the “extension” problem. It is shown through numericalsimulations that the proposed method is effective on solvingthe “extension” problem in multi-mode radar signal sorting,and can achieve higher correct sorting rate.
Complete Mitochondrial Genome of a Tongue Worm Armillifer agkistrodontis

Armillifer agkistrodontis (Ichthyostraca: Pantastomida) is a parasitic pathogen, only reported in China, which can cause a zoonotic disease, pentastomiasis. A complete mitochondrial (mt) genome was 16,521 bp comprising 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes, and 1 non-coding region (NCR). A phylogenetic tree drawn with the concatenated amino acid sequences of the 6 conserved PCGs (atp6, cox1-3, and nad2) showed that A. agkistrodontis and Armillifer armillatus constituted a clade Pentastomida which was a sister group of the Branchiura. The complete mt genome sequence of A. agkistrodontis provides important genetic markers for both phylogenetic and epidemiological studies of pentastomids.

Concrete bridge decks may suffer local damage such as delamination, cracking, reinforcement corrosion and spalling during service. Visual inspection and nondestructive evaluation (NDE) technologies are extensively used for monitoring damage in bridge decks. This paper presents a damage detection method for decks of concrete girder bridges using the frequency obtained from an actively excited vehicle. First, the solution for the frequency of the deck with a concentrated mass is derived with Rayleigh's method, where the bridge deck is regarded as a slab supported on four sides, and the test vehicle is simplified as a concentrated mass. The validity of the proposed method that uses the frequency change to detect the local damage is verified. Then, the damage detection procedure for bridge decks is proposed, and the numerical analysis is performed on a typical concrete girder bridge to prove the validity of the method. Finally, the damage detection experiment for the plywood plate verifies the effectiveness of the proposed method. The results of this study provide an effective method for detecting damage in the decks of concrete girder bridges, which is time-saving and easier to implement.
